O sistema de cruzamento da espécie arbórea de dossel da Mata Atlântica brasileira, Cryptocarya moschata, foi estudado a partir de material proveniente do Parque Estadual Carlos Botelho, São Miguel Arcanjo, São Paulo, Brasil. As taxas de cruzamento foram determinadas através de marcadores alozímicos obtidos de plântulas germinadas de coortes de sementes coletadas de 35 árvores. O valor médio da taxa de cruzamento de equilíbrio (estimador indireto) foi t^eq = 0.51. As estimativas das taxas de cruzamento uniloco e multilocos (estimadores diretos) foram t^s = 0.725± 0,041 e t^m = 0,884 ± 0,034, respectivamente, indicando um sistema de cruzamento predominantemente alogâmico. As taxas de cruzamento de árvores individuais variaram de 27 a 100 ( x¯ = 87,8) porcento, a partir de t^m calculado com as freqüências gênicas de pólen mantidas constantes ao nível populacional. A partir do modelo de par de irmãos (modelo de cruzamento correlacionado) de Ritland, a correlação entre duas progênies irmãs oriundas de autofecundação (r^s) e a correlação entre duas progênies irmãs oriundas de paternidade por exocruzamento (r^p) foram 35,7% e 99,0%, respectivamente. Esses resultados corroboram o fato de haver variação nas taxas de autocruzamento entre as diferentes árvores, podendo também indicar que quando há endogamia, a maior parte das sementes nas árvores são provavelmente irmãs-germanas.

The correlated matings in two populations (Selvíria - SEL and Paulo de Faria - PFA) of dioecious Myracrodruon urundeuva were studied in the Southwest of Brazil, by allozyme analysis of progeny arrays using the sibling-pair model. Open-pollinated seeds were collected from 25 to 30 trees within populations. Departure from random matings were evident from the differences in pollen and ovule allele frequencies. The high and significant correlation of paternity (SEL r̂p = 0.671 ± 0.074; PFA r̂p = 0.371 ± 0.062) and a low number of effective pollinating trees (ranging from 2 to 3) were detected in the populations, suggesting high proportion of full-sibs progenies. According to these results, the estimate of coancestry within families (θ = 0.209 - SEL; θ = 0.171 - PFA.) exceeded the expectation of the half-sib progenies (θ = 0.125). Result outcomes are discussed from a conservation and breeding point of view.

In the present study, allele frequency distributions for the 15 STR loci included in the PowerPlex® 16 Systems (Promega) were obtained from a sample of 55 unrelated individuals living in Araraquara region (SP, Brazil). The frequency of each allele for each locus tested, the exact test and the forensic and paternity parameters were calculated using POWERSTATS ver. 1.2 (Promega) and GENEPOP ver. 3.2 software. All loci are in the Hardy-Weinberg equilibrium and they reached a combined power discrimination of 0.999999999999999973 and combined power exclusion of 0.99999987, showing to be a powerful tool for paternity testing and individual identification in the population analyzed. © 2005 Elsevier B.V. Six microsatellite loci were used to quantify the mating system of two small fragmented populations (Selviria - SEL and Aparecida do Tabuado - APT, Mato Grosso do Sul State) and isolated trees in pastures, of the bat-pollinated tropical tree Hymenaea stignocarpa, growing in the Center-west region of Brazil. In SEL population, seeds were collected from 11 mother-trees; in APT, from three trees and, in the case of isolated trees, from six individuals growing at least 500 m apart in pastures. To investigate if there are differences on mating system between trees in populations and isolated trees, trees from populations were pooled as a group and, likewise, the isolated trees were pooled to another group. The outcrossing rate was higher in the populations (t̂ m= 0.873) than in isolated trees (t̂ m=0.857), but the difference was not significant. Significant and high differences between multi-locus and single-locus outcrossing rate were detected in populations (t̂ m- t̂ s=0.301, P<0.05) and isolated trees (t̂ m- t̂ s=0.276, P<0.05), suggesting mating between relatives. Higher paternity correlation was observed in trees from population (r̂ p=0.636) than in isolated trees (r̂ p=0.377), indicating the occurrence of some correlated matings and that part of offspring are full-sibs. It was not observed increased in self-fertilization rate in isolated trees in pastures. In general terms, the unique observed difference in mating system between populations and isolate trees was the high rate of correlated matings in trees from populations, due probably to the small distance among coespecifics and the pollinator behavior, visiting near trees.

Genetic population data for five X-STR (DXS6854, DXS7424, DXS101, DXS6808 and DXS7132) were obtained from Bauru population (São Paulo, Brazil). No deviations from the Hardy-Weinberg equilibrium were observed, with the exception of DXS101. The combined powers of discrimination in males and females were 0.99897253 and 0.99999120, respectively. These high values show the potential of this system in human identification and paternity testing. © 2008 Elsevier B.V. All rights reserved.

Latin-American Society of Forensic Genetics (SLAGF) Interlaboratory Quality Control Exercise (2010-2011) included the analysis of three bloodstain samples in FTA Classic Card (three persons, biologically unrelated) and one theoretical exercise. There were 56 participating laboratories from 13 Latin-American countries that belong to society, were reported 70 STRs, including autosomal and sex chromosome markers with consensus in 53 STRs with a rate in reporting errors of 2.3%. Fifty-six laboratories reported results in theoretical exercise with mistakes in calculation of IP for each marker. It is necessary to hold meetings to discuss the results of this exercise to reach conclusions and recommendations on all aspects of DNA forensics analysis and paternity test, to improve results and quality in the results of each laboratory. © 2011 Elsevier B.V.
