The fall armyworm Spodoptera frugiperda is one of the most important pests of maize. Various studies are conducted for their management, integrating chemical and biological control tactics as well as resistant plants. In order to offer alternatives for an efficient management of this pest with minimal use of pesticides, the technology of genetically modified plants resistant to insects has been widely studied. The aim of the present study was to evaluate the natural infestation of larvae of S. frugiperda and their injuries under field conditions in transgenic maize hybrids compared to their conventional isogenic counterparts at two sowing dates and two regions. The hybrids were planted in the off season of 2010 in Jaboticabal, SP, Brazil, and the summer of 2010/2011 in Jaboticabal, and Pindorama, SP, in a randomized block with seven treatments (hybrids) and four replications. Different levels of infestation of larvae occurred throughout the phenological development of plants in conventional and genetically modified hybrids with significant differences between the two groups in most evaluations. The hybrid 2B710HX was the least infested with caterpillars and had the least damaged leaf area. It follows that the Cry1F toxin was the most effective in protecting the plant in relation to other toxic proteins expressed by the other Bt hybrids against infestation and damage promoted by this pest, regardless of time of sowing.

The fall armyworm is a pest that feeds on various botanies species. The objective of this experiment was to study the biology of this pest in corn and cassava leaves. Caterpillars were collected in area under cultivation with cassava and maintained on artificial diet for two generations. Under controlled conditions in a climatic chamber (B.O.D) in the laboratory (25 degrees, 60 +/- 10% RH and photophase 14 hours) were evaluated daily 50 caterpillars in corn treatments and 50 in cassava, where duration and viability of the larval phase and pupal, weight of pupas were observed after 24 hours, deformation percentage of pupas and adults, longevity, fecundity and total life cycle. The viability of larvae fed on leaves of maize and cassava was 74% and 60%, respectively. The larval period of the insects was shorter in maize 16.89 days (seven instars) and cassava 20.08 days (six instars). The pupal phase lasted 11.42 days in cassava treatment and 10.87 in the maize. The pupal weight of females and males was higher in corn 204.91 mg and 198.97 mg, respectively. The biological cycle varied depending on the ingested food. Adult longevity lasted 9.88 days for insects fed on cassava leaves. Therefore, cassava affected the development of S. frugiperda.

Plantas transgênicas que expressam toxinas de Bacillus thuringiensis Berliner (Bt) têm sido amplamente utilizadas para o controle de Spodoptera frugiperda (J. E. Smith) no Brasil. Entretanto, a evolução da resistência é um dos maiores entraves para a continuidade do uso desta tecnologia. Para subsidiar programas de Manejo da Resistência de Insetos (MRI), foram conduzidos estudos para o aprimoramento dos programas de manejo da resistência de S. frugiperda a tecnologias Bt. Foram realizadas estudos para determinar a dominância funcional da resistência de S. frugiperda a tecnologias Bt mediante a avaliação da sobrevivência de larvas neonatas provenientes das linhagens de S. frugiperda resistentes ao milho Herculex® que expressa a proteína Cry1F (HX-R), ao milho YieldGard VT PRO™ que expressa as proteínas Cry1A.105 e Cry2Ab2 (VT-R), ao milho PowerCore™ que expressa as proteínas Cry1A.105, Cry2Ab2 e Cry1F (PW-R), e ao milho Agrisure Viptera™ que expressa a proteína Vip3Aa20 (Vip-R), além da linhagem suscetível (Sus) e de suas respectivas linhagens heterozigotas em diversas tecnologias de milho e algodão Bt. Posteriormente, um método prático para o monitoramento fenotípico da suscetibilidade a diferentes tecnologias de milho e algodão Bt foi testado a partir da avaliação da sobrevivência de larvas neonatas em folhas de plantas Bt em populações de S. frugiperda provenientes dos Estados do Rio Grande do Sul, Paraná, São Paulo, Goiás e Bahia na safra agrícola 2014/15. E por último, a estimativa da frequência de alelos de resistência de S. frugiperda a Vip3Aa20 foi validada pelo método de F1 screen. Em geral, observou-se alta mortalidade dos heterozigotos nas tecnologias Bt testadas, comprovando que a resistência de S. frugiperda a proteínas Bt é funcionalmente recessiva o que suporta a estratégia de refúgio em programas de MRI. Verificou-se também que linhagens resistentes a eventos que expressam proteínas Cry não sobrevivem em tecnologias que expressam proteína Vip. No monitoramento prático da suscetibilidade a tecnologias Bt, sobrevivência larval superior a 70% foi observada para populações de campo do Paraná, Goiás e Bahia no milho Herculex®. Em tecnologias de milho PowerCore™ e YieldGard VT PRO™ houve sobrevivência larval variando de 1,1 a 17,9%. Em contraste, não houve sobreviventes em tecnologias de milho Viptera™. Em algodão WideStrike® que expressa as proteínas Cry1Ac e Cry1F, sobrevivência acima de 41% foi observada para populações de campo de S. frugiperda. A sobrevivência larval em Bollgard II® que expressa as proteínas Cry1Ac e Cry2Ab2 variou de 14 a 40%. No algodão TwinLink® que expressa as proteínas Cry1Ab e Cry2Ae, a sobrevivência larval das populações foi menor que 20%. O método de F1 screen foi eficiente na detecção de alelos de resistência a Vip3Aa20 em populações de S. frugiperda provenientes de diferentes regiões produtoras de milho no Brasil na safra 2014/2015. De 263 isofamílias testadas, foram detectadas três isofamílias positivas oriundas do Paraná, Mato Grosso e Goiás. A frequência de resistência estimada a Vip3Aa20 variou de 0,0140 a 0,0367 nas populações avaliadas, sendo que a frequência total foi de 0,0076. Neste estudo, fornecemos informações para refinar as estratégias de MRI, além de introduzir novas técnicas para monitorar a resistência de S. frugiperda a tecnologias Bt no Brasil.

