Malaria is an endemic parasitosis and its causitive agent, Plasmodium, has a metabolism linked to iron supply. HFE is a gene with the polymorphisms C282Y and H63D, which are associated with a progressive iron accumulation in the organism leading to a disease called hereditary hemochromatosis. The aim of the present study was to determine the allelic and genotypic frequencies of the HFE gene polymorphisms in malaria patients and blood donors from the Brazilian Amazon region. We screened 400 blood donors and 400 malaria patients for the HFE C282Y and H63D polymorphisms from four states of the Brazilian Amazon region by polymerase chain reaction and restriction fragment length polymorphism analysis. We did not find any C282Y homozygous individuals, and the only five heterozygous individuals detected were from Pará State. The most frequent genotype in the North region of Brazil was the H63D heterozygote, in both study groups. Our results contribute to the concept that the Brazilian Amazon region should not be regarded as a single entity in South America. These polymorphisms did not influence the symptoms of malaria in the population studied, as neither severe signs nor high parasitemia were observed. Therefore, different hereditary hemochromatosis diagnostic and control measures must be developed and applied within its diverse locations. Investigations are currently being carried out in our laboratory in order to determine the importance of the coexistence of hereditary hemochromatosis in patients affected by parasitic diseases, such as malaria. ©FUNPEC-RP.

We looked for abnormal hemoglobins in blood samples sent for diagnosis of anemia. Identification of the hemoglobins was made using electrophoretic, chromatographic and molecular procedures. The 2020 blood samples were of patients from various regions of Brazil and from some other Latin American countries. Among the abnormal hemoglobins that we found, 3.5% are known to be rare, while 51% had an electrophoretic profile similar to that of Hb S at alkaline pH. Differentiation was possible only by combining electrophoretic and chromatographic methods. Hb Hasharon, an alpha globin chain mutant, was the most frequently found variant hemoglobin; it accounted for 14.3% of the abnormal DNA samples. The other abnormal hemoglobin phenotypes displayed distinct electrophoretic profiles; most of them migrated faster than Hb A. The frequencies of the different abnormal hemoglobin profiles that we found reflect the miscegenation of the Latin American population and indicate the importance of hemoglobin studies using various methods in combination for accurate diagnosis and appropriate counseling of carriers and their families.

There have been few studies on the mutations that cause heterozygous beta-thalassemia and how they affect the iron profile. One hundred and thirty-eight individuals were analyzed, 90 thalasemic β0 and 48 thalasemic β+, identified by classical and molecular methods. Mutations in the hemochromatosis (HFE) gene, detected using PCR-RFLP, were found in 30.4% of these beta-thalassemic patients; heterozygosity for H63D (20.3%) was the most frequent. Ferritin levels and transferrin saturation were similar in beta-thalassemics with and without mutations in the HFE gene. Ferritin concentrations were significantly higher in men and in individuals over 40 years of age. Transferrin saturation also was significantly higher in men, but only in those without HFE gene mutations. There was no significant difference in the iron profile among the β0 and β+ thalassemics, with and without HFE gene mutations. The frequency of ferritin values above 200 ng/mL in women and 300 ng/mL in men was also similar in β0 and β+ thalassemics (P > 0.72). Our conclusion is that ferritin levels are variable in the beta-thalassemia, trait regardless of the type of beta-globin mutation. Fetal hemoglobin (Hb F) is characteristic of the fetal development period. However, in some genetic conditions, such as hereditary persistence of fetal hemoglobin (HPFH) and delta-beta thalassemia (δβ-thalassemia), Hb F continues to be produced in adulthood. We evaluated the frequency of two mutations of HPFH, HPFH-1 and HPFH-2 African, and two mutations in δβ-thalassemia, Sicilian and Spanish, in a Brazilian population. Peripheral blood samples were collected from adults from hospitals and blood centers in southeast and northeast Brazil. These individuals were healthy and without complaints of anemia, but had increased Hb F. Samples were submitted to electrophoretic and chromatographic analyses to quantify Hb F values and, subsequently, to molecular analyses to verify the mutations. In the molecular analysis, 16 of the 60 samples showed a heterozygous profile for the HPFH mutations, two for HPFH-1 and 14 for HPFH-2. In the same sample set, three were heterozygous for Spanish δβ-thalassemia and none were heterozygous for Sicilian δβ- thalassemia. The Hb F values in the HPFH-2 heterozygotes differed from those previously reported for this mutation. In this group, the HPFH mutations were more frequent than the δβ-thalassemia mutations. The finding of these mutations in this Brazilian population reflects the mixing process that occurred during its formation.

