Maximizar la productividad en las plantaciones densas de olivo requiere de un manejo apropiado de la iluminación del canopeo. Pero el limitado conocimiento acerca de la respuesta del rendimiento a la radiación fotosintéticamente activa (PAR) dificulta su logro. Además, los periodos potencialmente críticos, donde el rendimiento sería sensible al sombreado, y el efecto de la poda sobre la respuesta a la PAR, han sido escasamente estudiados. Esta tesis se centró sobre las relaciones entre los determinantes y componentes del rendimiento, y la PAR, utilizando dos aproximaciones. En primer lugar, se caracterizó el ambiente lumínico en distintas posiciones de un seto en dos estaciones de crecimiento y se estableció su relación con los determinantes y componentes del rendimiento. Las yemas axilares e inflorescencias respondieron bilinealmente a la PAR aunque la poda alteró esta respuesta. El peso seco del fruto y la concentración de aceite respondieron linealmente a la irradiancia. En segundo lugar, se manipuló directamente el nivel de PAR (3 a 70 de PAR incidente) con redes mediasombra en árboles semi-aislados, focalizando las mediciones en una posición externa de la copa. Combinando ambos enfoques, se contrastaron las funciones de respuestas derivadas de distintas posiciones en el seto (primera aproximación) con aquellas obtenidas en una posición externa de la copa (segunda aproximación). Utilizando la segunda aproximación se demostró que la inducción floral, el cuaje y llenado del fruto son periodos críticos para el rendimiento bajo PAR limitante. También fue posible establecer que los frutos evidenciaron una mejor capacidad de recuperación después de retirar los tratamientos, cuando el periodo de post-sombreo precedió y/o aconteció durante la división celular del fruto. La información aquí generada podría mejorar el modelado de la estructura óptima del canopeo, que asegura una óptima intercepción de la PAR y un alto rendimiento.

Transpiration of two year-old olive trees of three different varieties, Arbequina, Cobrançosa and Galega (18 trees per variety), irrigated with three levels of salt (0, 80 or 200 mM NaCl) for about 90 days, was measured by a gravimetric method. To determine leaf area, each tree was photographed from the side against a white background and the total area of each projected image was determined with ImageJ software. To calibrate these area determinations, one tree of each variety was subsequently stripped of all its leaves and its total leaf area was accurately measured. A correlation was then obtained between the area on the photograph of this particular tree and the total area of the detached leaves of the same tree. Using the leaf area determined by this procedure, transpiration rates of the trees could be calculated. Knowing leaf and air temperatures and RH, it was possible to determine the difference in molar fraction of water between the leaf and the air. Using this and the values of the transpiration rate, stomatal conductance could be calculated (gs calc) and compared with the conductance measured on the same trees with a porometer (gs). Actual leaf area of a plant was 1,40 (Arbequina), 1,42 (Cobrançosa) or 1,24 (Galega) times the area measured with ImageJ on the photograph of the same plant. Leaf area of the trees, on average of all salt irrigations, was significantly higher on Arbequina (0,187 m2) then on the other two varieties (0,138 m2 or 0,148 m2, for Cobrançosa or Galega, respectively), but did not differ significantly in percentage of controls (0 salt). On average of all three varieties, leaf area was also higher on plants irrigated without salt (0,181 m2) than on plants exposed to 80 or 200 mM NaCl (0,152 m2 or 0,140 m2, respectively), which did not differ between them. The same significant difference was observed when leaf area was expressed as percentage of controls. Transpiration rate was significantly higher on Cobrançosa (1,17 mmol m-2 s-1), on average of all treatments, but there were no significant differences between Arbequina (1,08 mmol m-2 s-1) and Galega (0,82 mmol m-2 s-1). In percentage of controls, there were no significant differences between varieties. Salt reduced significantly the transpiration rate in all varieties, both the actual and percentual values, to about 50% or 30% of controls when exposed to 80 mM or 200 mM NaCl, respectively. Stomatal conductance (gs), assessed by porometry, was significantly higher in control plants, mainly in Cobrançosa (102 mmol m-2 s-1), then in Arbequina (77 mmol m-2 s-1) and the lower values were found in Galega (51 mmol m-2 s-1). Salt reduced gs, on average of the three varieties to 30% or 10% of controls on exposure to 80 mM or 200 mM NaCl, respectively. Calculated (gs calc) and measured (gs) values of stomatal conductance showed a close relation between them (0,967, R2 = 0,837) which indicates this non-destructive method to determine whole-plant leaf area to be reasonably accurate.

