The purpose of the study was to examine the relationships between intermittent high-intensity efforts (RAST) parameters and variables related to aerobic metabolism (anaerobic threshold; LAN, maximal oxygen uptake; VO2MAX and velocity correspondent to VO2MAX; iVO(2MAX)). Eight under-17 (U17) soccer players (16 +/- 1 years) participated in the study. The participants were submitted to a graded exercise test and six maximal sprints of 35m with 10 seconds of passive recovery between each effort (RAST). The RAST parameters were not significant correlated with VO2MAX and LAN. However absolute and relative mean power were significantly correlated with iVO(2MAX) (r=0.79 e r=0.85, respectively). Furthermore, the fatigue index and the relative peak power were significantly correlated with the iVO(2MAX) (r=-0,57 e r=0,73, respectively). In conclusion, the only aerobic variable correlated with performance in consecutive efforts with brief recovery periods, such as RAST, is iVO(2MAX).

Reactive oxygen species (ROS) are reactive molecules containing oxygen, that form as byproducts of aerobic metabolism, including immune system processes. Too much ROS may cause oxidative stress. In this study, we examined whether it can also limit the production of immune system compounds. To assess the relationship between antioxidant status and immunity we evaluated the effect of dietary supplementation with organic selenium, given at various levels for 10 days, on the antioxidant and immune system of the pacu fish (Piaractus mesopotamicus). Fish fed a diet containing 0.6 mg Se-yeast kg(-1) showed significant improvement in antioxidant status, as well as in hematological and immunological profiles. Specifically, they had the highest counts for catalase (CAT), glutathione peroxidase (GPx), glutathione S-transferase (GST), red blood cells, and thrombocytes; the highest leukocyte count (particularly for monocytes); and the highest serum lysozyme activity. There was also a positive correlation between GPx and lysozyme in this group of fish. These findings indicate that short-term supplementation with 0.6 mg Se-yeast kg(-1) reestablished the antioxidative status, allowing the production of innate components which can boost immunity without the risk of oxidative stress. This study shows a relationship between oxidative stress and immunity, and, from a practical perspective, shows that improving immunity and health in pacu through the administration of selenium could improve their growth performance.

Avaliaram-se os efeitos da cor do ambiente sobre o crescimento e a atividade da enzima antioxidante catalase (CAT) e da lactato desidrogenase (LDH) em tilápias do Nilo (n=24; 36,2±3,6g). Oito exemplares foram mortos para determinação da atividade basal das enzimas e os demais permaneceram isolados durante 14 dias sob espectro de luz branca ou azul (n=8 peixes/tratamento). A seguir os peixes foram submetidos a um estresse diário de confinamento de 90 minutos (15° ao 28° dia) e pesados semanalmente para cálculo da taxa de crescimento específico (TCE). A TCE negativa confirmou que o confinamento provocou estresse nos peixes, independentemente da cor do ambiente. O aumento da atividade da LDH no músculo vermelho dos peixes mantidos sob luz branca ou azul indicou mudança do metabolismo aeróbio para anaeróbio. O estresse reduziu a atividade da CAT no músculo branco dos peixes mantidos sob a luz branca ou azul. Na musculatura vermelha, esta redução ocorreu apenas nos animais mantidos sob a luz branca. O confinamento aumenta os processos metabólicos anaeróbios e é adequado para estudos sobre os efeitos do estresse. O espectro de luz azul não evita a redução do crescimento e a demanda energética anaeróbia em situações de estresse, mas preserva a atividade da CAT, contribuindo para o bem-estar da tilápia.

