High-resolution orbital and in situ observations acquired of the Martian surface during the past two decades provide the opportunity to study the rock record of Mars at an unprecedented level of detail. This dissertation consists of four studies whose common goal is to establish new standards for the quantitative analysis of visible and near-infrared data from the surface of Mars. Through the compilation of global image inventories, application of stratigraphic and sedimentologic statistical methods, and use of laboratory analogs, this dissertation provides insight into the history of past depositional and diagenetic processes on Mars. The first study presents a global inventory of stratified deposits observed in images from the High Resolution Image Science Experiment (HiRISE) camera on-board the Mars Reconnaissance Orbiter. This work uses the widespread coverage of high-resolution orbital images to make global-scale observations about the processes controlling sediment transport and deposition on Mars. The next chapter presents a study of bed thickness distributions in Martian sedimentary deposits, showing how statistical methods can be used to establish quantitative criteria for evaluating the depositional history of stratified deposits observed in orbital images. The third study tests the ability of spectral mixing models to obtain quantitative mineral abundances from near-infrared reflectance spectra of clay and sulfate mixtures in the laboratory for application to the analysis of orbital spectra of sedimentary deposits on Mars. The final study employs a statistical analysis of the size, shape, and distribution of nodules observed by the Mars Science Laboratory Curiosity rover team in the Sheepbed mudstone at Yellowknife Bay in Gale crater. This analysis is used to evaluate hypotheses for nodule formation and to gain insight into the diagenetic history of an ancient habitable environment on Mars.

The Danube is ca. 2850 km in length and is the second largest river in Europe. The Austrian part of the Danube falls 156 metres in altitude over its 351 km length and, since the early 1950s, the river has been developed into a power-generating waterway, so that the continuity of the river is now interrupted by ten impounded areas. Only two stretches of the original free-flowing river are left, the Wachau region (above river-km 2005, west of Vienna) and the region downstream from the impoundment at Vienna (river-km 1921). Most of the recent theories and concepts related to invertebrates, in the context of the ecology of running waters, are based on studies on small streams, whereas investigations of large rivers have played a minor role for a long time, mainly due to methodological difficulties. The authors' recent detailed studies on macroinvertebrates in the free-flowing section of the Danube below Vienna, provide an excellent opportunity to survey or restate scientific hypotheses on the basis of a large river. In this review the main interest focuses on the investigation of biodiversity, i.e. the number of species and their relative proportions in the whole invertebrate community, as well as major governing environmental factors. The article summarises the species composition, the important environmental variables at the river cross-section and the effect of upstream impoundment on the riverbed and its fauna.

Bio-orthogonal non-canonical amino acid tagging (BONCAT) is an analytical method that allows the selective analysis of the subset of newly synthesized cellular proteins produced in response to a biological stimulus. In BONCAT, cells are treated with the non-canonical amino acid L-azidohomoalanine (Aha), which is utilized in protein synthesis in place of methionine by wild-type translational machinery. Nascent, Aha-labeled proteins are selectively ligated to affinity tags for enrichment and subsequently identified via mass spectrometry. The work presented in this thesis exhibits advancements in and applications of the BONCAT technology that establishes it as an effective tool for analyzing proteome dynamics with time-resolved precision.

Chapter 1 introduces the BONCAT method and serves as an outline for the thesis as a whole. I discuss motivations behind the methodological advancements in Chapter 2 and the biological applications in Chapters 2 and 3.

Chapter 2 presents methodological developments that make BONCAT a proteomic tool capable of, in addition to identifying newly synthesized proteins, accurately quantifying rates of protein synthesis. I demonstrate that this quantitative BONCAT approach can measure proteome-wide patterns of protein synthesis at time scales inaccessible to alternative techniques.

In Chapter 3, I use BONCAT to study the biological function of the small RNA regulator CyaR in Escherichia coli. I correctly identify previously known CyaR targets, and validate several new CyaR targets, expanding the functional roles of the sRNA regulator.

In Chapter 4, I use BONCAT to measure the proteomic profile of the quorum sensing bacterium Vibrio harveyi during the time-dependent transition from individual- to group-behaviors. My analysis reveals new quorum-sensing-regulated proteins with diverse functions, including transcription factors, chemotaxis proteins, transport proteins, and proteins involved in iron homeostasis.

