The Farm Pond, University of Ife, was impounded by the construction of a dam, below the confluence of two small streams, between March and July, 1967. The pond is located at an altitude of 122.4 m. 0.D., its outline is an indented trapezium occupying 445 dkrn' at maximum fill. Natural food is fed into the pond by its two inlets, and the pond is often coloured a darkish brown due to the abundance of plankton. In 1969, on the recommendation by the Fishery Division, Ministry of Agriculture and Natural Resources, Ibadan, about four standard head-pans of poultry droppings per 40 dkm2 per month were spread uniformly over the pond during 1969 and 1970. Also four pounds of triple superphosphate per acre per month were spread for a period of six months. The pond was drained a number of times in 1970. However, the lowest average level of 1.37 m was recorded in April, 1970, before drainage began; and the maximum depth of 5.7 m was recorded in September and October, 1970.

One hundred forty-five cliff swallows, Petrochelidon pyrrhonota, were examined for cestodes in two separate surveys in Weld County, Colorado. Seven species representing four genera were found. Angularella audubonensis sp. n. is differentiated by the small size of the rostellar hooks (8.4-13.5 μm) and the medial relationship of the cirrus pouch to the osmoregulatory canals. Other species collected were Angularella beema, Anonchotaenia globata, Mayhewia ababili, Vitta magniuncinata, Vitta parvirostris and Vitta riparia. Seven new hosts and geographic distribution records were established for Colorado, and six new records were determined for North America.

The study of a collection of cestodes assigned to the genus Diplogonoporus Lönnberg, 1892 disclosed but two species, D. balaenopterae Lönnberg, 1892, and D. tetrapterus (von Siebold, 1848) (provis.). These cestodes occur characteristically in marine mammals but occasionally are found in terrestrial hosts; D. balaenopterae is recorded for the first time from the domestic dog, and it is concluded that D. grandis (Blanchard, 1894), from man, is conspecific with D. balaenopterae. The latter species is recorded for the first time from the humpback whale, Megaptera novaeangliae (Borowski). The relatively small D. tetrapterus, a common parasite of the Steller sea lion, Eumetopias jubata (Schreber), is reported for the first time from the sea otter, Enhydra lutris Linnaeus, and from the domestic mink, Mustela vison Schreber. Descriptions of representative specimens are presented, and the taxonomic status of other species assigned to Diplogonoporus is discussed. Although the diplogonadic organization of these cestodes is somewhat variable, it is nevertheless constant and serves to characterize the genus Diplogonoporus. The process of asexual reproduction by means of transverse subdivision of primary segments is described. This ability and the diplogonadic structure of these cestodes are considered to be adaptations that increase the production of eggs and thereby the probability of reproductive success in the marine habitat.

In spite of the importance of the various species of shrews to the biotic community, several aspects of their ecology remain very incompletely known. The helminths parasitic in the more conspicuous small mammals in North America have been studied more or less completely, but those occurring in shrews have been largely disregarded. This situation is particularly inappropriate when one considers that shrews often constitute the most abundant mammalian group in a given area. We have attempted to secure an adequate amount of shrew helminth material for study, and it is the purpose of this paper to present observations resulting from this work.

Among the cestodes, Echinococcus granulosus, Echinococcus multilocularis and Taenia solium represent the most dangerous parasites. Their larval stages cause the diseases cystic echinococcosis (CE), alveolar echinococcosis (AE) and cysticercosis, respectively, which exhibit considerable medical and veterinary health concerns with a profound economic impact. Others caused by other cestodes, such as species of the genera Mesocestoides and Hymenolepis, are relatively rare in humans. In this review, we will focus on E. granulosus and E. multilocularis metacestode laboratory models and will review the use of these models in the search for novel drugs that could be employed for chemotherapeutic treatment of echinococcosis. Clearly, improved therapeutic drugs are needed for the treatment of AE and CE, and this can only be achieved through the development of medium-to-high throughput screening approaches. The most recent achievements in the in vitro culture and genetic manipulation of E. multilocularis cells and metacestodes, and the accessability of the E. multilocularis genome and EST sequence information, have rendered the E. multilocularis model uniquely suited for studies on drug-efficacy and drug target identification. This could lead to the development of novel compounds for the use in chemotherapy against echinococcosis, and possibly against diseases caused by other cestodes, and potentially also trematodes.

