Glycosyl hydrolase family 1 beta-glucosidases are important enzymes that serve many diverse functions in plants including defense, whereby hydrolyzing the defensive compounds such as hydroxynitrile glucosides. A hydroxynitrile glucoside cleaving beta-glucosidase gene (Llbglu1) was isolated from Leucaena leucocephala, cloned into pET-28a (+) and expressed in E. coli BL21 (DE3) cells. The recombinant enzyme was purified by Ni-NTA affinity chromatography. The optimal temperature and pH for this beta-glucosidase were found to be 45 A degrees C and 4.8, respectively. The purified Llbglu1 enzyme hydrolyzed the synthetic glycosides, pNPGlucoside (pNPGlc) and pNPGalactoside (pNPGal). Also, the enzyme hydrolyzed amygdalin, a hydroxynitrile glycoside and a few of the tested flavonoid and isoflavonoid glucosides. The kinetic parameters K (m) and V (max) were found to be 38.59 mu M and 0.8237 mu M/mg/min for pNPGlc, whereas for pNPGal the values were observed as 1845 mu M and 0.1037 mu M/mg/min. In the present study, a three dimensional (3D) model of the Llbglu1 was built by MODELLER software to find out the substrate binding sites and the quality of the model was examined using the program PROCHEK. Docking studies indicated that conserved active site residues are Glu 199, Glu 413, His 153, Asn 198, Val 270, Asn 340, and Trp 462. Docking of rhodiocyanoside A with the modeled Llbglu1 resulted in a binding with free energy change (Delta G) of -5.52 kcal/mol on which basis rhodiocyanoside A could be considered as a potential substrate.

El presente trabajo se realizo en la comunidad de Pacora, San Francisco Libre, Managua, donde los habitantes tradicionalmente, han utilizado la crianza de ganado menor (cerdos, cabras, gallinas, patos, Peliguey y otros) para satisfacer en cierta medida sus necesidades alimenticias y económicas por la pobreza que caracteriza la comunidad. Con este estudio se presenta una opción de utilizar las tierras que no pueden ser aprovechadas con cultivos tradicionales, empleando especies forestales cuyo sistema radicular y funciones fisiológicas son muy diferente a los cultivos tradicionales, permitiendo su sobrevivencia, calidad nutritiva, el manejo agronómico y el potencial de incorporar estos forrajes en la dieta de animales domésticos. Se evalúa la producción de biomasa total y comestible, la sobrevivencia y los agentes biológicos que afectan a las especies de Quelite ( Cnidoscolus aconitifolium (Mill) L.M.Johnst), m arango (Moringa oleífera) Lam, y Leucaena (Leucaena leucocephala (Lam) de wit), estableciéndose en parcelas de 10.5 m 2 La mayor producción de biomasa verde total la obtuvo Moringa oleífera (Lam) con 15,991 kg ha -1 en la primera poda (7 meses de establecido el ensayo) y en la segunda poda (12 meses de establecido el ensayo) con 34,873 kg ha -1 . Esta misma especie mostró los mejores rendimientos de biomasa seca total en la primera poda con 4,181 kg ha -1 , pero en la segunda poda fue superada por la especie Leucaena leucocephala (Lam) de wit, con 6,782 kg ha -1 . La mayor producción de biomasa verde comestible en la primera poda la obtuvo Cnidoscolus aconitifolium (Mill) L. M.Johnst con 9,491 kg ha -1 y en la segunda poda con 25,553 kg ha -1 , Esta misma especie mostró los mayores rendimientos en biomasa seca comestible con 1,790 kg ha -1 en la primera poda y 5,817 kg ha -1 en la segunda poda. Los mayores porcentajes de sobrevivencia (100%), en la primera poda fueron obtenidos por la especie Leucaena leucocephala (Lam) de wit , Cinco meses después esta misma especie mostró rendimientos de 100% de sobrevivencia. Los agentes biológicos (insectos) encontrados en el ensayo no ocasionaron daños a las plantas que incurrieran en la producción y la calidad de la biomasa obtenida en el estudio.

