The BTEX (benzene, toluene, ethylbenzene and xylene) mixture is an environmental pollutant that has a high potential to contaminate water resources, especially groundwater. The bioremediation process by microorganisms has often been used as a tool for removing BTEX from contaminated sites. The application of biological assays is useful in evaluating the efficiency of bioremediation processes, besides identifying the toxicity of the original contaminants. It also allows identifying the effects of possible metabolites formed during the biodegradation process on test organisms. In this study, we evaluated the genotoxic and mutagenic potential of five different BTEX concentrations in rat hepatoma tissue culture (HTC) cells, using comet and micronucleus assays, before and after biodegradation. A mutagenic effect was observed for the highest concentration tested and for its respective non-biodegraded concentration. Genotoxicity was significant for all non-biodegraded concentrations and not significant for the biodegraded ones. According to our results, we can state that BTEX is mutagenic at concentrations close to its water solubility, and genotoxic even at lower concentrations, differing from some described results reported for the mixture components, when tested individually. Our results suggest a synergistic effect for the mixture and that the biodegradation process is a safe and efficient methodology to be applied at BTEX-contaminated sites. © 2012 Elsevier Ltd.

Abamectin (ABA), which belongs to the family of avermectins, is used as a parasiticide; however, ABA poisoning can impair liver function. In a previous study using isolated rat liver mitochondria, we observed that ABA inhibited the activity of adenine nucleotide translocator and FoF1-ATPase. The aim of this study was to characterize the mechanism of ABA toxicity in isolated rat hepatocytes and to evaluate whether this effect is dependent on its metabolism. The toxicity of ABA was assessed by monitoring oxygen consumption and mitochondrial membrane potential, intracellular ATP concentration, cell viability, intracellular Ca2+ homeostasis, release of cytochrome c, caspase 3 activity and necrotic cell death. ABA reduces cellular respiration in cells energized with glutamate and malate or succinate. The hepatocytes that were previously incubated with proadifen, a cytochrome P450 inhibitor, are more sensitive to the compound as observed by a rapid decrease in the mitochondrial membrane potential accompanied by reductions in ATP concentration and cell viability and a disruption of intracellular Ca2+ homeostasis followed by necrosis. Our results indicate that ABA biotransformation reduces its toxicity, and its toxic action is related to the inhibition of mitochondrial activity, which leads to decreased synthesis of ATP followed by cell death. © 2012 Elsevier Ltd.

Ethnopharmacological relevance Dragon's blood is a dark-red sap produced by species from the genus Croton (Euphorbiaceae), which has been used as a famous traditional medicine since ancient times in many countries, with scarce data about its safe use in humans. In this research, we studied genotoxicity and clastogenicity of Croton palanostigma sap using the comet assay and micronucleus test in cells of mice submitted to acute treatment. Material and methods HPLC analysis was performed to identify the main components of the sap. The sap was administered by oral gavage at doses of 300 mg/kg, 1000 mg/kg and 2000 mg/kg. For the analysis, the comet assay was performed on the leukocytes and liver cells collected 24 h after treatment, and the micronucleus test (MN) on bone marrow cells. Cytotoxicity was assessed by scoring 200 consecutive polychromatic (PCE) and normochromatic (NCE) erythrocytes (PCE/NCE ratio). Results and conclusion The alkaloid taspine was the main compound indentified in the crude sap of Croton palanostigma. The results of the genotoxicity assessment show that all sap doses tested produced genotoxic effects in leukocytes and liver cells and also produced clastogenic/aneugenic effects in bone marrow cells of mice at the two higher doses tested. The PCE/NCE ratio indicated no cytotoxicity. The data obtained suggest caution in the use of Croton palanostigma sap by humans considering its risk of carcinogenesis. © 2013 Elsevier B.V. Ethnopharmacological relevance Propolis is a bee product with numerous biological and pharmacological properties, such as immunomodulatory and anti-inflammatory activities. It has been used in folk medicine as a healthy drink and in food to improve health and prevent inflammatory diseases. However, little is known about its mechanism of action. Thus, the goal of this study was to verify the antioxidant activity and to explore the anti-inflammatory