Microsatellites, or simple sequence repeats (SSRs), have proven to be an important molecular marker in plant genetics and breeding research. The main strategies to obtain these markers can be through genomic DNA and from expressed sequence tags (ESTs) from mRNA/cDNA libraries. Genetic studies using microsatellite markers have increased rapidly because they can be highly polymorphic, codominant markers and they show heterozygous conserved sequences. Here, we describe a methodology to obtain microsatellite using the enrichment library of DNA genomic sequences. This method is highly efficient to development microsatellite markers especially in plants that do not have available ESTs or genome databases. This methodology has been used to enrich SSR marker libraries in Citrus spp., an important tool to genotype germplasm, to select zygotic hybrids, and to saturate genetic maps in breeding programs. © Springer Science+Business Media, LLC 2013.

A quitotriosidase foi a primeira quitinase humana descrita e sua função fisiológica ainda não está totalmente esclarecida. Entretanto, diversos estudos têm demonstrado sua participação como componente na resposta imune humana. Uma duplicação de 24pb no éxon 10 do gene chit1 promove uma mudança na matriz de leitura do RNAm com deleção de 87 nucleotídeos. Esta alteração produzirá uma proteína sem atividade catalítica. Esta condição é chamada de deficiência de quitotriosidase e apresenta uma frequência aproximada de 6% de homozigose para a duplicação em diferentes grupos étnicos. A malária é uma parasitose endêmica da região amazônica causada por protozoários do gênero Plasmodium cujos sintomas incluem febre, dor de cabeça e vômitos, o que induz a uma resposta imunológica característica com o objetivo de combater essa patologia. Os objetivos deste trabalho foram avaliar o comportamento da enzima quitotriosidase em pacientes acometidos por malária no estado do Pará e determinar a frequência da duplicação de 24pb no gene da quitotriosidase em uma amostra representativa. Foi realizada dosagem de quitotriosidase em 100 indivíduos sadios e 47 pacientes com malária para a análise. A análise molecular da duplicação de 24 pb foi realizada em 100 voluntários através de protocolo que incluiu as técnicas de extração de DNA, PCR e depois visualização em gel de agarose 2,5% para verificação dos fragmentos normais (homozigoto normal: 195pb) e com a duplicação de 24pb (homozigoto mutante: 219pb; heterozigoto: 219pb e 195pb). Este trabalho descreveu pela primeira vez na literatura científica a elevação dos níveis plasmáticos de quitotriosidase em pacientes acometidos por malária vivax em comparação com um grupo de indivíduos sadios. Não houve associação entre a parasitemia e os níveis plasmáticos de quitotriosidase nos pacientes com Malária. A análise molecular apresentou uma frequência de 72% de indivíduos homozigotos normais, 24% de indivíduos heterozigotos e 4% de homozigotos mutantes para duplicação de 24 pb. As frequências alélicas ficaram em torno de 84% para o alelo selvagem e 16% para o alelo mutante. Não foi encontrada correlação entre o genótipo e o fenótipo bioquímico (representado pelos níveis de quitotriosidase) no grupo controle.

