Systemic lupus erythematosus is a chronic autoimmune disease with multifactorial ethiopathogenesis. The complement system is involved in both the early and late stages of disease development and organ damage. To better understand autoantibody mediated complement consumption we examined ex vivo immune complex formation on autoantigen arrays. We recruited patients with SLE (n = 211), with other systemic autoimmune diseases (n = 65) and non-autoimmune control subjects (n = 149). Standard clinical and laboratory data were collected and serum complement levels were determined. The genotype of SNP rs1143679 in the ITGAM gene was also determined. Ex vivo formation of immune complexes, with respect to IgM, IgG, complement C4 and C3 binding, was examined using a functional immunoassay on autoantigen microarray comprising nucleic acids, proteins and lipids. Complement consumption of nucleic acids increased upon binding of IgM and IgG even when serum complement levels were decreased due to consumption in SLE patients. A negative correlation between serum complement levels and ex vivo complement deposition on nucleic acid autoantigens is demonstrated. On the contrary, complement deposition on tested protein and lipid autoantigens showed positive correlation with C4 levels. Genetic analysis revealed that the non-synonymous variant rs1143679 in complement receptor type 3 is associated with an increased production of anti-dsDNA IgG antibodies. Notwithstanding, homozygous carriers of the previously reported susceptible allele (AA) had lower levels of dsDNA specific IgM among SLE patients. Both the non-synonymous variant rs1143679 and the high ratio of nucleic acid specific IgG/IgM were associated with multiple organ involvement. In summary, secondary complement deficiency in SLE does not impair opsonization of nucleic-acid-containing autoantigens but does affect other antigens and potentially other complement dependent processes. Dysfunction of the receptor recognizing complement opsonized immune complexes promotes the development of class-switched autoantibodies targeting nucleic acids.

This study was undertaken to determine the effect of nutritional management of broodstock of Penaeus monodon on growth and maturation. Test specimens were obtained from a grow-out pond before attainment of maturity and were reared in hatchery tanks. Four types of dietary treatments (M1–M4) were given to separate batches that were run in duplicate. Feeding trials continued for five months. A diet with live bloodworm, bioencapsulated to contain tricalcic phosphate as its major component, was found to be the most efficient. Specimens of this particular batch assimilated food more efficiently, grew at a faster rate and attained maturity earlier than other groups. Bloodworm provided the lipid fractions for which there is no de novo synthesis in shrimp. A vitamina D, atualmente, é relacionada também ao metabolismo da glicose e o desenvolvimento de órgãos. Fêmeas de camundongos suíços (F0) foram alimentadas por uma das dietas experimentais: SC (dieta padrão) ou VitD- (dieta sem vitamina D). A prole de machos foi estudada nas idades: nascimento, 10 dias, desmame e seis meses, nas gerações F1 e F2. Avaliou-se a biometria [Massa Corporal (MC), Comprimento nasoanal (CNA) e Pressão Arterial (PA)], urina de 24 horas, glicemia e Teste Oral de Tolerância à Glicose (TOTG). Durante a eutanásia, o sangue foi coletado para análise bioquímica e os tecidos foram removidos para análise estereológica, morfométrica e Western blotting (WB). Não houve diferença de MC ao nascimento. Ao desmame, o grupo F2-VitD- teve maior MC que F2-SC (P=0,03) e aos seis meses, os grupos F1 e F2-VitD- tiveram MC mais elevada (P<0,05 vs SC). A PA foi crescente na prole VitD-, sendo maior em F1-VitD- (P=0,001). A glicemia e TOTG foram alterados somente na F1-VitD-, seguida de esteatose hepática (+99%), hipertrofia da ilhota pancreática (+40%) e elevação do triglicerídeo sanguíneo (P<0,01). O WB de fígado mostrou elevação de FAS (+18%, P<0,01), no grupo com esteatose. Curiosamente, embora a F2-VitD- tenha apresentado elevação de MC, somente o colesterol total fora alterado (P<0,05). Quanto à nefrogênese, houve 50% mais glomérulos imaturos em F1-VitD- que F1-SC (P<0,0001). Porém, na F2 houve aumento somente de 20% (P<0,001). Aos 10 dias, F1-VitD- teve 150% mais glomérulos imaturos e 25% mais glomérulos maduros que SC-F1 (P<0,0001). O WB de rim mostrou que a prole F1-VitD- apresentou maior expressão de renina, ao desmame e aos seis meses, enquanto que a expressão de podocina foi reduzida (P=0,0004). Não houve diferença na análise de WT1. A restrição materna em vitamina D altera a morfologia do pâncreas e fígado, com resistência à insulina, altera a expressão renal de importantes fatores, assim como retarda a maturação glomerular estendendo o período da nefrogênese, principalmente na geração F1.

Sex-specific demography and reproductive biology of stripey bass (Lutjanus carponotatus) (also known as Spanish flag snapper, FAO) were examined at the Palm and Lizard island groups, Great Barrier Reef (GBR).Total mortality rates were similar between the sexes. Males had larger L∞ at both island groups and Lizard Island group fish had larger overall L∞. Female:male sex ratios were 1.3 and 1.1 at the Palm and Lizard island groups, respectively. The former is statistically different from 1, but is unlikely significantly different in a biological sense. Females matured on average at 2 years of age and 190 mm fork length at both locations. Female gonadal lipid body indices peaked from August through October, preceding peak gonadosomatic indices in October, November, and December that were twice as great as in any other month. However, ovarian staging revealed 50% or more ovaries were ripe from September through February, suggesting a more protracted spawning season and highlighting the different interpretations that can arise between gonad weight and gonad staging methods. Gonadosomatic index increases slightly with body size and larger fish have a longer average spawning season, which suggests that larger fish produce greater relative reproductive output. Lizard Island group females had ovaries nearly twice as large as Palm Island group females at a given body size. However, it is unclear whether this reflects spatial differences akin to those observed in growth or effects of sampling Lizard Island group fish closer to their date of spawning. These results support an existing 250 mm minimum size limit for L. carponotatus on the GBR, as well as the timing of a proposed October through December spawning closure for the fishery. The results also caution against assessing reef-fish stocks without reference to sex-, size-, and location-specific biological traits.

The gonads of Otolithes cuvieri and Johnius elongatus are described in seven maturity stages. O. Cuvieri spawns once a year from April to September as evidence by ova diameter frequency distribution and GSI values. 50% maturity is attained at 210mm TL in males and 200mm TL in females. Fecundity ranged from 2387 to 104379 with a mean value of 33502. Log-Log relationship between fecundity and total lenght, body weight and ovary weight were determined. An overall sex ratio of 1.54:1.00 was unequal in favour of males. Johnius elongatus spawns twice a year from January-February to Aprile-May and from August to October as evidence by ova diameter frequency distribution and GSI values. 50% maturity is attained at 140-143mm TL in both sexes. Fecundity ranged from 4238 to 167669 with a mean value of 42818. Log-Log relationship between fecundity and total lenght, body weign and ovary weight were determined. An overall sex ratio of 1.00:1.20 was unequal in favour of females.