properties of propolis by addressing its intracellular mechanism of action. Caffeic acid was investigated as a possible compound responsible for propolis action. Materials and methods The antioxidant properties of propolis and caffeic acid were evaluated by using the 2,2-Diphenyl-1-picrylhydrazyl free radical (DPPH) scavenging method. To analyze the anti-inflammatory activity, Raw 264.7 macrophages were treated with different concentrations of propolis or caffeic acid, and nitric oxide (NO) production, a strong pro-inflammatory mediator, was evaluated by the Griess reaction. The concentrations of propolis and caffeic acid that inhibited NO production were evaluated on intracellular signaling pathways triggered during inflammation, namely p38 mitogen-activated protein kinase (MAPK), c-jun NH2-terminal kinase (JNK1/2), the transcription nuclear factor (NF)-κB and extracellular signal-regulated kinase (ERK1/2), through Western blot using specific antibodies. A possible effect of propolis on the cytotoxicity of hepatocytes was also evaluated, since this product can be used in human diets. Results Caffeic acid showed a higher antioxidant activity than propolis extract. Propolis and caffeic acid inhibited NO production in macrophages, at concentrations without cytotoxicity. Furthermore, both propolis and caffeic acid suppressed LPS-induced signaling pathways, namely p38 MAPK, JNK1/2 and NF-κB. ERK1/2 was not affected by propolis extract and caffeic acid. In addition, propolis and caffeic acid did not induce hepatotoxicity at concentrations with strong anti-inflammatory potential. Conclusions Propolis exerted an antioxidant and anti-inflammatory action and caffeic acid may be involved in its inhibitory effects on NO production and intracellular signaling cascades, suggesting its use as a natural source of safe anti-inflammatory drugs. © 2013 Elsevier B.V.

Para avaliar a importância da ultra-sonografia como método de diagnóstico por imagem na detecção de alterações ecográficas hepático-portais na hepatite por vírus C realizou-se esta investigação cujos objetivos foram: descrever os achados de marcadores ecográficos de alteração morfológica do fígado e do sistema porta; determinar a freqüência dessas alterações e investigar a presença de associação entre as alterações ultra-sonográficas hepáticas e portais e a infecção pelo vírus da hepatite C (VHC), em indivíduos portadores assintomáticos deste vírus, os quais foram selecionados entre pré-doadores de sangue da Fundação Hemopa, hemocentro de referência oficial no Estado do Pará, Brasil, no período de outubro a dezembro de 2003. O estudo foi realizado comparando-se os achados de um grupo com 30 participantes soropositivos para o VHC (grupo central) e de um grupo controle, composto de 38 participantes soronegativos. Ambos os grupos da pesquisa incluíram indivíduos assintomáticos, de ambos os sexos e com idade adequada para doação de sangue. A triagem para doação foi realizada no hemocentro, utilizando seus procedimentos de rotina. Procedeu-se pareamento dos participantes da pesquisa por sexo e idade. A amostra analisada evidenciou predomínio da faixa etária de 18 a 28 anos, correspondendo a 53% dos participantes do grupo central e 50% dos participantes do grupo controle. Hepatomegalia e hiperecogenicidade do parênquima, achados inespecíficos e encontrados comumente na esteatose hepática, foram mais freqüentes no grupo central, porém sem diferença estatística significante. Heterogenicidade textural, focos hipoecogênicos, alterações de visibilidade dos ramos venosos intra-hepáticos e esplenomegalia foram outros parâmetros mais freqüentes no grupo de infectados, também sem diferença estatística significante. Os participantes do grupo central apresentaram alterações com diferença estatística significante no parâmetro superfície hepática e na classificação por pontuação pelos escores de Lin, considerando-se os parâmetros superfície, parênquima, veias internas e índice esplênico. A totalização de pontos classificou os participantes nos padrões fino -80% no grupo central e 100% no grupo controle - heterogêneo -13,3% no grupo central e 0% no grupo controle - e nodular-6,7% no grupo central e 0% no grupo controle, demonstrando diferença estatística significante. Não foram observados em participantes de ambos os grupos achados de nódulos, massas, ascite, aumento de calibre das veias porta e esplênica e circulação colateral, podendo-se aventar que o predomínio da faixa etária mais jovem na população de pré-doadores pode ter minimizado os achados dos marcadores ecográficos, haja vista a tendência evolutiva lenta e silenciosa da infecção pelo VHC, com expressão tardia das alterações morfológicas de hepatite crônica, cirrose e carcinoma hepatocelular.

