Septic shock can occur as a result of Gram-negative or Gram-positive infection and involves a complex interaction between bacterial factors and the host immune system producing a systemic inflammatory state that may progress to multiple organ failure and death. Gram-positive bacteria are increasingly becoming more prevalent especially Staphylococcus epidermidis in association with indwelling devices. Lipopolysaccaride (LPS) is the key Gram-negative component involved in this process, but it is not clear which components of Gram-positive bacteria are responsible for progression of this often fatal disease. The aim of this thesis was to investigate the effect of bacterial components on the immune systems. Lipid S, a short chain form of lipoteichoic acid (LTA) found to be excreted from bacteria during growth in culture medium was examined along with other Gram-positive cell wall components: LTA, peptidoglycan (PG) and wall teichoic acids (WTA) and LPS from Gram-negative bacteria. Lipid S, LTA, PG and LPS but not WTA all stimulated murine macrophages and cell lines to produce significant amounts of NO, TNF-a, IL-6 and IL-1 and would induce fever and tissue damage seen in inflammatory diseases. Lipid S proved to be the most potent out of the Gram-positive samples tested. IgG antibodies in patients serum were found to bind to and cross react with lipid S and LTA. Anti-inflammatory antibiotics, platelet activating factor (PAF), PAF receptor antagonists and monoclonal antibodies (mAbs) directed to LTA, CD14 and toll-like receptors were utilised to modulate cytokine and NO production. In cell culture the anti-LTA and the anti-CD14 mAbs failed to markedly attenuate the production of NO, TNF-a, IL-6 or IL-1, the anti-TLR4 antibody did greatly inhibit the ability of LPS to stimulate cytokine production but not lipid S. The tetracyclines proved to be the most effective compounds, many were active at low concentrations and showed efficacy to inhibit both lipid S and LPS stimulated macrophages to produce NO.

1. S-adenosyl-L-methionine (SAMe) had no effect on cytochrome C reduction by superoxide generated from xanthine oxidase except at high concentrations. This was due to direct inhibition of the enzyme. 2. SAMe inhibited the neutrophil respiratory burst , measured by luminol enhanced chemiluminescence, to FMLP and zymosan A but not to PMA. 3. Adenosine and methylthioadenosine (MTA) inhibited the respiratory burst elicited by FMLP. 4. SAMe inhibited the phagocytosis of latex particles by neutrophils at high concentrations but methionine and S-adenosyl L-homocysteine had no effect. 5. Treatment with SAMe had no effect on cell infiltration or PGE2 production in 6-day air pouches. 6. Treatment with SAMe at the optimum dose of 50mg/kg inhibited the early phases of carrageenan induced rat hind paw inflammation but had a lesser effect on the secondary response. The antiinflammatory effect was sustained after inhibiton of polyamine synthesis. 7. SAMe increased liver putrescine levels in the presence and absence of inflammation Spermidine levels were increased in the presence of inflammation but spermine levels were unaffected by any of the treatments. 8. MT A and adenosine increased liver putrescine and spermidine levels 9. Treatment with SAMe had no effect on the polyamine status of blood. lO.Treatment with SAMe had no effect on the levels of glutathione in liver or blood. 11.SAMe and MTA inhibited histamine and platelet-activating factor (PAF) induced hind paw inflammation but had no effect on inflammation induced by dextran, zymosan, compound 48/80, 5-hydroxytryptamine, arachidonic acid or glucose oxidase. MTA was more effective than SAMe. 12. PAP-induced rat hind paw inflammation was inhibited by isoprenaline and verapamil. Combinations of these drugs with SAMe or MT A had no further enhancement of effect. 13. Incubation of rat PMNLs with [14c ] SAMe increased the intracellular levels of S-adenosyl-L-homocysteine in a dose dependent manner, but had no effect on the intracellular levels of SAMe, adenosine or MT A. 14. Previous research has indicated that schematic eyes incorporating aspheric surfaces but lacking gradient index are unable to model ocular spherical aberration and peripheral astigmatism simultaneously. This limits their use as wide-angle schematic eyes. This thesis challenges this assumption by investigating the flexibility of schematic eyes comprising aspheric optical surfaces and homogeneous optical media. The full variation of ocular component dimensions found in human eyes was established from the literature. Schematic eye parameter variants were limited to these dimensions. The levels of spherical aberration and peripheral astigmatism modelled by these schematic eyes were compared to the range of measured levels. These were also established from the literature. To simplify comparison of modelled and measured data, single value parameters were introduced; the spherical aberration function (SAF), and peripheral astigmatism function (PAF). Some ocular components variations produced a wide range of aberrations without exceeding the limits of human ocular components. The effect of ocular component variations on coma was also investigated, but no comparison could be made as no empirical data exists. It was demonstrated that by combined manipulation of a number of parameters in the schematic eyes it was possible to model all levels of ocular spherical aberration and peripheral astigmatism. However, the unique parameters of a human eye could not be obtained in this way, as a number of models could be used to produce the same spherical aberration and peripheral astigmatism, while giving very different coma levels. It was concluded that these schematic eyes are flexible enough to model the monochromatic aberrations tested, the absence of gradient index being compensated for by altering the asphericity of one or more surfaces.

