Hypercoiling polymers can be suited for application to living systems because they are similar in structure to the protein-based lipid assemblies found at fluid interfaces within the body. This leads to a range of exciting possibilities, not only in membrane transport applications but also in biosensors, drug delivery and mechanistic studies of biological membrane function. This study is focused in the study of the stability and suitability of nanostructures made of a hypercoiling polymer for drug delivery applications. The polymer poly (styrene-maleic acid) (PSMA) was combined with the phospholipid dimyristoylphosphatidylcholine (DMPC) to form amphiphilic nanostructures. The stability and suitability of these polymer-phospholipid nanocarriers for hydrophobic and hydrophilic molecules load and release was analyzed by several techniques. It was found that several of the studied molecules had a substantial effect on the surface charge and stability of the nanocarrier. It was also demonstrated that two types of nanocarriers, chemically modified and unmodified, were able to control the release of the molecules, especially in the case of hydrophobic compounds. In addition, as the hydrophobicity increased the release slowed down. These clear nanocarriers have the potential to behave very favorably at interfaces such as the tear lipid film were transparency is a requirement, giving a new way of controlled drug release in the eye.

The spectral distribution of solar radiation was studied under different sky conditions during a 15- month period in Miami, Florida (USA), and over a latitudinal gradient at solar maximum. Spectroradiometric scans were characterized for total irradiance (300- 3000 nm) and the relative energetic and photon contributions of the following wavelength regions: UV-B (300-320nm); UV-A (320-400nm); B (400-500rim); PAR (400-700 nm); R (600-700 nm); and FR (728- 732 rim). Notable results include: (i) significantly higher UV-A energy fluxes than currently in use for laboratory experiments involving the biological effects of this bandwidth (values ranged from 33.6 to 55.4 W/m 2 in Miami over the year); (ii) marked diurnal shifts in B:R and R:FR, with elevated R:FR values in early morning: (iii) a strong correlation between R: FR and atmospheric water content; and (iv) unusually high PAR values under direct sunlight with cloudy skies (2484 ~tmot/2 per s).

The extracellular matrix (ECM) provides a framework for cells and gives skin its tensile strength and elasticity. Loss of its integrity necessitates the clearing of damaged components and the deposition of firstly a provisional matrix and later remodelling of the ECM to support a functionally intact tissue. Matrix metalloproteinases (MMPs) are an important family of enzymes that function in the breakdown of the ECM and modulate the function of many biologically active molecules housed in the ECM. Through their enzymatic actions MMPs play a role in fundamental processes such as immune cell infiltration and ECM remodelling during wound repair. Their tight control is necessary for timely wound healing and excessive MMP activity participates in the development and persistence of chronic wounds, while reduced activity contributes to fibrosis. A number of inhibitors have been designed to target this activity and improve wound healing with limited success. Novel strategies are currently being investigated to improve wound healing by targeting MMP modulating molecules.

A comprehensive analysis of the crystal packing and the energetic features of a series of four biologically active molecules belonging to the family of substituted 4-(benzylideneamino)-3-(4-fluoro-3-phenoxyphenyl)-1H-1,2,4-triazole-5-(4 H)-thione derivatives have been performed based on the molecular conformation and the supramolecular packing. This involves the formation of a short centrosymmetric R-2(2)(8) NH...S supramolecular synthon in the solid state, including the presence of CH...S, CH...O, CH...N, CH...F, CH...Cl, CF...FC, CCl...ClC, and CH...pi intermolecular interactions along with pp stacking to evaluate the role of noncovalent interactions in the crystal. The presence of such synthons has a substantial contribution toward the interaction energy (-18 to -20 kcal/mol) as obtained from the PIXEL calculation, wherein the Coulombic and polarization contribution are more significant than the dispersion contribution. The geometrical characteristics of such synthons favor short distance, and the population of related molecules having these geometries is rare as has been obtained from the Cambridge Structural Database (CSD). Furthermore, their interaction energies have been compared with those present in our molecules in the solid state. The topological characteristics of the NH...S supramolecular synthon, in addition to related weak interactions, CH...N, CH...Cl, CF...FC, and CCl...ClC, have been estimated using the quantum theory of atoms in molecules (QTAIM). In addition, an analysis of the Hirshfeld surface and associated fingerprint plots of these four molecules also have provided a platform for the evaluation of the contribution of different atom...atom contacts, which contribute toward the packing of the molecules in solids.