Several epidemiological and experimental studies has been reported that lutein (LT) presents antioxidant properties. Aim of the present study was to investigate the protective effects of LT against oxidative stress and DNA damage induced by cisplatin (cDDP) in a human derived liver cell line (HepG2). Cell viability and DNA-damage was monitored by MU and comet assays. Moreover, different biochemical parameters related to redox status (glutathione, cytochrome-c and intracellular ROS) were also evaluated. A clear DNA-damage was seen with cDDP (1.0 mu M) treatment. In combination with the carotenoid, reduction of DNA damage was observed after pre- and simultaneous treatment of the cells, but not when the carotenoid was added to the cells after the exposure to cDDP. Exposure of the cells to cDDP also caused significant changes of all biochemical parameters and in co-treatment of the cells with LT, the carotenoid reverted these alterations. The results indicate that cDDP induces pronounced oxidative stress in HepG2 cells that is related to DNA damage and that the supplementation with the antioxidant LT may protect these adverse effects caused by the exposure of the cells to platinum compound, which can be a good predict for chemoprevention. Der Asialoglykoprotein-Rezeptor (ASGPR) vermittelt als integraler Bestandteil der Leberzellmembran die Endozytose von zirkulierenden Asialoglykoproteinen. Ziele dieser Arbeit waren proteinchemische Untersuchungen von funktionellem ASGPR aus humaner Leber aufgrund einer verbesserten Präparationsmethode und die rekombinante Darstellung der beiden Untereinheiten H1 und H2. In der denaturierenden SDS-PAGE erschienen H1 und H2 überwiegend als Monomere bei 46 und 50kD; nach Deglykosylierung ergaben sich Banden bei 34 und 32kD, wonach der Glykosidanteil etwa 28% beträgt. In der nicht-denaturierenden Größenausschluß-Chromatographie wurden im nativen ASGPR ausschließlich Trimere und Dimere gefunden. In Gegenwart von 2-Mercaptoethanol konnten funktionell eine aktive von einer nicht-aktiven Fraktion getrennt werden, wobei H2 in der nicht-aktiven Fraktion angereichert war, während sich H1 zu etwa gleichen Teilen in beiden Fraktionen befand. Durch zweidimensionale Auftrennung des deglykosylierten Rezeptors wurden auf Proteinebene vier Isoformen von H1 und zwei von H2 mit unterschiedlichen pI-Werten identifiziert. Der Vergleich von funktionellem ASGPR aus normaler Leber und den hepatischen Tumorzellinien HepG2 und Huh7 in der SDS-PAGE brachte Größenunterschiede von etwa sechs und vier Kilodalton hervor. Bei H1 konnte dies auf einen höheren Glykosylierungsgrad zurückgeführt werden, während H2 auch nach Behandlung mit N-GlykosidaseF ein größeres Molekulargewicht aufwies. Ein Antikörper gegen das Insertionspeptid im cytoplasmatischen Bereich einer Splice-Variante von H2 zeigte eine deutlich erhöhte Expression von H2 mit Insertion in Huh7-Zellen gegenüber natürlichem ASGPR. Da bisherige Kenntnisse über den humanen ASGPR vorwiegend aus kultivierten Hepatomzelllinien stammen, scheinen sie nicht ohne weiteres auf die Situation in normaler Leber übertragbar. Die Präparation von funktionellem H1 aus transfizierten cos7- und 293-Zellen führte zum gleichen Bandenmuster wie beim natürlichen ASGPR. Mit einem Enzymimmunoassay wurde die Eignung von rekombinantem H1 zur Detektion von Antikörpern gegen ASGPR in 177 von 178 Patientenseren gezeigt. Da durch Präinkubation mit rekombinantem Antigen die Reaktivität mit natürlichem Rezeptor inhibiert werden konnte, trägt H1 hauptsächlich die antigenen Stellen des ASGPR.

