The occurrence of pathogenic and endophytic species of Phyllosticta on cultivated Citrus in Australia was investigated by DNA sequence analysis of specimens held in plant pathology herbaria and culture collections. Sequences of the internal transcribed spacer region (ITS1, 5.8S, ITS2), and partial translation elongation factor 1-alpha (TEF) gene of 41 Phyllosticta-like isolates from Citrus were compared to those sequences from the type specimens of Phyllosticta recorded from around the world. Phylogenetic analysis resolved all the sequences of Australian accessions into two major clades. One clade corresponded to P. citricarpa, which causes citrus black spot disease. The other clade contained P. capitalensis, which is a known endophyte of Citrus and many other plant species. All included herbarium accessions previously designated as Guignardia mangiferae are now designated P. capitalensis. No Australian isolates were identified as the newly described pathogens of citrus P. citriasiana or P. citrichinaensis, or the endophytes Guignarida mangiferae, P. brazilianiae, or P. citribraziliensis. © 2013 Australasian Plant Pathology Society Inc.

Two new guignardones and one tricycloalternarene derivatives, named guignardone D, E (2-3) and tricycloalternarene F (4), and the known guignardone A (1) were isolated from Guignardia mangiferae, an endophytic fungus from the leaves of Viguiera arenaria (Asteraceae), after fermentation in Czapek medium. Structures were established on the basis of their spectroscopic data, including H-1 NMR, C-13 NMR, HMQC, HMBC and HRESI-MS. (C) 2012 Phytochemical Society of Europe. Published by Elsevier B.V. Neste trabalho testou-se o potencial antagonista de 16 fungos endofíticos isolados de videiras (Vitis vinifera L.), de castas representativas do Alentejo produzidas em modo de proteção integrada e em modo biológico, contra Guignardia bidwellii. Os isolados identificados após ITS-PCR e sequenciação pertencem aos géneros Epicoccum, Alternaria, Botrytis, Athelia, Phoma e Gibberella. Os isolados testados mostraram atividade antagonista contra G. bidwellii quer por inibição direta, quer através da produção de compostos voláteis, à exceção dos dois isolados de B. cinerea. No entanto, todos os isolados produziram alguns compostos voláteis com reconhecida atividade antimicrobiana, tais como benzaldeído, 3-metil-1-butanol e derivados de ácido propanoico. Foi ainda observado que seis dos isolados produziram também metabolitos não voláteis com capacidade de inibir o crescimento de G. bidwellii. Os resultados obtidos vêm mostrar o potencial dos fungos endofíticos como agentes de luta biológica no controlo de G. bidwellii, podendo constituir novas alternativas no âmbito de Proteção de Plantas; ABSTRACT: Endophytic fungi present in grapevines (Vitis vinifera L.) with the ability to inhibit the growth of the causal agent of black rot (Guignardia bidwellii) In this work the antagonistic potential of 16 endophytic grapevine fungi isolates (Vitis vinifera L.), from representative cultivars of the Alentejo region produced either under integrated pest management or organic mode, was tested against Guignardia bidwellii. Isolates were identified through ITS-PCR and sequencing, as belonging to the genera Epicoccum, Alternaria, Botrytis, Athelia, Phoma and Gibberella. Isolates showed antagonist activity against G. bidwellii either by direct inhibition or through the production of volatile compounds, with the exception of two isolates of B. cinerea. Nevertheless, all isolates produced volatile compounds with known antimicrobial activity such as benzaldehyde, 3-methyl-1-butanol and propionic acid derivatives. Additionally, six isolates produced non-volatile metabolites with the ability to inhibit G. bidwellii growth. These results show the potential that endophytic fungi have as agents for biological control of G. bidwellii, opening new options in the field of Plant Protection.

Several species of Phyllosticta (syn. Guignardia) have been described from orchids worldwide. A new species, Phyllosticta speewahensis, is proposed for a specimen isolated from leaf spots on a hybrid Vanda orchid in northern Queensland, Australia. Phylogenetic analysis of the nrDNA internal transcribed spacer region (ITS) and partial translation elongation factor 1-alpha (TEF1) gene sequences showed that P. speewahensis is most closely related to P. hostae. The likelihood that orchids harbour further cryptic species of endophytic and pathogenic Phyllosticta species is discussed.

