Monócitos e macrófagos são células da imunidade inata que desempenham importante papel na defesa contra infecções fúngicas, através do reconhecimento dos fungos, ativação e desenvolvimento de atividade fungicida. A função das células fagocitárias depende do seu estado de ativação, induzido principalmente pelo ambiente de citocinas, presentes durante a interação com o microrganismo. O presente projeto teve por objetivo avaliar o efeito da opsonização de células leveduriformes de Paracoccidioides brasiliensis com soro humano sobre a produção de peróxido de hidrogênio (H2O2) e atividade fungicida de monócitos humanos, estimulados ou não com Interferon-gama (IFN-γ), contra P. brasiliensis. Monócitos de sangue periférico, obtidos de indivíduos saudáveis, foram cultivados na ausência ou presença de IFN-γ por 24h a 37oC. A seguir, essas células foram desafiadas com a amostra Pb18 de P. brasiliensis por 60 min para determinação da produção de H2O2 e por 4h para a avaliação da atividade fungicida nas proporções fungomonócito de 1:50. A suspensão fúngica foi previamente incubada por 30 min a 37oC na ausência de soro (SS) ou na presença de pool de soro humano normal, inativado (SI) por aquecimento a 56oC ou soro humano fresco (SF) não submetido à inativação do sistema complemento, nas concentrações de 5%, 10%, 20% e 30% em meio de cultura RPMI. A atividade fungicida de monócitos contra a amostra Pb18 foi avaliada por plaqueamento dessas co-culturas em meio de cultivo BHIágar e determinação da recuperação de fungos viáveis após 14 dias de incubação a 36oC. A produção de H2O2 por monócitos foi determinada através da técnica de redução do vermelho de fenol. A Tuberculose (TB) é a principal causa de óbitos entre as doenças infecciosas causadas por um único agente. De acordo com a Organização Mundial da Saúde (OMS) o agente etiológico da TB no homem, o complexo Mycobacterium (M. tuberculosis, M. africanum, M. bovis) é responsável por cerca de 8 milhões de novas infecções e 3 milhões de mortes a cada ano no mundo. No começo da década de 80, a reemergência da TB em países em desenvolvimento deve-se à crescente incidência do Vírus da Imunodeficiência Humana (HIV), à falta de recursos para o tratamento desta doença e à proliferação de cepas resistentes a múltiplas drogas (MDR-TB). Esta situação criou a necessidade da busca por novos agentes antimicobacterianos capazes de reduzir o tempo de tratamento, melhorar a adesão dos pacientes ao mesmo e ser efetiva contra cepas MDR-TB. A via do chiquimato leva à biossíntese do corismato, o precursor de aminoácidos aromáticos, tirosina, triptofano e fenilalanina. A primeira reação na biossíntese de fenilalanina envolve a conversão de corismato a prefenato, catalisada pela corismato mutase. A segunda reação na biossíntese de fenilalanina é a descarboxilação e desidratação de prefenato a fenilpiruvato, catalisada pela prefenato desidratase. Embora ausente em mamíferos, esta via está presente em bactérias, algas, fungos, plantas e parasitos do Phyllum Apicomplexa. Esta rota é essencial em M. tuberculosis e, portanto, suas enzimas representam alvos potenciais para o desenvolvimento de novas drogas antimicobacterianas. O objetivo deste trabalho foi estudar o gene pheA da linhagem de M. tuberculosis H37Rv e seu produto, a enzima prefenato desidratase Para isso, DNA genômico de M. tuberculosis H37RV foi extraído e o gene pheA foi amplificado pela técnica de PCR, clonado no vetor de expressão pET-23a(+), seqüenciado e superexpresso em células de Escherichia coli BL21(DE3). Os resultados obtidos confirmaram a região predita para o gene pheA, que foi amplificado com sucesso, mostrando 963 pb, sendo que a presença de 10% dimetil sulfoxido (DMSO) mostrou ser essencial para permitir a desnaturação do DNA rico em bases G-C. Análise da seqüência nucleotídica pelo método de Sanger confirmou a identidade do gene clonado e demonstrou que nenhuma mutação foi introduzida pelos passos de PCR e clonagem. A enzima prefenato desidratase foi superexpressa em células de E. coli BL21(DE3) eletroporadas com pET-23a(+)::pheA. Análise por SDS-PAGE mostrou expressão significativa de uma proteína com aproximadamente 33kDa, estando de acordo com a massa molecular esperada para a prefenato desidratase. A proteína recombinante foi superexpressa sem a adição de IPTG, e a presença da proteína pôde ser detectada em todos os intervalos de tempo testados (6, 9 e 24 horas depois da OD600nm alcançar o valor de 0,5). Foi realizado ensaio enzimático com a prefenato desidratase de acordo com Gething et al. (1976) utilizando prefenato de bário como substrato e coeficiente de extinção molar de 17.500 a 320 nm para calcular a concentração de fenilpiruvato. Houve um aumento de 1766 vezes na atividade específica da prefenato desidratase no extrato bruto da proteína recombinante em relação ao controle, no qual o vetor pET23a(+) sem o gene pheA foi introduzido em células de E. coli BL21(DE3).