Gegenstand dieser Arbeit war die Untersuchung, welche Rolle endogen gebildete oxidative DNA-Modifikationen bei der Kanzerogenese spielen. Dazu wurden Cockayne Syndrom B-knockout-Mäuse (Csb-/-), 8-Hydroxyguanin-DNA-Glykosylase-knockout-Mäuse (Ogg1-/-) und Csb-/-/Ogg1-/- Mäuse generiert, die das bakterielle lacI-Gen (Big Blue®) tragen und somit für in vivo Mutationstests eingesetzt werden können. Die Ergebnisse zeigen, dass es in den Lebern der Ogg1-/- Mäuse zu einem 2,1-fachen und in Csb-/-/Ogg1-/- Mäusen zu einem statistisch signifikanten 3,3-fachen Anstieg der Mutationsfrequenz kommt. Die gefundene Erhöhung der Mutationsfrequenz war vor allem auf eine Erhöhung der G:C zu T:A Transversionen zurückzuführen, die typischerweise aus nicht repariertem 8 Hydroxyguanin (8-oxoG) entstehen. Aus mechanistischer Sicht verdeutlichen die Ergebnisse, dass OGG1 das primäre Abwehrsystem gegen oxidative DNA-Modifikationen darstellt und dass das CSB-Protein einen Ausfall von OGG1, selbst in nicht transkribierter DNA, teilweise kompensieren kann. Aus der Korrelation der gefundenen oxidativen DNA-Schäden - bestimmt mittels Alkalischer Elution und der bakteriellen Formamidopyrimidin-DNA-Glykosylase (Fpg-Protein) - mit der Mutationsfrequenz konnte abgeleitet werden, dass bereits weniger als 0,2 Fpg-sensitive DNA-Modifikationen pro 1 Million Basenpaare ausreichen, die spontane Mutationsfrequenz in vivo zu verdoppeln. Zur Untersuchung, welche Rolle die erhöhte Mutationsfrequenz bei der Krebsentstehung spielt, wurden Csb-/-/Ogg1-/- und Wildtyp-Mäuse mit dem Peroxisomenproliferator und spezifischem Leberpromotor WY-14,643 behandelt um spontan initiierte Hepatozyten zur Proliferation anzuregen. Als Endpunkt einer malignen Entartung wurde das Auftreten von Glucose-6-Phosphatase positiven und negativen Läsionen beobachtet. Es zeigte sich, dass Csb-/-/Ogg1-/- Mäuse signifikant mehr enzymveränderte Läsionen in ihren Lebern aufwiesen, als die Wildtyp-Kontrollen. Die Ergebnisse verdeutlichen, dass endogen gebildete oxidative DNA-Modifikationen und daraus resultierende Mutationen grundsätzlich einen erheblichen Anteil zur hohen spontanen Krebsinzidenz in der Bevölkerung leisten könnten.

