Due to the increasing search for alternative sources of natural rubber (NR) whose properties are similar to Hevea brasiliesis, several sources have been studied in the past few years. Among them, Mangabeira (Hancornia speciosa Gomes), which is native to Amazon rainforest and other regions of Brazil, has a potential as another viable rubber source. As a continuation of a series of comparative studies between Hancornia and Hevea (clone RRIM 600) these two species by our research team, their thermal behavior was analyzed by thermogravimetry (TG) using Flynn-Wall-Ozawa's approach in order to obtain kinetic parameters (reaction order, pre-exponential factor and activation energy) of the decomposition process. Results indicated that the thermal behavior of NR from Hancornia was comparable to Hevea with some differences observed as follows: reaction order for Hancornia was higher than for Hevea at the beginning of degradation and very close for temperatures over 350 A degrees C; activation energy and pre-exponential factor had the same trend, i.e., increased with increasing degree of conversion remaining almost constant between 20 and 70% and then increasing for higher degrees, although Hevea was slightly more thermally stable than Hancornia. These major influences in the degradation process in the early stage are attributed to differences in non-rubber constituents present in these two species.

A mangabeira (Hancornia speciosa Gomes) destaca-se por possuir um grande potencial como planta frutífera e produtora de borracha. As dificuldades encontradas no seu processo de propagação por meio de sementes, devido, principalmente, à baixa taxa de germinação e à recalcitrância, valorizam a busca por soluções alternativas para a produção de mudas dessa espécie, de maneira rápida e eficiente. Objetivou-se, neste trabalho, realizar o estudo da germinação de sementes de mangabeira em condições in vitro, tendo como precedente a obtenção de explantes, para posterior utilização no cultivo in vitro. Neste estudo foram avaliados os efeitos de diferentes meios de cultura, concentrações de sacarose e GA3 e de três níveis de pH na germinação da mangabeira. Frutos maduros foram coletados, passaram por processo de beneficiamento e tiveram suas sementes retiradas e utilizadas como explantes. Maior porcentagem de germinação de sementes de mangabeira in vitro foi obtida com a utilização dos meios de cultura WPM e MS/2, suplementados com 15,0 g L-1 de sacarose, 0,2 mg L-1 de GA3 e com pH corrigido para 5,8.

Este trabalho teve como objetivo estudar o efeito das poliaminas espermidina e espermina no crescimento de calos Hancornia speciosa Gomes. Calos com 0,5 cm de diâmetro foram inoculados em meio Murashige & Skoog (1962) (MS) a 50% + 100 mg L-1 de caseína hidrolisada + 200 mg L-1 de levedura de cerveja, variando os tratamentos:A: 1 mmol de espermina + 2 mg L-1 de 2,4-D (ácido 2,4 diclorofenoxiacético) + 0,5 mg L-1 de NAA (ácido naftalenoacético); B: 1 mmol de espermidina + 2 mg L-1 de 2,4-D + 0,5 mg L-1 de NAA; C: 2 mg L-1 de 2,4-D + 0,5 mg L-1 de NAA. Não houve influência das poliaminas no crescimento dos calos. observou-se, nos calos tratados com espermidina, maior concentração celular de putrescina (582,37 µg g mf-1) aos 60 dias, maior teor de espermidina (502,54 µg g mf-1) e espermina (868,53 µg g mf-1) aos 40 dias de cultivo, quando se aplicou a própria poliamina. Conclui-se que a aplicação exógena de poliaminas em Hancornia speciosa não proporciona aumento no crescimento de calos. A oxidação promovida por longos períodos de cultivo in vitro induz aumento nos níveis de putrescina.

Hancornia speciosa Gomes (Apocynaceae), popularly known as ‘mangabeira’, has been used in folk medicine to treat inflammatory disorders, hypertension, dermatitis, diabetes, liver diseases and stomach disorders. Regarding the Hancornia speciosa fruits, the ethnobotany indicates its use especially for treating inflammation and tuberculosis. However, no study has been done so far to prove such biological activities. The objective was evaluation anti-inflammatory activity from the fruits of Hancornia speciosa Gomes (mangabeira). Aqueous extract was prepared by decoction, subsequently submitted the liquid-liquid fractionation. The secondary metabolites were identified by high performance liquid chromatography coupled with detector diode array (HPLC-DAD) and liquid chromatography diode array detector coupled with mass spectrometry (LC-DAD-MS). The anti-inflammatory properties of the aqueous extract, dichloromethane (CH2Cl2), ethyl acetate (EtOAc) and n-butanol (n-BuOH) fractions of the fruits from H. speciosa, as well as rutin and chlorogenic acid were investigated using in vitro and in vivo models. In vivo tests comprised the xylene-induced ear edema that was measured the formation of edema, carrageenan-induced peritonitis was evaluated the total leukocytes at 4h and zymosan-induced air pouch was measured the total leukocytes and differential cell count at 6, 24 and 48 hours, whereas in vitro tests were evaluated levels of cytokines IL-1β, IL-6, IL-12 and TNF-α using ELISA obtained of carrageenan-induced peritonitis model. The results showed the presence of rutin and chlorogenic acid were detected in the aqueous extract from H. speciosa fruits by HPLC-DAD and LC-DAD-ME. Furthermore, the aqueous extracts and fractions, as well as rutin and chlorogenic acid significantly inhibited the xilol-induced ear edema and reduced cell migration in the animal models such as carrageenan-induced peritonitis and zymosan-induced air pouch. In addition, reduced levels of cytokines IL-1β, IL-6, IL-12 and TNF-α were observed. This is the first study that demonstrated the anti-inflammatory effect of aqueous extract from Hancornia speciosa fruits against different inflammatory agents in animal models, suggesting that their bioactive molecules, especially rutin and chlorogenic acid contributing, at least in part, to the anti-inflammatory effect of aqueous extract. These findings support the widespread use of Hancornia speciosa in popular medicine and demonstrate that this aqueous extract has therapeutic potential for the development of a herbal drugs with anti-inflammatory properties.

