The use of cover crops is a fundamental strategy to the weed management in Southern Brazil. In highly infested areas, the herbicides use is increasing, which increases the costs of the crops production as well as the environmental contamination. Oat and velvet bean plants havecontrasting characteristics regarding to residues decomposition speed and the capacity to immobilize Nitrogen in the soil, providing distinct results of weeds suppression throughout the time, and therefore, requiring distinct management strategies before, during, and after the corn crop establishment. The general objective of the experiment was to evaluate the environmental dynamics of the herbicide atrazine, the corn grain yield, and the efficiency of the weed control, considering areas with distinct history regarding the use of mulching, levels of straw and rates of atrazine. For this, the experiment was carried out in two parts: in the first part, two trials with the corn crop were established, one using oat and the other using velvet bean as cover crops. The experimental design used for both field trials was randomized complete blocks arrangement with four replications. The factor A was constituted by four levels of straw (0; 0.75x; 1.5x; 3x) and the factor B was constituted by four rates of the herbicide atrazine (0; 2100; 4200; 8400 g a i. ha-1). Soil samples were collected for greenhouse trialsto determine the persistence. Atrazine leaching evaluation was performed by chromatography using samples collected over the soil profile.In the field, the weed density, the fresh and dry weight and the yield of the corn were evaluated. In the greenhouse trials, the main variables evaluated were plant height and injury caused by the herbicide toxicity. In the second part, soils with distinct covering history were sampled, and the mineralization and sorption studies, both with 14C-atrazine, were conducted in the laboratory. The experimental design was randomized complete blocks arrangement with four replications. The results from the field experiment show that the high levels of straw above ground, isolated, were not efficient to control completely the weeds, and that high levels of velvet bean`s straw decreased the corn potential yield. The greenhouse trials showed that high levels of oat straw prevent the scape of atrazine to soil, this effect of oat straw upon the herbicide availability on soil was detected up to 12 days after spraying. The half-life of atrazine sprayed over oat straw varied from 7 to 14 days after spraying, while the half-life of atrazine sprayed over velvet bean varied from 5 to 14 days after spraying. Increasing oat straw levels presents the capacity to reduce the lixiviation of atrazine in the soil profile, however, this effect was not verified when using velvet bean straw, because the herbicide was not detected in the soil profile, at 21 days after spraying. The chromatographic analysis indicate thatthe atrazine concentrates closer to the soil surface regardless of amount of straw, not being detected deeper than 8 cm in the soil. The accumulated mineralization of 14C-arazine sprayed over V. sativa is superior if compared to soils with S. cereale or non-covered soils. The sorption coefficient of atrazine is superior when sprayed over straw than over the soil.

A descriptive study was developed in order to compare indoor and outdoor air contamination caused by fungi and particles in seven poultry units. Twenty eight air samples of 25 litters were collected through the impaction method on malt extract agar. Air sampling and particles concentration measurement were done in the interior and also outside premises of the poultries’ pavilions. Regarding the fungal load in the air, indoor concentration of mold was higher than outside air in six poultry units. Twenty eight species / genera of fungi were identified indoor, being Scopulariopsis brevicaulis (40.5%) the most commonly isolated species and Rhizopus sp. (30.0%) the most commonly isolated genus. Concerning outdoor, eighteen species/genera of fungi were isolated, being Scopulariopsis brevicaulis (62.6%) also the most isolated. All the poultry farms analyzed presented indoor fungi different from the ones identified outdoors. Regarding particles’ contamination, PM2.5, PM5.0 and PM10 had a statistically significant difference (Mann-Whitney U test) between the inside and outside of the pavilions, with the inside more contaminated (p=.006; p=.005; p=.005, respectively). The analyzed poultry units are potential reservoirs of substantial amounts of fungi and particles and could therefore free them in the atmospheric air. The developed study showed that indoor air was more contaminated than outdoors, and this can result in emission of potentially pathogenic fungi and particles via aerosols from poultry units to the environment, which may post a considerable risk to public health and contribute to environmental pollution.