Geographical isolation and polyploidization are central concepts in plant evolution. The hierarchical organization of archipelagos in this study provides a framework for testing the evolutionary consequences for polyploid taxa and populations occurring in isolation. Using amplified fragment length polymorphism and simple sequence repeat markers, we determined the genetic diversity and differentiation patterns at three levels of geographical isolation in Olea europaea: mainland-archipelagos, islands within an archipelago, and populations within an island. At the subspecies scale, the hexaploid ssp. maroccana (southwest Morocco) exhibited higher genetic diversity than the insular counterparts. In contrast, the tetraploid ssp. cerasiformis (Madeira) displayed values similar to those obtained for the diploid ssp. guanchica (Canary Islands). Geographical isolation was associated with a high genetic differentiation at this scale. In the Canarian archipelago, the stepping-stone model of differentiation suggested in a previous study was partially supported. Within the western lineage, an east-to-west differentiation pattern was confirmed. Conversely, the easternmost populations were more related to the mainland ssp. europaea than to the western guanchica lineage. Genetic diversity across the Canarian archipelago was significantly correlated with the date of the last volcanic activity in the area/island where each population occurs. At the island scale, this pattern was not confirmed in older islands (Tenerife and Madeira), where populations were genetically homogeneous. In contrast, founder effects resulted in low genetic diversity and marked genetic differentiation among populations of the youngest island, La Palma.

Endophytic fungi live inside plants, apparently do not cause any harm to their hosts and may play important roles in defense and growth promotion. Fungal growth is a routine practice at microbiological laboratories, and the Potato Dextrose Agar (PDA) is the most frequently used medium because it is a rich source of starch. However, the production of potatoes in some regions of the world can be costly. Aiming the development of a new medium source to tropical countries, in the present study, we used leaves from the guarana (a tropical plant from the Amazon region) and the olive (which grows in subtropical and temperate regions) to isolate endophytic fungi using PDA and Manihot Dextrose Agar (MDA). Cassava (Manihot esculenta) was evaluated as a substitute starch source. For guarana, the endophytic incidence (EI) was 90% and 98% on PDA and MDA media, respectively, and 65% and 70% for olive, respectively. The fungal isolates were sequenced using the ITS- rDNA region. The fungal identification demonstrated that the isolates varied according to the host plant and media source. In the guarana plant, 13 fungal genera were found using MDA and six were found using PDA. In the olive plant, six genera were obtained using PDA and 4 were obtained using MDA. The multivariate analysis results demonstrated the highest fungal diversity from guarana when using MDA medium. Interestingly, some genera were isolated from one specific host or in one specific media, suggesting the importance of these two factors in fungal isolation specificity. Thus, this study indicated that cassava is a feasible starch source that could serve as a potential alternative medium to potato medium.

This PhD thesis is related to the evolution of phenolic composition of olive fruit to demonstrate the relationship between the raw material sources and the compounds responsible for the healthy and sensory properties of virgin olive oil, and to investigate the mechanisms involved in the synthesis and/or in the degradation of the phenolic fraction. On the basis of phenolic compositions of twelve Italian olive cultivars (Bianchella, Canino, Coratina, Dolce di Andria, Dritta, Frantoio, Leccino, Moraiolo, Nocellara del Belice, Nocellara Etnea, Rosciola and Tendellone) analysed, some significative qualitative and quantitative differences were observed. The main results were utilized, during the following analysis of the genetic expressions of olive cultivars, to determine the genes directly involved in the synthesis and the degradation of the phenolic fraction during the different fruit ripening stages. On the basis of the results about the evolution of phenolic compounds of olive drupes a research program aimed to identify the genes involved in the biosynthesis pathways of fruit secoiridoids, was developed by the CNR-Institute of Plant Genetics, Perugia.