Numerose evidenze sperimentali hanno dimostrato il contributo delle cellule staminali di derivazione midollare nei processi di rigenerazione epatica dopo danno tissutale. E’ cresciuto pertanto l’interesse sul loro potenziale impiego in pazienti con cirrosi. Questo studio si propone di valutare la fattibilità e la sicurezza della reinfusione intraepatica di cellule staminali midollari autologhe CD133+ in 12 pazienti con insufficienza epatica terminale definita da un punteggio di Model for End Stage of Liver Disease (MELD) compreso tra 17 e 25. L’efficacia in termini di funzionalità epatica rappresenta un obiettivo secondario. Previa mobilizzazione nel sangue periferico mediante somministrazione di granulocyte-colony stimulating factor (G-CSF) alla dose di 7,5 mcg/Kg/b.i.d. e raccolta per leucoaferesi, le cellule CD133+ altamente purificate vengono reinfuse in arteria epatica a partire da 5x104/Kg fino a 1x106/kg. Nei tre giorni successivi si somministra G-CSF per favorire l’espansione e l’attecchimento delle cellule. Durante la mobilizzazione, la reinfusione e nei 12 mesi successivi i pazienti sono sottoposti a periodici controlli clinici, laboratoristici e strumentali e ad attenta valutazione di effetti collaterali. Lo studio è tuttora in corso e ad oggi, 11 pazienti sono stati sottoposti a reinfusione e 4 hanno completato i 12 mesi di follow-up. Il G-CSF è stato ben tollerato e ha consentito di ottenere una buona espansione cellulare. Dopo la reinfusione sono stati documentati un ematoma inguinale e due episodi transitori di encefalopatia portosistemica. Durante il follow-up 4 pazienti sono stati trapiantati e 2 sono morti. Non è stata osservata alcuna modificazione significativa degli indici di funzione epatica. Questi risultati preliminari confermano la possibilità di mobilizzare e reinfondere un numero adeguato di cellule staminali di derivazione midollare in pazienti con malattia epatica in stadio terminale.

Myeloid cell leukemia-1 (Mcl-1) ist ein anti-apoptotisches Mitglied der Bcl-2-Proteinfamilie. Als solches ist es in der Lage, die mitochondriale Aktivierung während der Apoptose zu hemmen. Dadurch schützt es Zellen bei zellulärem Stress (wie z.B. Differenzierung, Proliferation oder Virusinfektion) vor Apoptoseinduktion. Aufgrund dieser Eigenschaft ist es unabkömmlich während der Embryogenese und in verschiedenen hämatopoetischen Zellpopulationen. Des Weiteren ist Mcl-1 als Protoonkogen in verschiedenen humanen Tumorentitäten verstärkt exprimiert und kann so zu einer verminderten Apoptosesensitivität von Tumorzellen beitragen. Auch primäre humane Hepatozyten können nach Mcl-1-Induktion durch Wachstumsfaktorbehandlung gegenüber CD95-vermittelter Apoptose geschützt werden. Daher sollte untersucht werden, welche Bedeutung Mcl-1 im hepatozellulären Karzinom (HCC) und in der gesunden Leber einnimmt. Hierzu wurde zunächst humanes HCC-Gewebe hinsichtlich der Expression von Mcl-1 untersucht. Es konnte gezeigt werden, dass Mcl-1 sowohl auf mRNA- als auch auf Protein-Ebene in HCC-Gewebe verstärkt exprimiert ist im Vergleich zu benachbartem Normalgewebe. Auch in verschiedenen HCC-Zelllinien konnte eine starke Mcl-1-Expression nachgewiesen werden. Diese war vor allem über den PI3K/Akt-Signalweg reguliert. Eine Hemmung dieses Signalwegs führte zu einer Reduktion der Mcl-1-Expression und so zu einer Sensitivierung der Zellen gegenüber verschiedenen Chemotherapeutika und zielgerichteten Therapien. Des Weiteren wurde die Mcl-1-Expression spezifisch durch RNA-Interferenz gehemmt. Auch hier konnte gezeigt werden, dass