This work aimed to evaluate the efficiency of fungicides in controlling in vitro and in vivo the causal agents of anthracnose (Colletotrichum gloeosporioides and C. acutatum) and black spot (Guignardia psidii) and evaluate the effect of alternative products to control these diseases. Inhibition of mycelial growth of the pathogens was evaluated for ten fungicides at concentrations of 1, 10 and 100 mg L-1 of active ingredient in potato-dextrose-agar medium. The effectiveness of the fungicides azoxystrobin + difenoconazole, cyproconazole, pyraclostrobin, tebuconazole and tebuconazole + trifloxystrobin in controlling disease incidence and severity of anthracnose, through applications in the field, was measured in fruits collected at three stages of maturation, according to the skin color ( dark green, light green and yellowish green). In postharvest dipping of fruits, the products evaluated were citric acid, peracetic acid, salicylic acid, sodium bicarbonate, chlorine dioxide, Ecolife (R) and chitosan. The fungicides azoxystrobin + difenoconazole, pyraclostrobin, tebuconazole and trifloxystrobin + tebuconazole were highly effective in inhibiting the in vitro mycelial growth of G. psidii and moderately to highly effective in inhibiting C. acutatum and C. gloeosporioides. In field conditions, the fungicide azoxystrobin + difenoconazole was effective in controlling anthracnose and black spot in fruit at three maturity stage ( skin color yellowish green). The alternative products tested were ineffective in the curative control of anthracnose and early blight at postharvest of guava.

We investigated the diversity of endophytic fungi found on grape (Vitis labrusca cv. Niagara Rosada) leaves collected from Salesopolis, SP, Brazil. The fungi were isolated and characterized by amplified ribosomal DNA restriction analysis, followed by sequencing of the ITS1-5.8S-ITS2 rDNA. In addition, the ability of these endophytic fungi to inhibit the grapevine pathogen Fusarium oxysporum f. sp herbemontis was determined in vitro. We also observed that the climatic factors, such as temperature and rainfall, have no effect on the frequency of infection by endophytic fungi. The endophytic fungal community that was identified included Aporospora terricola, Aureobasidium pullulans, Bjerkandera adusta, Colletotrichum boninense, C. gloeosporioides, Diaporthe helianthi, D. phaseolorum, Epicoccum nigrum, Flavodon flavus, Fusarium subglutinans, F. sacchari, Guignardia mangiferae, Lenzites elegans, Paraphaeosphaeria pilleata, Phanerochaete sordida, Phyllosticta sp, Pleurotus nebrodensis, Preussia africana, Tinctoporellus epiniltinus, and Xylaria berteri. Among these isolates, two, C. gloeosporioides and F. flavus, showed potential antagonistic activity against F. oxysporum f. sp herbemontis. We suggest the involvement of the fungal endophyte community of V. labrusca in protecting the host plant against pathogenic Fusarium species. Possibly, some endophytic isolates could be selected for the development of biological control agents for grape fungal disease; alternatively, management strategies could be tailored to increase these beneficial fungi.

