Glycosylation represents an important modification that regulates biological processes in tissues relevant for disease pathogenesis in systemic sclerosis (SSc), including the endothelium and extracellular matrix. Whether patients with SSc develop antibodies to carbohydrates is not known.

A convenient methodology has been developed for the selective removal of the anomeric acyl group of carbohydrate derivatives using HCI04-Si02 under acidic reaction conditions. Anomeric benzoyl groups can also be removed selectively following similar reaction conditions. The yields were excellent in all cases.

A series of di- and trisaccharides containing galactofuranose were synthesized using HClO4-SiO2 catalyzed glycosylation and ring transformation for the conversion of galactopyranose to galactofuranose. Yields in glycosylations and ring transformation reactions were excellent.

The presence of soluble carbohydrates in the cambial zone, either from sugars recently produced during photosynthesis or from starch remobilized from storage organs, is necessary for radial tree growth. However, considerable uncertainties on carbohydrate dynamics and the consequences on tree productivity exist. This study aims to better understand the variation in different carbon pools at intra-annual resolution by quantifying how cambial zone sugar and starch concentrations fluctuate over the season and in relation to cambial phenology. A comparison between two physiologically different species growing at the same site, i.e., the evergreen Picea abies Karst. and the deciduous Larix decidua Mill., and between L. decidua from two contrasting elevations, is presented to identify mechanisms of growth limitation. Results indicate that the annual cycle of sugar concentration within the cambial zone is coupled to the process of wood formation. The highest sugar concentration is observed when the number of cells in secondary wall formation and lignification stages is at a maximum, subsequent to most radial growth. Starch disappears in winter, while other freeze-resistant non-structural carbohydrates (NSCs) increase. Slight differences in NSC concentration between species are consistent with the differing climate sensitivity of the evergreen and deciduous species investigated. The general absence of differences between elevations suggests that the cambial activity of trees growing at the treeline was not limited by the availability of carbohydrates at the cambial zone but instead by environmental controls on the growing season duration.

The current studies were undertaken to examine the effect of retinoic acid (RA)-induced differentiation of the murine embryonal carcinoma cell line, F-9, on the glycosylation of specific cellular glycoproteins and on the expression of two members of the family of endogenous lactoside-binding lectins. It was found that RA-induced differentiation of these cells into cells with the properties of primitive endoderm results in the increased fucosylation of 3 glycoproteins with molecular weights of 175 (gp175), 250 (gp250), and 400 (pg400) kDa. These three fucose-containing glycoproteins can be considered as new markers of differentiation in this system. The increased fucosylation of these glycoproteins preceded the 3-fold increase in fucosyltransferase (FT) activity that was seen upon RA-induced differentiation of these cells, indicating that an increase in fucosyltransferase activity alone cannot explain the increased fucosylation of these glycoproteins.^ The effect of RA and Ch55, a chalcone carboxylic acid with retinoid-like properties, induced differentiation of a variety of murine embryonal carcinoma cell lines on the activities of both FT and sialyltransferase (ST) was examined. The effect of differentiation on the activities of both glycosyltransferases was modulated and most probably is dependent upon the differentiation pathway that is triggered by the retinoids for each of the embryonal carcinoma cell lines.^ Two glycoproteins, Lysosomal Associated Membrane Glycoproteins 1 and 2 (LAMP-1 and LAMP-2) were examined in more detail during the course of RA-induced differentiation of F-9 cells. Both the levels and glycosylation of both glycoproteins are increased following differentiation of these cells. Differentiation results in the increased binding of $\sp{125}$l-labelled L-phytohemagglutinin to bind to LAMP-1 which indicates increased GlcNAc $\beta$1,6 branching of the oligosaccharide side chains.^ We found that RA-induced differentiation of F-9 cells results in the decreased expression of the 34 kDa lectin 24 h after addition of the retinoid to the medium. Additionally, 48 h of RA-treatment results in the increased expression of the 14.5 kDa lectin. By indirect immunofluorescence we were able to colocalize the 14.5 kDa lectin and laminin which suggests that laminin may be a ligand for the lectin in the F-9 cells. We analyzed 10 core samples of Pleistocene and Pliocene sediment for residual carbohydrates. All yielded positive results for total carbohydrates and acid-extractable glucose. We also detected galactose, mannose, arabinose, xylose, and traces of ribose and fucose in the Pleistocene samples. In the Pliocene samples we found only rare mannose. Only one Pleistocene sample yielded measurable cellulose and amylose.

Selected parts of ten frozen core samples from Holes 482A, 482B, 483A, and 485A, Leg 65 of the Deep Sea Drilling Project (DSDP), were analyzed for residual carbohydrates in order to determine the provenance and history of the organic material in the sediments. The samples, which represented silty-clay, shale, and nannofossil- chalk sediments, were analyzed for water-soluble monosaccharides, acid-soluble monosaccharides, and for starch and cellulose. Most samples yielded positive results for acid-extractable (polymeric) arabinose, fucose, xylose, mannose, galactose, and glucose. Amylose was detected in seven of the samples, whereas cellulose was found in only one. Possible explanations for the relatively high levels of free sugars are suggested in the conclusions to this chapter.

Patterns of distribution and variations of group and monosaccharide compositions of carbohydrates in suspended matter of the Pacific Ocean were studied. It is shown that carbohydrate content of surface ocean waters depends on reproduction of organic matter by phytoplankton. Water-insoluble polysaccharides (average 77.9% of total) predominate in composition of carbohydrates in suspended matter. Water-soluble polysaccharides and oligosaccharides were detected in considerably smaller quantities (average 12.4 and 7.3% respectively). Free monosaccharides were not detected. The main sugars in all isolated groups of carbohydrates of suspended matter are hexoses, which account for 90.8% in oligosaccharides, 64.9% in water-soluble polysaccharides, and 69.8% in water-insoluble polysaccharides. Determination of monosaccharide composition of carbohydrates in suspension showed that apparently they basically consist of mixture of reserve and structural polysaccharides (or their residues) of phytoplankton organisms.