Overall, this work describes how to use BONCAT to perform quantitative, time-resolved proteomic analysis and demonstrates that these measurements can be used to study a broad range of biological processes.

Cyclopids, exactly in the same way as daphnids, significant component in the nutrition of plankton-f and the young of the majority of fishes. It is established that the food spectrum of cyclopids is extremely broad: daphnids, planarians, Copepodite stages of copepods (cannibalism), rotifers, protists, bacteria, phytoplankton and so on. It is clear that the problem of studying these or other components of feeding in the general food spectrum can be definitely resolved only after obtaining exact quantitative data on the feeding of cyclopids. This article attempts to fill the gap in the study of the quantitative side of the feeding of cyclopias; in it is investigated the size of the 24-hour ration of cyclopids feeding on protists, the dependence of the ration on some factors of the external medium, and the difference of 24-hour consumption per unit weight of tody with two species of cyclopids (Cyclops strenuus and Cyclops viridis).

Pressupondo que o conhecimento sobre a doença renal crônica (DRC) e seu tratamento, possibilita ao cliente entendimento e aceitação para conviver com esse agravo, favorecendo comportamentos de autocuidado, delimitou-se os problemas: Qual é a qualidade de vida de clientes com DRC submetidos à hemodiálise? Quais são as necessidades de orientação de enfermagem para o autocuidado desses clientes visando à promoção de sua qualidade de vida? Objetivos específicos: Identificar as características sóciodemográficas e nosológicas de clientes com DRC, em hemodiálise, associando às suas necessidades de orientação de enfermagem para o autocuidado; Identificar a qualidade de vida desses clientes, aplicando o questionário de Kidney Disease Quality of Life Short Form (KDQOL-SF); Relacionar as necessidades de orientação de enfermagem para o autocuidado com a qualidade de vida dos clientes com DRC em terapia de hemodiálise. Descreve-se como marco referencial a Teoria do Autocuidado de Orem, concepções de autocuidado e de qualidade de vida. Pesquisa descritiva, quantitativa, através da entrevista individual realizada na Unidade de Diálise da Enfermaria de Nefrologia do Hospital Universitário Pedro Ernesto da Universidade do Estado do Rio de Janeiro, no período de agosto de 2008 a maio de 2009. Foram sujeitos de pesquisa 43 clientes. Foram utilizados: formulário para caracterização da clientela e levantamento das necessidades de autocuidado e o questionário KDQOL-SF para mensurar a qualidade de vida dos sujeitos. Resultados: Os clientes com doença renal crônica em terapia de hemodiálise são, em sua maioria, do sexo masculino (55%) e mantém união estável (81%); situando-se 39,53%, na faixa etária de 45 a 65 anos e 79,07% na categoria de aposentados. 37,54% têm ensino fundamental. Quanto às características nosológicas, 74,42% possuem hipertensão arterial, encontrando-se 83,72% em hemodiálise, há menos de um ano. A qualidade de vida desses clientes, avaliada pelo KDQOL-SF, obteve os menores escores nas dimensões: limitações causadas por problemas da saúde física; condição de trabalho; limitações causadas por problemas da saúde emocional; capacidade funcional e sobrecarga imposta pela doença renal. Relacionando esse resultado com o obtido no questionário para avaliação das necessidades de orientação de enfermagem para o autocuidado tem-se: problemas da saúde física relacionado com terapia nutricional, ingestão de líquidos, complicações da hemodiálise, anticoagulação e prática de atividade física; relacionadas a problemas de saúde emocional tem-se a associação a grupos e a atividades de lazer; e relacionada à capacidade funcional e sobrecarga da doença renal tem-se a prática de atividade física. Conclui-se que a enfermagem, além de administrar a realização das sessões de hemodiálise, tem papel fundamental na educação à saúde dos clientes, familiares e/ou acompanhantes. O apoio do enfermeiro ao cliente no processo de enfrentamento e tratamento da DRC, contribui para que este adquira habilidade nas ações de autocuidado e consequentemente favoreça sua qualidade de vida.
