Metabolism in Psammomys obesis, a polygenic animal model of obesity and type 2 diabetes is associated with dysregulated nocturnal fat oxidation in diabetic animals. Furthermore, a new gene called AGT-203 has been identified. Evidence indicates that AGT-203 is involved in abnormal glucose metabolism leading to the proposition that AGT-203 is a new candidate gene for type 2 diabetes.

Creatine is an important molecule involved in providing energy to the body. Its major stores are in skeletal muscle. The creatine transporter protein (CreaT) mRNA is believed to be responsible for the uptake of the majority of creatine in skeletal muscle. This thesis examined factors that might have affected the expression of the creatine transporter in skeletal muscle.

The majority of falls occur during routine ambulation and transfer tasks required for basic mobility such as rapid stopping. The study found that in comparison with young adults older adults more frequently used two steps to terminate walking. This is due to increased onset times in the rear leg muscles and a lower frequency of ankle and hip extensor recruitment in the front leg.

The results of this thesis indicate that vegetarianism and prior exercise can increase skeletal muscle creatine loading. The mechanisms by which these phenomena occur remain unknown but may be related to changes in the gene expression of a cellular pump within the muscle known as the "creatine transporter".

The effects of exercise on novel signalling enzymes in skeletal muscle of humans was investigated. It was shown that exercise increased the activity of a calcium and calmodulin activated kinase. High-intensity, but submaximal, exercise increased the activity of some but not all isoforms of protein kinase C, a lipid-activated kinase family. These findings suggest that these enzymes may be part of the signalling process leading to beneficial adaptation to repeated exercise as well as the control of function within skeletal muscle during exercise.

This work examined the effects of a novel dairy fatty acid conjugated linoleic acid (CLA) and its effects on muscle wasting in advanced cancer. Results showed a positive anti-inflammatory role of CLA on the supression of tumour growth and established a model for studying the action of CLA in human muscle-wasting conditions.

Fifteen trained male cyclists had muscle biopsies and performed cycling tests to determine if relationships exist between the oxygen uptake response and various intramuscular variables. It was found that muscle oxidative capacity is better able to explain the oxygen uptake response during high intensity cycling than muscle fiber distribution.

Objective : To determine the level of neck strength decrement and the rate of strength recovery of the neck muscles after a single bout of specific neck conditioning exercise in both males and females.

Hypothesis : A decrement in neck strength may be evident after a bout of strengthening exercise.

Design : Intervention study with pre-and-post design.

Setting : Biomechanics laboratory.

Participants : Twenty healthy participants (10 male and 10 female, mean ± standard deviation age 22 ± 1.2 years).

Main Outcome Measures : Participants performed a single bout of neck strengthening exercise. Neck strength testing using an isokinetic dynamometer was performed pre and at five time points (1 h, one, three, five and seven days) post-exercise to assess the level of neck strength decrement and neck strength recovery rate from pre-exercise levels.

Results : Statistically significant (p ≥ 0.036) decreases in neck extension strength were recorded in all participants 1 h and one day post-exercise. The level of neck extension strength returned to pre-exercise levels three days post-exercise and surpassed pre-exercise levels five and seven days post-exercise. The male participants' neck flexion strength decrement and recovery followed a similar pattern to that displayed in neck extension but more variability in neck flexion strength recovery rates were recorded in the female participants in this study.

Conclusion : The consistent strength recovery times for the male participants recorded in this study idealise the prescription of neck strengthening exercises in a periodised fashion. More investigation needs to be instigated for the female neck musculature as consistent strength recovery rates were not identified in this study.