[EN] The aim of this study was to determine the influence of activity performed during the recovery period on the aerobic and anaerobic energy yield, as well as on performance, during high-intensity intermittent exercise (HIT). Ten physical education students participated in the study. First they underwent an incremental exercise test to assess their maximal power output (Wmax) and VO2max. On subsequent days they performed three different HITs. Each HIT consisted of four cycling bouts until exhaustion at 110% Wmax. Recovery periods of 5 min were allowed between bouts. HITs differed in the kind of activity performed during the recovery periods: pedaling at 20% VO2max (HITA), stretching exercises, or lying supine. Performance was 3-4% and aerobic energy yield was 6-8% (both p < 0.05) higher during the HITA than during the other two kinds of HIT. The greater contribution of aerobic metabolism to the energy yield during the high-intensity exercise bouts with active recovery was due to faster VO2 kinetics (p< 0.01) and a higher VO2peak during the exercise bouts preceded by active recovery (p < 0.05). In contrast, the anaerobic energy yield (oxygen deficit and peak blood lactate concentrations) was similar in all HITs. Therefore, this study shows that active recovery facilitates performance by increasing aerobic contribution to the whole energy yield turnover during high-intensity intermittent exercise.

In der vorliegenden Arbeit untersuchte ich die Diversität und die sauerstoffabhängige Expression der Globine von Karpfenfischen. Mit Globin X konnte ein fünfter Globintyp identifiziert werden, dessen Vorkommen auf Fische und Amphibien beschränkt ist. Globin X wird sowohl auf mRNA- als auch auf Proteinebene in zahlreichen Geweben exprimiert. Zur Aufklärung der genauen Funktion müssen noch weitere Analysen durchgeführt werden. Phylogenetische Untersuchungen ergaben eine ursprüngliche Verwandtschaft zwischen Neuroglobin und Globin X und deuten darauf hin, dass der letzte gemeinsame Vorfahre der Protostomia und Deuterostomia bereits zwei verschiedene Globintypen besessen hat. Im Zebrabärbling und im Goldfisch konnte ich eine Myoglobin-Expression neben dem Herzen auch in Hirn, Kieme, Leber und Niere nachweisen und somit zeigen, dass Myoglobin nicht nur im Muskelgewebe lokalisiert ist. Des Weiteren konnte eine hirnspezifische Myoglobin-Isoform im Goldfisch identifiziert werden, deren Funktion noch unklar ist und weiterer Untersuchungen bedarf. Das Vorhandensein der zweiten Isoform ist innerhalb der Cyprinidae (Karpfenfische) aufgrund einer Genomduplikation bei den Cyprininae (Kärpflinge) auf diese Unterfamilie beschränkt. Durch Hypoxieexperimente konnte gezeigt werden, dass die Expression der Globine von der Intensität des Sauerstoffmangels abhängig ist und gewebe- und artspezifisch erfolgt. Im Zebrabärbling wurde eine Abnahme der Hämoglobin- und Globin X-Konzentration beobachtet, während das Cytoglobin-Expressionsniveau nahezu unverändert blieb. Im Fall von Myoglobin und Neuroglobin konnte zum ersten Mal gezeigt werden, dass die hypoxieinduzierte Zunahme der mRNA-Menge auch mit einer verstärkten Expression des jeweiligen Proteins korreliert ist. Im Vergleich dazu war die Veränderung der Expression der meisten Globine im Goldfisch gering, lediglich Myoglobin wurde im Fischkörper auf mRNA-Ebene nach Hypoxie deutlich verstärkt exprimiert. Durch einen Vergleich der konstitutiven Neuroglobin-Expression beider Karpfenfische konnte in Auge und Hirn des hypoxietoleranten Goldfisches eine 3- bzw. 5-fach höhere Neuroglobin-Konzentration als im hypoxiesensitiven Zebrabärbling nachgewiesen werden. Meine Ergebnisse stützen somit die Hypothese, dass Neuroglobin eine myoglobinähnliche Funktion einnimmt und den aeroben Stoffwechsel im neuronalen Gewebe auch unter Sauerstoffmangel aufrechterhält.