We describe a large Brazilian consanguineous kindred with 3 clinically affected patients with a Thomsen myotonia phenotype. They carry a novel homozygous nonsense mutation in the CLCN1 gene (K248X). None of the 6 heterozygote carriers show any sign of myotonia on clinical evaluation or electromyography. These findings confirm the autosomal recessive inheritance of the novel mutation in this family, as well as the occurrence of phenotypic variability in the autosomal recessive forms of myotonia. We ascertained a Brazilian family with nine individuals affected by autosomal dominant nonsyndromic sensorineural hearing loss. The bilateral hearing loss affected mainly mid-high frequencies, was apparently stable with an early onset. Microsatellites close to the DFNA8/DFNA12 locus, which harbors the TECTA gene, showed significant multipoint lod scores (32) close to marker D11S4107. Sequencing of the exons and exon-intron boundaries of the TECTA gene in one affected subject revealed the deletion c.5383 + 5delGTGA in the 5' end of intron 16, that includes the last two bases of the donor splice site consensus sequence. This mutation segregates with deafness within the family. To date, 33 different TECTA mutations associated with autossomal dominant hearing loss have been described. Among them is the mutation reported herein, first described by Hildebrand et al. (2011) in a UK family. The audioprofiles from the UK and Brazilian families were similar. In order to investigate the transcripts produced by the mutated allele, we performed cDNA analysis of a lymphoblastoid cell line from an affected heterozygote with the c.5383 + 5delGTGA and a noncarrier from the same family. The analysis allowed us to identify an aberrant transcript with skipping of exon 16, without affecting the reading frame. One of the dominant TECTA mutations already described, a synonymous substitution in exon 16 (c.5331 G<A), was also shown to affect splicing resulting in an aberrant transcript lacking exon 16. Despite the difference in the DNA level, both the synonymous substitution in exon 16 (c.5331 G<A) and the mutation described herein affect splicing of exon 16, leading to its skipping. At the protein level they would have the same effect, an in-frame deletion of 37 amino-acids (p.S1758Y/G1759_N1795del) probably leading to an impaired function of the ZP domain. Thus, like the TECTA missense mutations associated with dominant hearing loss, the c5383 + 5delGTGA mutation does not have an inactivating effect on the protein. (C) 2012 Elsevier B.V. Background: The Interleukin 28B (IL28B) rs12979860 polymorphisms was recently reported to be associated with the human T-cell leukemia virus type 1 (HTLV-1) proviral load (PvL) and the development of the HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP). Methods: In an attempt to examine this hypothesis, we assessed the association of the rs12979860 genotypes with HTLV-1 PvL levels and clinical status in 112 unrelated Brazilian subjects (81 HTLV-1 asymptomatic carriers, 24 individuals with HAM/TSP and 7 with Adult T cell Leukemia/Lymphoma (ATLL)). Results: All 112 samples were successfully genotyped and their PvLs compared. Neither the homozygote TT nor the heterozygote CT mutations nor the combination genotypes (TT/CT) were associated with a greater PvL. We also observed no significant difference in allele distribution between asymptomatic carriers and patients with HTLV-1 associated HAM/TSP. Conclusions: Our study failed to support the previously reported positive association between the IL28B rs12979860 polymorphisms and an increased risk of developing HAM/TSP in the Brazilian population.

