Cyclophosphamide (CY) was used to evaluate the effect on the immune system of sheep. Castred adult rams were divided into 3 groups, with 6 animals each one. Group I (day 0) and Group II (day 1) were treated with CY (40 mg/kg, single dose, IV), and Group III was not treated and remained as control. All groups were immunized on day 0 with B19 brucellosis vaccine. on day 6, all animals were bled and serum agglutination test for brucellosis antibodies detection was performed. During 7 days blood lymphocyte counts and electrophoresis gammaglobulin dosage were daily performed. The results showed statistical decrease of immune response. Low serum titers of brucellosis antibodies were found in Groups I and II, and lymphopenia and hypogammaglobulinemia were also found in these groups. A high mortality rate (40%) occurred in the treated animals.

Aims The macrophage migration inhibitory factor (MIF) is an intracellular inhibitor of the central nervous system actions of angiotensin II on blood pressure. Considering that angiotensin II actions at the nucleus of the solitary tract are important for the maintenance of hypertension in spontaneously hypertensive rats (SHRs), we tested if increased MIF expression in the nucleus of the solitary tract of SHR alters the baseline high blood pressure in these rats.Methods and resultsEight-week-old SHRs or normotensive rats were microinjected with the vector AAV2-CBA-MIF into the nucleus of the solitary tract, resulting in MIF expression predominantly in neurons. Rats also underwent recordings of the mean arterial blood pressure (MAP) and heart rate (via telemetry devices implanted in the abdominal aorta), cardiac- and baroreflex function. Injections of AAV2-CBA-MIF into the nucleus of the solitary tract of SHRs produced significant decreases in the MAP, ranging from 10 to 20 mmHg, compared with age-matched SHRs that had received identical microinjections of the control vector AAV2-CBA-eGFP. This lowered MAP in SHRs was maintained through the end of the experiment at 31 days, and was associated with an improvement in baroreflex function to values observed in normotensive rats. In contrast to SHRs, similar increased MIF expression in the nucleus of the solitary tract of normotensive rats produced no changes in baseline MAP and baroreflex function.ConclusionThese results indicate that an increased expression of MIF within the nucleus of the solitary tract neurons of SHRs lowers blood pressure and restores baroreflex function. © 2012 Published on behalf of the European Society of Cardiology. Existe uma diversidade de espécies de Leishmania prevalentes na região Amazônica associadas à LTA configurando a etiologia múltipla da doença e, apesar do conhecimento da elevada ocorrência desta protozoose na Mesorregião do Baixo Amazonas, à oeste do Estado do Pará, quase nada era sabido sobre os agentes etiológicos da doença na referida área. Nesse sentido, o presente trabalho propôs-se a caracterizar por eletroforese de isoenzimas as amostras de Leishmania isoladas de pacientes procedentes da Mesorregião do baixo Amazonas, verificando a existência da correlação geográfica das espécies encontradas com a sua distribuição regional previamente conhecida, e ainda, verificando a presença de variação intraespecífica. A caracterização das 43 amostras de Leishmania foi feita por eletroforese em gel de amido utilizando sete sistemas enzimáticos (6PGDH, PGM, G6PD, MPI, GPI, ASAT E ALAT), comparando seus perfis eletroforéticos com os perfis das sete cepas-referência das espécies conhecidas da região. As amostras foram testadas previamente por imunofluorescência indireta com o uso de um painel com 23 anticorpos monoclonais (sistema biotina-avidina) apenas como uma triagem. A caracterização isoenzimática das amostras permitiu o seguinte resultado: 11 (25,28%) amostras de L. (V) braziliensis, 20 (46,50%) de L.(V) guyanensis, 2 (4,60%) de L.(L.) amazonensis, 4 (9,30%) de L.(V) shawi e 6 (13,95%) de L.(V) lainsoni. A eletroforese isoenzimática apresentou elevado poder discriminatório para a identificação das amostras estudadas, permitindo concluir que esta técnica representa uma importante ferramenta para a caracterização dos parasitos do gênero Leishmania. Nas cepas de L. (V) braziliensis observou-se pela primeira vez na Mesorregião do Baixo Amazonas a ocorrência de variação intraespecífica revelada pela presença de três serodemas. Nas cepas de L. (V) guyanensis observou-se a presença de duas variantes, uma que apresentou reatividade com o monoclonal B 19 (espécie-específico), porém com variação nas enzimas 6PGDH e PGM, e a Segunda, sem reatividade para este monoclonal e com perfis eletroforéticos semelhantes ao da cepa-referência L. (V) guyanensis especialmente nas enzimas 6PGDH, porém com tribandas, e na PGM, consideradas os melhores marcadores enzimáticos pelo seu elevado poder discriminatório. Dessa forma, descreveu-se pela primeira vez a ocorrência de diferentes espécies de Leishmania dermotrópicas na Mesorregião do Baixo Amazonas, as quais já tem registro na região norte do Brasil, sugerindo a transmissão simpátrica das espécies encontradas na referida área estudada.

