Patterns of the biosynthesis ofmajor metabolites of the oleaginous yeast Cryptococcus curvatus NRRL Y-1511 were investigated during cultivation on sugar-based media. When lactose or sucrose was employed as substrate under nitrogen-limited conditions, the yeast strain accumulated high quantities of intra-cellular total sugars (ITS) at the beginning of fermentation (up to 68% w/w), with ITS values progressively decreasing to 20%, w/w, at the end of the fermentation. Decrease in ITS content and consumption of extracellular lactose led to a subsequent rise in lipid accumulation, reaching 29.8% in dry cell weight at 80 g/L of initial lactose concentration. Lactose was a more favorable substrate for lipid production than sucrose. In nitrogen-excess conditions, ITS were produced in significant quantities despite the continuous presence of nitrogen in the medium. Growth on lactose was not followed by secretion of extra-cellular b-galactosidase. High quantities of extra-cellular invertase were observed during growth on sucrose. The composition of ITS was highly influenced by the sugar used as substrate. Cellular lipids contained mainly palmitic and to lesser extent linoleic and stearic acids. This is the first report in the literature that demonstrates the interplay between the biosynthesis of intra-cellular total sugars and lipid synthesis for oleaginous yeast strains.

The therapeutic efficacy of amphotericin B and voriconazole alone and in combination with one another were evaluated in immunodeficient mice (BALB/c-SCID) infected with a fluconazole-resistant strain of Cryptococcus neoformans var. grubii. The animals were infected intravenously with 3 x 10(5) cells and intraperitoneally treated with amphotericin B (1.5 mg/kg/day) in combination with voriconazole (40 mg/kg/days). Treatment began 1 day after inoculation and continued for 7 and 15 days post-inoculation. The treatments were evaluated by survival curves and yeast quantification (CFUs) in brain and lung tissues. Treatments for 15 days significantly promoted the survival of the animals compared to the control groups. Our results indicated that amphotericin B was effective in assuring longest-term survival of infected animals, but these animals still harbored the highest CFU of C. neoformans in lungs and brain at the end of the experiment. Voriconazole was not as effective alone, but in combination with amphotericin B, it prolonged survival for the second-longest time period and provided the lowest colonization of target organs by the fungus. None of the treatments were effective in complete eradication of the fungus in mice lungs and brain at the end of the experiment.

The increased incidence of infections caused by the opportunistic pathogen Cryptococcus neoformans, which mainly affects immunocompromised patients but can also infect immunocompetent individuals, has needed additional studies on this micro-organism`s pathogenicity and factors related to virulence, such as enzyme production, for a better understanding of the aetiology of cryptococcosis. The aim of this study was to verify the applicability of non-denaturing PAGE for analysis of laccases by quantification of the amount of melanin pigment produced by clinical and environmental strains of C. neoformans. After incubation of the gel with the substrate L-dopa, strains produced melanin spots of a bright brown to black colour. Quantification of these spots was performed by densitometry analysis and the amount of melanin produced was calculated and compared among the strains. All strains showed laccase activity. Serotype B strains showed a higher melanin intensity than serotype A strains. Over half of the clinical strains (56.2%) showed the lowest melanin intensities, suggesting that melanin production may not be the main virulence factor against host defence. The clinical strain ICB 88 revealed two melanin spots on the gel, indicating the presence of two laccase isoforms. The environmental strains showed the highest values of melanin intensity, which may be related to previous exposure to environmental stress conditions.

Nitrogen uptake and metabolism are essential to microbial growth. Gat1 belongs to a conserved family of zinc finger containing transcriptional regulators known as GATA-factors. These factors activate the transcription of Nitrogen Catabolite Repression (NCR) sensitive genes when preferred nitrogen sources are absent or limiting. Cryptococcus neoformans GAT1 is an ortholog to the Aspergillus nidulans AreA and Candida albicans GAD genes. In an attempt to define the function of this transcriptional regulator in C. neoformans, we generated null mutants (gat1 Delta) of this gene. The gat 1 mutant exhibited impaired growth on all amino acids tested as sole nitrogen sources, with the exception of arginine and proline. Furthermore, the gat1 mutant did not display resistance to rapamycin, an immunosuppressant drug that transiently mimics a low-quality nitrogen source. Gal is not required for C. neoformans survival during macrophage infection or for virulence in a mouse model of cryptococcosis. Microarray analysis allowed the identification of target genes that are regulated by Gat1 in the presence of proline, a poor and non-repressing nitrogen source. Genes involved in ergosterol biosynthesis, iron uptake, cell wall organization and capsule biosynthesis, in addition to NCR-sensitive genes, are Gat1-regulated in C. neoformans. This study was addressed to investigate the composition and antifungal activity of essential oils from leaves of Piperaceae species (Piper aduncum, Piper amalago, Piper cernuum, Piper diospyrifolium, Piper crassinervium, Piper gaudichaudianum, Piper solmsianum, Piper regnellii, Piper tuberculatum, Piper umbelata and Peperomia obtusifolia) against Candida albicans, C. parapsilosis, C. krusei and Cryptococcus neoformans. The essential oils from P. aduncum, P. gaudichaudianum and P. solmsianum showed the highest antifungal activity against Cryptococcus neoformans with the MIC of 62.5, 62.5 and 3.9 mg.mL-1, respectively. The oil from P. gaudichaudianum showed activity against C. krusei with MIC of 31.25 mg.mL-1.

