Respiratory syncytial virus (RSV) is an important cause of severe upper and lower respiratory disease in infants and in the elderly. There are 2 main RSV subtypes A and B. A recombinant vaccine was designed based on the central domain of the RSV-A attachment G protein which we had previously named G2Na (aa130–230). Here we evaluated immunogenicity, persistence of antibody (Ab) response and protective efficacy induced in rodents by: (i) G2Na fused to DT (Diphtheria toxin) fragments in cotton rats. DT fusion did not potentiate neutralizing Ab responses against RSV-A or cross-reactivity to RSV-B. (ii) G2Nb (aa130–230 of the RSV-B G protein) either fused to, or admixed with G2Na. G2Nb did not induce RSV-B-reactive Ab responses. (iii) G2Na at low doses. Two injections of 3 µg G2Na in Alum were sufficient to induce protective immune responses in mouse lungs, preventing RSV-A and greatly reducing RSV-B infections. In cotton rats, G2Na-induced RSV-reactive Ab and protective immunity against RSV-A challenge that persisted for at least 24 weeks. (iv) injecting RSV primed mice with a single dose of G2Na/Alum or G2Na/PLGA [poly(D,L-lactide-co-glycolide]. Despite the presence of pre-existing RSV-specific Abs, these formulations effectively boosted anti-RSV Ab titres and increased Ab titres persisted for at least 21 weeks. Affinity maturation of these Abs increased from day 28 to day 148. These data indicate that G2Na has potential as a component of an RSV vaccine formulation.

A live, cold-passaged (cp) candidate vaccine virus, designated respiratory syncytial virus (RSV) B1 cp-52/2B5 (cp-52), replicated efficiently in Vero cells, but was found to be overattenuated for RSV-seronegative infants and children. Sequence analysis of reverse-transcription–PCR-amplified fragments of this mutant revealed a large deletion spanning most of the coding sequences for the small hydrophobic (SH) and attachment (G) proteins. Northern blot analysis of cp-52 detected multiple unique read-through mRNAs containing SH and G sequences, consistent with a deletion mutation spanning the SH:G gene junction. Immunological studies confirmed that an intact G glycoprotein was not produced by the cp-52 virus. Nonetheless, cp-52 was infectious and replicated to high titer in tissue culture despite the absence of the viral surface SH and G glycoproteins. Thus, our characterization of this negative-strand RNA virus identified a novel replication-competent deletion mutant lacking two of its three surface glycoproteins. The requirement of SH and G for efficient replication in vivo suggests that selective deletion of one or both of these RSV genes may provide an alternative or additive strategy for developing an optimally attenuated vaccine candidate.

HRSV is one of the most important pathogens causing acute respiratory tract diseases as bronchiolitis and pneumonia among infants. HRSV was isolated from two distinct communities, a public day care center and a public hospital in Sao Jose do Rio Preto - SP, Brazil. We obtained partial sequences from G gene that were used on phylogenetic and selection pressure analysis. HRSV accounted for 29% of respiratory infections in hospitalized children and 7.7% in day care center children. On phylogenetic analysis of 60 HRSV strains, 48 (80%) clustered within or adjacent to the GA1 genotype; GA5, NA1, NA2, BA-IV and SAB1 were also observed. SJRP GA1 strains presented variations among deduced amino acids composition and lost the potential O-glycosilation site at amino acid position 295, nevertheless this resulted in an insertion of two potential O-glycosilation sites at positions 296 and 297. Furthermore, a potential O-glycosilation site insertion, at position 293, was only observed for hospital strains. Using SLAC and MEME methods, only amino acid 274 was identified to be under positive selection. This is the first report on HRSV circulation and genotypes classification derived from a day care center community in Brazil.

Background: Human respiratory syncytial virus (HRSV) is one of the major etiologic agents of respiratory tract infections among children worldwide. Methodology/Principal Findings: Here through a comprehensive analysis of the two major HRSV groups A and B (n = 1983) which comprise of several genotypes, we present a complex pattern of population dynamics of HRSV over a time period of 50 years (1956-2006). Circulation pattern of HRSV revealed a series of expansions and fluctuations of co-circulating lineages with a predominance of HRSVA. Positively selected amino acid substitutions of the G glycoprotein occurred upon population growth of GB3 with a 60-nucleotide insertion (GB3 Insert), while other genotypes acquired substitutions upon both population growth and decrease, thus possibly reflecting a role for immune selected epitopes in linkage to the traced substitution sites that may have important relevance for vaccine design. Analysis evidenced the co-circulation and predominance of distinct HRSV genotypes in Brazil and suggested a year-round presence of the virus. In Brazil, GA2 and GA5 were the main culprits of HRSV outbreaks until recently, when the GB3 Insert became highly prevalent. Using Bayesian methods, we determined the dispersal patterns of genotypes through several inferred migratory routes. Conclusions/Significance: Genotypes spread across continents and between neighboring areas. Crucially, genotypes also remained at any given region for extended periods, independent of seasonal outbreaks possibly maintained by re-infecting the general population.