A library of 426 FDA-approved drugs was screened for in vitro activity against E. multilocularis metacestodes employing the phosphoglucose isomerase (PGI) assay. Initial screening at 20 µM revealed that 7 drugs induced considerable metacestode damage, and further dose-response studies revealed that bortezomib (BTZ), a proteasome inhibitor developed for the chemotherapy of myeloma, displayed high anti-metacestodal activity with an EC50 of 0.6 µM. BTZ treatment of E. multilocularis metacestodes led to an accumulation of ubiquinated proteins and unequivocally parasite death. In-gel zymography assays using E. multilocularis extracts demonstrated BTZ-mediated inhibition of protease activity in a band of approximately 23 kDa, the same size at which the proteasome subunit beta 5 of E. multilocularis could be detected by Western blot. Balb/c mice experimentally infected with E. multilocularis metacestodes were used to assess BTZ treatment, starting at 6 weeks post-infection by intraperitoneal injection of BTZ. This treatment led to reduced parasite weight, but to a degree that was not statistically significant, and it induced adverse effects such as diarrhea and neurological symptoms. In conclusion, the proteasome was identified as a drug target in E. multilocularis metacestodes that can be efficiently inhibited by BTZ in vitro. However, translation of these findings into in vivo efficacy requires further adjustments of treatment regimens using BTZ, or possibly other proteasome inhibitors.

The literature associated with descriptions and definitions of the sucker-like attachment organs in trypanorhynchs, termed either bothria or bothridia, is reviewed. There are descriptions of 14 trypanorhynch species representing 10 families. In none of these trypanorhynchs was a membrane separating the attachment organ from the scolex parenchyma described, one of the definitions used to distinguish bothria from bothridia. Transmission electron microscopy of the bothria of the trypanorhynch species Nybelinia queenslandensis Beveridge & Jones, 1998 (Tentaculariidae) and Otobothrium mugilis Hiscock, 1954 (Otobothriidae) also failed to show any membranous structure separating the surface of the attachment organ from the cestode parenchyma. The sucker-like attachment organs of trypanorhynchan cestodes appear, therefore, to be bothria rather than bothridia. As a result, changes in the terminology of related features of the scolex are proposed here. Henceforth, the pars bothridialis should be referred to as the pars bothrialis and the bothridial pits should be referred to bothrial pits.

DNA approaches are now being used routinely for accurate identification of Echinococcus and Taenia species, subspecies and strains, and in molecular epidemiological surveys of echinococcosis/taeniasis in different geographical settings and host assemblages. The publication of the complete sequences of the mitochondrial (int) genomes of E. granulosus, E. multilocularis, T solium and Asian Taenia, and the availability of mtDNA sequences for a number of other taeniid genotypes, has provided additional genetic information that can be used for more in depth phylogenetic and taxonomic studies of these parasites. This very rich sequence information has provided a solid molecular basis, along with a range of different biological, epidemiological, biochemical and other molecular-genetic criteria, for revising the taxonomy of the genus Echinococcus and for estimating the evolutionary time of divergence of the various taxa. Furthermore, the accumulating genetic data has allowed the development of PCR-based tests for unambiguous identification of Echinococcus eggs in the faeces of definitive hosts and in the environment. Molecular phylogenies derived from mtDNA sequence comparisons of geographically distributed samples of T solium provide molecular evidence for two genotypes, one being restricted to Asia, with the other occurring in Africa and America. Whether the two genetic forms of T solium differ in important phenotypic characteristics remains to be determined. As well, minor DNA sequence differences have been reported between isolates of T saginata and Asian Taenia. There has been considerable discussion over a number of years regarding the taxonomic position of Asian Taenia and whether it should be regarded as a genotype, strain, subspecies or sister species of T saginata. The available molecular genetic data do not support independent species status for Asian Taenia and T saginata. What is in agreement is that both taxa are closely related to each other but distantly related to T solium. This is important in public health terms as it predicts that cysticercosis in humans attributable to Asian Taenia does not occur, because cysticercosis is unknown in T saginata. Temporal and spatial patterns in parasite assemblages were examined to evaluate the degree of movement and connectivity of post-recruitment life-history stages of a large, non-diadromous tropical estuarine teleost, king threadfin Polydactylus macrochir, collected from 18 locations across northern Australia. Ten parasites types (juvenile stages of two nematodes and seven cestodes, and adults of an acanthocephalan) were deemed to be suitable for use as biological tags, in that they were considered to have a long residence time in the fish, were relatively easy to find and were morphologically very different to each other which aided discrimination. Univariate and discriminant function analysis of these parasites revealed little difference in temporal replicates collected from five locations, suggesting that the parasite communities were stable over the timeframes explored. Univariate, discriminant function, and BrayCurtis similarity analyses indicated significant spatial heterogeneity, with BrayCurtis classification accuracies ranging from 55 to 100% for locations in north-western and northern Australia, 24 to 88% in the Gulf of Carpentaria, and 39 to 88% on the east coast of Queensland. Few differences were observed among locations separated by <200 km. The observed patterns of parasite infection are in agreement with concurrent studies of movement and connectivity of P. macrochir in that they indicate a complex population structure across northern Australia. These results should be considered when reviewing the management arrangements for this species.