La mosca blanca del fresno, Siphoninus phillyreae (Haliday) , es una especie polífaga invasiva que causa graves daños en sus hospedadores, entre ellos el olivo (Olea europaea L.). El uso del hospedador por parte de S. phillyreae fue observado en tres cultivares de olivo (Arauco, Arbequina y Aloreña) en el norte de la provincia de La Rioja (Argentina) durante 2007 y 2008. De cada cultivar se muestrearon seis plantas infestadas y de cada planta se tomaron ocho hojas. De cada hoja se registró la abundancia de adultos y estados inmaduros de la mosca blanca que fueron comparadas entre los cultivares mediante modelos lineales generales y mixtos. Los resultados obtenidos mostraron diferencias significativas entre las densidades de adultos y ninfas de S. phillyreae en los distintos cultivares de olivo analizados. Dichos resultados indican que este insecto realiza un uso diferente de los cultivares de olivo, siendo Arauco y Arbequina las variedades hospedadoras más utilizadas en la zona de estudio.

O objetivo do presente estudo foi avaliar a atividade antioxidante de extrato de folhas de oliveira (EFO) (Olea europaea L.) por diferentes metodologias analíticas in vitro e in situ, para verificação de efeito em sistemas biológicos. O extrato foi obtido a partir de folhas secas de oliveira, previamente micronizadas, em metanol/água (80/20%) na proporção 1:20 (m/v), após remoção de compostos solúveis em n-hexano. Após liofilização, no EFO foi avaliado o poder redutor por Folin-Ciocalteau, conteúdo de flavonoides totais, teor de oleuropeina, poder de redução do íon férrico (FRAP) e atividade antioxidante sobre DPPHo, ABTSo+, ânion superóxido (O2o-), ácido hipocloroso (HOCl) e óxido nítrico (NOo). O extrato foi também avaliado quanto ao efeito protetor sobre danos oxidativos em eritrócitos humanos. O ácido ascórbico foi utilizado como referência. O experimento foi repetido seis vezes (n = 6) e os ensaios realizados em duplicata. O poder redutor do extrato e o conteúdo de flavonoides totais e oleuropeína foram 131,7 ± 9,4 mg equivalente de ácido gálico/g extrato seco (ms), 19,4 ± 1,3 mg equivalente de quercetina/g ms e 25,5 ± 5,2 mg oleuropeína/g ms, respectivamente. O ensaio de FRAP apresentou 281,8 ± 22,8 mg equivalente de trolox/g ms. O EFO foi efetivo na inibição dos radicais DPPHo e ABTSo+, dependente da concentração de extrato, com valores de IC50 de 13,8 ± 0,8 e 16,1 ± 1,2 µg/mL, respectivamente. Com relação à atividade antioxidante sobre espécies reativas de importância biológica, o EFO apresentou forte capacidade de inibição de O2o- (IC50 = 52,6 ± 2,1 µg/mL) e NOo (IC50 = 48,4 ± 6,8 µg/mL), quando comparado ao ácido ascórbico. Porém, a inibição de HOCl não foi tão eficiente (IC50 = 714,1 ± 31,4 µg/mL). O EFO inibiu a hemólise induzida em eritrócitos de maneira dependente da concentração (IC50 = 7,8 ± 1,1 µg/mL), assim como a peroxidação lipídica e a formação de meta-hemoglobina, com valores de IC50 de 38,0 ± 11,7 e 186,3 ± 29,7 µg/mL, respectivamente. Os resultados obtidos neste estudo sugerem que extrato de folhas de oliveira possui efetiva atividade antioxidante em sistemas biológicos, pelo efeito sequestrador de determinadas espécies reativas que participam dos processos bioquímicos, e pela prevenção de danos oxidativos em eritrócitos humanos. Portanto, sua ingestão pode estar relacionada com a prevenção de estresse oxidativo in vivo, com consequentes benefícios à saúde.

Reforestation projects in semiarid lands often yield poor results. Water scarcity, poor soil fertility, and structure strongly limit the survival and growth of planted seedlings in these areas. At two experimental semiarid sites, we evaluated a variety of low-cost planting techniques in order to increase water availability to plants. Treatments included various combinations of traditional planting holes; water-harvesting microcatchments; stone or plastic mulches; small waterproof sheets to increase water harvesting; dry wells; buried clay pots; and deep irrigation. Some of these treatments were also combined with addition of composted biosolids. Waterproof sheets significantly enhanced water harvesting (43%) and soil moisture in the planting hole (40%), especially for low-intensity rainfall events. Treatment effects on the survival and growth of Olea europaea seedlings varied between experimental sites. At the most water-limited site, clay pots, and dry wells improved seedling survival, while no treatment enhanced seedling growth. At the least water-stressed site, the application of composted sludge significantly improved seedling growth. We conclude that nutrient-mediated stress is subordinate to water stress in arid and semiarid environments, and we suggest modifications on the microsite scale to address these limiting conditions in Mediterranean drylands.

