The studies of this work aimed to determine the labile fractions of manganese (Mn) in natural and drainage water samples collected around the Osamu Utsumi uranium mine, located in the municipality of Caldas, south-central region of Poços de Caldas- MG, using the technique of diffusion gradient in thin films (DGT). The DGT devices were mounted with Chelex-100 resin, polyacrylamide-agarose hydrogel (conventional porosity) and cellulose acetate membrane. The device were deployed up to 48 hours in six water samples collected from different areas around the uranium mine (075, 076, 022-E, 025-E, 014, and 041). The DGT devices immersed in each sample were gradually removed after 4, 8, 12, 24 and 48 hours. The pH of the samples ranged from 3.0 to 10.5, which influenced the lability and the sampling of the analyte by the Chelex-100 resin. The results showed a linear relationship between accumulated mass and sampling time (immersion curve) for samples 014 and 025-E (pH between 6 and 8) suggesting the ability of the DGT technique for sampling the analyte. The results obtained for samples 075 and 076 (pH<5) and samples 041 and 022-E (pH around 10) were characterized by nonlinear relationships. The values obtained by DGT were compared with Solid Phase Extraction (SPE) technique using Chelex-100. For samples 014 and 025-E, there was a good agreement between the results obtained by both techniques

Colostrum intake in newborn goat kids is essential for the acquisition of immunoglobulins (Ig) and influencing development of gastrointestinal mucosa. The present study investigated small intestine structure in the postnatal goat kid fed lyophilized bovine colostrum, an alternative source of antibodies to small ruminants, or goat colostrum using scanning electron microscopy technique. At 0,7 and 14 h of life 15 male newborns received 5% of body weight of lyophilized bovine colostrum (LBC) and 14 goat colostrum (GC), both with 55 mg/mL of IgG. Samples of duodenum, medium jejunum and ileum were collected at 18, 36 and 96 h of life. Three animals were sampled at birth without colostrum intake (0 h). The enteric tissues were analyzed for villi density (villi/cm(2)) and morphological characteristics. The villi density did not differ between treatment, sampling time and intestinal segments (P>0.05). The morphological characteristics were not different between LBC and GC in all segments. Duodenal villi were fingerlike, thick and short, and with different heights. Duodenal folds could also be verified. Frequent anastomoses in all sampling times were observed in this segment. In the jejunum, fingerlike villi, thin and thick, of different heights were observed in all sampling times as well as leaf-shaped villi. Vacuoles with colostrum were observed in the jejunum of goats sampled at 18 h of life. In ileum, fingerlike villi were observed in all sampling times. At 0 and 96 h of life, thick and low villi were verified while at 18 and 36 h the villi showed different heights and widths. At all sampling times, regularly cell extrusion processes were observed with grouped cells at the apex of the ileum villi and with isolated cells along the villi. In the first 4 days of goat kids' life the small intestine structure was unaffected by different sources of colostrum, goat or lyophilized bovine, and by the replacement of fetal enterocytes, which are able to absorb macromolecules, by adult-type ones. Neospora caninum is an apicomplexan protozoan parasite that is one of the most important infectious causes of abortion in both dairy and beef cattle in many countries. The objectives of this longitudinal study were to determine the prevalence, rates of vertical and horizontal transmission of N. caninum and hazard for culling of N. caninum-seropositive animals in three Brazilian dairy herds. Blood samples from all animals were collected nine times at each of the three farms over a two-year period. Serum was tested for antibodies against N. caninum using the indirect fluorescent antibody test with a cutoff value of 1:100. The percentage of N. caninum-positive samples at each sampling time ranged at Farm 1 from 3.32% to 11.71%, at Farm 11 from 3.90% to 22.06% and at Farm III from 3.90% to 22.06%. The number of positive serological reactions varied in relation to the number of repeated samples taken from individual animals at each farm. In all herds, there was a high degree (P < 0.05) of association between the N. caninum serological status of dams and daughters. The seropositive conversion rate was estimated as 0.37%, 3.00% and 6.94% per 100 cow-years at Farms I, II and III, respectively. The seronegative conversion rate was estimated as 31.58% and 11.11% per 100 cow-years at Farms 1 and III, respectively. In all herds, there was no difference (P > 0.05) in the culling rate between the cattle that were seropositive cattle and seronegative for N. caninum infection. The results from this study confirm the importance of vertical transmission in the epidemiology of N. caninum. Although a few positive seroconversions indicated horizontal transmission, it does not appear to be the major route of infection for N. caninum. (C) 2012 Elsevier B.V. All rights reserved.