Moulds may produce a diversity of toxins such as aflatoxins, ochratoxins, trichothecenes, zearalenone, fumonisins and others. Although toxicological, environmental and epidemiological studies have addressed the problem of these toxins one by one, more than one mycotoxin are found usually in the same contaminated food. Risk assessment for humans potentially exposed to multimycotoxins suffers very much from the lack of adequate food consumption data. Furthermore, for a given mycotoxin, synergism and antagonism with other mycotoxins, found in the same food commodities, are not taken into account. Aflatoxin B1 and ochratoxin A belong to the most frequently occurring mycotoxins. This has repeatedly been demonstrated, however, normally, the risk resulting from their simultaneous occurrence is not considered. A descriptive study was developed to monitor air fungal contamination in one hospital food unit.

A descriptive study was developed to compare air and surfaces fungal contamination in ten hospitals’ food units and two food units from companies. Fifty air samples of 250 litres through impaction method were collected from hospitals’ food units and 41 swab samples from surfaces were also collected, using a 10 by 10 cm square stencil. Regarding the two companies, ten air samples and eight surface samples were collected. Air and surface samples were collected in food storage facilities, kitchen, food plating and canteen. Outdoor air was also collected since this is the place regarded as a reference. Simultaneously, temperature, relative humidity and meal numbers were registered. Concerning air from hospitals’ food units, 32 fungal species were identified, being the two most commonly isolated genera Penicillium sp.

The presence of weeds decreases the crop yield. Among the alternatives to reduce the crop yield loss, it can be included to increase the competitive ability of the crop and the chemical control of the weeds. A research program was developed in the course of Agronomy at Federal Technological University at Paraná, Campus Pato Branco - PR, during the years 2015/16, with the objectives evaluating if gibberellin inhibitors increase the competitive ability of bean plants, making them insensitive to the initialism, extending the period prior to weed-crop interference. Evaluate the tolerance of common bean plants to the herbicide ethoxysulfuron and investigate the existence of relationship between the plant mass and the level of tolerance of the plants to the herbicide. Evaluate the effect of increasing doses of ethoxysulfuron on morphological characteristics, yield components and grain yield of the bean cultivars IPR Tangará and IPR Andorinha. Evaluate the effect of increasing doses of ethoxysulfuron on the development of IAC Imperador and the community of weeds present in the area. Elucidate the mechanism that confers tolerance to bean plants to the herbicide ethoxysulfuron. The results indicate that gibberellin inhibitors were not effective in increasing periods of weed-crop coexistence. Trinexapac-ethyl increased 20% the grain yield of bean plants. It was observed high variability as the response of bean cultivars to the herbicide ethoxysulfuron, however, despite high doses (200 g ha-1), it was not observed death of the plants. The field results indicate that when the ethoxysulfuron dose is 83.2 g ha-1, the reduction in grain yield can reach 40% with the cultivar IPR Tangará and 30% in the cultivar IPR Andorinha. However, respectively for each cultivar cited, ethoxysulfuron at 17 and 12 g ha-1 are enough to reduce 10% of grain yield. Evaluating the control of weeds within the bean crop cultivar IAC Imperador with the herbicide ethoxysulfuron, it was observed that doses at 20 g ha-1 are enough to control soybean and Ipomoea spp. plants. But, due to the level of plant injury, the crop grain yield increase was not sufficient to match the one observed on the weed-free untreated control. The mechanism of tolerance of bean plants to ethoxysulfuron appears to be the herbicide degradation.