Se realizó un estudio preliminar de dos variedades de Leucaena leococephala (Lam) de Wit, en un ensayo en condiciones de secano en la Unidad experimental “Las Mercedes” propiedad del ISCA, durante el periodo de Agosto de 1978 a Enero de 1989. Se utilizó un diseño completo al Azar (DCA); unifactorial no balanceado. Los tratamientos utilizados fueron: tratamiento 1. Cv Cunningham 200.000pta /ha; tratamiento 2. Cv Cunningham -150.000 ptas/ha tratamiento 3. Cv k -17-502 -2000.000 ptas/HA. Cada parcela experimental tenía un tamaño de 6 metros cuadrados: Y se efectuaron 5 cortes cada 75 días, 5 meses después de establecido el ensayo. A las muestras obtenida se le midió el rendimiento, composición química y relación porcentual hoja tallo: Los rendimientos obtenidos fueron: 22.82 ton/ha/año, , 4,78 kg/ha/año,5,770kg/ha/año en el tratamiento 2; 18.37ton/ha/año, 4,290 kg/ha/año, 5,250kg/ha/año para materia seca, proteína bruta respectivamente. No existiendo diferencia significativa (p<0.05) entre los tratamientos. Los porcentajes de hojas encontrados son: para el tratamiento 1,57% tratamiento 2,61%; tratamiento 3,62%

El cultivo en callejones o “Alley croppin” es un sistema de producción que se ascia árboles de rápido crecimiento con cultivos alimenticios anuales como: maíz, frijol, sorgo, etc. (Sánchez y salinas 1983). En este sistema los cultivos anulaes crecen entre hileras de árboles leñosas o árboles siendo podados periódicamente durante la época de cultivo para prevenir la sobra y suministrar abono verde al cultivo acompañante. El presente trabajo de tesis, tiene como objetivo evaluar el comportamiento de las especies leguminosas arbórea Gliricidia y Leucaena hasta su establecimiento en el sistema de cultivo en callejones. Se encontró a nivel de vivero que las especies leguminosas obtuvieron una germinación de 88.5% Gliricidia Sepium, el crecimiento promedio a los 50 DDS fue de 44.23cm y 44.65 cm por ambas especies respectivamente. A nivel de plantación la sobrevivencia registrada a los 90 y 180 DDS fue de 98% y 90% para Leucaena y con un crecimiento promedio de 86 y 140 en ambos mediciones para en del establecimiento de dicha especia al sistema, Gliricidia registro a una sobrevivencia de 83% y 67.5% a los 90 y 180 DDS, mientras el Crecimiento seco se realizó a las 90 DDS con un promedio de 77 cm debido al ataque de roedores que ocasionó serios daños a este especie.

Con el objetivo de caracterizar el comportamiento inicial de Moringa oleifera y profundizar en las ventajas de esta especie, en comparación con otra forrajera de rápido crecimiento (Leucaena leucocephala); se llevo a cabo en el vivero de la Facultad de Recursos Naturales un ensayo con diseño completamente aleatorio con dos tratamientos, y 20 repeticiones. Se realizó un conteo diario de germinación por un periodo de tres semanas (21 días); mientras patrones de crecimiento fueron determinados a partir de evaluaciones semanales de diámetro basal y raíz, longitud de tallo y raíz, y numero de raíces secundarias; así como producción de biomasa expresada en peso seco y peso fresco de la parte área y radicular, de una muestra de 8 plantas destruidas semanalmente. Los resultados muestran ventajas de la especie M. oleifera para la mayoría de variables; solo superado en el número de raíces secundarias de L .leucocephala. Los valores relacionados a la calidad o potencial de establecimiento sugieren pocos problemas de establecimiento en campo para las especies M. oleifera y L. leucocephala como producto del rápido crecimiento, y aun con la desigual proporción en la relación tallo-raíz, y biomasa.

A-ten-week feeding trial was carried out to evaluate the growth and survival rate of Oreochromis niloticus fed with varying percentage levels of Leucaena leucocephala leaf meal based diets. The substitution rates of L.leucocephala leaf meal for groundnut cake in the various diets were 0% - Diet 1, 25% - Diet 2, 50% - Diet 3 and 75% - Diet 4. Ten fries with an average weight of 0.44g were stocked at the rate of 10 fish per bowl and fed at 5% body weight. Diet 1 with 0% inclusion of Leucaena leaf meal gave a significant difference (P>0.05) in growth and survival rate compared with diets 2, 3 and 4. The water quality parameters recorded were appropriate for fish culture

Among plant protein ingredients,ipil ipil (Leucaena leucocephala) leafmeal (ILLM) is considered the most nutritive plant protein source after soybean meal in aquatic feeds. That was proven in a 21-day experiment conducted to assess the response of juvenile Monosex Nile tilapia Oreochromis niloticus with four iso-nitrogenous formulated diets: One control diet was formulated based on fishmeal, one on soybean meal and one on rice bran, ipil ipil leafmeal was also included in experimental diets.