Las respuestas de los componentes del rendimiento y de la calidad del aceite a la temperatura aún no han sido estudiadas en olivo ni en otras oleaginosas que acumulan aceite principalmente en mesocarpo (cerca de 95 por ciento del total en el caso de olivo). La información disponible en olivo se basa en correlaciones ligadas a variaciones en ubicación geográfica, altitud y años en los que pudieron covariar con la temperatura otros factores (radiación, disponibilidad hídrica, nutrición, etc). El objetivo de esta tesis fue evaluar el rol de la temperatura durante el crecimiento de frutos de olivo sobre el peso seco de los mismos, su contenido y concentración de aceite y las proporciones de ácidos grasos del aceite. Se realizaron experimentos manipulativos aplicados a ramas fructíferas creciendo a campo, utilizando cámaras transparentes con control y registro de temperatura. Los rangos de temperaturas medias alcanzados (entre 15-32 ºC) fueron mucho mayores a los rangos de 1 y 4 ºC logrados en estudios correlativos y permitieron generar relaciones funcionales para el peso seco del fruto, su concentración de aceite, y la proporción de ácidos grasos en el aceite. El contraste entre aplicaciones de temperaturas mayores o menores al ambiente durante períodos largos (114 días) y cortos (30 días) demostraron que tanto la proporción de aceite del fruto como la de ácido oleico en el aceite de fruto entero (mesocarpo + semilla) disminuye al aumentar la temperatura. Si bien en ambas estructuras del fruto la temperatura tuvo un efecto negativo sobre la concentración de aceite, las respuestas a la temperatura de la proporción de ácidos grasos de la semilla y el mesocarpo, medidos en forma separada, difirieron entre sí. Los patrones de respuesta de las proporciones de ácidos grasos en el aceite de semilla mostraron similitudes con las conocidas para especies oleaginosas de semilla como girasol. Por contraste, en el aceite de mesocarpo de olivo la proporción de ácido oleico disminuyó con el aumento de la temperatura, patrón opuesto a lo manifestado en semillas de olivo y en las de oleaginosas anuales. En esta tesis también se evaluó el efecto de la temperatura mínima nocturna sobre el crecimiento de los frutos y la calidad del aceite. Los resultados sugieren que la temperatura mínima nocturna y la amplitud térmica diarias son las dimensiones del régimen térmico diario con los que mejor se asociaron los cambios en el peso seco del fruto, la proporción de aceite y las proporciones de ácidos grasos en el mismo. Los resultados de esta tesis sirven para guiar la selección de sitios de plantación de nuevos olivares basados en los registros térmicos zonales, y para explicar los desajustes de la proporción de ácidos grasos del aceite de oliva producido en el NOA y la normativa del Consejo Oleícola Internacional (COI). Las relaciones funcionales entre las variables respuesta y la temperatura definidas podrán incorporarse en modelos de simulación del rendimiento y calidad que se desarrollen en el futuro. Tomados en conjunto con la tendencia al aumento de la temperatura, producto del calentamiento global, los resultados permitirán estimar la magnitud de los impactos negativos que podrían afectar el rendimiento del cultivo y la calidad del aceite.