Zellen mit unterdrückter Mcl-1-Expression deutlich sensitiver gegenüber verschiedenen Apoptose-induzierenden Substanzen reagierten. Eine kombinierte Hemmung der Mcl-1-Expression und der PI3-Kinase führte schließlich zu einer nochmals verstärkten Sensitivierung. Im Gegensatz dazu führte eine Überexpression von Mcl-1 zu einer Hemmung der Apoptoseinduktion. Im zweiten Teil der Arbeit wurde eine Mauslinie etabliert, welche spezifisch in Hepatozyten kein Mcl-1 exprimiert, um so die Bedeutung von Mcl-1 für die Leber in vivo zu untersuchen. Es zeigte sich, dass Mcl-1flox/flox-AlbCre-Mäuse bereits im Alter von acht Wochen eine verminderte Lebergröße aufweisen. Dies wurde verursacht durch spontane Apoptoseinduktion in den Mcl-1 negativen Hepatozyten. Hierdurch kam es zu einer Leberschädigung, ersichtlich durch erhöhte Transaminasenwerte, erhöhte Caspase-3-Aktivierung, und Schädigung der Gewebsstruktur. Zudem war als kompensatorischer Effekt die Zellproliferation erhöht, ohne dass sich jedoch das Lebergewicht an das von Kontrolltieren anglich. Interessanterweise kam es in Mcl-1flox/flox-AlbCre-Mäusen als Folge der chronischen Leberschädigung zur Entwicklung einer Leberfibrose, ersichtlich durch eine verstärkte Collageneinlagerung. Weiterhin reagierten Mcl-1flox/flox-AlbCre-Mäuse wesentlich empfindlicher gegenüber Todesrezeptor-vermittelter Apoptose. Diese Daten zeigen zum einen, dass Mcl-1 zur Apoptoseresistenz von HCC-Zellen beitragen kann. Zielgerichtete Therapien, welche die Expression von Mcl-1 hemmen, könnten folglich für die Therapie des HCCs von Interesse sein. Des Weiteren konnte in dieser Arbeit zum ersten Mal gezeigt werden, dass Mcl-1 ein zentraler anti-apoptotischer Faktor für Hepatozyten in vivo ist.

E’ cresciuto pertanto l’interesse sul loro potenziale impiego in pazienti con cirrosi. Questo studio si proponeva di valutare la fattibilità e la sicurezza della reinfusione intraepatica di cellule staminali midollari autologhe CD133+ in 12 pazienti con insufficienza epatica terminale. Previa mobilizzazione nel sangue periferico mediante somministrazione di granulocyte-colony stimulating factor (G-CSF) alla dose di 7,5 mcg/Kg/b.i.d. e raccolta per leucoaferesi (solo se la concentrazione di CD133 + SC era > 8/μL), le cellule CD133+ altamente purificate sono state reinfuse in arteria epatica a partire da 5x104/Kg fino a 1x106/kg. Nei tre giorni successivi è stato somministrato G-CSF per favorire l’espansione e l’attecchimento delle cellule. Durante la fase della mobilizzazione e quella della reinfusione sono stati eseguiti saggi biologici quali: caratterizzazione fenotipica delle SC circolanti, saggi clonogenici, valutazione della concentrazione sierica del Hepatocyte Growth Factor (HGF), Stromal-Derived Factor-1 (SDF-1) ed il Vascular-Endotelial Growth Factor (VEGF) e caratterizzazione fenotipica delle CD133+SC purificate. Fino ad oggi sono stati reinfusi 12 pazienti. Questi dati preliminari suggeriscono che è possibile mobilizzare e reinfondere un numero considerevole di SC autologhe CD133+ altamente purificate in pazienti con ESLD . Gli studi biologici mostrano che: il numero di progenitori ematopoietici ed endoteliali circolanti è aumentato dopo il trattamento con G–CSF; le SCs CD133+ altamente purificato esprimono marcatori emopoietici ed endoteliali; la concentrazione sierica di HGF, SDF-1, VEGF e la capacità clonogenica di progenitori emopoietici sono aumentati durante la mobilitazione e nelle fasi di reinfusione; il potenziale clonogenico dei progenitori endoteliali mostra espressione variabile.