La revisión de la literatura nos muestra que la depresión es comúnmente comórbida con alcoholismo, esta relación ha sido siempre objeto de interés clínico y científico. El presente estudio tiene como objeto determinar la frecuencia de la Depresión en pacientes alcohólicos hospitalizados, siendo evaluados al tercer día de hospitalización y luego de transcurridos 30 días de encontrarse en abstinencia alcohólica. Es un estudio epidemiológico de tipo longitudinal. Participaron 102 pacientes del CRA. Se aplicó el cuestionario de síntomas diseñado por la OMS [SQR] para diagnóstico de depresión y la escala de Hamilton-D para determinar la severidad del cuadro a los pacientes que resultaron positivos para la enfermedad. En la primera evalución la frecuncia de la depresión fue de 29.4 por ciento y de los cuales el 93.3 por ciento corresponden a una depresión mayor. Transcurridos treinta días, se realizó una nueva valoración, disminuyendo considerablemente la frecuencia de la enfermedad a 18.2 por ciento y de los cuales el 100 por cien es una depresión mayor. Los antecendentes personales y familiares de alcoholismo y depresión juegan un papel importante en la presencia de esta patología. Antes de iniciar un tratamiento antidepresivo se debe realizar un análisis profundo de la historis clínica del paciente, para no tratar depresiones que puedan remitir espontánemante con abstiencia alchólica de alugnos días.

Nos dias de hoje a contaminação dos solos e águas subterrâneas com pesticidas através da agricultura tornou-se um problema. Problema este, considerado ainda maior nas áreas onde o abastecimento de água potável é quase totalmente à base de água subterrânea, causando deste modo risco para a saúde humana devido à exposição directa de possíveis resíduos de pesticidas na água potável. É certo que a persistência dos pesticidas no solo é importante para obter um bom controlo sob as ervas daninhas durante a sua época de crescimento, contudo o uso desses pesticidas contamina não só o solo como as águas superficiais. As questões acerca do uso de pesticidas na actualidade continuarão a persistir, uma vez que existem muitos factores e características inerentes a este processo que necessitam de ser abordadas e mais importante que isso estudadas, como por exemplo a sua degradação e toxicidade. Neste trabalho efectuou-se o encapsulamento de pesticidas em moléculas de β – ciclodextrina (β-CD). O que se pretende com este encapsulamento, é aumentar a hidrofilicidade do pesticida de forma a garantir que este persista o tempo suficiente permitindo um bom controlo das ervas daninhas, tendo sempre em conta as preocupações inerentes ao uso dos pesticidas, como por exemplo a dificuldade de biodegradação. O estudo centrou-se em torno de dois dos pesticidas mais utilizados em Portugal: MCPA e Bentazona. Estes herbicidas foram encapsulados individualmente na β-CD formando assim complexos, mais solúveis e eventualmente mais estáveis quimicamente garantindo uma redução dos efeitos dos pesticidas no meio ambiente. Este estudo foi dividido essencialmente em duas partes: a síntese e caracterização dos complexos pesticida-β-CD e posteriormente a avaliação da estabilidade química em solução aquosa e da solubilidade dos complexos formados. A utilização de diversas técnicas analíticas nomeadamente DSC, FTIR, Espectrofotometria de UV, HPLC e Electroquímica permitiram concluir que o pesticida MCPA encapsula pela acção da β-CD aquando do complexo formado em solução etanólica e numa proporção estequiométrica MCPA:β-CD de 1:2 respectivamente. Obteve-se para as várias soluções estudadas, todas elas com concentrações diferentes de β-CD, uma constante de estabilidade de 102,4. No caso da Bentazona, os resultados preliminares obtidos indiciam claramente a formação de um complexo com a β-CD para o complexo formado em solução etanólica.

A mucosa intestinal é a primeira barreira biológica encontrada pelas micotoxinas presentes nos alimentos, sendo a patulina, uma micotoxina produzida por fungos do género Penicillium spp., uma preocupação particular atendendo a que a exposição humana a esta micotoxina pode conduzir a efeitos imunológicos, neurológicos e gastrointestinais. Considerando estes efeitos para a saúde, o presente estudo tem como objetivos a avaliação do efeito tóxico da exposição intestinal a patulina, bem como a determinação do potencial efeito protetor da coadministração de patulina e cisteína na membrana intestinal, utilizando para o efeito células Caco-2. A integridade da membrana intestinal foi determinada pela medição da resistência elétrica transepitelial (TEER). Os resultados evidenciaram um decréscimo acentuado nos valores de TEER após 24 horas de exposição celular a 95 μM de patulina. Para as concentrações mais reduzidas verificou-se uma redução máxima inferior a 25% após 24 horas de exposição. A coadministração de patulina (95 μM) e cisteína (40 μM) revelou um decréscimo nos valores de TEER. O tratamento com cisteína em concentrações superiores ( 400 μM) revelou efeito protetor da membrana intestinal, tendo em conta os valores de TEER. Estes resultados contribuem para uma avaliação do risco mais precisa associada à exposição a contaminantes alimentares.