In the work underlying this thesis solid-phase microextraction (SPME) was evaluated as a passive sampling technique for organophosphate triesters in indoor air. These compounds are used on a large scale as flame-retarding and plastizicing additives in a variety of materials and products, and have proven to be common pollutants in indoor air. The main objective of this work was to develop an accurate method for measuring the volatile fraction. Such a method can be used in combination with active sampling to obtain information regarding the vapour/particulate distribution in different indoor environments. SPME was investigated under both equilibrium and non-equilibrium conditions and parameters associated with these different conditions were estimated. In Paper I, time-weighted average (TWA) SPME under dynamic conditions was investigated in order to obtain a fast air sampling method for organophosphate triesters. Among the investigated SPME coatings, the absorptive PDMS polymer had the highest affinity for the organophosphate triesters and was consequently used in all further work. Since the sampling rate is dependent on the agitation conditions, the linear airflow rates had to be carefully considered. Sampling periods as short as 1 hour were shown to be sufficient for measurements in the ng-μg m-3 range when using a PDMS 100-μm fibre and a linear flow rate above 7 cm s-1 over the fibre. SPME under equilibrium conditions is rather time-consuming, even under dynamic conditions, for slowly partitioning compounds such as organophosphate triesters. Nevertheless, this method has some significant advantages. For instance, the limit of detection is much lower compared to 1 h TWA sampling. Furthermore, the sampling time can be ignored as long as equilibrium has been attained. In Paper II, SPME under equilibrium conditions was investigated and evaluated for organophosphate triester vapours. Since temperature and humidity are closely associated with the distribution constant a simple study of the effect of these parameters was performed. The obtained distribution constants were used to determine the air levels in a common indoor environment. SPME and parallel active sampling on filters yielded similar results, indicating that the detected compounds were almost entirely associated with the vapour phase To apply dynamic SPME method in the field a sampler device, which enables controlled linear airflow rates to be applied, was constructed and evaluated (Paper III). This device was developed for application of SPME and active sampling in parallel. A GC/PICI-MS/MS method was developed and used in combination with active sampling of organophosphate triesters in indoor air (Paper IV). The combination of MS/MS and the soft ionization achieved with methanol as reagent gas yielded high selectivity and detection limits comparable to those provided by GC with nitrogen-phosphorus detection (NPD). The method limit of detection, when sampling 1.5 m3 of air, was in the range 0.1-1.4 ng m-3. In Paper V, the developed MS method was used in combination with SPME for indoor air measurements. The levels detected in the investigated indoor environments range from a few ng to μg m-3. Tris(2-chloropropyl) phosphate was detected at a concentration as high as 7 μg m-3 in a newly rebuilt lecture room.