Heparin has been used as an anticoagulant drug for more than 70 years. The global distribution of contaminated heparin in 2007, which resulted in adverse clinical effects and over 100 deaths, emphasizes the necessity for safer alternatives to animal-sourced heparin. The structural complexity and heterogeneity of animal-sourced heparin not only impedes safe access to these biologically active molecules, but also hinders investigations on the significance of structural constituents at a molecular level. Efficient methods for preparing new synthetic heparins with targeted biological activity are necessary not only to ensure clinical safety, but to optimize derivative design to minimize potential side effects. Low molecular weight heparins have become a reliable alternative to heparin, due to their predictable dosages, long half-lives, and reduced side effects. However, heparin oligosaccharide synthesis is a challenging endeavor due to the necessity for complex protecting group manipulation and stereoselective glycosidic linkage chemistry, which often result in lengthy synthetic routes and low yields. Recently, chemoenzymatic syntheses have produced targeted ultralow molecular weight heparins with high-efficiency, but continue to be restricted by the substrate specificities of enzymes.

To address the need for access to homogeneous, complex glycosaminoglycan structures, we have synthesized novel heparan sulfate glycopolymers with well-defined carbohydrate structures and tunable chain length through ring-opening metathesis polymerization chemistry. These polymers recapitulate the key features of anticoagulant heparan sulfate by displaying the sulfation pattern responsible for heparin’s anticoagulant activity. The use of polymerization chemistry greatly simplifies the synthesis of complex glycosaminoglycan structures, providing a facile method to generate homogeneous macromolecules with tunable biological and chemical properties. Through the use of in vitro chromogenic substrate assays and ex vivo clotting assays, we found that the HS glycopolymers exhibited anticoagulant activity in a sulfation pattern and length-dependent manner. Compared to heparin standards, our short polymers did not display any activity. However, our longer polymers were able to incorporate in vitro and ex vivo characteristics of both low-molecular-weight heparin derivatives and heparin, displaying hybrid anticoagulant properties. These studies emphasize the significance of sulfation pattern specificity in specific carbohydrate-protein interactions, and demonstrate the effectiveness of multivalent molecules in recapitulating the activity of natural polysaccharides.

A indução da síntese de moléculas biologicamente ativas é conhecida como uma estratégia de defesa do organismo vegetal às condições adversas, onde plantas medicinais e/ou aromáticas são foco de pesquisa na terapêutica médica, na indústria farmacêutica, de cosméticos e de alimentos. A possibilidade de que os hidrocarbonetos de petróleo presentes no solo possam agir como agentes elicitores do metabolismo vegetal tem despertado interesse científico. O presente trabalho teve como objetivo avaliar o desenvolvimento in vivo e in vitro de Ruta graveolens L. (Rutaceae), considerando as respostas morfológicas e metabólicas na presença de petróleo e derivados. Além disso, verificar o potencial de desenvolvimento dessa espécie em áreas contaminadas, com base na hipótese de que tais substâncias podem influenciar na produção de substâncias de interesse, sem que haja redução significativa da biomassa vegetal. Foram realizados experimentos de (i) germinação in vivo em solo contaminado e parcialmente biorremediado, assim como solo com contaminação recente com óleo cru (0,8 % e 2,8 %) e diesel comercial S50 (0 % a 6,0 %), sendo que neste último foi avaliado o desenvolvimento e a produção de biomassa das plântulas; (ii) germinação in vitro em meio MS contaminado com fenantreno (1,0; 5,0 e 10,0 mg/L de meio) ou benzo[a]pireno (0,001; 0,010 e 0,100 mg/L de meio), sendo avaliado o desenvolvimento e a produção de biomassa; e (iii) resposta fisiológica de plantas oriundas do estoque in vitro à exposição de fenantreno ou benzo[a]pireno. R. graveolens teve redução significativa da germinação em solos contaminados, principalmente os solos parcialmente biorremediados pelo do processo de bioestímulo; não teve redução significativa da germinação in vivo nos solos contaminados com óleo cru, biorremediado e contaminado com diesel, porém o desenvolvimento pósgerminação foi afetado. Na exposição in vitro aos hidrocarbonetos, não houve alteração significativa na germinação nem na capacidade de desenvolvimento e produção de biomassa nas concentrações testadas de até 10 mg/L de fenantreno e até 0,1 mg/L de benzo[a]pireno. Em relação à propagação in vitro, foi possível a multiplicação de plantas em meio de cultura sem suplementação hormonal, na presença dos contaminantes. A presença de fenantreno e benzo[a]pireno in vitro não promoveu mudanças significativas nos níveis de compostos fenólicos e flavonoides totais produzidos, não havendo também alteração no perfil fitoquímico da espécie, quanto à presença de rutina. Pelo exposto, conclui-se que a espécie tem potencial para uso em biomonitoramento de solos contaminados , considerando a sua capacidade de produzir metabólitos e tolerar a presença dos hidrocarbonetos selecionados, sendo indicada ao cultivo em áreas contaminadas nos níveis investigados