Fungal polysaccharides have received a great deal of attention due to itsbecause of their potential use in a wide rangegreat variety fromof industries. Some studies have demonstrated that polysaccharides extracted offrom basidiomycetes they have presented significant properties as anti-inflammatory, antimicrobial, antioxidant and anti-tumoral properties. In spite of thisDespite these potential properties, these mushrooms have not been insufficiently investigated, and the great number of antibiotics number produced forby these organisms suggests that they canmay be a new source of bioactives composites source. In tThe present work, reports onlated the chemical composition, potential antioxidant, antiinflammatory and citotoxycity of extracted polymers extracted offrom the fruits bodies of the fungiius Geastrum saccatum and Polyporus dermoporus, native mushrooms of the Atlantic forest inof the state of the Rio Grande do Norte, Brazil. The Cchemical analyses had revealed ademonstrated text of total sugar rates of 65% and 49%, and proteins of 7.0% for in extracts of G. saccatum and P. dermoporus extracts, respectively. The analyses ofNMR spectroscopy of RMN had demonstrated that these extracts are composites forof a complex involving β- glucans and- proteins complex. The inhibition of the formation of superoxide radicals formation was of 88.4% in G. saccatum and 83.3% in P. dermoporus, and 75 and 100% for inhibition of hydroxyls radicals inhibition. TopicalThe topic application of extracts the 10, 30 and 50 mg/kg extract in BALBc mice with cutaneous inflammation induced byfor croton oil demonstrated to inhibitedion of ear edema of ear and cells polimorfonuclears cells atin the inflamed siteplace, being this reply more effective in lower concentrations being more effective. The evaluation of the glucans of G. saccatum and P. dermoporus glucans under induced pleurisy for carrageenan-induced pleurisya of showed the antiinflammatory action of these composites., being analyzed tThe frame number in the pleural exudates and thedosage of nitric oxide dosage was also analyzed. The cytotoxic action of these polymers was analyzed throughthrough the mitochondrial function (MTT). The incubation of the glucans with mononuclear cells of the peripheral blood demonstrated that the extracted glucans extracted fromof G. saccatum havepossess a moderate cytotoxic action. These results suggest that these mushrooms possess polymers formed byfor a complex glucana-protein complex, with antiinflammatory and antioxidant actions

Globulins fractions of legume seeds of Crotalaria pallida, Erytrina veluntina and Enterolobium contortisiliquum were isolated and submitted to assays against serine, cysteine and aspartic proteinases, as also amylase present in midgut of C. maculatus and Z. subfasciatus. Hemagglutination assays indicated presence of a lectin in E. veluntina globulin fractions. This lectin had affinity to human erythrocytes type A, B and O. Vicilins were purified by chromatography on Sephacryl S-300 followed of a chromatography on Sephacryl S-200, which was calibrated using protein markers. Vicilins from C. pallida (CpV) and E. veluntina (EvV) seeds had a molecular mass of 124.6 kDa and E. contortisiliquum a molecular mass of 151kDa. Eletrophoresis in presence of SDS showed that CpV was constituted by four subunities with apparent molecular mass of 66, 63, 57 and 45 kDa, EvV with three subunities with apparent molecular mass of 45kDa and EcV four subunities, two with 37.1 kDa and two with 25.8 kDa. Non denaturantig eletrophoresis displayed single bands with high homogeneity, where CpV had lower acidic behavior. All vicilins are glycoproteins with carbohydrate contents at 1 to1.5%. Bioassays were done to detect deleterious effects of vicilins against C. maculatus and Z. subfasciatus larvae. CpV, EvV and EcV exhibited a WD50 of 0.28, 0.19 and 1.03%; LD50 0.2, 0.26, and 1.11% respectively to C. maculatus. The dose responses of CpV, EvV and EcV to Z. subfasciatus were: WD50 of 0.12, 0.14, 0.65% and LD50 of 0.09, 0.1, and 0.43% respectively. The mechanism of action of these proteins to bruchids should be based on their properties of bind to chitin present in mid gut of larvae associated with the low digestibility of vicilin. In assays against phytopatogenous fungus, only EcV was capable of inhibit F. solani growth at concentrations of 10 and 20 µg and its action mechanism should be also based in the affinity of EcV to chitin present in the fungi wall