Immune responses have the important function of host defense and protection against pathogens. However, the immune response also causes inflammation and host tissue injury, termed immunopathology. For example, hepatitis B and C virus infection in humans cause immunopathological sequel with destruction of liver cells by the host's own immune response. Similarly, after infection with lymphocytic choriomeningitis virus (LCMV) in mice, the adaptive immune response causes liver cell damage, choriomeningitis and destruction of lymphoid organ architecture. The immunopathological sequel during LCMV infection has been attributed to cytotoxic CD8(+) T cells. However, we now show that during LCMV infection CD4(+) T cells selectively induced the destruction of splenic marginal zone and caused liver cell damage with elevated serum alanin-transferase (ALT) levels. The destruction of the splenic marginal zone by CD4(+) T cells included the reduction of marginal zone B cells, marginal zone macrophages and marginal zone metallophilic macrophages. Functionally, this resulted in an impaired production of neutralizing antibodies against LCMV. Furthermore, CD4(+) T cells reduced B cells with an IgM(high)IgD(low) phenotype (transitional stage 1 and 2, marginal zone B cells), whereas other B cell subtypes such as follicular type 1 and 2 and germinal center/memory B cells were not affected. Adoptive transfer of CD4(+) T cells lacking different important effector cytokines and cytolytic pathways such as IFNγ, TNFα, perforin and Fas-FasL interaction did reveal that these cytolytic pathways are redundant in the induction of immunopathological sequel in spleen. In conclusion, our results define an important role of CD4(+) T cells in the induction of immunopathology in liver and spleen. This includes the CD4(+) T cell mediated destruction of the splenic marginal zone with consecutively impaired protective neutralizing antibody responses.

To understand how the serum amyloid A (SAA) genes are regulated, the cis-acting elements and trans-acting factors involved in the regulation of mouse SAA3 and rat SAA1 genes expression during inflammation were analyzed.^ To identify DNA sequences involved in the liver-specific expression of the mouse SAA3 gene, the 5$\sp\prime$ flanking region of this gene was analyzed by transient transfection studies. Results suggest that C/EBP, a liver-enriched transcription factor, plays an important role for the enhanced expression of the mouse SAA3 gene in hepatocytes.^ Transfection studies of the regulation of the expression of rat SAA1 gene indicated that a 322 bp fragment ($-$304 to +18) of the gene contains sufficient information for cytokine-induced expression of the reporter gene in a liver cell-specific manner. Further functional analysis of the 5$\sp\prime$ flanking region of the rat SAA1 gene demonstrated that a 65 bp DNA fragment ($-$138/$-$73) can confer cytokine-inducibility onto a heterologous promoter both in liver and nonliver cells. DNase I footprint and gel retardation assays identified five putative cis-regulatory elements within the 5$\sp\prime$ flanking region of the gene: one inducible element, a NF$\kappa$B binding site and four constitutive elements. Two constitutive elements, footprint regions I and III, were identified as C/EBP binding sites with region III having over a 10-fold higher affinity for C/EBP binding than region I. Functional analysis of the cis-elements indicated that C/EBP(I) and C/EBP(III) confer liver cell-specific activation onto a heterologous promoter, while sequences corresponding to the NF$\kappa$B element and C/EBP(I) impart cytokine responsiveness onto the heterologous promoter. These results suggest that C/EBP(I) possesses two functions: liver-specific activation and cytokine responsiveness. The identification of two cytokine responsive elements (NF$\kappa$B and C/EBP(I)), and two liver-specific elements (C/EBP(I) and C/EBP(III)) implies that multiple cis-acting elements are involved in the regulation of the expression of the rat SAA1 gene. The tissue-specific and cytokine-induced expression of rat SAA1 gene is likely the result of the interactions of these cis-acting elements with their cognate trans-acting factors as well as the interplay between the different cis-acting elements and their binding factors.