Brown rot caused by Monilinia laxa and Monilinia fructigena is considered one of the most important diseases affecting Prunus species. Although some losses can result from the rotten fruits in the orchard, most of the damage is caused to fruits during the post-harvest phase. Several studies reported that brown rot incidence during fruit development highly varies; it was found that at a period corresponding to the the pit hardening stage, fruit susceptibility drastically decreases, to be quickly restored afterwards. However the molecular basis of this phenomenon is still not well understood. Furthermore, no difference in the rot incidence was found between wound and un-wound fruits, suggesting that resistance associated more to a specifc biochemical response of the fruit, rather than to a higher mechanical resistance. So far, the interaction Monilinia-peach was analyzed through chemical approaches. In this study, a bio-molecular approach was undertaken in order to reveal alteration in gene expression associated to the variation of susceptibility. In this thesis three different methods for gene expression analysis were used to analyze the alterations in gene expression occurring in peach fruits during the pit hardening stage, in a period encompassing the temporary change in Monilinia susceptibility: real time PCR, microarray and cDNA AFLP techniques. In 2005, peach fruits (cv.K2) were weekly harvested during a 19-week long-period, starting from the fourth week after full bloom, until full maturity. At each sampling time, three replicates of 5 fruits each were dipped in the M.laxa conidial suspension or in distilled water, as negative control. The fruits were maintained at room temperature for 3 hours; afterwards, they were peeled with a scalpel; the peel was immediately frozen in liquid nitrogen and transferred to -80 °C until use. The degree of susceptibility of peach fruit to the pathogen was determined on 3 replicates of 20 fruits each, as percentage of infected fruits, after one week at 20 °C. Real time PCR analysis was performed to study the variation in expression of those genes encoding for the enzymes of the phenylpropanoid pathway (phenylalanine ammonia lyase (PAL), chalcone synthase (CHS), cinnamate 4-hydroxylase (C4H), leucoanthocyanidine reductase (LAR), hydroxycinnamoyl CoA quinate hydroxycinnamoyl transferase (HQT) and of the jasmonate pathway, such as lipoxygenase (LOX), both involved in the production of important defense compounds. Alteration in gene expression was monitored on fruit samples of a period encompassing the pit hardening stage and the corresponding temporary resistance to M.laxa infections, weekly, from the 6thto the 12th week after full bloom (AFB) inoculated with M. laxa or mock-inoculated. The data suggest a critical change in the expression level of the phenylpropanoid pathway from the 7th to the 8th week AFB; such change could be directly physiologically associated to the peach growth and it could indirectly determine the decrease of susceptibility of peach fruit to Monilinia rot during the subsequent weeks. To investigate on the transcriptome variation underneath the temporary loss of susceptibility of peach fruits to Monilinia rot, the microarray and the cDNA AFLP techniques were used. The samples harvested on the 8th week AFB (named S, for susceptible ones) and on the 12th week AFB (named R, for resistant ones) were compared, both inoculated or mock-inoculated. The microarray experiments were carried out at the University of Padua (Dept. of Environmental Agronomy and Crop Science), using the μPEACH1.0 microarray together with the suited protocols. The analysis showed that 30 genes (corresponding to the 0.6% of the total sequences (4806) contained in the μPeach1.0 microarray) were found up-regulated and 31 ( 0.6%) down regulated in RH vs. SH fruits. On the other hand, 20 genes (0.4%) were shown to be up-regulated and 13 (0.3%) down-regulated in the RI vs. SI fruit. No genes were found differentially expressed in the mock-inoculated resistant fruits (RH) vs. the inoculated resistant ones (RI). Among the up-regulated genes an ATP sulfurylase, an heat shock protein 70, the major allergen Pru P1, an harpin inducing protein and S-adenosylmethionine decarboxylase were found, conversely among the down-regulated ones, cinnamyl alcohol dehydrogenase, an histidine- containing phosphotransfer protein and the ferritin were found. The microarray experimental results and the data indirectly derived, were tested by Real Time PCR analysis. cDNA AFLP analysis was also performed on the same samples. 339 transcript derived fragments considered significant for Monilinia resistance, were selected, sequenced and classified. Genes potentially involved in cell rescue and defence were well represented (8%); several genes (12.1%) involved in the protein folding, post-transductional modification and genes (9.2%) involved in cellular transport were also found. A further 10.3% of genes were classified as involved in the metabolism of aminoacid, carbohydrate and fatty acid. On the other hand, genes involved in the protein synthesis (5.7%) and in signal transduction and communication (5.7%) were found. Among the most interesting genes found differentially expressed between susceptible and resistant fruits, genes encoding for pathogenesis related (PR) proteins were found. To investigate on the association of Monilinia resistance and PR biological function, the major allergen Pru P1 (GenBank accession AM493970) and its isoform (here named Pru P2), were expressed in heterologous system and in vitro assayed for their anti-microbial activity. The ribonuclease activity of the recombinant Pru P1 and Pru P2 proteins was assayed against peach total RNA. As the other PR10 proteins, they showed a ribonucleolytic activity, that could be important to contrast pathogen penetration. Moreover Pru P1 and Pru P2 recombinant proteins were checked for direct antimicrobial activity. No inhibitory effect of Pru P1 or Pru P2 was detected against the selected fungi.