Criptococose é uma levedurose que acomete o homem e vários animais, podendo ocorrer em indivíduos imunocompetentes, mas freqüentemente está associada a um estado de comprometimento imunológico. Atualmente a etiologia desta micose é atribuída a três espécies: Cryptococcus neoformans, C. grubii (isolados no solo rico em fezes de pombos), e C. gattii (isolado nos eucaliptos). A via mais freqüente de contaminação é a inalatória, com posterior colonização do trato respiratório superior, podendo atingir aos alvéolos e desenvolver a sintomatologia respiratória, ou ocorrer disseminação hematógena com possível comprometimento do Sistema Nervoso Central. É notável o aumento na procura de animais de companhia nos grandes centros urbanos e, também, se verifica uma significativa população de pombos nas cidades, alojados em igrejas, prédios, parques e praças. A exposição dos cães a locais possivelmente contaminados e a saúde destes animais constituise uma preocupação e, visto que existem relatos na literatura, ainda que poucos, sobre a criptococose em cães, realizamos uma pesquisa sobre a presença da levedura nesta espécie. Este trabalho teve como objetivo verificar a ocorrência do Cryptococcus em cães com sintomatologia respiratória e/ou neurológica, atendidos no Hospital de Clínicas Veterinárias da Universidade Federal do Rio Grande do Sul (UFRGS), localizado na cidade de Porto Alegre, Brasil. A amostragem foi composta por 112 cães. Realizou-se o exame direto, com nigrosina, do líquido cefalorraquidiano (n=17); o cultivo da secreção nasal (112), do sangue total (112) e do líquido cefalorraquidiano (n=17) em ágar níger (Guizotia abissynica), com incubação a 370C durante dez dias; e o teste de aglutinação em látex, utilizando-se o teste Crypto-LA (Wampole), com o soro (n=112) e o líquido cefalorraquidiano (n=17) Para o teste de aglutinação em látex foi realizado, também, tratamento com Pronase em 32 (28,57%) amostras de soro e 4 (23,52%) amostras de líquido cefalorraquidiano. Os resultados do exame direto, do cultivo e do teste de aglutinação em látex foram negativos para o Cryptococcus em todas as amostras testadas. Não se excluiu a possibilidade de ter ocorrido resultados falsos negativos, por não ter sido realizado um lavado nasal para coleta de material para cultivo e, também, porque o Cryptococcus poderia estar presente em pequenas quantidades no organismo e/ou ser pouco capsulado e, portanto, não sendo detectado através do teste de aglutinação em látex. Apesar dos resultados desta amostragem, é plausível a suposição que deva existir a ocorrência da infecção pelo Cryptococcus, embora haja a falta de uma suspeita clínica desta enfermidade, posto que, na região de abrangência do estudo existe uma população canina constantemente exposta ao risco por coabitarem com uma grande população de pombos.

A yeast strain (CBS 8902) was isolated from the nest of a leaf-cutting ant and was shown to be related to Cryptococcus humicola. Sequencing of the D1/D2 region of the 26S ribosomal DNA and physiological characterization revealed a separate taxonomic position. A novel species named Cryptococcus haglerorum is proposed to accommodate strain CBS 8902 that assimilates n-hexadecane and several benzene compounds. Physiological characteristics distinguishing the novel species from some other members of the C. humicola complex are presented. The phylogenetic relationship of these strains to species of the genus Trichosporon Behrend is discussed.

We studied the effects of Amphotericin B (AmB) on Cryptococcus neoformans using different viability methods (CFUs enumeration, XTT assay and propidium iodide permeability). After 1 h of incubation, there were no viable colonies when the cells were exposed to AmB concentrations >= 1 mg/L. In the same conditions, the cells did not become permeable to propidium iodide, a phenomenon that was not observed until 3 h of incubation. When viability was measured in parallel using XTT assay, a result consistent with the CFUs was obtained, although we also observed a paradoxical effect in which at high AmB concentrations, a higher XTT reduction was measured than at intermediate AmB concentrations. This paradoxical effect was not observed after 3 h of incubation with AmB, and lack of XTT reduction was observed at AmB concentrations higher than 1 mg/L. When stained with dihydrofluorescein, AmB induced a strong intracellular oxidative burst. Consistent with oxidative damage, AmB induced protein carbonylation. Our results indicate that in C. neoformans, Amphotericin B causes intracellular damage mediated through the production of free radicals before damage on the cell membrane, measured by propidium iodide uptake. The growth of the yeast Cryptococcus laurentii in complete broth for yeast and in liquid and semi-solid sugar cane vinasse media supplemented with several nitrogen and phosphorus sources was analysed in order to evaluate its potential utilization as biomass producer from vinasse. The trials were performed in a 1-liter fermentor, at 30-degrees-C, under magnetic agitation for 48 hours, and initial pH 5.0 for the liquid media. The parameters analysed were biomass, protein, and final pH. For the semi-solid media, agitated flasks under rotational agitation for 24 hours, at 30-degrees-C, and initial pH 5.0 were used. The results obtained showed that the yeast did not grow well in liquid sugar cane vinasse media, both supplemented or not, in comparison to the complete broth for yeast. The protein content was also lower in liquid sugar cane vinasse media. The medium pH did not alter markedly during the cultivation. The best results were obtained in semi-solid sugar cane vinasse media with supplementation, probably due to the known polysaccharide production by this species, which allows a better survival to solid substrates.