ABSTRACT Given the decline of shallow-water red coral populations resulting from over-exploitation and mass mortality events, deeper populations below 50 metres depth (mesophotic populations) are currently the most harvested; unfortunately, very little is known about their biology and ecology. The persistence of these populations is tightly linked to their adult density, reproductive success, larval dispersal and recruitment. Moreover, for their conservation, it is paramount understand processes such as connectivity within and among populations. Here, for the first time, genetic variability and structuring of Corallium rubrum populations collected in the Tyrrhenian Sea ranging from 58 to 118 metres were analyzed using ten microsatellite loci and two mitochondrial markers (mtMSH and MtC). The aims of the work were 1) to examine patterns of genetic diversity within each geographic area (Elba, Ischia and Praiano) and 2) to define population structuring at different spatial scales (from tens of metres to hundreds of kilometres). Based on microsatellite data set, significant deviations from Hardy-Weinberg equilibrium due to elevated heterozygote deficiencies were detected in all samples, probably related to the presence of null alleles and/or inbreeding, as was previously observed in shallow-water populations. Moreover, significant levels of genetic differentiation were observed at all spatial scale, suggesting a recent isolation of populations. Biological factors which act at small spatial scale and/or abiotic factors at larger scale (e.g. summer gyres or absence of suitable substrata for settlement) could determine this genetic isolation. Using mitochondrial markers, significant differences were found only at wider scale (between Tuscany and Campania regions). These results could be related to the different mutation rate of the molecular makers or to the occurrence of some historical links within regions. A significant isolation by distance pattern was then observed using both data sets, confirming the restricted larval dispersal capability of the species. Therefore, the hypothesis that deeper populations may act as a source of larvae helping recovery of threatened shallow-water populations is not proved. Conservation strategies have to take into account these results, and management plans of deep and currently harvested populations have to be defined at a regional or sub regional level, similarly to shallow-water populations. Nevertheless, further investigations should be needed to understand better the genetic structuring of this species in the mesophotic zone, e.g. extending studies to other Mediterranean deep-water populations.

Ziel der vorliegenden Arbeit war es, ein innovatives System zur Herstellung einer konditional/reversiblen SCL-knock-out Maus zu etablieren. Es wurde deshalb ein neuer knock-in/knock-out Ansatz mit Hilfe des Tet-Systems gewählt. Im Rahmen dieser Arbeit war es möglich für die Generierung der konditionalen SCL-knock-out Maus essentielle Effektor- und Respondermauslinien zu etablieren und zu charakterisieren. Durch knock-in der rtTA-cDNA in den SCL-Lokus wurde eine Effektormauslinie hergestellt, welche in allen untersuchten hämatopoietischen Geweben rtTA exprimierte.Die zur Generierung der SCL-knock-out Maus notwendigen transgenen Responderlinien wurden ebenfalls generiert. Diese Linien wurden in einem Funktionsassay auf ihre Regulierbarkeit rekombinanter SCL-Expression untersucht. Dabei wurde eine Linie identifiziert, welche stringent/exogen regulierbar war.Effektor- und Responderlinie konnten etabliert und durch adäquate Kreuzung eine heterozygote Effektor/Responderlinie generiert werden. Die Rückkreuzung dieser Linie bei gleichzeitiger Gabe von DOX sollte in der konditionalen/reversiblen SCL-knock-out Maus resultieren. Bei Ende dieser Arbeit wurde allerdings in keinem Fall ein Rescue des letalen SCL Nullhintergrunds durch die rtTA-vermittelte rekombinante SCL-Expression beobachtet. Die in dieser Arbeit bereits hergestellten und verbesserten neuen Effektorkonstrukte sowie das Zurückgreifen auf alle vorhandenen SCL-Respondermäuse können in Zukunft zur erfolgreichen Etablierung eines exogen regulierbaren konditional/reversiblen SCL knock-out führen.