Amphibian defensive skin secretions are complex, species-specific cocktails of biologically active molecules, including many uncharacterized peptides. The study of such secretions for novel peptide discovery is time-limited, as amphibians are in rapid global decline. While secretion proteome analysis is non-lethal, transcriptome analysis has until now required killing of specimens prior to skin dissection for cDNA library construction. Here we present the discovery that polyadenylated mRNAs encoding dermal granular gland peptides are present in defensive skin secretions, stabilized by endogenous nucleic acid-binding amphipathic peptides. Thus parallel secretory proteome and transcriptome analyses can be performed without killing the specimen in this model amphibian system--a finding that has important implications in conservation of biodiversity within this threatened vertebrate taxon and whose mechanistics may have broader implications in biomolecular science.

Compostos do tipo pirazol e 1,2,3-triazol encontram-se presentes em inúmeras moléculas biologicamente ativas. Muitos fármacos atualmente comercializados ou em fase de estudos clínicos contêm na sua estrutura base núcleos de pirazol ou 1,2,3-triazol. Por isso, estes compostos têm sido alvo de intensa pesquisa na procura de novas moléculas com potenciais aplicações medicinais e agroquímicas. Nesta dissertação são descritas novas vias de síntese de novos compostos do tipo pirazol e 1,2,3-triazol. No primeiro capítulo apresenta-se uma breve revisão bibliográfica sobre a atividade biológica, ocorrência natural e métodos de síntese de pirazóis e seus derivados. O segundo capítulo foca-se na síntese de (E)-2-estiril-3-halo-4H-cromen-4-onas e sua transformação em 3(5)-aril-5(3)-[2-(2-hidroxifenil)-2-oxoetil-1H-pirazóis. Em primeiro lugar faz-se uma revisão bibliográfica sobre as (E)-2-estiril-4H-cromen-4-onas e a sua semelhança estrutural com as flavonas, a sua importância e ocorrência natural e métodos de síntese. São ainda abordadas as metodologias mais utilizadas para a síntese de derivados halogenados de (E)-2-estiril-4H-cromen-4-onas. Seguidamente são apresentados e discutidos os resultados da síntese de (E)-3-bromo-2-estiril-4H-cromen-4-onas através da reação de 5-aril-3-hidroxi-1-(2-hidroxifenil)penta-2,4-dien-1-onas com NBS, sob irradiação com micro-ondas, tendo sido estabelecida uma nova metodologia mais eficiente, rápida e regiosseletiva para a síntese de (E)-3-bromo-2-estiril-4H-cromen-4-onas, na ausência de solvente. São igualmente apresentados os resultados da síntese regiosseletiva de (E)-2-estiril-3-iodo-4H-cromen-4-onas através da reação de 5-aril-3-hidroxi-1-(2-hidroxifenil)penta-2,4-dien-1-onas com NIS e TFA/TFAA/NaOAc. Em ambos os métodos de halogenação desenvolvidos, obtiveram-se como produtos secundários as (E)-2-estiril-4H-cromen-4-onas correspondentes. Seguidamente é apresentado o estudo da reação de (E)-2-estiril-3-halo-4H-cromen-4-onas com hidrato de hidrazina. Ao contrário do esperado, obtiveram-se os 3(5)-aril-5(3)-[2-(2-hidroxifenil)-2-oxoetil-1H-pirazóis através de uma reação de adição conjugada 1,6-, de hidrazina à posição C- da cromona com consequente abertura do anel, seguida de uma adição conjugada 1,4- intramolecular. Estes resultados demonstraram que esta reação segue um mecanismo diferente daquele que está reportado na literatura para a reação de (E)-2-estiril-4H-cromen-4-onas não halogenadas em C-3 com hidrato de hidrazina. No terceiro capítulo apresenta-se uma breve revisão bibliográfica sobre as propriedades, aplicações e metodologias de síntese de 1,2,3-triazóis, dando mais relevância às reações de cicloadição 1,3-dipolar e de “click-chemistry”. Seguidamente descrevem-se os resultados obtidos na reação de (E)-5(3)-estiril-3(5)-(2-hidroxifenil)-1H-pirazóis com a azida de sódio para obtenção de díades pirazol-1,2,3-triazol. No entanto esta reação deu origem a novos 5(3)-(2-aril-2-azidoetil)-3(5)-(2-hidroxifenil)-1H-pirazóis e não às díades pirazol-1,2,3-triazol pretendidas. Como o resultado não foi o esperado, desenvolveu-se outra metodologia de síntese, que envolve, num primeiro, a reação de (E)-2-estiril-4H-cromen-4-onas com azida de sódio, dando origem a 5(4)-aril-4(5)-(cromon-2-il)-1H-1,2,3-triazóis. No passo seguinte, efetuou-se a reação destes compostos com hidrato de hidrazina tendo ocorrido a formação das diades 5(4)-aril-4(5)-[3(5)-(2-hidroxifenil)-1H-pirazol-5(3)-il]-1H-1,2,3-triazol pretendidas. No quarto capítulo, estudou-se a reatividade de (E)-5(3)-estiril-3(5)-(2-hidroxifenil)-1H-pirazóis em reações de iodação com vista à obtenção de 4-iodo-1H-pirazóis. Apresenta-se uma breve revisão bibliográfica sobre os diferentes métodos descritos na literatura para a iodação de compostos heterocíclicos aromáticos, nomeadamente para a obtenção de 4-iodo-1H-pirazóis. Dos vários sistemas de iodação testados, o sistema oxidativo I2/CAN foi o que deu melhores resultados na iodação dos (E)-5(3)-estiril-3(5)-(2-hidroxifenil)-1H-pirazóis. Este método permitiu iodar a posição C-4 do núcleo de pirazol apenas para os derivados que possuem o grupo nitro ou o átomo de cloro no anel do grupo estirilo, obtendo-se o 3(5)-(2-hidroxifenil)-4-iodo-5(3)-(4-nitrofenil)vinil-1H-pirazol e o 5(3)-(4-clorofenil)vinil)-3(5)-(2-hidroxi-5-iodofenil)-4-iodo-1H-pirazol; no entanto, para os restantes derivados, verificou-se apenas a iodação nas posições ativadas do anel fenólico. Todos os novos compostos sintetizados foram caraterizados estruturalmente recorrendo a estudos de espetroscopia de ressonância magnética nuclear (RMN) mono e bidimensionais. Sempre que possível, para uma caraterização estrutural mais completa, foram efetuados espetros de massa (EM) e análises elementares ou espetros de massa de alta resolução (EMAR) para todos os novos compostos sintetizados. Finalmente são apresentadas as conclusões gerais deste trabalho e perspetivas futuras.