A study was carried out to determine the effect of 10 or 20% leaves or seeds in the diet of Penaeus monodon , and the extent to which local ipil-ipil (Leucaena leucocephala ) could replace head shrimp meal. A brief description is given of the experimental methodology, and details of composition of the diet, proximate chemical composition of the diets, mean weight gain and survival of Penaeus monodon larvae fed shrimp head meal and ipil-ipil as protein sources, are presented. Mean weight gains for all groups were poor and not statistically significant. Survival rates for those fed 10% ipil-ipil were significantly higher than those fed 20% diets. Wherever the survival rate was high, mean weight gain was low and vice versa. The presence of the toxic alkaloids mimosine in ipil-ipil could have caused the low survival rate.

Penaeus monodon juveniles were fed diets containing fish meal, shrimp head meal and ipil-ipil leaves soaked and unsoaked, local and peruvian varieties. Mean weight gain at the end of 8 wk was significantly highest among those given the diet containing commercial ipil-ipil leaves. Gain in length followed the same pattern as mean weight gains. Among the diets containing ipil-ipil leaves there was a direct relationship in the amount of mimosine in the diet and the survival rate, the lower the amount of mimosine (due to soaking) the higher the survival rate. The Results thus indicate the beneficial effect of the addition of commercial ipil-ipil leaves to the diets of prawns, providing the mimosine content is kept low by soaking. A reduce in costs is also obtained, since 1kg of shrimp head meal or fish costs more than 2 or 4 tons, respectively, than that of ipil-ipil foliage.

The study aimed to find a cheap and practical method of extracting mimosine from Leucaena leucocephala, otherwise known as ipil-ipil in the Philippines. L. leucocephala leaves are used in cattle, poultry and swine feed and have been tried as a food ingredient in some fish diets. While it contains relatively high amount of protein, its use as feed has been limited because of the presence of toxic substance, mimosine. Findings revealed that soaking the leaves in water was highly efficient for the extraction of mimosine, the longer the duration of soaking the more mimosine was extracted. On the other hand, 87 % of the juveniles Penaeus monodon fed with diets containing L. leucocephala leaves soaked for 24 hours survived, much higher compared to those that were fed with unsoaked leaves for eight weeks.

This study aimed to evaluate the effects of salinity in the growth and nitrogen fixation in leucaena. Plants were cultivated in Leonard pots containing clean and sterilized sand. The treatments were distributed in a completely randomized design, in a 3x3 factorial, with five replicates. The first factor corresponded to no inoculation and inoculation with Bradyrhizobium sp SEMIA 6070 and SEMIA 6153 strains. The second factor corresponded to NaCl concentrations: 0, 25 and 50 mol m(-3). Were analyzed the height and dry matter accumulation, the number of nodules and nitrogen in plants. The inoculation of plants with the strain SEMIA 6070 provided higher growth and greater nitrogen accumulation in plants thar were not subject to salinity. Nodulation was not affected by salinity. Inoculation with SEMIA 6153 strain provided greater tolerance to salinity.

Excessive salt can to promote water soil retention, reducing your availability to plants. Besides, they can interfere in protoplasm metabolism. The plants ability to survive in salinity conditions is an important factor to geographic distribution and agriculture in salinized regions. The objective of this work was to verify the effects of the NaCl in the growth, dry matter distribution and N, K, Ca, Mg, Na, and Cl content in young plants. The experiment was conducted in 'Leonard' pots, with nutrient solutions, with NaCl ( 0, 25, 50 and 100 mmol L(-1)). Increasing of NaCl concentration reduces growth and total dry matter. There was an increase of N and K content, in shoots, and N and Mg content in roots. There was an reduction in Mg content in shoots, and reduction of Ca content in roots with increasing of NaCl. Leucaena plants were inefficient to exclude Na and Cl, in shoots principally.