A proteinaceous trypsin inhibitor was purified from Crotalaria pallida seeds by ammonium sulphate fractionation, affinity chromatography on immobilized Trypsin-Sepharose and TCA precipitation. The trypsin inhibitor, named ITC, had Mr of 32.5 kDa by SDS-PAGE and was composed by two subunits with 27.7 and 5.6 kDa linked by disulphide bridges, a typical characteristic of Kunitz-Inhibitor family. ITC was stable until 50°C, and at 100°C its residual activity was of about 60%. Also, ITC was stable at pHs 2 to 12. The inhibition of trypsin by ITC was non-competitive, with a Ki of 8,8 x 10-7M. ITC inhibits weakly other serine proteinases such as chymotrypsin and elastase. The inhibition of papain (44% of inhibition), a cysteine proteinase was an indicative of the bi-functionality of ITC. In vitro assays against digestive proteinases from several Lepdoptera, Diptera and Coleoptera pests were made. ITC inhibited in 100% digestive enzymes of Ceratitis capitata (fruit fly), Spodoptera frugiperda and Alabama argillacea, the last one being a cotton pest. It also inhibited in 74.4% Callosobruchus maculatus (bean weevil) digestive enzymes, a Coleoptera pest. ITC, when added in artificial diet models, affected weakly the development of C. capitata larvae and it had a WD50 of 2.65% to C. maculatus larvae

The objective of this work was to evaluate the biological variables of Spodoptera frugiperda on species of cover crops. The experiments were conducted in laboratory and greenhouse using the following species: sunflower (Helianthus annuus), sun hemp (Crotalaria juncea), brachiaria (Urochloa decumbens e Urochloa ruziziensis), millet (Pennisetum americanum), black oat (Avena stringosa), white lupin (Lupinus albus), forage turnip (Rafanus sativus) and maize (Zea mays). In laboratory the S. frugiperda larval survival varied from 57%, on L. albus, to 93% on H. annuus and the survival of the pre-imaginal phase varied from 45% on U. decumbens to 81.6% on Z. mays. On C. juncea the larval biomass was lower and the development period of the young and larval stage was higher. The adaptation index was less on C. juncea in greenhouse and laboratory. In greenhouse the larval survival at 14 days was similar for all plants and at 21 days was the lowest on C. juncea. There was less accumulation of biomass at 14 days on C. juncea and at 21 days on C. juncea and A. stringosa. Regarding damage, C. juncea presented less susceptibility to Spodoptera frugiperda attack, which together with the other evaluated parameters, indicated this plant as the most appropriate for soil cover before cultivation of maize.