Le développement ainsi que l’amélioration des différentes techniques de purification sont des défis importants pour la chimie d’aujourd’hui. Certaines des méthodes actuelles, tel que le greffage d’un réactif sur un support solide permettant d’accéder à un produit pur par simple filtration du milieu, comportent toutefois certains inconvénients. En effet, les propriétés de solubilité de ces polymères rendent la mise en œuvre des réactions plus difficiles. C’est dans ce contexte que le groupe du Pr. Charette a rapporté l’utilisation de réactifs liés à un sel de tétraarylphosphonium (TAP). Ces sels peuvent être solubilisés dans un solvant tel que le dichlorométhane et aisément retirés du milieu réactionnel par précipitation à l’aide d’éther diéthylique (Chapitre 1). L’un des objectifs de cette thèse a donc été lié à la découverte de deux méthodes complémentaires qui, jusqu’à présent, sont considérées comme des méthodes de choix pour la synthèse des sels de TAP fonctionnalisés (Chapitre 2). L’une d’entre elles est utilisée par Soluphase inc., une entreprise qui commercialise ces sels de TAP. L’efficacité des sels en tant que support dans la synthèse de petites molécules a été démontrée lors de la synthèse d’un produit naturel, la (–)-coniine (Chapitre 3). L’isolement des intermédiaires synthétiques instables par simple précipitation à l’aide d’un support de TAP a permis de rendre cette synthèse plus efficace que celle déjà connue. Dans le deuxième volet de cette thèse, plusieurs problèmes reliés à la synthèse de dérivés cyclopropaniques 1,1-disubstitués ont été étudiés. Ces derniers font partie intégrale de plusieurs produits naturels et de médicaments. Cependant, leur formation par une réaction de cyclopropanation d’alcènes utilisant des réactifs diazoïques possédant deux groupements de type accepteur n’est pas tâche facile (Chapitre 4). En effet, cette réaction souffre d’un faible contrôle diastéréosélectif. Par le fait même, très peu de méthodologies de synthèse ont rapporté l’utilisation de ce type de réactifs diazoïques dans des réactions de cyclopropanation stéréosélectives. L’étude du mécanisme de la réaction de cyclopropanation catalysée au Rh(II) a proposé des indices favorisant un modèle ayant des précédents dans la littérature (Chapitre 5). Ces études nous ont mené à la découverte de la «capacité trans-directrice» du groupement amide lors des réactions de cyclopropanation d’oléfines. Nous avons donc utilisé cette propriété afin de résoudre plusieurs problèmes rencontrés dans la littérature. Nous avons montré qu’elle permet l’accès à des dérivés cyclopropaniques possédant deux groupements carboxyliques géminaux avec des sélectivités élevées (Chapitre 6). Ces produits étaient accessibles que par des séquences synthétiques nécessitant plusieurs étapes. De plus, nous avons démontrés que ces nouveaux dérivés cyclopropaniques sont des outils synthétiques fort utiles dans la synthèse de produits naturels d’intérêt biologique. Cette formidable «capacité trans-directrice» du groupement amide nous a permi de résoudre le problème de la synthèse asymétrique de dérivés carboxyliques α-cyano cyclopropaniques (Chapitre 7). De plus, ce projet nous a menées à la découverte de l’effet de divers additif achiraux permettant d’augmenter la sélectivité dans certaines réactions. Cette réaction possède une vaste étendue et l’utilité de ces nouveaux dérivés cyclopropaniques a été démontrée par plusieurs transformations de groupements fonctionnels.