In this work it is assessed the performance of Portland cement-based mortar to the grouting of type II ceramic plates with the addition of unusable tire rubber powder. It is presented a bibliographical review about the subject in which is done the theoretical and methodological foundation of the whole investigative process. The analyzed universe comprises a sample of mortar to the grouting of conventional ceramic plates type II (reference sample) and five more samples to the grouting of ceramic plates type II, which were made up of the addition of unusable tire rubber powder in the respective proportion (in mass) of 4%, 8%, 12%, 16% and 20%. These mortar samples were subject to the trials of determination of the consistency index (Brazilian Standard NBR 13276:2005), water retention ( Brazilian Standard NBR 14992 Attachment B:2003), permeability in 240 minutes (Brazilian Standard NBR 14992 Attachment G:2003), absorption of water by immersion (Brazilian Standard NBR 9781:2013), resistance to compression (Brazilian Standard NBR14992 Attachment D: 2003), resistance to traction in the flexion (Brazilian Standard NBR 13279:2005), resistance of traction adherence (Brazilian Standard NBR 14081 part 4:2012) and hardened mass density (Brazilian Standard NBR 13280: 2005). It has been found out from the analyzes of the results in the trial the following situation: the reference mortar used met the established requirements in the norms of specifications corresponding to only six from the eight parameters assessed in the research; the mortar with addition of 4,0% of tire rubber powder met the established requirements corresponding to only the resistance to compression and the resistance of adherence to traction. Thus, the other kinds of mortar with addition of 8,0 %, 12,0 %, 16,0 % and 20,0 % of tire rubber powder met the requirements of specifications corresponding to only the resistance to compression and the resistance of adherence to traction. This result concludes that the adding of tire rubber powder does not grant improvement to the mortar of type II grouting to the laying of ceramic plates.

This thesis takes liberation to be supreme knowledge of the unity underlying the world of multiplicity. This knowledge is always already attained, so all are eternally liberated, but it is unrecognized in ordinary experience. We will look at the Bhagavad-Gītā to consider why this is so. When Arjuna saw Kṛṣṇa’s imperishable Self, he saw all beings standing as one in Kṛṣṇa; thus, he was confronted by supreme knowledge. But he was overwhelmed with fear and confusion and took refuge in blindness. I argue that Arjuna was not prepared to face recognition because he was unpractised in non-attachment. Attached to his subjectivity, he trembled in the face of unity. The supreme goal is standing firm in recognition while living in the world.

The unsteady laminar incompressible boundary-layer attachment-line flow on a flat plate with attached cylinder with heat and mass transfer has been studied when the free stream velocity, mass transfer and surface wall temperature vary arbitrarily with time. The governing partial differential equations with three independent variables have been solved numerically using an implicit finite-difference scheme. The heat transfer was found to be strongly dependent on the Prandtl number, variation of wall temperature with time and dissipation parameter (for large times). However, the free stream velocity distribution and mass transfer affect both the heat transfer and skin friction.