Hidrocarbonetos Aromáticos Policíclicos (HAPs) e seus derivados Halogenados (HHAPs) são conhecidos como um conjunto de contaminantes ambientais, classificados como potencialmente tóxicos, mutagénicos e carcinogénicos, sendo um assunto de extrema importância e preocupação para a saúde pública. Diversos estudos têm reportado o potencial efeito tóxico destes compostos após longos períodos de exposição, pelo uso de métodos colorimétricos como o ensaio de brometo de 3-[4,5- dimetil-tiazol-2-il]-2,5-difeniltetrazólio (MTT) e vermelho neutro. Contudo, apesar de estes métodos clássicos de avaliação da citotoxicidade in vitro permitirem, de forma rápida, identificar as propriedades nocivas dos compostos, não permitem compreender os mecanismos moleculares subjacentes responsáveis pela sua toxicidade. Deste modo, no presente estudo, para avaliar os efeitos à exposição aos compostos Pireno (Pir) e seu derivado resultante da desinfeção de águas por bromação, 1-BromoPireno (1-BrPir), foram utilizados outros métodos complementares como avaliação do stress oxidativo e vários fenómenos associados à morte celular programada. Os resultados obtidos comprovam não só a existência de efeitos citotóxicos dos poluentes quando acumulados, como também possíveis efeitos genotóxicos. Assim, pelos métodos complementares, foi possível a identificação de alvos moleculares passiveis de ação farmacológica, o que pode constituir o primeiro passo para o desenho de estratégias terapêuticas que visem prevenir ou tratar os danos provocados por estes poluentes no Homem.

Os Hidrocarbonetos Aromáticos Policíclicos (HAPs) são contaminantes persistentes em meio aquoso. Estes compostos são conhecidos pelas suas propriedades carcinogénicas, mutagénicas e genotóxicas. O principal objetivo deste trabalho consistiu na avaliação das potencialidades de subprodutos da indústria corticeira, como adsorventes alternativos para a remoção de cinco HAPs em meio aquoso: benzo(a)pireno, benzo(ghi)perileno, benzo(b)fluoranteno, benzo(k)fluoranteno e indeno(1,2,3-cd)pireno. A metodologia analítica para quantificar os HAPs envolveu a preparação das amostras, através da técnica de extração em fase sólida (SPE), e a quantificação dos compostos analisados por cromatografia líquida com detetor de fluorescência (LC-FLD). O método foi otimizado e validado, obtendo-se limites de quantificação de 0,004 μg/L para todos os HAPs. Os estudos incidiram na utilização de uma amostra de cortiça, pó de aglomerado de cortiça expandida (PACE), obtida por aglutinação de cortiça em condições hidrotérmicas, a qual nos estudos preliminares revelou desempenho semelhante aos carvões ativados. Com exceção do benzo(ghi)perileno, os resultados mostram que o processo de adsorção dos HAPs na amostra PACE segue uma cinética de pseudo-segunda ordem e as isotérmicas ajustam-se ao modelo de Langmuir.