Expression of biologically active molecules as fusion proteins with antibody Fc can substantially extend the plasma half-life of the active agent but may also influence function. We have previously generated a number of fusion proteins comprising a complement regulator coupled to Fc and shown that the hybrid molecule has a long plasma half-life and retains biological activity. However, several of the fusion proteins generated had substantially reduced biological activity when compared with the native regulator or regulator released from the Fc following papain cleavage. We have taken advantage of this finding to engineer a prodrug with low complement regulatory activity that is cleaved at sites of inflammation to release active regulator. Two model prodrugs, comprising, respectively, the four short consensus repeats of human decay accelerating factor (CD55) linked to IgG4 Fc and the three NH2-terminal short consensus repeats of human decay accelerating factor linked to IgG2 Fc have been developed. In each, specific cleavage sites for matrix metalloproteinases and/or aggrecanases have been incorporated between the complement regulator and the Fc. These prodrugs have markedly decreased complement inhibitory activity when compared with the parent regulator in vitro. Exposure of the prodrugs to the relevant enzymes, either purified, or in supernatants of cytokine-stimulated chondrocytes or in synovial fluid, efficiently cleaved the prodrug, releasing active regulator. Such agents, having negligible systemic effects but active at sites of inflammation, represent a paradigm for the next generation of anti-C therapeutics.

In this work we realized and experimental and theoretical study of the N-alkylation of nitroimidazoles. The N-alkyl-2-methyl-nitroimidazoles correspond to biologically active molecules, obtained by reaction of 2-methyl-5-nitroimidazole and different alkyl halides. This reaction showed the formation of a mixture of isomeric products in different proportions, denominated like N-alkyl-2-methyl-4-nitroimidazole and N-alkyl-2-methyl-5-nitroimidazole, respectively. The reaction suggestes the formation of a tautomeric equilibrium, which generates two nucleophilic sites susceptible to electrophilic attack by the alkyl halide. The local nucleophilic reactivity of the nitroimidazole nng is determined using local reactivity indices such as the Fukui function and the electrostatic potential, besides the electronic localization function (ELF). The Fukui function was integrated for each atom using partition schemes based on analysis of Mulliken charges and natural bond orbital (NBO). Finally the reaction profiles were assessed. The results show a minor difference in the local reactivity. Nevertheless a significant difference in energy barriers is observed explaining the formation of an isomeric product over another. These results agree quite well with the experimental data.

Silk contains a fibre forming protein, fibroin, which is biocompatible, particularly after removing the potentially immunogenic non-fibroin proteins. Silk can be engineered into a wide range of materials with diverse morphologies. Moreover, it is possible to regenerate fibroin with a desired amount of crystallinity, so that the biodegradation of silk materials can be controlled. These advantages have sparked new interest in the use of silk fibroin for biomedical applications, including tissue engineering scaffolds and carriers for sustained release of biologically active molecules. This article summarizes the current research related to the formation of silk materials with different morphologies, their biocompatibility, and examples of their biomedical applications. Recent work on the preparation of silk particles by mechanical milling and their applications in silk composite scaffolds is also discussed.