Olive (Olea europaea L.), one of the main crops in the Mediterranean basin, is mainly propagated by cuttings, a classical propagation method that relies on the ability of the cuttings to form adventitious roots. While some cultivars are easily propagated by this technique, some of the most interesting olive cultivars are considered difficult-to-root which poses a challenge for their preservation and commercialization. Therefore, increasing the current knowledge on adventitious root formation is extremely important for species like olive. This research focuses on evaluating the role of free auxins and oxidative enzymes on adventitious root formation of two olive cultivars with different rooting ability - ‘Galega vulgar’ (difficult-to-root) and ‘Cobrançosa’ (easy-to-root). In this context, free auxin levels and enzyme activities were determined in in vitro-cultured ‘Galega vulgar’ microshoots and in semi-hardwood cuttings of cvs. ‘Galega vulgar’ and ‘Cobrançosa’. To attain this goal, an analytical method for the quantification of free indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) was developed, which is based on dispersive liquid-liquid microextraction followed by microwave derivatization (DLLME-MAD) and gas chromatography-mass spectrometry (GC/MS) analysis. The developed method was validated in terms of linearity, recovery, limit of detection (LOD) and limit of quantification (LOQ) and proved to be useful in the analysis of two very different types of plant tissues. The results from auxin quantification in olive samples point at a relationship between free auxin levels and rooting ability of both microshoots and semihardwood cuttings. A defective IBA-IAA conversion, resulting in a peak of free IAA during initiation phase, seems to be associated with low rooting ability. Likewise, differences in the activity of oxidative enzymes also appear to be related with rooting ability. Higher polyphenol oxidases (PPO) activity is likely related with an easyto- root behavior, while the opposite is true for peroxidases (POX) (including IAA oxidase (IAAox)) activity. A possible hypothesis for adventitious root formation in olive microcuttings is presented herein for the first time. Free auxins, oxidative enzymes, alternative oxidase (AOX) and reactive oxygen species (ROS) are some of the factors that may be involved in this highly complex physiological process. Interestingly, while temporal changes in auxin levels were similar between microshoots and semihardwood cuttings, the conclusions obtained from enzyme activity results in microshoots didn’t translate to semi-hardwood tissues, showing the emerging need for adaptation of classical agronomical research studies to modern techniques; Resumo: Procurando compreender o papel das auxinas e enzimas oxidativas na formação de raízes adventícias em cultivares de oliveira (Olea europaea L.) A oliveira (Olea europaea L.) é uma das principais culturas da bacia Mediterrânica e é propagada maioritariamente por estacaria, um processo altamente dependente da capacidade das estacas para formar raízes adventícias. Enquanto algumas cultivares são fáceis de propagar desta forma, algumas das cultivares de oliveira mais interessantes são consideradas difíceis de enraizar, o que dificulta a sua preservação e comercialização e torna extremamente importante aprofundar o conhecimento sobre o enraizamento adventício desta espécie. Este trabalho foca-se na avaliação do papel das auxinas livres e das enzimas oxidativas na formação de raízes adventícias em duas cultivares de oliveira com diferente capacidade de enraizamento - ‘Galega vulgar’ (difícil de enraizar) e ‘Cobrançosa’ (fácil de enraizar). Neste contexto, determinaram-se os níveis de auxinas livres e as actividades de enzimas oxidativas em microestacas de ‘Galega vulgar’ cultivadas in vitro bem como em estacas semi-lenhosas das cvs. ‘Galega vulgar’ e ‘Cobrançosa’. Para tal foi necessário desenvolver uma metodologia analítica para a quantificação de ácido indol-3-acético (IAA) e ácido indol-3-butírico (IBA), baseada em microextracção dispersiva líquido-líquido (DLLME) seguida de derivatização em microondas (MAD) e análise por cromatografia gasosa acoplada a espectrometria de massa (GC/MS). O método desenvolvido foi validado em termos de linearidade, recuperação, limite de detecção (LOD) e limite de quantificação (LOQ), e mostrou-se eficaz na análise de dois tipos de tecidos vegetais bastante diferentes. Os resultados da análise de auxinas em amostras de oliveira apontam para uma possível relação entre os níveis de auxinas livres e a capacidade de enraizamento, tanto em microestacas como em estacas semi-lenhosas. Uma conversão IBA-IAA deficiente, que resulta num pico de IAA durante a fase de iniciação, parece estar associada à baixa capacidade de enraizamento. Por outro lado, a capacidade de enraizamento também parece estar relacionada com diferenças na actividade de enzimas oxidativas. Comportamentos fáceis de enraizar estão associados a actividade mais elevada das polifenoloxidases (PPO), enquanto o oposto é verdade para a actividade das peroxidases (POX) (incluindo a IAA oxidase (IAAox)). Neste trabalho propõe-se pela primeira vez uma possível explicação para o enraizamento adventício em microestacas de oliveira. Auxinas livres, enzimas oxidativas, oxidase alternativa (AOX) e espécies reactivas de oxigénio (ROS) são alguns dos factores envolvidos neste processo fisiológico altamente complexo. Curiosamente, enquanto as alterações temporais nos níveis de auxinas foram semelhantes entre microestacas e estacas semi-lenhosas, o mesmo não se observou relativamente à actividade enzimática, o que mostra a necessidade de adaptação dos estudos agronómicos tradicionais às técnicas correntes.