Das Enterobakterium Escherichia coli sowie das Bodenbakterium Bacillus subtilis können C4-Dicarbonsäuren als aerobe Kohlenstoffquelle zur Energiekonservierung nutzen. Die Regulation des C4-Dicarboxylatstoffwechsels erfolgt in E. coli und B. subtilis durch das Zweikomponentensystem DcuSREc bzw. DctSRBs, bestehend aus einer Sensorkinase und einem Responseregulator. Diese kontrollieren die Expression des C4-Dicarboxylat-Transporters DctA. Der Sensor DcuSEc benötigt für seine Funktion im aeroben Stoffwechsel den Transporter DctA als Cosensor. Für das DctSRBs-System gibt es Hinweise aus genetischen Untersuchungen, dass DctSBs das Bindeprotein DctBBs und möglicherweise auch DctABs als Cosensoren für seine Funktion benötigt. In dieser Arbeit sollte ein direkter Nachweis geführt werden, ob DctBBs und DctABs gemeinsam oder nur jeweils eine der Komponenten als Cosensoren für DctSBs fungieren. Sowohl für DctBBs als auch für DctABs wurde eine direkte Protein-Protein-Interaktion mit DctSBs durch zwei in vivo Interaktionsmethoden nachgewiesen. Beide Methoden beruhen auf der Co-Reinigung der Interaktionspartner mittels Affinitätschromatographie und werden je nach Affinitätssäule als mSPINE oder mHPINE (Membrane Strep/His-Protein INteraction Experiment) bezeichnet. Die Interaktion von DctSBs mit DctBBs wurde zusätzlich über ein bakterielles Two-Hybrid System nachgewiesen. Nach Coexpression mit DctSBs interagieren DctABs und DctBBs in mSPINE-Tests gleichzeitig mit der Sensorkinase. DctSBs bildet somit eine sensorische DctS/DctA/DctB-Einheit in B. subtilis und das Bindeprotein DctBBs agiert nur als Cosensor, nicht aber als Transport-Bindeprotein. Eine direkte Interaktion zwischen dem Transporter DctABs und dem Bindeprotein DctBBs besteht nicht. Transportmessungen belegen, dass der DctA-vermittelte Transport von [14C]-Succinat unabhängig ist von DctBBs. Außerdem wurde untersucht, ob Zweikomponentensysteme aus anderen Bakteriengruppen nach einem ähnlichen Schema wie DcuSREc bzw. DctSRBs aufgebaut sind. Das thermophile Bakterium Geobacillus kaustophilus verfügt über ein DctSR-System, welches auf genetischer Ebene mit einem Transporter des DctA-Typs und einem DctB-Bindeprotein geclustert vorliegt. Die Sensorkinase DctSGk wurde in E. coli heterolog exprimiert und gereinigt. Diese zeigt in einer E. coli DcuS-Insertionsmutanten Komplementation der DcuS-Funktion und besitzt dabei Spezifität für die C4-Dicarbonsäuren Malat, Fumarat, L-Tartrat und Succinat sowie für die C6-Tricarbonsäure Citrat. In Liposomen rekonstituiertes DctSGk zeigt Autokinase-Aktivität nach Zugabe von [γ-33P]-ATP. Der KD-Wert für [γ-33P]-ATP der Kinasedomäne von DctSGk liegt bei 43 μM, die Affinität für ATP ist damit etwa 10-fach höher als in DcuSEc.

The mechanisms causing brain damage after acute subdural hematoma (SDH) are poorly understood. A decrease in cerebral blood flow develops immediately after the hematoma forms, thus reducing cerebral oxygenation. This in turn may activate mitochondrial failure and tissue damage leading to ionic imbalance and possibly to cellular breakdown. The purpose of this study was to test whether a simple therapeutic measure, namely increased fraction of inspired oxygen (FiO2 100), and hence increased arterial and brain tissue oxygen tension, can influence brain glucose and lactate dynamics acutely after subdural hematoma in the rat. Twenty-five male Sprague-Dawley anesthetized rats were studied before, during and after induction of the SDH in two separate groups. The Oxygen group (n = 10) was ventilated with 100% oxygen immediately after induction of the SDH. The Air group (n = 10) was ventilated during the entire study with 21% oxygen. Brain microdialysate samples were analyzed for glucose and lactate. All rats were monitored with femoral arterial blood pressure catheters, arterial blood gas analysis, arterial glucose, lactate and end tidal CO2 (EtCO2). Five male Sprague-Dawley rats were sham operated to measure the effect of oxygen challenge on glucose-lactate dynamics without injury. Arterial oxygen tension in the Oxygen group was 371 +/- 30 mmHg and was associated with significantly greater increase in dialysate lactate in the first 30 min after induction of SDH. Dialysate glucose initially dropped in both groups, after SDH, but then reverted significantly faster to values above baseline in the Oxygen group. Changes in ventilatory parameters had no significant effect on dialysate glucose and lactate parameters in the sham group. Extracellular dialysate lactate and glucose are influenced by administration of 100% O2 after SDH. Dialysate glucose normalizes significantly quicker upon 100% oxygen ventilation. We hypothesize that increased neural tissue oxygen tension, in presence of reduced regional CBF, and possibly compromised mitochondrial function, after acute SDH results in upregulation of rate-limiting enzyme systems responsible for both glycolytic and aerobic metabolism. Similar changes have been seen in severe human head injury, and suggest that a simple therapeutic measure, such as early ventilation with 100% O2, may improve cerebral energy metabolism, early after SDH. Further studies to measure the generation of adenosine triphosphate (ATP) are needed to validate the hypothesis.

