Organs developing as appendages of the ectoderm are initiated from epithelial thickenings called placodes. Their formation is regulated by interactions between the ectoderm and underlying mesenchyme, and several signalling molecules have been implicated as activators or inhibitors of placode formation. Ectodysplasin (Eda) is a unique signalling molecule in the tumour necrosis factor family that, together with its receptor Edar, is necessary for normal development of ectodermal organs both in humans and mice. We have shown previously that overexpression of the Eda-A1 isoform in transgenic mice stimulates the formation of several ectodermal organs. In the present study, we have analysed the formation and morphology of placodes using in vivo and in vitro models in which both the timing and amount of Eda-A1 applied could be varied. The hair and tooth placodes of K14-Eda-A1 transgenic embryos were enlarged, and extra placodes developed from the dental lamina and mammary line. Exposure of embryonic skin to Eda-A1 recombinant protein in vitro stimulated the growth and fusion of placodes. However, it did not accelerate the initiation of the first wave of hair follicles giving rise to the guard hairs. Hence, the function of Eda-A1 appears to be downstream of the primary inductive signal required for placode initiation during skin patterning. Analysis of BrdU incorporation indicated that the formation of the epithelial thickening in early placodes does not involve increased cell proliferation and also that the positive effect of Eda-A1 on placode expansion is not a result of increased cell proliferation. Taken together, our results suggest that Eda-A1 signalling promotes placodal cell fate during early development of ectodermal organs.

Development of ectodermal appendages, such as hair, teeth, sweat glands, sebaceous glands, and mammary glands, requires the action of the TNF family ligand ectodysplasin A (EDA). Mutations of the X-linked EDA gene cause reduction or absence of many ectodermal appendages and have been identified as a cause of ectodermal dysplasia in humans, mice, dogs, and cattle. We have generated blocking antibodies, raised in Eda-deficient mice, against the conserved, receptor-binding domain of EDA. These antibodies recognize epitopes overlapping the receptor-binding site and prevent EDA from binding and activating EDAR at close to stoichiometric ratios in in vitro binding and activity assays. The antibodies block EDA1 and EDA2 of both mammalian and avian origin and, in vivo, suppress the ability of recombinant Fc-EDA1 to rescue ectodermal dysplasia in Eda-deficient Tabby mice. Moreover, administration of EDA blocking antibodies to pregnant wild type mice induced in developing wild type fetuses a marked and permanent ectodermal dysplasia. These function-blocking anti-EDA antibodies with wide cross-species reactivity will enable study of the developmental and postdevelopmental roles of EDA in a variety of organisms and open the route to therapeutic intervention in conditions in which EDA may be implicated.

Background: Gynaecomastia in male goats is characterized by abnormal development of the mammary gland. Enlarged udder may be observed cranially to the scrotum, which can occasionally reach the size of the testicles. The udder may carry functional glands and impair the animal's reproductive performance and welfare. The case of a successful surgical treatment of gynaecomastia in a high reproductive performance Saanen buck-goat is reported in the present study.Material, Methods & Results: The animal was admitted presenting significant augmentation of the mammary glands, which was clinically diagnosed as gynaecomastia. The male goat owned optimal phenotypic characteristics for the Saanen breed, which had been producing high performance descendents. The mammary glands had been impairing the goat's locomotion and sexual performance. Manual milking resulted in great amount of milk secretion. The animal presented anorexia and impaired sexual performance. After clinical and laboratorial evaluation, the animal was submitted to radical mastectomy. An elliptic skin incision was performed around each mammary gland. Subcuticular blunt dissection was accomplished to isolate the mammarian tissue from the abdominal muscular layer and the spermatic chord. The excised mass was sampled for histological assessment. Subcuticular layer and skin closure was carried in a routine fashion. Hygienization of the surgical wound was performed with 2,5% PVP-I solution for ten days. Additionally, an association of penicillin G benzathine and streptomycin, and fluxinin meglumine were also given. The surgical procedure was successfully accomplished without any peroperative complication. The excised mass was sampled for anatomic/histological assessment. Macroscopically, the left mammary gland presented 22 cm in length, 12 cm wide and 26 cm in diameter. The right gland presented 16 cm in length, 7 cm wide and 13,5 cm in diameter. The microscopic assessment revealed hyperplasia of the glandular ducts. No abnormalities resembling malignant mammary neoplasms or degeneration were observed. At the end of the treatment, the animal was completely recovered. The animal convalesced satisfactorily and surgical wound healed completely within the first 10 days post-op. The goat was not culled and returned to normal reproductive activity. Within 12 months of follow-up, the animal was able to produce high milk yield performance progenies.Discussion: This case report presented relevant aspects of the surgical management of gynaecomastia, especially to veterinary practitioners dealing with milk goats. Gynaecomastia is not as common as other reproductive disorders in domestic animals. In opposition to the findings of the present study, other trials revealed that gynaecomastia usually does not affect fertility, libido, ejaculate parameters and sexual performance of goats. However, it is important to consider that neoplasic disorders such as mammary adenocarcinoma may be present, even though these are rare complications. Last but not least, the decision making on mastectomy in the present study was crucial in order to reestablish the animal's welfare and its functionality in the farms reproduction program.