The activity of oxidative enzymes and the levels of free auxins were determined during adventitious root formation in olive explants. Rooting trials were performed both with in vitro-cultured micro shoots of the cultivar ‘Galega Vulgar’, treated with indole-3-butyric acid (IBA) and with salicylhydroxamic acid(SHAM) + IBA, as well as with semi-hardwood cuttings of the cultivars ‘Galega Vulgar’ (difficult-to-root)and ‘Cobrançosa’ (easy-to-root), treated with IBA. The auxin (IBA) was used in all experiments as a rooting promoter, while SHAM was used in micropropagation trials as rooting inhibitor, providing a negative control. Free indole-3-acetic acid (IAA) and IBA concentrations were determined in microshoots, as well as in semi-hardwood cuttings, throughout the rooting period at pre-established time-points. At the sametime-points, the enzymatic activity of polyphenol oxidases (PPO), peroxidases (POX), and IAA oxidase(IAAox) was evaluated in the microshoots. Microshoots treated with SHAM + IBA revealed higher POX and IAAox activity, as well as lower PPO activity, than those treated only with IBA. IAA levels were higher in IBA-treated microshoots during induction phase, but lower during early initiation phase. Incontrast, free IBA levels were higher in microshoots treated with SHAM + IBA during induction, but lower during initiation. A similar pattern of free auxin levels was observed in semi-hardwood cuttings of the two contrasting cultivars under evaluation. The similarities found on the auxin patterns of microshoots treated with SHAM and those of semi-hardwood cuttings of the difficult-to-root olive cultivar allow considering SHAM a reliable control for when simulation of a difficult-to-root behavior is necessary. The inhibitory effect of SHAM in root formation could be related with 1) the inhibition of alternative oxidase(AOX), leading to a down regulation of phenylpropanoid biosynthetic pathways, which would decrease the concentration of phenolic substrates for PPO; 2) an increase in IAAox activity resulting in lower free IAA levels or; 3) a defective conversion of IBA into IAA.

A oliveira tem sido multiplicada ao longo dos tempos por métodos convencionais de propagação vegetativa como a enxertia e a estacaria lenhosa e semilenhosa. No entanto, estes métodos revelam-se lentos ou ineficientes para determinadas cultivares. No caso da cv. ‘Galega Vulgar’, ainda com grande expressão no olival português, e de difícil enraizamento por estacaria semilenhosa, tem sido usada a micropropagação de modo a contornar essas limitações e assim obter um elevado número de plantas em curto período de tempo. O custo final de produção por este processo ainda é elevado, podendo comprometer a sua aplicação a nível comercial. Grande parte dos custos estão relacionados com a fase de enraizamento in vitro que carece de ambiente estéril e condições de assépsia para a sua execução. Com vista a uma redução de custos associados a esta fase de produção, pretendeu-se com este trabalho testar a viabilidade do enraizamento ex vitro, na ausência de condições de assépsia. Este método poderá permitir uma significativa redução da mão-de-obra, ao mesmo tempo que facilitará a aclimatização das plantas e a obtenção de um sistema radicular de melhor qualidade. Compararam-se as taxas de enraizamento in vitro (controlo), com as obtidas ex vitro. Foram utilizados explantes provenientes de dois clones da cv. ‘Galega Vulgar’, (cl. 1441 e cl. 2022) cultivados e mantidos in vitro há vários anos no Laboratório de Melhoramento e Biotecnologia da Universidade de Évora. Para além do clone foi avaliada a influência do tipo de estaca (basal e apical), da hormona de enraizamento (AIB e ANA), da sua concentração (540 e 3000 ppm) e ainda de dois substratos, Preformas Jiffy® e pastilhas de fibra coco. Os melhores resultados foram obtidos com o clone 1441 em pastilhas de fibra de coco prensada, com o uso de estacas basais. Quanto à auxina, não se observaram diferenças significativas entre a utilização de ANA na concentração de 540 ppm e AIB na concentração de 3000 ppm. A aclimatização das plantas foi conseguida com taxas elevadas de sucesso, independentemente do tratamento utilizado. Conclui-se que a aplicação do método de enraizamento ex vitro simplifica procedimentos e mantém taxas de enraizamento elevadas, conduzindo assim a uma efetiva redução de tempo e custos associados; Simplifying procedures for in vitro propagation