Hancornia speciosa Gomes é uma espécie conhecida popularmente no Brasil como mangabeira, cujo fruto apresenta alto valor nutricional. O conhecimento sobre a sua fisiologia é ainda escasso, principalmente no que se refere ao desenvolvimento inicial. Dessa forma, o objetivo do presente trabalho foi avaliar os efeitos de diferentes níveis de déficit hídrico sobre o padrão de crescimento, fluorescência de clorofila e relações hídricas em mudas de mangabeira. Foi utilizado um esquema fatorial (tratamentos x época de avaliação) com quatro tratamentos hídricos com base na capacidade de campo (CC) (80%, 60%, 40% e 20%), com cinco repetições. Foram avaliados a altura das plantas, número de folhas, diâmetro do caule, produção e partição de biomassa, eficiência quântica do fotossistema II (PSII), potencial hídrico (?w), teor relativo de água (TRA) e teor de carboidratos, proteínas e prolina. O déficit hídrico severo (20% CC) levou a uma redução no crescimento e alterou o padrão de partição de biomassa nas mudas. No entanto, as relações hídricas não foram significativamente afetadas, pois as mudas mantiveram altos valores de ?w e TRA, sem acúmulos significativos nos teores de solutos orgânicos quando cultivadas com 20%CC. Além do mais, a eficiência quântica do PSII não foi afetada pelos diferentes regimes hídricos, sugerindo que não houve fotoinibição devido ao estresse hídrico. A mudança no padrão de crescimento, com um incremento no aprofundamento das raízes e redução no crescimento da parte aérea parece ser a principal estratégia das mudas de H. speciosa para a manutenção da hidratação dos tecidos durante períodos de déficit hídrico.

Hancornia speciosa Gomes is a fruit tree native from Brazil that belongs to Apocinaceae family, and is popularly known as Mangabeira. Its fruits are widely consumed raw or processed as fruit jam, juices and ice creams, which have made it a target of intense exploitation. The extractive activities and intense human activity on the environment of natural occurrence of H. speciosa has caused genetic erosion in the species and little is known about the ecology or genetic structure of natural populations. The objective of this research was the evaluation of the genetic diversity and genetic structure of H. speciosa var. speciosa. The genetic variability was assessed using 11 allozyme loci with a sample of 164 individuals distributed in six natural populations located in the States of Pernambuco and Alagoas, Northeastern Brazil. The results showed a high level of genetic diversity within the species (e= 0.36) seeing that the most of the genetic variability of H. speciosa var. speciosa is within its natural populations with low difference among populations ( or = 0.081). The inbreeding values within ( = -0.555) and among populations ( =-0.428) were low showing lacking of endogamy and a surplus of heterozygotes. The estimated gene flow ( m ) was high, ranging from 2.20 to 13.18, indicating to be enough to prevent the effects of genetic drift and genetic differentiation among populations. The multivariate analyses indicated that there is a relationship between genetic and geographical distances, which was confirmed by a spatial pattern analysis using Mantel test (r = 0.3598; p = 0.0920) with 1000 random permutations. The high genetic diversity index in these populations indicates potential for in situ genetic conservation.

Direct flow injection electrospray ionization ion trap tandem mass spectrometry (ESI-IT-MS/MS) was used to investigate the polyphenolic compounds present in an infusion from the barks of Hancornia speciosa Gom. (Apocynaceae), a native Brazilian plant popularly known as 'mangabeira', used as a source of nutrition and against gastric disorders. After a simple sample filtration pretreatment the characteristic fingerprint of the infusion was performed in negative ion ESI mode in a few minutes. At low capillary-voltage activation, the deprotonated molecules ([M-H](-)) were observed and using collision-induced dissociation the product ion spectra showed the presence of a homologous series of B-type proanthocyanidins, as well as another series containing their respective C-glycosylated derivatives, with a degree of polymerization from 1 up to 6 units of interlinked catechins. Therefore, direct flow injection allowed us to identify the key compounds without preparative isolation of the components. Copyright (C) 2007 John Wiley & Sons, Ltd.