Biopsy by fine needle aspiration together with microbiological examination and scanning electron microscopy were evaluated in diagnosis of clinical bovine mastitis in a Prototheca zopfii outbreak. Fine needle aspiration was performed in 21 mammary quarters from ten Holstein cows presenting clinical mastitis caused by P. zopfii. The algae were previously identified in the microbiological examination of milk collected from these cows. Material aspirated from these 21 mammary glands was submitted to cytological staining (Gram, Giemsa and/or Shor staining). Fine needle aspiration enabled cytological identification of the algae in these 21 mammary glands, from which P. zopfii was isolated in the milk. Simultaneously, five mammary fragments collected by fine needle aspiration from these 21 mammary glands presenting clinical mastitis were also submitted to microbiological examination. P. zopfii was also isolated from these five fragments. Scanning electron microscopy technique also identified three of these five P. zopfii strains isolated from mammary fragments collected by cytological aspiration. These results suggest that fine needle aspiration may be an alternative method for the diagnosis of clinical mastitis.

Routine diagnosis methods used in bovine mastitis were studied in 55 mares in lactation. The findings of strip cup test, California Mastitis Test-CMT, electronic somatic cell count-CCS, microbiological culture, and in vitro antimicrobial susceptibility profile of isolates were discussed. Streptococcus spp., Staphylococcus spp, and enterobacteria were the most common microorganisms isolated in health and CMT-positive mammary glands. Staphylococcus aureus and Arcanobacterium pyogenes were identified in two mares presenting clinical mastitis. Mean somatic cell count of eight mares without presence of microorganisms in milk was 247.57x10³/mL and 1.621,86x10³/mL in 47 mares with positive microbiological culture. Moderate concordance (63.8%) between positive reactions in CMT (1 to 3+) and microbiological culture was observed. Amicacin (78.9%), ceftiofur (74.7%), sulpha-trimetoprim (69,0%) and norfloxacin (69.0%), were the most effective drugs, while resistance of isolates was mainly observed against penicillin (64.8%), gentamycin (35.2%), azithromycin (35.2%), enrofloxacin (28.2%), and florfenicol (28.2%).

Prototheca zopfii has been considered one of the most important causes of environmental mastitis in Brazil. These algae are refractory to conventional therapy and cause great damage to the mammary gland. The present study evaluated the in vitro algaecide effect of sodium hypochlorite and iodine based antiseptics on 27 P. zopfii strains isolated from the milk of cattle. Low concentrations of sodium hypochlorite (0.0390625-0.15625%) and iodine (0.15625-0.625%) were effective against the isolates. These antiseptics may be recommended for hygiene routines, pre and postdipping and cauterization of bovine mammary glands infected by P. zopfii. (C) 2009 Elsevier Ltd. All rights reserved.

