Today's scientific interest in tissue engineering for organ transplantations and regeneration from stem cells, allied with recent observations on biostimulation of tissues and cells by laser radiation, stands as a strong motivation for the present work, in which we examine the effects of the low power laser radiation onto planarians under regenerative process. To investigate those effects, a number of 60 amputated worms were divided in three study groups: a control group and two other groups submitted to daily 1 and 3 min long laser treatment sections at similar to 910 W/m(2) power density. A 685 nm diode laser with 35 mW optical power was used. Samples were sent to histological analysis at the 4th, the 7th and the 15th (lays after amputation. A remarkable increase in stem cells counts for the fourth day of regeneration was observed when the regenerating worms was stimulated by the laser radiation. Our findings encourage further research works on the influence of optical radiation onto stem cells and tissue regeneration. (c) 2005 Elsevier B.V. All rights reserved.

In this work we have:investigated the growth and differentiation of bone marrow stem cells in mice bearing Ehrlich ascites tumor-and treated with three dose-regimens of Dicyclopentadienyldichlorotitanium (IV) (DDCT). We also: studied the presence of colony stimulating factors In the serum of PDCT-treated animals as well-as the effects-of the drug on the survival of the tumor-bearing mice. The-results demonstrated that the myelosuppression developed in the tumor-bearing animals is prevented by the administration:of 1, 2 or 3 doses of 15 mg/kg DDCT. In the treatment with three doses, however, 23 % of the animals died. Moreover, DDCT treatment in normal animals resulted in increased numbers of CFU-GM. We observed the presence of stimulating factors in the serum of drug-treated animals which induced the growth and differentiation of bone marrow progenitor cells from normal animals in vitro. on the other hand, in vitro addition of the drug to these cultures had no effect. Thus, we conclude that the drug protects against the myelosuppression induced by the tumor and that this protection may be related to an indirect action of the drug. Emphysema is characterized by destruction of alveolar walls with loss of gas exchange surface and consequent progressive dyspnea. This study aimed to evaluate the efficiency of cell therapy with bone marrow mononuclear cells (BMMC) in an animal model of elastase-induced pulmonary emphysema. Emphysema was induced in C57Bl/J6 female mice by intranasal instillation of elastase. After 21 days, the mice received bone marrow mononuclear cells from EGFP male mice with C57Bl/J6 background. The groups were assessed by comparison and statistically significant differences (p & 0. 05) were observed among the groups treated with BMMC and evaluated after 7, 14 and 21 days. Analysis of the mean linear intercept (Lm) values for the different groups allowed to observe that the group treated with BMMC and evaluated after 21 days showed the most significant result. The group that received no treatment showed a statistically significant difference when compared to other groups, except the group treated and evaluated after 21 days, evidencing the efficacy of cell therapy with BMMC in pulmonary emphysema. © 2012 Springer Science+Business Media New York.

Embryonic stem cells are pluripotent and able to generate all cell types of the body, being the most promising cells to the study for regenerative medicine. Following the differentiation path, there are adult stem cells, which are committed with a specific cell lineage, creating limitations on their application. On the extreme opposite of embryonic stem cells there are the induced pluripotent stem cells, originated from a somatic cell after genetic reprogramming. Induced pluripotent stem cells are a recent science discovery and may substitute the use of embryonic stem cells in future research. But with nowadays knowledge, the use of adult stem cells and induced pluripotent stem cells are limited due to high expenses and long time process demand. Moreover, the development achieved on all kinds of stem cells study are, in some part, due to the study of embryonic stem cells, what makes the study of these cell type still mandatory. © Todos os direitos reservados a.

A zona subgranular (ZSG) do giro denteado (GD) de mamíferos adultos é conhecida por produzir constantemente novos neurônios. A busca por novas moléculas que possam modular a formação de novas células neurais são bastante atuais. Visto que a Amazônia é conhecida mundialmente pela sua biodiversidade, com um potencial pouco explorado de fármacos naturais derivados de plantas medicinais típicas da região. O trabalho buscou investigar o efeito neurogênico do extrato aquoso (EA) da Physalis angulata e da substância purificada Fisalina D sobre as células-tronco do GD do hipocampo de camundongos adultos. Os camundongos machos (BALB/c), 6 a 8 semanas de idade foram divididos em quatro grupos experimentais: controle e tratados com EA ou substância purificada. Os animais receberam diferentes doses do extrato (0,1; 1 e 5 mg/Kg) e/ou substância purificada (5mg/Kg) ou salina (grupo controle), 5 horas depois uma única dose de 5-Bromodeoxiuridina (BrdU) [50mg/kg]. Em seguida, os animais foram sacrificados 24 horas ou 7 dias após a administração do BrdU. Os cérebros foram coletados e cortes coronais do hipocampo (40 μm) foram realizados para contagem das células BrdU-positivas no GD hipocampal. Para avaliação estatística realizamos análise de variância (ANOVA) das médias amostrais seguida pelo pós-teste t de Student. O EA promoveu um aumento significativo do número de células BrdU positivas no GD dos grupos tratados em relação ao grupo controle [Controle, 92±24 (n=9); 0,1mg/Kg, 160±22 (n=4); 1mg/Kg, 310±5 (n=4); 5mg/Kg, 501±24 (n=3)] nos animais sacrificados 24 horas após administração do BrdU. Quando os animais foram sacrificados 7 dias após administração do BrdU, o número de células BrdU+ no GD também foi maior no grupo tratado em relação ao controle [Controle, 107±7 (n=4); 5mg/Kg, 145±23 (n=4)]. Usando a substância purificada, Fisalina D, também observamos um aumento do número de células BrdU+ no GD do grupo tratado com a droga em relação ao grupo controle [Controle, 92±24 (n=9); Fisalina D, 5mg/Kg, 316±37 (n=3)]. Este resultado sugere que o EA e a sustância purificada, na dose de 5 mg/Kg, estimulam a proliferação de células BrdU-positivas na ZSG do GD do hipocampo de camundongos adultos.