The Gulf of Aqaba represents a small scale, easy to access, regional analogue of larger oceanic oligotrophic systems. In this Gulf, the seasonal cycles of stratification and mixing drives the seasonal phytoplankton dynamics. In summer and fall, when nutrient concentrations are very low, Prochlorococcus and Synechococcus are more abundant in the surface water. This two populations are exposed to phosphate limitation. During winter mixing, when nutrient concentrations are high, Chlorophyceae and Cryptophyceae are dominant but scarce or absent during summer. In this study it was tried to develop a simulation model based on historical data to predict the phytoplankton dynamics in the northern Gulf of Aqaba. The purpose is to understand what forces operate, and how, to determine the phytoplankton dynamics in this Gulf. To make the models data sampled in two different sampling station (Fish Farm Station and Station A) were used. The data of chemical, biological and physical factors, are available from 14th January 2007 to 28th December 2009. The Fish Farm Station point was near a Fish Farm that was operational until 17th June 2008, complete closure date of the Fish Farm, about halfway through the total sampling time. The Station A sampling point is about 13 Km away from the Fish Farm Station. To build the model, the MATLAB software was used (version 7.6.0.324 R2008a), in particular a tool named Simulink. The Fish Farm Station models shows that the Fish Farm activity has altered the nutrient concentrations and as a consequence the normal phytoplankton dynamics. Despite the distance between the two sampling stations, there might be an influence from the Fish Farm activities also in the Station A ecosystem. The models about this sampling station shows that the Fish Farm impact appears to be much lower than the impact in the Fish Farm Station, because the phytoplankton dynamics appears to be driven mainly by the seasonal mixing cycle.

In der Vergangenheit wurde die Wichtigkeit von Iodverbindungen im Bezug auf die Aerosolbildung in Küstennähe wiederholt bestätigt. Durch Photolyse von flüchtigen iodorganischen Verbindungen (VOIs) können in der Atmosphäre Iodatome gebildet werden. Diese hochreaktiven Radikale wiederum können mit Ozon und/oder OH-Radikalen reagieren. Es werden so unter anderem schwerflüchtige Iodoxide gebildet, die in die Partikelphase übergehen können. Um ein Verständnis für die Mechanismen und chemischen Reaktionen zu bekommen, die zur Bildung von iodhaltigen Aerosolpartikeln führen, müssen auch Vorläufersubstanzen qualitativ und quanitativ bestimmt werden. Ob diese Reaktionen und chemischen Verbindungen auch über dem offenen Ozean einen Beitrag zu Aerosolbildung und somit zur Beeinflussung des weltweitem Klimas leisten, soll in dem EU-Projekt MAP geklärt werden, diese Arbeit ist Teil dieses Projekts. Im Rahmen dieser Arbeit wurde eine Methode entwickelt, die es zum einen möglich macht, anorganisches Iod in Meerwasser zu bestimmen. Zum anderen sollte eine Methode entwickelt werden, um elementares Iod in der maritimen Atmosphäre zu bestimmen. Es wurde eine Derivatisierungsmethode entwickelt, die es möglich macht elementares Iod in Anwesenheit von Stärke, a-Cyclodextrin oder RAMEA zu derivatisieren. Die Derivatisierung erfolgt zu 4-Iodo-N,N-Dimethylanilin. Durch Extraktion wird der Analyt in die organische Phase überführt. Die Quantifizierung erfolgt anschließend