Ectomycorrhizal associations are poorly known from tropical lowlands of South America. Recent field trips to the reserve Parque Estadual das Dunas in Natal, in Rio Grande do Norte state, Brazil, revealed a undocumented community of ectomycorrhizal fungi. This type of Mycorrhizal association is frequently in the north hemisphere in temperate and boreal forests. The aim of this work is to analyze the occurrence of ectotrophic areas in atlantic rainforest. Collections along and around the trails in the reserve revealed six genera of putatively ECM fungi which belong to the basidiomycete, Amanitaceae, Boletaceae, Russulaceae, Entolomataceae, and Sclerodermataceae family which are poorly documented in Brazil. Plants belonging to Myrtaceae, Polygonaceae, Leguminosae/Caesalpinioideae, Erythroxylaceae, Malphigiaceae, Bromeliaceae, Loganiaceae, Sapotaceae e Celastraceae were found living next to the species of fungi analized. Our results suggest that the area studied is an ectotrophic environment which shows high diversity of putatively ECM fungi and some plants probably host ECM. The tropical lands are a potential focus to study reinforced by the new records of Scleroderma in Brazil and Northwest of Brazil

Gasteroids fungi are characterized by the basidiospores maturation inside the basidioma, from which spores liberation occurs in a passive manner. These fungi were once seen as a well definite class of Basidiomycota, but nowadays they are considered an artificial assemblage, because the organisms have independent evolutionary histories forming a polyphyletic group with a vast morphological variety. Despite their diversity, studies with this group in the tropics are incipient, and the phylogenetic relationships of the species from temperate climate remain unknown. Thus, this work aimed to elucidate the phylogenetic relationships of gasteroids fungi from the Geastrales and Phallales orders, with the inclusion of tropical and temperate species, and with these analyses suggest a systematic position of species like Aseroë floriformis and Phallus roseus, as well as to verify if the lignicolous habit can indicate parental relationship in the Geastrum genus. For this, basidiomata were collected at Atlantic rain forest areas, during the rainy season, and the specimen identification followed specific literature for gasteroid fungi. The phylogenetic analyses were performed with Maximum Parsimony and Bayesian Analysis, making use of RPB2 and 28S nuclear genes and atp6 mitochondrial gene. It could be observed on the Phallales dendogram, that Aseroë floriformis did not cluster with A. rubra, and that it has an anterior divergence from all others species of the family Clathraceae used in this analysis, assuming a basal position in the clade. Phallus roseus, which once was recognized as Itajahya, has previous divergence from the group formed by Phallus species. At the Geastrales dendogram, in the group corresponding to Geastrum genus, it could be observed that species with lignicolous habitat clustered in a clade with high support values. So, the results suggest the creation of a new genus to accommodate A. floriformis, and the revalidation of Itajahya, as well as it can be affirmed that the lignicolous habitat on the Geastrum genus in fact indicates parental relationships, and that it has arised only once at the evolutionary history of the genus