Listeria and Salmonella are important foodborne pathogens normally associated with the shrimp production chain. This study investigated the potential of Salmonella Typhimurium, Salmonella Senftenberg, and Listeria monocytogenes (Scott A and V7) to attach to and colonize shrimp carapace. Attachment and colonization of Listeria and Salmonella were demonstrated. Shrimp abdominal carapaces showed higher levels of bacterial attachment (P < 0.05) than did head carapaces. Listeria consistently exhibited greater attachment (P < 0.05) than did Salmonella on all surfaces. Chitinase activity of all strains was tested and found not to occur at the three temperatures (10, 25. and 37 degrees C) tested. The surface physicochemical properties of bacterial cells and shrimp carapace were Studied to determine their role in attachment and colonization. Salmonella had significantly (P < 0.05) more positive (-3.9 and -6.0 mV) cell surface charge than Listeria (-18 and -22.8 mV) had. Both bacterial species were found to be hydrophilic (<35%) when measured by the bacterial adherence to hydrocarbon method and by contact angle (theta) measurements (Listeria, 21.3 and 24.8 degrees, and Salmonella, 14.5 and 18.9 degrees). The percentage of cells retained by Pheryl-Sepharose was lower for Salmonella (12.8 to 14.8%) than it was for Listeria (26.5 to 31.4%). The shrimp carapace was found to be hydrophobic (theta = 74.5 degrees), and a significant (P < 0.05) difference in surface roughness between carapace types was noted. There was a linear correlation between bacterial cell Surface charge (r(2) = 0.95) and hydrophobicity (r(2) = 0.85) and initial attachment (P < 0.05) of Listeria and Salmonella to carapaces. However, the same properties Could not be related to subsequent colonization.

O psiquiatra suíço Carl Gustav Jung (1875-1961) é um dos principais nomes da psicologia e da psicoterapia do século XX. Algumas de suas maiores contribuições teórico-metodológicas são as idéias de realidade psíquica, complexo, arquétipo (inconsciente coletivo), processo de individuação, método dialético, método construtivo e imaginação ativa. A psicologia analítica de Jung, ao longo de sua formação, foi influenciada por diversas disciplinas, dentre elas a etnologia (ciências sociais). Este trabalho buscou dar continuidade a este processo de construção epistêmica, mediante exame das concepções de Jung por intermédio da teoria do ator-rede (TAR), uma importante corrente da sociologia contemporânea. Pretendeu-se também saber se a psicologia analítica se mantém atual ou se já é uma teoria e prática clínica anacrônicas. O principal autor relacionado à TAR a quem se recorreu neste trabalho foi o sociólogo francês Bruno Latour. De sua perspectiva, o acordo moderno, disjuntor de Natureza e Cultura, é insuficiente para explicar a complicação inerente às entidades que compõem a realidade. Para escapar das armadilhas conceptuais da modernidade, Latour opera com constructos tais como coletivo (social), ator-rede, proposição, vínculo e plasma. Além do pensamento de Latour, este trabalho valeu-se das idéias sociológicas de Gabriel Tarde e da influenciologia etnopsicanalítica de Tobie Nathan, aproveitando-se da afinidade teórica que compartilham com Latour. Nathan, por desenvolver uma prática em psicoterapia, permitiu propor à psicologia clínica de Jung determinadas questões que o enfoque mais estritamente sociológico de Latour não possibilitava. Uma vez expostas as concepções de Latour, Tarde e Nathan, apresentaram-se os elementos da psicologia analítica com os quais se esperava que elas fossem compatíveis. Concluiu-se que, apesar das diferenças, muitas aproximações são plausíveis entre psicologia analítica e TAR. Constatou-se que a concepção de Jung de um psiquismo multifacetado, em devir, cujos componentes se relacionam de diferentes maneiras, é comparável à noção de ator-rede trabalhada por Latour e à monadologia de Tarde. Verificou-se também que a abordagem pragmática e construtiva identificada na psicoterapia junguiana é em muitos aspectos análoga à prática da etnopsicanálise. Assim, foi possível afirmar que a TAR e a psicologia analítica podem formar aliança.

Toward the development of an in vitro cultivation of marine sponge cells for sustainable production of bioactive metabolites, the attachment characteristics of marine sponge cells of Hymeniacidon perleve on three types of microcarriers, Hillex, Cytodex 3, and glass beads, were studied. Mixed cell population and enriched cell fractions of specific cell types by Ficoll gradient centrifugation (6%/8%/15%/20%) were also assessed. Cell attachment ratio (defined as the ratio of cells attached on microcarrier to the total number of cells in the culture) on glass beads is much higher than that on Cytodex 3 and Hillex for both mixed cell population and cell fraction at Ficoll 15-20% interface. The highest attachment ratio of 41% was obtained for the cell fraction at Ficoll 15-20% interface on glass beads, which was significantly higher than that of a mixed cell population (18%). The attachment kinetics on glass beads indicated that the attachment was completed within 1 h. Cell attachment ratio decreases with increase in cell-to-microcarrier ratio (3-30 cells/bead) and pH (7.6-9.0). The addition of serum and BSA (bovine serum albumin) reduced the cell attachment on glass beads.
