Determination of snake-necked turtle Hydromedusa tectifera (Cope, 1870) (Testudines: Chelidae) plasma protein concentrations by refractometry and the biuret method.

The purpose of this study was to evaluate the accuracy of hand-held refractometer in determining plasma protein concentrations in South American snake-necked turtle (Hydromedusa tectifera) as compared with the standard biuret method. The results indicated that plasma protein values may be accurately determined in snake- necked turtle with a hand-held refractometer.

Endoparasites new to the Neotropical freshwater turtle, Mesoclemmys vanderhaegei (Bour 1973) (Testudines, Chelidae), from central Brazil.

Information on endoparasites infecting Neotropical turtles is scarce. The present paper reports the occurrence of endoparasites in three adult individuals of Vanderhaege's Toad-Headed Turtle (Mesoclemmys vanderhaegei). The records include an undescribed nematode species in the genus Spiroxys, not previously reported for the family Chelidae, and the first South American record of a monogenetic trematode of the genus Neopolystoma.

Radioautographic study of the seasonal distribution of leukocytes in turtles Phrynops hilarii (Chelonia Chelidae)

The aim of this study was to present a morphological description of the leukocytes of Phrynops hilarii turtles according to the seasonal distribution of these cells and to show their replacement in the blood circulation using a radioautographic method. Five animals of both sexes weighing 600-1200 g were used. The animal's blood was aspirated, smeared on glass slides, and stained with the Romanowsky stain, and 500 cells of each animal were counted during each season. The results obtained were analyzed statistically by analysis of variance followed by the Bonferrom test (NCSS), with the level of significance set at p < 0.05. The radioautographic analysis of turtle blood exposed to 1000 mu Ci of H-3-thymidine and developed after 30 days showed a large number of silver grains incorporated into the cells, except for basophils, with cell renewal occurring every seven days. Quantitative data demonstrated a seasonal influence on the distribution of some leukocyte types, with the following p values: heterophils (p = 0.0007), basophils (p = 0.0002), monocytes (p = 0.0016), eosinophils (p = 0.0073). However, using this statistical method, it was not possible to detect a significant difference related to seasonal influence on lymphocytes (p = 0. 16295) or thrombocytes (p = 0. 1046). Using this experimental animal model, a seasonal influence on the distribution of some leukocyte types was observed, and the radioautographic method revealed a cell renewal system occurring every seven days, except for basophils. Atividade peroxidásica em basófilos de Phrynops geoffroanus (Testudines Chelidae)

As peroxidases, presentes nos peroxissomos e lisossomos, pertencem às oxidases e atuam como catalítico para o peróxido de hidrogênio (H2O2), posteriormente decomposto pela oxidação de cossubstratos, evitando danos celulares.(¹) Foi aplicada a técnica da peroxidase(2) em esfregaços sanguíneos de Phrynops geoffroanus, comparando com sangue humano, para avaliação da atividade e controle da reação. O esfregaço sanguíneo humano apresentou marcações em neutrófilos, fagócitos com muitos lisossomos e peroxissomos (Figura 1). Nos esfregaços sanguíneos de Phrynops geoffroanus, as marcações apresentaram-se nos basófilos (Figura 2), que representam de 10% a 25% dos leucócitos de quelônios e possuem grande número de granulações citoplasmáticas,(3) sugerindo a presença de grande quantidade de enzimas e organelas como lisossomos e peroxissomos, possivelmente associadas a sua participação em reações imunes. A atividade peroxidásica representa resposta do organismo a ações ambientais danosas, servindo como marcador biológico.

Anatomy of the male reproductive system of Phrynops geoffroanus (Testudines: Chelidae).

The reproductive system of male Phrynops geoffroanus adults is macroscopically described and the variation in testicular biometry is evaluated. A pair of oval testes is connected by the efferent ductules to the epididymis, which continue as deferent ducts, which emerge in the penis. The volume of the gonads showed the highest averages during spring and summer months. GSI varied significantly throughout the year, with the highest averages observed in the months that correspond to the end of spring and the beginning of summer, when the reproduction of the species takes place, and the lowest averages were seen in winter, suggesting a cyclical testicular activity.

Characterization of blood mononuclear phagocytes in Phrynops hilarii (Chelonia Chelidae)

The localization of peroxidase activity in different cell regions is used as a criterion for the classification of the stage of maturation of mammalian mononuclear phagocytes with a positive peroxidase reaction indicating the presence of monoblasts, promonocytes, monocytes and macrophages. In this study it was evaluated the peroxidase activity of blood mononuclear phagocytes of this turtle detected at different stages of differentiation. The present observations suggest that, in turtles, the differentiation of mononuclear phagocytes occur in the blood circulation, in contrast to animals, where only are monocytes in circulating blood and macrophage differentiation occurs in other body compartments. © 2007 Sociedad Chilena de Anatomía.

Courtship behavior of Mesoclemmys vanderhaegei (Bour, 1973) (Testudines: Chelidae) under natural conditions in the Brazilian Cerrado

Mesoclemmys vanderhaegei is a Neotropical chelid lacking basic natural history studies. Based on a film made in December 2004, we describe the courtship behavior in this species. Field observations were conducted at Chapada dos Guimarães, Mato Grosso State, southwestern Brazil, in an oligotrophic stream amidst the savannah like Cerrado formation. We provide a schematic drawing of the behavioral sequences during courtship, as well as a flow diagram describing sequential responses of the female to the male behavioral patterns. Mesoclemmys vanderhaegei courtship behavior is similar to that described for other freshwater chelonians.

New records and distribution extensions of three species of Mesoclemmys Gray, 1863 (Testudines: Chelidae) in Mato Grosso state, Brazil, with observations on terrestrial movements

Distribution patterns of most of the 20 Neotropical freshwater turtles belonging to the family Chelidae are usually based on few locality records. We here report on vouchered records of three species of the chelid genus Mesoclemmys in the Brazilian state of Mato Grosso. Presence of M. vanderhaegei is confirmed for the Amazon River Basin, and M. gibba and M. raniceps are recorded in Mato Grosso for the first time. © 2012 Check List and Authors.
