A pH indicator film based on cassava starch plasticized with sucrose and inverted sugar and incorporated with grape and spinach extracts as pH indicator sources (anthocyanin and chlorophyll) has been developed, and its packaging properties have been assessed. A second-order central composite design (2(2)) with three central points and four star points was used to evaluate the mechanical properties (tensile strength, tensile strength at break, and elongation at break percentage), moisture barrier, and microstructure of the films, and its potential as a pH indicator packaging. The films were prepared by the casting technique and conditioned under controlled conditions (75% relative humidity and 23 degrees C), at least 4 days before the analyses. The materials were exposed to different pH solutions (0, 2, 7, 10, and 14) and their color parameters (L*, a*, b*, and haze) were measured by transmittance. Grape and spinach extracts have affected the material characterization. Film properties (mechanical properties and moisture barrier) were strongly influenced by extract concentration presenting lower results than for the control. Films containing a higher concentration of grape extract presented a greater color change at different pH`s suggesting that anthocyanins are more effective as pH indicators than chlorophyll or the mixture of both extracts. The crude cell-free medium from a culture of Kluyveromyces marxianus var. bulgaricus was immobilized in a gelatin-water support, with an immobilization yield of 82.60% for inulinase activity. The optimum pH for both free and immobilized inulinase was the same (3.5) and the optimum temperatures were 55 degrees C for the free and 60 degrees C for the immobilized enzyme. The Arrhenius plots were linear and activation energies were 56.20 (free enzyme) and 20.27 kj/mol K (immobilized enzyme). The kinetic parameters were calculated by Lineweaver-Burk plots and the V-max and K-m were 37.60 IU/mg protein and 61.83 mM for the free inulinase and 31.45 IU/mg protein and 149.28 mM for the immobilized enzyme, respectively. The operational stability of the immobilized inulinase was studied in a continuous fixed-bed column reactor for 33 days, at the end of which the sucrose conversion was 58.12%. (c) 2008 Elsevier Ltd. The development in the area of creates was studied of 14 nuclei with four mass off cells from the division of nine beehives of africanized Apis mellifera honeybees, distributed in two treatments: TPL - nuclei fed with inverted sugar + 3.5ml of the vitaminic amino acid supplement (Promotor L® ), composition for six nuclei and TAI - nuclei fed with composed inverted sugar for eight nuclei. The nuclei had been fed weekly in individual feeder's type tray, and the evaluations carried through in four periods, totalizing 74 days. The treatments had not presented significant difference, being that, number the TPL presented area of creates inferior to TAI (233.63 versus. 273.02cm 2, respectively). How much to the periods the four was superior (P<0.05) to the first and as second, being that the third did not present significant difference (P<0.05) in relation to the others. The use of the TAI was economically more favorable in relation to the TPL in R$0.21 in relation to the cost for production of 1kg of food.

A criação de abelhas sem ferrão é antiga nas Américas. No entanto, a atividade ainda esta subdesenvolvida, necessitando de melhorias nas técnicas de manejo. Este estudo avaliou alternativas nutricionais para a abelha amazônica Melipona flavolineata, visando substituição da alimentação com mel e pólen fermentado. As seguintes alternativas nutricionais foram comparadas com o mel (T1): xarope de açúcar invertido com minerais (T2) e este xarope acrescido de solução de aminoácidos e vitaminas nas seguintes concentrações: 0,5% (T3), 1,0% (T4) e 10% (T5). Ao pólen fermentado (T1) foram comparados alimentos fermentados com inóculo de pólen fermentado de M. flavolineata, constituídos de: pólen apícola (T2); levedo (T3); e extrato de soja, em duas diferentes concentrações de proteínas (T4, 12% e T5, 18%). O valor nutricional das dietas foi baseado no consumo diário, no peso das operárias, no tamanho dos acinos das glândulas hipofaringeas e no tamanho dos oócitos. Avaliaram-se, tambem, os custos dos ingredientes das dietas. Os resultados obtidos indicaram que dentre as alternativas para o mel, não houve diferenças estatísticas entre o controle e todos os tratamentos. Foram encontradas diferenças apenas entre os tamanhos dos acinos das operárias alimentadas com os tratamentos 2 e 4, sendo o tratamento 4 superior. Os custos dos ingredientes indicaram o tratamento 2 como sendo mais barato que os demais. Dentre as alternativas para o sabura foram encontradas maiores diferenças. O consumo do tratamento 4 foi significativamente superior ao do T3, indicando maior aceitação do extrato de soja em relação ao levedo. Em relação aos demais parâmetros avaliados, o tratamento 5 foi superior a todos os demais e seu custo foi muito inferior ao do T2 e semelhante ao dos T3 e T4. Concluiu-se que as melhores alternativas para a nutrição de M. flavolineata sao o xarope de acucar invertido com minerais, e o pólen fermentado semi-artificial à base de extrato de soja. Colônias recém-formadas, com 500-600 abelhas, devem receber 10 a 20 ml de xarope e 25 g de pólen fermentado semi-artificial semanalmente.