A produção in vitro de embriões (PIV) é uma biotecnologia utilizada para aumentar o potencial reprodutivo de animais geneticamente superiores, os embriões produzidos in vitro são de qualidade inferior aos produzidos in vivo, por isso técnicas tentam melhorar os índices de embriões produzidos in vitro. Uma técnica é o sistema de co-cultivo com células somáticas que removem metabólitos tóxicos e protegem contra o stress oxidativo. As células-tronco mesenquimais derivadas de tecido adiposo (CTA) são células multipotentes que segregam fatores de crescimento e citocinas. As células-tronco foram utilizadas em co-cultivo in vitro de embriões bovinos em diferentes concentrações com o objetivo de melhorar o protocolo de PIVE. CTAs foram submetidas à diferenciação em três linhagens mesenquimais, e foi realizada a imunofenotipagem de marcadores específicos de membrana das CTMs. A taxa de clivagem foi avaliada no segundo dia após a fertilização e taxa de blastocistos no sétimo dia, quando foram armazenados para contagem do número total de células e expressão gênica. Os resultados foram analisados por ANOVA, Teste-t e pós-teste de Fisher, adotando um nível de significância de 5%. O tratamento do co-cultivo com CTAs influenciou significativamente a formação de blastocisto, o número total de células de embriões e a expressão gênica correlacionada a pluripotência e metabolismo de carboidratos. Estes resultados mostraram aumento da taxa de produção e qualidade dos embriões produzidos in vitro em co-cultivo com CTAs em relação ao co-cultivo com células da granulosa. Os resultados deste trabalho indicam também que a presença constante de CTAs em co-cultivo é superior ao condicionamento com CTAs. Os efeitos verificados das CTAs podem ocorrer através de fatores solúveis ou via exossomos secretados pelas CTAs. Estudos futuros são necessários para esclarecer a possível via causadora dos efeitos positivos verificados neste trabalho pelas CTAs em co-cultivo.

BACKGROUND Chronic obstructive pulmonary disease is a major inflammatory disease of the airways and an enormous therapeutic challenge. Within the spectrum of chronic obstructive pulmonary disease, pulmonary emphysema is characterized by the destruction of the alveolar walls with an increase in the air spaces distal to the terminal bronchioles but without significant pulmonary fibrosis. Therapeutic options are limited and palliative since they are unable to promote morphological and functional regeneration of the alveolar tissue. In this context, new therapeutic approaches, such as cell therapy with adult stem cells, are being evaluated.OBJECTIVE This article aims to describe the follow-up of up to 3 years after the beginning of a phase I clinical trial and discuss the spirometry parameters achieved by patients with advanced pulmonary emphysema treated with bone marrow mononuclear cells.METHODS Four patients with advanced pulmonary emphysema were submitted to autologous infusion of bone marrow mononuclear cells. Follow-ups were performed by spirometry up to 3 years after the procedure.RESULTS The results showed that autologous cell therapy in patients having chronic obstructive pulmonary disease is a safe procedure and free of adverse effects. There was an improvement in laboratory parameters (spirometry) and a slowing down in the process of pathological degeneration. Also, patients reported improvements in the clinical condition and quality of life.CONCLUSIONS Despite being in the initial stage and in spite of the small sample, the results of the clinical protocol of cell therapy in advanced pulmonary emphysema as proposed in this study, open new therapeutic perspectives in chronic obstructive pulmonary disease. It is worth emphasizing that this study corresponds to the first study in the literature that reports a change in the natural history of pulmonary emphysema after the use of cell therapy with a pool of bone marrow mononuclear cells.

The purpose of this work was to validate isolation methods for sheep mesenchymal stem cells (MSC) from different sources and to explore the hypothesis that MSC exhibit markers of the same phenotype independent from tissue source. Cells derived from ovine bone marrow, synovial membrane and adipose tissue were characterized using the following markers: CD44, CD45, CD11b and MHC-I. The isolated MSC were cultivated, went through osteogenic, chondrogenic and adipogenic differentiation, and were characterized by flow cytometry using mouse anti-ovine CD44, CD45 and MHC-I monoclonal antibody (mAb), and mouse anti-bovine CD11b mAb. Ovine MSC from all three sources differentiated under chondorgenic, osteogenic and adipogenic conditions. Also, MSC from the three tissues were found to express CD44 and MHC-I but lack of CD11b and CD45. The results obtained revealed that our isolation methods for the different tissues tested are valid and that MSC from the three sources studied have same immunophenotic characteristics.