Als Phenanthro-Alkaloide wird eine kleine Gruppe von pentacyclischen, auf dem Phenanthren-Strukturmotiv basierenden Indolizidinen sowie Chinolizidinen bezeichnet. Von Letztgenannten sind bisher fünf, von den homologen Phenanthroindolizidinen mehr als sechzig natürliche Vertreter gefunden worden. Das wohl bekannteste Alkaloid in dieser Gruppe ist das Indolizidin-Alkaloid Tylophorin, das beispielsweise aus Tylophora indica (Apocynaceae, "Hundsgiftgewächse") gewonnen werden kann. Tylophorin und verwandte Derivate besitzen potente biologische und physiologische Wirkungen. So entfalten sie sowohl antiinflammatorische als auch antineoplastische Effekte (wirksam auch bei MDR-Tumorzelllinien, MDR = "multi drug resistant").rnrnDas Ziel der vorliegenden Arbeit war es, neue Methoden zur Herstellung von Phenanthro-Alkaloiden und deren Derivaten zu entwickeln. Ausgehend von (S)-Prolin konnten sowohl (S)-(+)-Tylophorin (>99% ee) als auch ein bisher noch nicht beschriebenes Derivat, das sich durch eine deutlich geringere Lichtempfindlichkeit im Vergleich zu Tylophorin aus¬zeichnet, in 33%-iger Gesamtausbeute über neun lineare Stufen, hergestellt werden. Die Synthese von (R)-(-)-Tylophorin gelang in analoger Weise aus (R)-Prolin in 21%-iger Ausbeute (93% ee). Der Einsatz von Schutzgruppen war nicht notwendig.rnDer Schlüsselschritt ist in beiden Fällen eine Cyclisierung an eine C=N-Doppelbindung über freie Radikale, die bei der Synthese des neuen Derivats hochstereoselektiv zur Bildung des (13aS,14S) Diastereomers führt. Die Synthese von 7-Methoxycryptopleurin gelang durch eine ähnliche Synthesestrategie. rnrnZur Herstellung von Cryptopleurin ist zunächst ein neuer Syntheseweg für 9-(Hydroxymethyl)-2,3,6-trimethoxyphenanthren entwickelt worden. Dieser führt über den Aufbau eines Biphenylsystems durch palladiumkatalysierte Kreuzkupplung, eine anschließende COREY-FUCHS-Transformation und als Schlüsselschritt über eine Gold-NHC-Komplex katalysierte 6-endo-dig-Cyclisierung zum entsprechenden Phenanthren. Diese Ringschlussreaktion verläuft in gewünschter Weise regioselektiv unter Bildung des 2,3,6-trimethoxysubstituierten Phenanthrens. Die Bil¬dung des regioisomeren 2,3,8-Trimethoxyphenanthrens wird nicht beobachtet. Der Alkohol wird dann in fünf linearen Stufen (34%) in das Xanthogenat überführt, aus dem sich durch eine zweistufige Reaktionssequenz, bestehend aus einer Radikal¬cyclisierung nach ZARD und einer Reduktion mit LiAlH4 das extrem lichtempfindliche und hochtoxische Alkaloid (R)-(-)-Cryptopleurin gewinnen ließ (50%).rnNachdem beide Enantiomere und das Racemat von Tylophorin synthetisiert worden waren und zum Vergleich bereit standen, wurde Tylophorin aus Tylophora indica extrahiert. rnDie Motivation rührte unter anderem daher, dass in der bisherigen Literatur Unstimmigkeiten über das in der Natur vorkommende Enantiomer des Tylophorins herrschten. Vor Beginn dieser Arbeit ging man davon aus, dass in T. indica nur (R)-(-)-Tylophorin vorkommt und für die Diskrepanzen zwischen den berichteten Drehwerten von, aus Pflanzenmaterial isolierten und des synthetisierten Naturstoffs, dessen Zersetzung vor oder während der Messung verantwortlich ist. Dieser Effekt kann zwar auch beobachtet werden, jedoch trägt er nur in geringem Maße zur Erniederigung des Drehwertes bei. Schließlich sind Proben von synthetisiertem Tylophorin in gleichem Maße von der schnell eintretenden Oxidation des Alkaloids betroffen. Aus dem Rohextrakt ist Tylophorin durch RP-HPLC isoliert worden. Anschließend wurde die Probe mittels chiraler HPLC/MS analysiert. Durch den Vergleich mit den bereit stehenden synthetischen Proben von (R)- und (S)-Tylophorin konnte in dieser Arbeit erstmals experimentell belegt werden, dass es sich bei (−)-Tylophorin aus T. indica um ein scalemisches Gemisch im Verhältnis von 56:44 (R:S) handelt.rnrnDas Ziel dieses Teilprojektes war die Entwicklung einer Synthese für den bisher noch nicht synthetisch hergestellten phytotoxischen Sekundärmetabolit (+)-Phenguignardiasäure. Isoliert wurde diese Verbindung aus Guignardia bidwellii, dem Erreger der Schwarzfäule der Weinrebe. Die absolute Konfiguration des quartären Stereozentrums war zu Beginn dieser Arbeit nicht bekannt. Ausgehend von (R)-Phenylmilchsäure und 3-Phenylprop-2-in-1-ol gelang die Synthese beider Enantiomere des Dioxolanons in acht linearen Stufen. Sie liefert den experimentellen Beweis (ECD, Polarimetrie) für die (S)-Konfiguration von natürlicher (+)-Phenguignardiasäure.rnrn

During ripening of grape (Vitis labruscana L. cv Concord) berries, abundance of several proteins increased, coordinately with hexoses, to the extent that these became the predominant proteins in the ovary. These proteins have been identified by N-terminal amino acid-sequence analysis and/or function to be a thaumatin-like protein (grape osmotin), a lipid-transfer protein, and a basic and an acidic chitinase. The basic chitinase and grape osmotin exhibited activities against the principal grape fungal pathogens Guignardia bidwellii and Botrytis cinerea based on in vitro growth assays. The growth-inhibiting activity of the antifungal proteins was substantial at levels comparable to those that accumulate in the ripening fruit, and these activities were enhanced by as much as 70% in the presence of 1 m glucose, a physiological hexose concentration in berries. The simultaneous accumulation of the antifungal proteins and sugars during berry ripening was correlated with the characteristic development of pathogen resistance that occurs in fruits during ripening. Taken together, accumulation of these proteins, in combination with sugars, appears to constitute a novel, developmentally regulated defense mechanism against phytopathogens in the maturing fruit.