O trabalho teve como objetivo desenvolver uma alimentação artificial protéica a base de extrato de soja (saburá artificial), e avaliar o seu efeito sobre a razão sexual, longevidade de operárias e desenvolvimento de colônias recém-divididas de Melipona fasciculata além de verificar a adaptação da espécie dentro de casas de vegetação. O saburá artificial aqui desenvolvido é constituído de 50g de extrato de soja, 20g de saburá fresco e 60ml de xarope de açúcar invertido (60%). Foi utilizado anilina para colorir o saburá artificial na tentativa de rastrear e verificar o consumo pelas operárias dentro das colônias. Foram utilizadas cinco colônias, das quais três receberam somente saburá e duas o saburá artificial. Não houve diferença significativa entre a produção de rainhas e operárias nos dois tratamentos e nos dois casos não houve produção de machos. As operárias que nasceram de caixas alimentadas com saburá artificial apresentaram maior longevidade e menor peso ao nascer. Estas caixas ainda iniciaram o processo de construção de células e postura mais cedo que as caixas alimentadas com saburá, contudo, suas rainhas apresentaram menor taxa de oviposição diária. O alimento a base de extrato de soja (saburá artificial) não afetou negativamente colônias recém divididas de M. fasciculata. Nos primeiros dias, as abelhas passaram a maior parte do seu tempo no topo da casa-de-vegetação tentando fugir, somente após o terceiro dia houve redução na mortalidade das operárias. Não houve diferença significativa, ao longo de cinco dias, entre a mortalidade de operárias em caixas transferidas, durante a noite e durante o dia, para dentro da casa-de-vegetação. A anilina se mostrou uma excelente ferramenta para controlar os alimentos manuseados e consumidos pela colônia.

Chemically inducible gene switches can provide precise control over gene expression, enabling more specific analyses of gene function and expanding the plant biotechnology toolkit beyond traditional constitutive expression systems. The alc gene expression system is one of the most promising chemically inducible gene switches in plants because of its potential in both fundamental research and commercial biotechnology applications. However, there are no published reports demonstrating that this versatile gene switch is functional in transgenic monocotyledonous plants, which include some of the most important agricultural crops. We found that the original alc gene switch was ineffective in the monocotyledonous plant sugar cane, and describe a modified alc system that is functional in this globally significant crop. A promoter consisting of tandem copies of the ethanol receptor inverted repeat binding site, in combination with a minimal promoter sequence, was sufficient to give enhanced sensitivity and significantly higher levels of ethanol inducible gene expression. A longer CaMV 35S minimal promoter than was used in the original alc gene switch also substantially improved ethanol inducibility. Treating the roots with ethanol effectively induced the modified alc system in sugar cane leaves and stem, while an aerial spray was relatively ineffective. The extension of this chemically inducible gene expression system to sugar cane opens the door to new opportunities for basic research and crop biotechnology.

Guanine rich sequences adopt a variety of four stranded structures, which differ in strand orientation and conformation about the glycosidic bond even though they are all stabilised by Hoogsteen hydrogen bonded guanine tetrads. Detailed model building and molecular mechanics calculations have been carried out to investigate various possible conformations of guanines along a strand and different possible orientations of guanine strands in a G-tetraplex structure. It is found that for an oligo G stretch per se, a parallel four stranded structure with all guanines in anti conformation is favoured over other possible tetraplex structures. Hence an alternating syn-anti arrangement of guanines along a strand is likely to occur only in folded back tetraplex structures with antiparallel G strands. Our study provides a theoretical rationale for the observed alternation of glycosidic conformation and the inverted stacking arrangement arising from base flipover, in antiparallel G-tetraplex structures and also highlights the various structural features arising due to different types of strand orientations. The molecular mechanics calculations help in elucidating the various interactions which stabilize different G-tetraplex structures and indicate that screening of phosphate charge by counterions could have a dramatic effect on groove width in these four stranded structures.