A batata está sujeita a stresses bióticos (doenças, pragas, infestantes) e abióticos (condições climáticas, secura, excesso de água, ou fitotoxicidade provocada por herbicidas, entre outros). Os adubos foliares contendo nutrientes (macro e micronutrientes) poderão contribuir para uma melhor nutrição da planta, para além de exercer uma ação benéfica ao nível do potencial hídrico da folha e sua proteção contra os diversos inimigos bióticos. Instalou-se um ensaio em Marinhais na variedade de batata de indústria Lady Rosetta, para avaliar o efeito da adubação foliar de uma gama de adubos foliares “Humigel”, contendo, entre outros nutrientes, o enxofre e o potássio, numa situação de fitotoxidade causada pela aplicação de um herbicida. O ensaio foi instalado com um dispositivo experimental de parcelas totalmente aleatórias, com 4 repetições. Os tratamentos foram a aplicação de “Humigel” e sem aplicação (testemunha). A produção comercial nas parcelas com “Humigel” foi significativamente mais elevada (48 192 kg/ha) do que nas parcelas testemunha (43 458 kg/ha). Não se registaram diferenças significativas ao nível do número de tubérculos (comercial e total) por planta, nem ao nível dos parâmetros de qualidade (peso específico e teste de fritura).

Este proyecto de investigaci?n presenta un an?lisis ex-ante del impacto socioecon?mico de introducir una yuca tolerante a herbicidas en el departamento del Cauca. Para la estimaci?n del cambio en el margen neto de ganancia de los agricultores en el primer a?o de adopci?n de la tecnolog?a (entendido como la diferencia entre el ingreso bruto y los costos totales de producci?n) se emple? un enfoque de presupuestos parciales y diferentes escenarios de an?lisis, entre ellos, un an?lisis estoc?stico que permite considerar la incertidumbre (a trav?s de la asignaci?n de distribuciones de probabilidad) en las variables de la estructura de costos de producci?n que se ven afectadas con la tecnolog?a, otorgando mayor realismo a los resultados. Por otro lado, se calcularon los potenciales beneficios sociales de esta tecnolog?a y la distribuci?n de estos entre productores y consumidores utilizando un modelo de excedentes econ?micos. Los resultados indican que una variedad de yuca tolerante a herbicida permitir? reducir, en promedio, 12,6% los costos totales de producci?n por hect?rea. Adem?s, traer? beneficios econ?micos positivos tanto a productores como a consumidores, no obstante la TIR de la inversi?n del proyecto est? apenas por encima de la tasa de descuento, indicando que la probabilidad de que una instituci?n privada est? interesada en invertir en este tipo de tecnolog?as para el cultivo de yuca no es muy alta. El an?lisis de sensibilidad de los principales par?metros del modelo de excedentes econ?micos permiti?, por un lado, obtener resultados m?s robustos y, por otro lado, aportar informaci?n para futuros an?lisis o planes de acci?n para reducir las posibles amenazas y potencializar beneficios de esta tecnolog?a.

There is a growing concern within public health about mycotoxin involvement in human diseases, namely those related to children. The MycoMix project (2012-2015), funded by the Portuguese Foundation for Science and Technology, gathered a multidisciplinary team aiming at answering several questions: 1) Are Portuguese children exposed daily to one or several mycotoxins through food? 2) Can this co-exposure affect children´s health? and 3) Are there interaction effect between mycotoxins? Mycomix results revealed that Portuguese children (< 3 years old, n=103) are exposed to multiple mycotoxins through food consumption. Cumulative risk assessment results revealed a potential health concern for the high percentiles of intake, specially for aflatoxins which are carcinogenic compounds. This fact assumes particular importance considering the interactive effects found in in vitro bioassays. These results highlight the need for a more accurate approach to assess the human exposure to mycotoxins6. Within the Mycomix project the assessment of mycotoxin exposure was based on calculations combining mycotoxin data in food with population data on food consumption. This approach does not consider some aspects as the inter-individual metabolism variation, the exposure through sources other than food and the heterogeneous distribution of mycotoxins in food. Exposure assessment of mycotoxins in Portuguese population through biomarkers is still missing and further studies are urgent to be developed. The European Human Biomonitoring Initiative (EHBMI), a proposal within the European Joint Programme, aims to advance the understanding of the extent of exposure to environmental chemicals across Europe and the impact on human health, by gathering national expertise in human biomonitoring domain. At national level Mycomix project uncovered the potential health risk of exposure of Portuguese children to multiple mycotoxins. The risk assessment expertise acquired within Mycomix, namely in analysis and toxicology of chemical mixtures, will be brought together as a contribute to EHBMI objectives.

