The topic of my doctoral thesis is to demonstrate the usefulness of incorporating tonal and modal elements into a pitch-web square analysis of Béla Bartók's (1881-1945) opera, 'A kékszakállú herceg vára' ('Duke Bluebeard's Castle'). My specific goal is to demonstrate that different musical materials, which exist as foreground melodies or long-term key progressions, are unified by the unordered pitch set {0,1,4}, which becomes prominent in different sections of Bartók's opera. In Bluebeard's Castle, the set {0,1,4} is also found as a subset of several tetrachords: {0,1,4,7}, {0,1,4,8}, and {0,3,4,7}. My claim is that {0,1,4} serves to link music materials between themes, between sections, and also between scenes. This study develops an analytical method, drawn from various theoretical perspectives, for conceiving superposed diatonic spaces within a hybrid pitch-space comprised of diatonic and chromatic features. The integrity of diatonic melodic lines is retained, which allows for a non-reductive understanding of diatonic superposition, without appealing to pitch centers or specifying complete diatonic collections. Through combining various theoretical insights of the Hungarian scholar Ernő Lendvai, and the American theorists Elliott Antokoletz, Paul Wilson and Allen Forte, as well as the composer himself, this study gives a detailed analysis of the opera's pitch material in a way that combines, complements, and expands upon the studies of those scholars. The analyzed pitch sets are represented on Aarre Joutsenvirta's note-web square, which adds a new aspect to the field of Bartók analysis. Merocyanine dyes that exhibit antithetic cyaninelike behaviour and giant first-order hyperpolarisability (beta) values have been designed. These cyanine-type dyes open up an intriguing route towards molecular-based electrooptic materials as well as new second-harmonic generation dyes for imaging.

The principal aim of this study was to investigate the possibility of transference to Escherichia coli of β-lactam resistance genes found in bacteria isolated from ready-to-eat (RTE) Portuguese traditional food. From previous screenings, 128 β-lactam resistant isolates (from different types of cheese and of delicatessen meats), largely from the Enterobacteriaceae family were selected and 31.3% of them proved to transfer resistance determinants in transconjugation assays. Multiplex PCR in donor and transconjugant isolates did not detect bla CTX, bla SHV and bla OXY, but bla TEM was present in 85% of them, while two new TEMs (TEM-179 and TEM-180) were identified in two isolates. The sequencing of these amplicons showed identity between donor and transconjugant genes indicating in vitro plasmid DNA transfer. These results suggest that if there is an exchange of genes in natural conditions, the consumption of RTE foods, particularly with high levels of Enterobacteriaceae, can contribute to the spread of antibiotic resistance.

Ce mémoire examine le rapport du visible à l’invisible dans l’œuvre du cinéaste hongrois Béla Tarr afin de montrer comment cette œuvre fait sa plus grande place, quoique le plus souvent sous la forme d’une absence-présence, à la question du néant et de la mort. À travers une étude tripartite, tour à tour concernant le traitement cinématographique et thématique du monde, du temps et du personnage et de leurs fonctions, l’objectif de cette recherche est celui de montrer comment cette œuvre soustractive interroge le rapport de l’existant à un monde qui fait défaut. C’est la promesse, en partie telle que théorisée par Peter Sloterdijk, qui permettra ultimement d’éclairer la démarche tragique des personnages tarriens : lutte contre la mort au moyen d’un constant effort de soi et ce, jusqu’à l’épuisement radical. La notion de personnage-figurant développée dans ce mémoire se propose d’articuler ensemble les valeurs figuratives et thématiques communes aux différents personnages qui traversent l’œuvre afin d’en mieux comprendre, malgré les ruptures stylistiques et narratives, la continuité. Enfin, le dialogue engagé ici entre l’œuvre du cinéaste et certains concepts-clé de la tradition philosophique (le monde, le soi, le temps) entend montrer comment la première ouvre des pistes pertinentes pour une réflexion sur le sujet contemporain et son époque.

Antimicrobial drug resistance is a global challenge for the 21st century with the emergence of resistant bacterial strains worldwide. Transferable resistance to beta-lactam antimicrobial drugs, mediated by production of extended-spectrum beta-lactamases (ESBLs), is of particular concern. In 2004, an ESBL-carrying IncK plasmid (pCT) was isolated from cattle in the United Kingdom. The sequence was a 93,629-bp plasmid encoding a single antimicrobial drug resistance gene, bla(CTX-M-14). From this information, PCRs identifying novel features of pCT were designed and applied to isolates from several countries, showing that the plasmid has disseminated worldwide in bacteria from humans and animals. Complete DNA sequences can be used as a platform to develop rapid epidemiologic tools to identify and trace the spread of plasmids in clinically relevant pathogens, thus facilitating a better understanding of their distribution and ability to transfer between bacteria of humans and animals.