über die Analyse mit GC/MS und externer Kalibrierung. Die absolute Nachweisgrenze für Iod in Wasser beträgt 0,57nmol, für Iodid 0,014nmol und für Iodat 0,115nmol. Die absoluten Nachweisgrenzen für Iod in Anwesenheit eines Absorptionsmittel betragen für Stärke 0,24nmol, für a-Cyclodextrin 0,9nmol und für RAMEA 0,35nmol. Die Analysenmethoden wurden zunächst im Labor entwickelt und anschließend zur Analyse von Realproben verwendet. An verschiedenen Orten wurden Meerwasserproben (auf der Celtic Explorer und in der Nähe der Mace Head Messstation) genommen und deren Iod-, Iodid- und Iodatgehalt bestimmt. Keine der Proben enthielt elementares Iod. Iodid konnte in allen Proben detektiert werden. In Proben, die auf dem offenen Ozean an Bord der Celtic Explorer genommen wurden variierte die Menge zwischen 12µg/L und 90µg/L. Auffällig war hierbei, dass die Proben, die in Küstennähe genommen wurden höhere Iodidkonzentrationen aufwiesen. Ein Einfluss der Küste und der dort vorhandenen Makroalgen ist sehr wahrscheinlich. Meerwasserproben, die in der Nähe der MHARS genommen wurden wiesen höhere Konzentrationen und einen größeren dynamischen Bereich der Iodidkonzentrationen auf. Die Konzentrationen variierten von 29µg/L bis 630 µg/L. Der Iodatgehalt der Meerwasserproben wurde ebenfalls bestimmt. 1µg/L bis 90µg/L Iodat konnte in den Proben vom offenen Ozean detektiert werden. Die Küstenproben wiesen mit 150µg/L bis 230µg/L deutlich höhere Iodatkonzentrationen auf. Es konnte kein Zusammenhang zwischen der Tageszeit und den Iodid- oder Iodatkonzentrationen gefunden werden. Es konnte ebenso kein Zusammenhang zwischen der Fluoreszenz des Meerwassers und den Iodid- oder Iodatkonzentrationen gefunden werden. Auf der Celtic Explorer, wie auch in Mace Head wurden außerdem beschichtete Denuder zur Anreicherung von elementarem Iod aus Luft eingesetzt. Die Denuder, die auf dem Schiff verwendet wurden waren mit Stärke bzw. mit a-CD beschichtet. Die mit Stärke beschichteten Denuder geben so einen Überblick über die Iodkonzentration in Luft über einen längeren Zeitraum (ca. 2-3h), während die mit Cyclodextrin beschichteten Denuder die Iodkonzentration in der letzten halben Stunde der Probennahme widerspiegeln. In fast allen Denudern, die mit Stärke beschichtet waren, konnte mehr Iod nachgewiesen werden, als in denen, die mit a-CD beschichtet waren. Im Allgemeinen konnten in den Proben höhere Iodkonzentrationen gefunden werden, die nachts genommen wurden. Der Grund hierfür liegt in der sehr hohen Photolyserate des elementaren Iods während des Tages. Ein Zusammenhang zwischen der Konzentration von VOIs und dem Iodgehalt konnte nicht gefunden werden. Anhand der genommen Denuderproben von Mace Head konnte festgestellt werden, dass die Iodkonzentration in Denudern, deren Probenahme während Ebbe beendet wurde hoch deutlich höher sind, als die in anderen Denudern. Das lässt sich dadurch erklären, dass Makroalgen während Ebbe in direktem Kontakt zur Luft sind und somit mehr Iod in der Luft zu finden ist. Eine wichtige Frage, die im Zusammenhang mit der Iodchemie in maritimer Umgebung steht konnte im Rahmen dieser Arbeit geklärt werden. In der maritimen Grenzschicht über dem Nordatlantik konnte elementares Iod detektiert werden, d.h. es deutet sich an, dass Iod auch auf dem offenen Ozean einen Beitrag zur Partikelbildung liefern kann und es sich nicht ausschließlich um einen Küsteneffekt handelt.