Historically, the health risk of mycotoxins had been evaluated on the basis of single-chemical and single-exposure pathway scenarios. However, the co-contamination of foodstuffs with these compounds is being reported at an increasing rate and a multiple-exposure scenario for humans and vulnerable population groups as children is urgently needed. Cereals are among the first solid foods eaten by child and thus constitute an important food group of their diet. Few data are available relatively to early stages child´s exposure to mycotoxins through consumption of cereal-based foods. The present study aims to perform the cumulative risk assessment of mycotoxins present in a set of cereal-based foods including breakfast cereals (BC), processed cereal-based foods (PCBF) and biscuits (BT), consumed by children (1 to 3 years old, n=75) from Lisbon region, Portugal. Children food consumption and occurrence of 12 mycotoxins (aflatoxins, ochratoxin A, fumonisins and trichothecenes) in cereal-based foods were combined to estimate the mycotoxin daily intake, using deterministic and probabilistic approaches. Different strategies were used to treat the left censored data. For aflatoxins, as carcinogenic compounds, the margin of exposure (MoE) was calculated as a ratio of BMDL (benchmark dose lower confidence limit) and aflatoxin daily exposure. For the remaining mycotoxins, the output of exposure was compared to the dose reference values (TDI) in order to calculate the hazard quotients (HQ, ratio between exposure and a reference dose). The concentration addition (CA) concept was used for the cumulative risk assessment of multiple mycotoxins. The combined margin of exposure (MoET) and the hazard index (HI) were calculated for aflatoxins and the remaining mycotoxins, respectively. Main results revealed a significant health concern related to aflatoxins and especially aflatoxin M1 exposure according to the MoET and MoE values (below 10000), respectively. HQ and HI values for the remaining mycotoxins were below 1, revealing a low concern from a public health point of view. These are the first results on cumulative risk assessment of multiple mycotoxins present in cereal-based foods consumed by children. Considering the present results, more research studies are needed to provide the governmental regulatory bodies with data to develop an approach that contemplate the human exposure and, particularly, children, to multiple mycotoxins in food. The last issue is particularly important considering the potential synergistic effects that could occur between mycotoxins and its potential impact on human and, mainly, children health.

The intestinal mucosa is the first biological barrier encountered by natural toxins, and could possibly be exposed to high amounts of dietary mycotoxins. Patulin (PAT), a mycotoxin produced by Penicillium spp. during fruit spoilage, is one of the best known enteropathogenic mycotoxins able to alter functions of the intestine (Maresca et al., 2008). This study evaluated the effects of PAT on barrier function of the gut mucosa utilizing the intestinal epithelial cell model Caco-2, and scrutinized immunomodulatory effects using human peripheral blood mononuclear cells (PBMC) and human blood monocyte-derived dendritic cells (moDCs) as test systems. PAT exposure reduced Caco-2 cell viability at concentrations above 12 mM. As expected, the integrity of a polarized Caco-2 monolayer was affected by PAT exposure, as demonstrated by a decrease in TER values, becoming more pronounced at 50 mM. No effects were detected on the expression levels of the tight junction proteins occludin, claudin-1 and claudin-3 at 50 mM. However, the expression of zonula occludens-1 (ZO-1) and myosin light chain 2 (MLC2) declined. Also, levels of phospho-MLC2 (p-MLC2) increased after 24 h of exposure to 50 mM of PAT. T cell proliferation was highly sensitive to PAT with major effects for concentrations above 10 nM of PAT. The same conditions did not affect the maturation of moDC. PAT causes a reduction in Caco-2 barrier function mainly by perturbation of ZO-1 levels and the phosphorylation of MLC. Low doses of PAT strongly inhibited T cell proliferation induced by a polyclonal activator, but had no effect on the maturation of moDC. These results provide new information that strengthens the concept that the epithelium and immune cells of the intestinal mucosa are important targets for the toxic effects of food contaminants like mycotoxins