A clinical Klebsiella pneumoniae isolate carrying the extended-spectrum beta-lactamase gene variants bla(SHV-40), bla(TEM-116) and bla(GES-7) was recovered. Cefoxitin and ceftazidime activity was most affected by the presence of these genes and an additional resistance to trimethoprim-sulphamethoxazole was observed. The bla(GES-7) gene was found to be inserted into a class 1 integron. These results show the emergence of novel bla(TEM) and bla(SHV) genes in Brazil. Moreover, the presence of class 1 integrons suggests a great potential for dissemination of bla(GES) genes into diverse nosocomial pathogens. Indeed, the bla(GES-7) gene was originally discovered in Enterobacter cloacae in Greece and, to our knowledge, has not been reported elsewhere.

As beta-lactamases são produzidas por bactérias gram-positivas e gram-negativas. Cerca de 40% a 50% das mulheres desenvolveram um infecção urinária durante a sua vida adulta. O objetivo o presente trabalho foi realizar a caracterização dos genes Bla SHV, Bla TEM e Bla CTX-M produtoras de beta-lactamases de espectro estendido em E. coli e Klebsiella spp isoladas de gestante com infecção do trato urinário atendidas na Unidade Básica de Saúde de Imperatriz - MA no período de maio a agosto de 2012. Participaram do presente estudo 50 de mulheres grávidas maiores de 18 anos, que apresentaram sintomas com caracterização clínica de infecção do trato urinário (ITU), referenciadas no ambulatório na Unidade Básica de Saúde Imperatriz - MA. Foi coletada urina, para realização da urinocultura e antibiograma, posteriormente foi realizado a Reação em Cadeia da Polimerase (PCR) detectar a presença dos genes Bla SHV, Bla TEM e Bla CTX-M produtores das enzimas Beta-lactamases. A média de idade destes pacientes foi de 21 anos, sendo 42% estando na faixa etária de 18 a 25 anos, 70% destas tem residência fixa em Imperatriz e os outros 30% são oriundos de outros municípios. Entre estas, 24% das voluntárias já apresentaram ITU durante a gravidez e fora do estado gravídico e 80% delas afirmaram fazer o uso de antibióticos sem prescrição médica. Das 12 amostras com crescimento microbiológico positivo, 11 foram positivas para Escherichia coli com resultados do antibiograma que apresentaram resistência para o antibiótico cefalotina, em 91,7%. Sendo isolado, uma cepa de Klebsiella ssp, e esta apresentou resistência a todos os antibióticos testados. Pela análise dos resultados da PCR das amostras isoladas, os três genes Bla SHV, Bla TEM e Bla CTX-M, não foram observadas nas amostras P9, P11 e P12, porém nas demais amostras houve a ocorrência de pelo menos um dos genes. Este estudo confirmou a presença dos três tipos de genes (Bla SHV, Bla TEM e Bla CTX-M) entre as amostras estudada.

The rise of extended-spectrum beta-lactamase (ESBL)-producing Escherichia coli in food-producing animals is a growing concern for public health. We investigated ESBL producers isolated from broiler chickens in Brazil and characterized 19 CTX-M-2-producing E.coli. The ISCR1 was detected upstream of the chromosome-located gene bla(CTX-M-2), associated with sul-1 type integron structure. CTX-M-2-producing E.coli exhibited different PFGE-types and phylogenetic groups, showing a non-clonal dissemination. The sequence types found (ST93, ST155 and ST2309) have been associated with humans and animals worldwide. Herein, we report the chromosomal location of bla(CTX-M-2) on E.coli, highlighting the risks of multidrug-resistant bacteria in food-producing animals.

Broad-host-range plasmid pRIO-5, harboring the extended-spectrum beta-lactamase bla(BES-1) gene in Serratia marcescens, was fully sequenced. Analysis of the 12,957-bp sequence of this IncP6-type plasmid revealed that the bla(BES-1) gene was associated with two copies of the insertion sequence IS26. The promoter responsible for the bla(BES-1) expression was hybrid, made of a - 35 box located inside the inverted repeat of IS26 and a - 10 box inside a remnant of an insertion sequence.