OBJECT: Early impairment of cerebral blood flow in patients with severe head injury correlates with poor brain tissue O2 delivery and may be an important cause of ischemic brain damage. The purpose of this study was to measure cerebral tissue PO2, lactate, and glucose in patients after severe head injury to determine the effect of increased tissue O2 achieved by increasing the fraction of inspired oxygen (FiO2). METHODS: In addition to standard monitoring of intracranial pressure and cerebral perfusion pressure, the authors continuously measured brain tissue PO2, PCO2, pH, and temperature in 22 patients with severe head injury. Microdialysis was performed to analyze lactate and glucose levels. In one cohort of 12 patients, the PaO2 was increased to 441+/-88 mm Hg over a period of 6 hours by raising the FiO2 from 35+/-5% to 100% in two stages. The results were analyzed and compared with the findings in a control cohort of 12 patients who received standard respiratory therapy (mean PaO2 136.4+/-22.1 mm Hg). The mean brain PO2 levels increased in the O2-treated patients up to 359+/-39% of the baseline level during the 6-hour FiO2 enhancement period, whereas the mean dialysate lactate levels decreased by 40% (p < 0.05). During this O2 enhancement period, glucose levels in brain tissue demonstrated a heterogeneous course. None of the monitored parameters in the control cohort showed significant variations during the entire observation period. CONCLUSIONS: Markedly elevated lactate levels in brain tissue are common after severe head injury. Increasing PaO2 to higher levels than necessary to saturate hemoglobin, as performed in the O2-treated cohort, appears to improve the O2 supply in brain tissue. During the early period after severe head injury, increased lactate levels in brain tissue were reduced by increasing FiO2. This may imply a shift to aerobic metabolism.

Traditionally, critical swimming speed has been defined as the speed when a fish can no longer propel itself forward, and is exhausted. To gain a better understanding of the metabolic processes at work during a U(crit) swim test, and that lead to fatigue, we developed a method using in vivo (31)P-NMR spectroscopy in combination with a Brett-type swim tunnel. Our data showed that a metabolic transition point is reached when the fish change from using steady state aerobic metabolism to non-steady state anaerobic metabolism, as indicated by a significant increase in inorganic phosphate levels from 0.3+/-0.3 to 9.5+/-3.4 mol g(-1), and a drop in intracellular pH from 7.48+/-0.03 to 6.81+/-0.05 in muscle. This coincides with the point when the fish change gait from subcarangiform swimming to kick-and-glide bursts. As the number of kicks increased, so too did the Pi concentration, and the pH(i) dropped. Both changes were maximal at U(crit). A significant drop in Gibbs free energy change of ATP hydrolysis from -55.6+/-1.4 to -49.8+/-0.7 kJ mol(-1) is argued to have been involved in fatigue. This confirms earlier findings that the traditional definition of U(crit), unlike other critical points that are typically marked by a transition from aerobic to anaerobic metabolism, is the point of complete exhaustion of both aerobic and anaerobic resources.