This chapter reports on a framework that has been successfully used to analyze the e-business capabilities of an organization with a view to developing their e-capability maturity levels. This should be the first stage of any systems development project. The framework has been used widely within start-up companies and well-established companies both large and small; it has been deployed in the service and manufacturing sectors. It has been applied by practitioners and consultants to help improve e-business capability levels, and by academics for teaching and research purposes at graduate and undergraduate levels. This chapter will provide an account of the unique e-business planning and analysis framework (E-PAF) and demonstrate how it works via an abridged version of a case study (selected from hundreds that have been produced). This will include a brief account of the three techniques that are integrated to form the analysis framework: quality function deployment (QFD) (Akao, 1972), the balanced scorecard (BSC) (Kaplan & Norton, 1992), and value chain analysis (VCA) (Porter, 1985). The case study extract is based on an online community and dating agency service identified as VirtualCom which has been produced through a consulting assignment with the founding directors of that company and has not been published previously. It has been chosen because it gives a concise, comprehensive example from an industry that is relatively easy to relate to.

The city of Natal has a significant daylight availability, although it use isn’t systematically explored in schools architecture. In this context, this research aims to determine procedures for the analysis of the daylight performance in school design in Natal-RN. The method of analysis is divided in Visible Sky Factor (VSF), simulating and analyzing the results. The annual variation of the daylight behavior requires the adoption of dynamic simulation as data procedure. The classrooms were modelled in SketchUp, simulated in Daysim program and the results were assessed by means of spreadsheets in Microsoft Excel. The classrooms dimensions are 7.20mx 7.20m, with windows-to-wall-ratio (WWR) of 20%, 40% and 50%, and with different shading devices, such as standard horizontal overhang, sloped overhang, standard horizontal overhang with side view protection, standard horizontal overhang with a dropped edge, standard horizontal overhang with three horizontal louvers, double standard horizontal overhang, double standard horizontal overhang with three horizontal louvers, plus the use of shelf light in half the models with WWR of 40% and 50%. The data was organized in spreadsheets, with two intervals of UDI: between 300lux and 2000 lux and between 300lux and 3000lux. The simulation was performed with the weather file of 2009 to the city of NatalRN. The graphical outputs are illuminance curves, isolines of UDI among 300lux and 2000 lux and tables with index of occurrences of glare and to an UDI among 300lux 3000lux. The best UDI300-2000lux performance was evidenced to: Phase 1 (models with WWR of 20%), Phase 2 (models with WWR of 40% and 50% with light shelf). The best UDI300-3000lux performance was evidenced to: Phase 1 (models with WWR of 20% and 40% with light shelf) and Phase 2 (models with WWR of 40% and 50% with light shelf). The outputs prove that the daylight quality mainly depends on the shading system efficacy to avoid the glare occurrence, which determines the daylight discomfort. The bioclimatic recommendations of big openings with partial shading (with an opening with direct sunlight) resulted in illuminances level higher than the acceptable upper threshold. The improvement of the shading system percentage (from 73% to 91%) in medium-size of openings (WWR 40% and 50%) reduced or eliminate the glare occurrence without compromising the daylight zone depth (7.20m). The passive zone was determined for classrooms with satisfactory daylight performance, it was calculated the daylight zone depth rule-of-thumb with the ratio between daylight zone depth and the height of the window for different size of openings. The ratio ranged from 1.54 to 2.57 for WWR of 20%, 40% and 50% respectively. There was a reduction or elimination of glare in the passive area with light shelf, or with awning window shading.

Este trabalho visa analisar o potencial do sombreamento vegetal no edifício para promover conforto térmico, luminoso e eficiência energética, a partir de simulações computacionais nos softwares DesignBuilder e Daysim. Foram simuladas diferentes combinações de fator de céu visível (FCV), transparência da copa vegetal e percentual de abertura da fachada (PAF) para edificação residencial térrea em Nata/RN, a fim de quantificar os impactos e propor recomendações projetuais. Os modelos foram analisados por meio do método de conforto adaptativo indicado pela ASHRAE Standard 55 (ASHRAE, 2010), classificação do nível de eficiência energética do Regulamento Técnico da Qualidade para o Nível de Eficiência Energética de Edificações Residenciais (RTQ-R), resultados de Daylight autonomy (DA) e uniformidade da luz natural para as exigências lumínicas de 100-300-500 lux. Os resultados demonstram grande potencial para integração da vegetação na edificação, principalmente para os fatores de céu médio e grande, e falta de coerência do RTQ-R para classificar as edificações da Zona bioclimática 08 pelo método de simulação.