Background: Infectious diarrhea can be caused by bacteria, viruses, or protozoan organisms, or a combination of these. The identification of co-infections in dogs is important to determine the prognosis and to plan strategies for their treatment and prophylaxis. Although many pathogens have been individually detected with real-time polymerase chain reaction (PCR), a comprehensive panel of agents that cause diarrhea in privately owned dogs has not yet been established. The objective of this study was to use a real-time PCR diarrhea panel to survey the frequencies of pathogens and co-infections in owned dogs attended in a veterinary hospital with and without diarrhea, as well the frequency in different countries. Feces samples were tested for canine distemper virus, canine coronavirus, canine parvovirus type 2 (CPV-2), Clostridium perfringens alpha toxin (CPA), Cryptosporidium spp., Giardia spp., and Salmonella spp. using molecular techniques.Results: In total, 104 diarrheic and 43 control dogs that were presented consecutively at a major private veterinary hospital were included in the study. Overall, 71/104 (68.3%) dogs with diarrhea were positive for at least one pathogen: a single infection in 39/71 dogs (54.9%) and co-infections in 32/71 dogs (45.1%), including 21/32 dogs (65.6%) with dual, 5/32 (15.6%) with triple, and 6/32 (18.8%) with quadruple infections. In the control group, 13/43 (30.2%) dogs were positive, all with single infections only. The most prevalent pathogens in the diarrheic dogs were CPA (40/104 dogs, 38.5%), CPV-2 (36/104 dogs, 34.6%), and Giardia spp. (14/104 dogs, 13.5%). CPV-2 was the most prevalent pathogen in the dual co-infections, associated with CPA, Cryptosporidium spp., or Giardia spp. No statistical difference (P = 0.8374) was observed in the duration of diarrhea or the number of deaths (P = 0.5722) in the presence or absence of single or co-infections.Conclusions: Diarrheic dogs showed a higher prevalence of pathogen infections than the controls. Whereas the healthy dogs had only single infections, about half the diarrheic dogs had co-infections. Therefore, multiple pathogens should be investigated in dogs presenting with diarrhea. The effects of multiple pathogens on the disease outcomes remain unclear because the rate of death and the duration of diarrhea did not seem to be affected by these factors.

Porcine parvovirus (PPV) is associated with reproductive failure and it has been found worldwide, including Brazil. Many diagnostic procedures are used for its detection, for example, immunofluorescence, HA, HI and PCR and this is an important technique because it is very specific and sensitive. In this work, the presence of PPV in fetuses from swine farms with reproductive problems was detected by PCR. All of 170 samples from aborted fetuses, mummies or stillborns were sampled by PCR with primers designed to VP2 region of PPV and c-myc (endogenous control). Only 142 samples (83,53%) were positive for c-myc and among them six samples (4,22%) were positive for PPV which were tested in HA. In this test, erythrocytes suspension 1% was used and three samples (50%) agglutinated but they presented low titer (4). For this work, porcine parvovirus was detected in the samples analyzed by PCR and HA. It is important to emphasize the use of endogenous control when material with elevated degree of autolysis is examined

Several alloys present the shape memory effect and among them, the equiatomic NiTi alloy, nitinol, is the most important one. It is usually used in several engineering applications and also in biomedical devices, in the fabrication of orthodontic wire, stents and Judet staples. Although a considerable amount of these biomedical devices is utilized in Brazil and a fraction of it is already made here, all nitinol used is bought abroad. Thus, it is important to develop the necessary know-how to fabricate NiTi wire and sheet. It would mean less importation with job creation and wealth generation for the country. In this work nitinol was obtained powder metallurgy from elemental powders of Ti and Ni using uniaxial compression and uniaxial compression followed by isostatic compression. The final densities achieved were determined by the Archimedes method. The precipitation of intermetallic secondary phases was studied and the samples were characterized by metallographic analysis, optical microscopy and X-ray diffraction. Results indicated that 50 hours sintering route showed a low amount of intermetallics, and no trace of unreacted powder. XRD and metallography at room temperature indicated B19’ as the predominant phase, which corresponds to martensite. Although density results showed little dispersion, the most dense sample was compacted under uniaxial compression and presented 4.8 g/cm3, corresponding to 20.84% porosity. Density variation was considered normal to the measurement process and independent of the compaction mode

Metagenomics was used to characterize viral genomes in clinical specimens of horses with various organ-specific diseases of unknown aetiology. A novel parvovirus as well as a previously described hepacivirus closely related to human hepatitis C virus and equid herpesvirus 2 were identified in the cerebrospinal fluid of horses with neurological signs. Four co-infecting picobirnaviruses, including an unusual genome with fused RNA segments, and a divergent anellovirus were found in the plasma of two febrile horses. A novel cyclovirus genome was characterized from the nasal secretion of another febrile animal. Lastly, a small circular DNA genome with a Rep gene, from a virus we called kirkovirus, was identified in the liver and spleen of a horse with fatal idiopathic hepatopathy. This study expands the number of viruses found in horses, and characterizes their genomes to assist future epidemiological studies of their transmission and potential association with various equine diseases.

Brucellosis remains as a public health concern worldwide. In domestic animals, the disease is characterized by reproductive disorders in male and female. Besides extensive use of serological tests and recent development of molecular biology techniques, microbiological culture of Brucella species is yet considered a “gold standard” method for diagnosis. Here, semen of 335 bovine bulls was subjected simultaneously to microbiological culture in Brucella agar, Farrell media, and CITA media to evaluate comparatively the best selective media for isolation of Brucella sp. Among all 335 samples, B. abortus B19 strain was isolated from semen of five (1.49%) bulls using the three selective media. However, Farrell media was considered the best selective media for microbiological diagnosis, because of allowed isolation of B. abortus B19 strain from bull semen without bacterial commensal or fungal contamination of plates.