As espécies de entomopatógenos que causam impacto em populações de hospedeiros são muitas. Entretanto, informações básicas sobre sua biologia, identificação e relação entre o sistema patógeno-hospedeiro são escassas. Este estudo teve por objetivo identificar os fungos associados a Parlatoria ziziphus (Lucas)em pomares citrícolas do município de Taiúva, SP. Foram realizadas coletas mensais, em dois pomares de laranja (Citrus sinensis Osbeck), variedade Pera, de janeiro de 1995 a fevereiro de 1996. Amostraram-se 15 árvores em cada pomar, retirando-se 16 folhas por árvore. Estas foram levadas ao laboratório, para determinação dos fungos associados à cochonilha. Para a identificação dos fungos, consideraram-se aspectos da estrutura reprodutiva e do esporo. Foram identificadas cinco espécies, citadas como patógenos de diaspidídeos por vários autores: Fusarium coccophilum (Desm.) Wr. & Rg., Nectria flammea (Tul.) Dingley (fase sexual de F. coccophilum), Tetracrium coccicolum Hohnell, Podonectria coccicola (Ellis & Everhart) Petch (fase sexual de T. coccicolum) e Myriangium duriaei Mont. & Berk. Fusarium coccophilum foi a espécie mais freqüente e a mais prevalente, na maioria das coletas realizadas, em ambas as áreas amostradas.

Gasteroid fungi include several distinct lineages of basidiomycetes that were grouped by presenting some striking features in common like angiocarpic development and passive release of basidiospores. For a long period these fungi were accomodate in Gasteromycetes class. However, biochemistry and molecular studies showed the polyphyly of this group and curriently this class lies devoid of taxonomic value. These organisms influence the ecology of the various biomes, however, are poorly studied and knowledge of their diversity in neotropical ecosystems remains insufficient, despite studies that have been developed in recent years. The Brazilian semiarid region has many areas with an unexplored mycobiota. Ceará State has many areas of extreme biological importance and for this study three protected areas were chosen: Área de Proteção Ambiental da Serra da Ibiapaba, Parque Nacional de Ubajara and Área de Proteção Ambiental de Baturité. Therefore, the aim of this study was to inventory gasteroid fungi ocorring in these three areas in the semiarid region of Ceará. The expeditions, herborization and analysis of specimes were based in traditional methodology used to identify gasteroid fungi. Were identified 16 species belong to 5 families: Agaricaceae, Clathraceae, Geastraceae, Nidulariceae and Phallaceae. Morganella nuda Alfredo & Baseia is recorded for the second time in the world, Blumenavia angolensis (Welw. & Curr) Dring and Mutinus elegans (Mont.) E. Fisch. corresponds to a first record in the Brazilian Northeastern. Except for Abrachium floriforme (Baseia & Calonge) Baseia & T.S. Cabral and Geastrum lloydianum Rick, all remaining species are new records for Ceará, increasing the list of gasteroid fungi in the region of 3 for 17 species. Identified species were deposited in the collection of the Herbarium of Universidade Federal do Rio Grande do Norte. Although these areas have proved conducive to the study of gasteroid fungi, more efforts are still needed to increase knowledge of these fungi in the region

Three species of filamentous fungi, Aspergillus niger, Penicillium fellutanum and Mucor hiemalis, were selected and cultivated in vinasse media with different addition of molasses, pasteurized to 85°C for 30 minutes and with pH = 5.0. The microorganisms, previously adapted to the respective medium for 48 hours, from a solution of 107 spores.ml-1, were cultivated in pure and mixed cultures in Erlenmeyer vessel of 500ml, to 30°C, with constant agitation of 170 rpm, for 24, 48 and 72 hours, with four repetition for each samples. The biomass was separated by vacuum filtration in filter Whatman #1 and dried in oven at 105°C until right weight, the obtained liquid was submited to COD analysis. The datas were statistically analysed using a response surface methodology, to improve the effect on the molasses proportion and culture time, in the biomass production by microorganism in research. According to the obtained results (5.02% of molasses, 55.59h, 70% of spores solution of A. niger and 30% of spores solution of P. fellutanum), cultivating was carried out in Microferm Fermentor New Brunswick for 48 hours at 300 rpm, aired at 1v/v/m, using 5 liters of medium added with 5.0% of molasses on the conditions above described. The average of the results obtained (6.81g.l-1) was higher than the confidence interval (5.937 ; 6.369) and was inside the prediction interval (4.471 ; 7.834) both of them significant at 95% by the statistical test employed.