of olive “Olea europaea L.” Abstract: The olive tree has been multiplied throughout the ages by conventional methods of vegetative propagation such as grafting and wood or softwood cuttings. These propagation methods are somehow inefficient for certain cultivars. For the CV. ‘Galega Vulgar‘, still with great expression in Portuguese olive orchards, propagation has been attempted by in vitro culture in order to circumvent these limitations and so obtain a large number of plants in short time period. The final production fees associated to this process are still high which may compromise its application to a commercial level. Most of this process fees are related to the in vitro rooting phase which lacks sterile and aseptic conditions for its implementation. Aiming to reduce the costs associated with this production phase, this work tested the feasibility of the ex vitro rooting in the absence of aseptic conditions, which can allow a significant reduction of the manpower involved and an easier plant acclimatization due to its transplant with a balled-root system. In vitro rooting rates (control) were compared with those obtained with the ex vitro experiments. Explants from two clones of the cv. ‘Galega Vulgar‘ (cl. 1441 and cl. 2022), grown and maintained in vitro for several years in the Laboratory of Biotechnology and Plant Breeding of the University of Évora, were used in the trials. In addition to the clone, the effect of the cutting type (basal and apical), the rooting hormone (AIB and ANA), their concentration (540 and 3000 ppm) and two substrates, Preformas Jiffy ® and pressed coco fiber pellets, were also evaluated. The best results were obtained with the clone 1441, when rooted in pressed coco fiber pellets, using basal cuttings. Under this conditions no significant differences were observed between the use of ANA at 540 ppm or AIB in the 3000 ppm. Acclimatization of plants was achieved with high rates of success, regardless of the treatment used. It can be concluded that the application of the ex vitro rooting method allows to maintain high rooting rates, contributing for an effective reduction of time and fees of the rooting process.

Background Epidemiological studies have shown a reduced incidence of cardiovascular disease in the Mediterranean population attributed to the consumption of dietary olive oil rich in antioxidants. This has lead to increased interest in the antioxidant properties of other phenolic compounds of olive tree products. It has been suggested that olive leaf extract may also have health benefits due to its antioxidant and anti-inflammatory activities. Antioxidants can prevent the effects of oxidative metabolism by scavenging free radicals and decreasing the hyperactivity of platelets associated with the development of occlusive thrombosis. No studies to date have investigated the effects of olive leaf extract on platelet function to our knowledge. Improved understanding of the antioxidant properties of olive leaf extract and its effect on platelet function could lead to improved cardiovascular health. Objective The current study used an olive leaf extract prepared from the Olea europaea L. tree. The aim was to determine if polyphenols in olive leaf extract would reduce platelet activity and, to establish an optimal dose in vitro that would reduce platelet aggregation and ATP release. Design Eleven subjects with normal platelet counts (150–400 x 109/L) were recruited for the current in vitro study. Olive leaf extract was added to citrated whole blood to obtain five concentrations ranging from 5.4 ug/mL to 54.0 ug/mL for a dose response curve. Baseline samples, without olive leaf extract were used as a negative control for each subject. After 2 hours incubation with olive leaf extract samples were analyzed for platelet aggregation and ATP release from platelets stimulated by the addition of collagen. Results Whole blood analysis (n=11) showed a clear dose-dependant reduction in platelet aggregation with the increasing olive leaf extract concentrations (p<0.0001). There was also a similar decrease in ATP release from collagen stimulated platelets (p=0.02). Conclusion In the current study the olive leaf extract obtained from Olea europaea L. inhibited platelet aggregation and ATP release from collagen stimulated platelets in vitro. This study suggests olive leaf extract may prevent occlusive thrombosis by reducing platelet hyperactivity.
