Seals must manage their energy reserves carefully while they fast on land to ensure that they go to sea with sufficient fuel to sustain them until they find food. Glucocorticoids (GCs) have been implicated in the control of fuel metabolism and termination of fasting in pinnipeds. Here we tested the hypothesis that dexamethasone, an artificial GC, increases fat and protein catabolism, and induces departure from the breeding colony in wild, fasting grey seal pups. A single intramuscular dose of dexamethasone completely suppressed cortisol production for 24–72 h, demonstrating activation of GC receptors. In experiment 1, we compared the effects of a single dose of dexamethasone or saline administered 10 days after weaning on fasting mass and body composition changes, cortisol, blood urea nitrogen (BUN) and glucose levels, and timing of departure from the colony. In experiment 2, we investigated the effects of dexamethasone on short-term (5 days) changes in mass loss, body composition and BUN levels. In experiment 1, dexamethasone induced a short-lived increase in mass loss, but there was no difference in timing of departure between dexamethasone- and saline-treated pups (N=10). In experiment 2, dexamethasone increased protein and water loss and prevented a decrease in BUN levels (N=11). Our data suggest changes in cortisol contribute to regulation of protein catabolism in fasting seal pups, irrespective of the sex of the animal, but do not terminate fasting. By affecting the rate of protein depletion, lasting changes in cortisol levels could influence the amount of time seal pups have to find food, and thus may have important consequences for their survival.

Le climat continental et froid de la Béringie lors de la glaciation du Wisconsinien a conduit à la formation d’une forme relique de pergélisol syngénétique nommé yedoma. Ces dépôts ont permis la préservation d’indicateurs environnementaux très diversifiés qui peuvent être employés pour reconstituer la dynamique climatique et écologique de la Béringie avant le dernier maximum glaciaire. À ce jour, peu d’études ont été réalisées au nord de la chaîne de montagnes Brooks (Alaska) et l’hétérogénéité écologique régionale de la Béringie Est lors de la glaciation du Wisonsinien reste mal définie. Ce mémoire porte sur une reconstitution paléoenvironnementale de plus de 39 ka du nord de l’Alaska réalisée à partir de sédiments provenant du Yedoma de la rivière Itkillik. Les objectifs sont (1) de reconstituer l’histoire de la végétation avec l’analyse pollinique; (2) de reconstituer les températures de juillet, le contraste de température saisonnier et l’ensoleillement de juillet avec la technique des analogues modernes et (3) de mettre les données biogéochimiques et glaciologiques du site en lien avec le climat reconstitué. L’étude montre que vers 35 ka BP (Interstade du Wisconsinien Moyen), des conditions climatiques semblables à l’actuel ont favorisé l’accumulation de tourbe riche en carbone organique. À partir de 29,7 ka BP, les températures de juillet reconstituées diminuent, alors que la continentalité du climat semble augmenter. Le contenu en glace des sédiments est plus alors plus faible et la pluie pollinique devient dominée par Poaceae, Artemisia et autres herbacés non graminoïdes. Ces indicateurs suggèrent des conditions environnementales plus xériques qu’aujourd’hui. Les anomalies isotopiques de 18O, 2H et l’excès de deutérium confirment un épisode d’avancée glaciaire (Wisconsinien Tardif). Après 17,9 ka BP (Tardiglaciaire), les températures de juillet et le contraste saisonnier augmentent. Les valeurs de contenu en carbone organique des sédiments sont plus élevées et la plus grande disponibilité en eau favorise l’établissement d’un couvert herbacé moderne dominé par les Cyperaceae.

OBJECTIVE: Low levels of daily energy expenditure (insufficient physical activity and increased sedentary time) have been associated with adverse health outcomes in young people. The Multimedia Activity Recall for Children and Adolescents (MARCA) is a computerized, self-report, use-of-time tool that can assess daily energy expenditure. The study aim was to validate the MARCA for the estimation of energy expenditure in young people, using the criterion standard doubly labeled water. MATERIALS/METHODS: Over a 15 day assessment period, 32 participants (10-18 years) completed the MARCA and underwent a doubly labeled water protocol. Indirect calorimetry was used to assess resting metabolic rate. Total daily energy expenditure (TEE) and activity-related energy expenditure (AEE) were estimated from both the MARCA and doubly labeled water. Association and agreement between methods for TEE and AEE were assessed using Spearman correlations and Bland-Altman plots, respectively. RESULTS: Compared to doubly labeled water, the MARCA over-estimated TEE by an average of 50 kcal/day (limits of agreement -1 589 to 1 490 kcal/day) and under-estimated AEE 105 kcal/day (limits of agreement -1 404 to 1 614 kcal/day). The MARCA showed strong correlation with doubly labeled water for TEE (rho=0.70, p<0.0001) and moderate correlation for AEE (rho=0.56, p=0.0009). CONCLUSIONS: Overall, the MARCA indicated moderate validity for the assessment of daily TEE and AEE. The wide limits of agreement indicate the MARCA has greater utility for group-level rather than individual-level estimates.

OBJECTIVES: The detailed structure of brain-derived Aβ amyloid fibrils is unknown. To approach this issue, we investigate the molecular architecture of Aβ(1-40) fibrils grown in either human cerebrospinal fluid solution, in chemically simple phosphate buffer in vitro or extracted from a cell culture model of Aβ amyloid plaque formation. METHODS: We have used hydrogen-deuterium exchange (HX) combined with nuclear magnetic resonance, transmission electron microscopy, seeding experiments both in vitro and in cell culture as well as several other spectroscopic measurements to compare the morphology and residue-specific conformation of these different Aβ fibrils. RESULTS AND CONCLUSIONS: Our data reveal that, despite considerable variations in morphology, the spectroscopic properties and the pattern of slowly exchanging backbone amides are closely similar in the fibrils investigated. This finding implies that a fundamentally conserved molecular architecture of Aβ peptide fold is common to Aβ fibrils.

The cold season in the Arctic extends over eight to nine months during which ecosystem gas exchange and water balance of arctic plants have been largely unexplored. The overall objective of this thesis was to examine two critical gaps in our knowledge about tundra cold season processes – ecosystem respiration at very low temperatures and water uptake during the winter-spring transition. I determined the temperature response of ecosystem respiration of tundra monoliths down to temperatures as low as can be expected under snow-covered conditions (-15 °C). Temperature responses fit the Arrhenius function well with Q10 values over the range of -15 to 15 °C varying from 6.1 to 4.8. I used deuterium-enriched water (2H2O) as a tracer to evaluate water uptake of evergreen plants at snowmelt when soils are largely frozen. The results revealed that evergreen plants take up water under snow cover, possibly via roots but undoubtedly by foliar uptake.