This report describes the production of cytotoxic necrotizing factor (CNF) by an Escherichia coli strain isolated from clinical bovine mastitis with clinical signs of toxemia The animal had hemorrhages and necrosis of the mammary glands, and died within 24 hours after the onset of clinical signs. In addition to CNF identification, alpha-haemolysin and siderophores production were also characterized in this strain. This report reinforce the association of CNF and alpha-haemolysin production in E. coli virulence associated with clinical cases of severe bovine mastitis.

The present work aimed to evaluate udders of Bergamasca ewes and their correlation with milk yield in a mix system of milk yield. Seventy-seven ewes were fed with balanced concentrate starting 20 days before lambing until the end of the experiment. The lambs stayed with their mothers in pastures during the day and were separated at night. They returned to their mothers after the morning milking and were weaned at 45 days of age. Forty-eight hours after lambing, ewes were machine milked once daily at 7 am and the milk yield was recorded for a period of 60 days. Measurements of circumference, depth and width of the udder, and width and length of teats, at 30 and 60 days, were taken. A higher average daily yield of commercial milk was observed after lambs weaning (0.509 vs. 0.435 kg/ewe/day) than before. In the same way, the correlations between udder depth, circumference and width and milk yield were positive and significant only after weaning (0.74, 0.75 and 0.62, respectively). Udder measures had positive correlations with milk yield and can be used in programs of milk yield improvement. (c) 2007 Elsevier B.V. All rights reserved.

Uma técnica empregando fluoresceína sódica 5% e luz ultravioleta foi desenvolvida para a identificação dos trajetos dos vasos linfáticos superficiais das glândulas mamárias em dez cadelas. A fluoresceína foi injetada por via intradérmica ao redor da base de cada mamilo e cada mama foi observada após 5, 15, 30 e 60 minutos. em cada animal, primeiramente, foram avaliadas as mamas torácica cranial, abdominal cranial e inguinal das cadeias direita e esquerda e 48 horas após as mamas torácica caudal e abdominal caudal das cadeias direita e esquerda. de um total de 97 mamas injetadas, em 8 a fluoresceína não foi captada pelos linfáticos. O tempo de 30 minutos foi o mais adequado para a visualização total dos trajetos. Mostrou ser um método simples, rápido e inócuo de verificação in vivo de vasos linfáticos.

As neoplasias mamarias constituem aproximadamente 50% dos tumores diagnosticados em cadelas. Apesar dosharmónios sexuais femininos desempenharem papel fundamental no desenvolvimento desses tumores em mamíferos, o valor da supressão hormonal pela ovário-histerectomia como auxiliar no tratamento do tumor de mama em caninos permanece controverso. Discute-se ainda se a realização da ovário-histerectomia após o diagnóstico influencia ou não o crescimento e progressão do tumor na glândula afetada ou em outras glândulas mamarias. O objetivo desta revisão é discutir alguns aspectos relacionados à influência hormonal na etiologia de tumores mamarias em cadelas, assim como o valor terapêutico da castração, quando realizada no momento da mastectomia.

Estudaram-se os resultados obtidos no exame clínico da glândula mamária de 3.191 vacas mestiças e no "California Mastitis Test" (CMT) em 12.764 amostras de leite colhidas destes animais, relacionando-os com o estádio de lactação, o número de partos e a localização das afecções. O exame clínico revelou que 50 quartos apresentavam mastite e 231 mostravam-se atrofiados ou afuncionais. A aplicação do CMT indicou alterações de secreção em 1.741 quartos. A localização das afecções nos quartos foi estatisticamente semelhante, o mesmo ocorrendo com a incidência de mastite clínica com relação ao estádio de lactação. Quanto ao número de partos, não houve diferença significativa em relação à incidência de mastite clínica, porém a ocorrência de casos subclínicos foi estatisticamente superior em animais de uma a seis crias. O percentual de quartos afuncionais foi significativamente maior nos animais, acima de quatro partos.