Im Rahmen dieser Doktorarbeit wurde in zwei Schwerpunktanalysen mit eine Teil- und Gesamtdatensatz die Untersuchung der Hybridisierung zwischen den beiden Microcebus-Arten M. murinus und M. griseorufus im Ökoton Südostmadagaskars umfangreich und vertieft untersucht. Für die genetischen Analysen wurden die maternal vererbte mitochondriale Hypervariable Region I (HVR 1) und neun nukleäre biparental vererbte Mikrosatellitenmarker eingesetzt. Als weiterer Datensatz wurden morphometrische Daten verwendet. Für die erste Schwerpunktanalyse wurde ein bereits vorhandener Teildatensatz (Hapke 2005 & Gligor 2006) mit Daten von insgesamt 162 Individuen aus neun Populationen der Dornbuschzone, der Übergangswaldzone und des Küstenwaldgebietes eingesetzt. In der zweiten Schwerpunktanalyse wurde eine umfangreiche Untersuchung der Microcebus griseoruus-M. murinus- Hybridzone vorgenommen. Für diese detaillierte Charakterisierung der Hybridzone wurde eine ausgedehnte und fein auflösende Probennahme in einem als Kernzone definierten Bereich, der die gesamte Übergangswaldzone und die dazu benachbarten Dornbuschgebiete umfasste, durchgeführt. Die morphometrischen und genetischen Daten der neu beprobten Individuen dieser Kernzone wurden mit den Daten des Teildatensatzes und weiteren Daten aus Küstenwaldpopulationen (Hapke 2005) zu einem Gesamtdatensatz zusammengefasst. Die Integration des Teildatensatzes in den Gesamtdatensatz erforderte umfassende und zeitintensive Labor- und Analysearbeiten, die im Rahmen dieser Doktorarbeit durchgeführt wurden. Der Gesamtdatensatz umfasste insgesamt 569 Individuen der Gattung Microcebus aus 29 Untersuchungsstandorten. Die mit beiden Datensätzen durchgeführte Analyse morphometrischer Daten zeigte deutlich, dass die Mehrzahl der Individuen aus der Übergangswaldzone einen intermediären Morphotyp aufweist. Durch die mit den Daten des Teildatensatzes durchgeführten Bayes’schen Clusteranalysen und Assignment-Tests, das vornehmlich in den Populationen der Übergangszone beobachtete signifikante Kopplungsungleichgewicht und Heterozygotendefizit, die festgestellte Verteilung der mitochondrialen Haplotypen und das kontrastierende Muster zwischen nukleären Mikrosatellitengenotypen und mitochondrialen Haplotypen in den Übergangswaldpopulationen konnte erstmals das Vorkommen einer Hybridzone zwischen Microcebus-Arten wissenschaftlich fundiert festgestellt werden. Die Ergebnisse dieser Schwerpunktanalyse wurden in der Fachzeitschrift Molecular Ecology publiziert (Gligor et al. 2009). Die in der ersten Schwerpunktanalyse festgestellte Hybridzone konnte durch die zweite Schwerpunktanalyse mit den genetischen und morphometrischen Daten des Gesamtdatensatzes nicht nur bestätigt werden, sondern auch auf die gesamte Übergangswaldzone erweitert werden. Ferner wurden starke Hinweise auf eine Hybridisierung beider Microcebus-Arten an einigen Dornbuschstandorten der Kernzone gefunden. Durch die große Datenmenge des Gesamtdatensatzes, vor allem aus der Kernzone des Untersuchungsgebietes, war es möglich eine fundierte Charakterisierung der Microcebus griseoruus-M. murinus- Hybridzone durchzuführen. Die Übereinstimmung der Hybridzone mit dem beobachteten Vegetationsmosaik zusammen mit den Ergebnissen der PCA, der PCoA und der Bayes’schen Clusteranalyse sprechen für das Modell der „Mosaik Hybridzone“, während die Einzelbetrachtung der mosaikartig verteilten intermediären Übergangswälder eine hohe Abundanz der Hybride aufzeigte und somit eher das „Bounded Hybrid Superiority model“ unterstützt. Der gewählte geographische Beprobungsmaßstab könnte somit einen Einfluss auf die beobachtete Struktur einer Hybridzone haben. Eines der markantesten Muster in der Hybridzone ist das stark kontrastierende cyto-nukleäre Muster. Der seit ca. 3000 Jahren fortschreitende Klimawandel in Südmadagaskar und die damit verbundene Expansion des Verbreitungsgebietes der Art Microcebus griseorufus nach Osten, das in dieser Arbeit festgestellte „male-biased dispersal“ bei M. griseorufus und der Einfluss exogener Selektion sprechen stark für eine massive asymmetrische nukleäre Genintrogression von M. griseorufus-Allelen in M. murinus-Populationen, verbunden mit einer potentiellen Verdrängung der Art M. murinus aus der Übergangswaldzone. In den jeweiligen Kerngebieten Dornbusch und Küstenwald bleibt jedoch die Diskretheit beider Arten gewahrt.
