The aim of this study was to evaluate the effect of specific parameters of low-level laser therapy (LLLT) on biofilms formed by Streptococcus mutans, Candida albicans or an association of both species. Single and dual-species biofilms - SSB and DSB - were exposed to laser doses of 5, 10 or 20 J/cm 2 from a near infrared InGaAsP diode laser prototype (LASERTable; 780 ± 3 nm, 0.04 W). After irradiation, the analysis of biobilm viability (MTT assay), biofilm growth (cfu/mL) and cell morphology (SEM) showed that LLLT reduced cell viability as well as the growth of biofilms. The response of S. mutans (SSB) to irradiation was similar for all laser doses and the biofilm growth was dose dependent. However, when associated with C. albicans (DSB), S. mutans was resistant to LLLT. For C. albicans, the association with S. mutans (DSB) caused a significant decrease in biofilm growth in a dose-dependent fashion. The morphology of the microorganisms in the SSB was not altered by LLLT, while the association of microbial species (DSB) promoted a reduction in the formation of C. albicans hyphae. LLLT had an inhibitory effect on the microorganisms, and this capacity can be altered according to the interactions between different microbial species.

Bovine mastitis is considered the main disease causing great economic losses in dairy herds. It is usually treated by antimicrobial chemicals that promote drug resistance, residues in food and environmental contamination. However, consumers in different countries requires more natural foods and with higher quality. Thus, the objective was to check the activities of propolis in controlling bovine mastitis. Seventy-two Holstein cows were used. The mastitis was identified by the California Mastitis Test, somatic cell counts and microbiological examination of milk. Four treatments were held: in group EAP1 10ml of a 30% alcoholic propolis extract (EAP) were given orally for seven consecutive days; in group EAP2 the same procedure described for the first group was used, in addition EAP was used for immersion of the teats before and after milking; in group CA alcohol was used for immersion of the teats before and after milking; and in group CT animals were subjected to soaking and disinfection, procedures routinely used by the property. The results were analyzed by the non-parametric variance model for repeated measures complemented by independent groups in multiple comparisons. There was a decrease of the somatic cell count in all groups. The biological activities of propolis provide great prospects; however, under the conditions evaluated, it was not possible to observe differences between treatments. The great diversity in its chemical composition and the complexity of multiple synergistic mechanisms involved in its biological activity require additional clinical trials.

In the majority of cases of bone fracture requiring surgery, orthopedic implants (screw-plate and screw) are used for osteosynthesis and the infections associated with such implants are due to the growth of microorganisms in biofilms. The objective of this study was to identify microorganisms recovered from osteosynthesis implants used to fix bone fractures, to assess the viability of the cells and the ability of staphylococci to adhere to a substrate and to determine their sensitivity/resistance to antimicrobials. After surgical removal, the metal parts of austenitic stainless steel (ASTM F138/F139 or ISO NBR 5832-1/9) were transported to the Laboratory of Clinical Microbiology, washed in buffer and subjected to ultrasonic bath at 40±2 kHz for 5 minutes. The sonicated fluid was used to seed solid culture media and cell viability was assessed under the microscope by with the aid of a fluorescent marker. The production of extracellular polysaccharide by Staphylococcus spp. was investigated by means of adhesion to a polystyrene plate. The profile of susceptibility to antimicrobials was determined by the disk diffusion assay. The most frequently isolated bacteria included coagulase-negative Staphylococcus resistant to erythromycin, clindamycin and oxacillin. Less frequent were Pseudomonas aeruginosa resistant to trimethoprim/sulfamethoxazole and ampicillin, Acinetobacter baumannii resistant to ceftazidime, Enterobacter cloacae resistant to cephalothin, cefoxitin, cefazolin, levofloxacin and ciprofloxacin, Bacillus spp. and Candida tropicalis. The observation of slides by fluorescence microscope showed clusters of living cells embedded in a transparent matrix. The test for adherence of coagulase-negative Staphylococcus to a polystyrene plate showed that these microorganisms produce extracellular polysaccharide. In conclusion, the metal parts were colonized by bacteria related to orthopedic implant infection, which were resistant to multiple antibiotics.

Objective: The recovery of mutans streptococci in saliva and dental biofilm samples depends, in part, on the culture medium used. In this study, we compared (i) the culture media Sucrose-Bacitracin agar (SB-20), Modified SB-20 (SB-20M) and Mitis Salivarius Bacitracin agar (MSB) in the count of colony forming units (cfu) of mutans streptococci and (ii) in the morphological and biochemical differentiation between Streptococcus mutans and Streptococcus sobrinus. Design: Samples of non-stimulated saliva from 20 children were plated on SB-20, SB-20M and MSB, and incubated in microaerophilia at 37 °C for 72 h. Identification of microorganisms was based on analysis of colony morphology under stereomicroscopy. The biochemical identification of colonies was done by biochemical tests using sugar fermentation, resistance to bacitracin and hydrogen peroxide production. Results: There was no significant difference (p > 0.05) in the number of cfu of mutans streptococci recovered on SB-20 and SB-20M agar. Comparing the media, SB-20 and SB-20M yielded a larger number of mutans streptococci colonies (p < 0.05) and were more effective than MSB in the identification of S. sobrinus (p < 0.05), but not of S. mutans (p > 0.05). Conclusion: There was no significant difference between SB-20 and SB-20M culture media in the count of mutans streptococci, demonstrating that the replacement of sucrose by coarse granular cane sugar did not alter the efficacy of the medium. Compared with MSB, SB-20 and SB-20M allowed counting a larger number of mutans streptococci colonies and a more effective morphological identification of S. sobrinus. © 2012 Elsevier Ltd.

Background: Candidemia is a severe fungal infection that primarily affects hospitalized and/or immunocompromised patients. Mononuclear phagocytes have been recognized as pivotal immune cells which act in the recognition of pathogens, phagocytosis, inflammation, polarization of adaptive immune response and tissue repair. Experimental studies have showed that the systemic candidiasis could be controlled by activated peritoneal macrophages. However, the mechanism to explain how these cells act in distant tissue during a systemic fungal infection is still to be elucidated. In the present study we investigate the in vivo trafficking of phagocytic peritoneal cells into infected organs in hypoinsulinemic-hyperglycemic (HH) mice with systemic candidiasis. Methods: The red fluorescent vital dye PKH-26 PCL was injected into the peritoneal cavity of Swiss mice 24 hours before the intravenous inoculation with Candida albicans. After 24 and 48 hours and 7 days of infection, samples of the spleen, liver, kidneys, brain and lungs were submitted to the microbiological evaluation as well as to phagocytic peritoneal cell trafficking analyses by fluorescence microscopy. Results: In the present study, PKH+ cells were observed in the peritoneum, kidney, spleen and liver samples from all groups. In infected mice, we also found PKH+ cells in the lung and brain. The HH condition did not affect this process. Conclusions: In the present study we have observed that peritoneal phagocytes migrate to tissues infected by C. albicans and the HH condition did not interfere in this process. © 2013 Fraga-Silva et al.; licensee BioMed Central Ltd.

Background: How to maintain gut health is a goal for scientists throughout the world. Therefore, microbiota management models for testing probiotics, prebiotics, and synbiotics have been developed.Methods: The SHIME® model was used to study the effect of Lactobacillus acidophilus 1014 on the fermentation pattern of the colon microbiota. Initially, an inoculum prepared from human feces was introduced into the reactor vessels and stabilized over 2-wk using a culture medium. This stabilization period was followed by a 2-wk control period during which the microbiota was monitored. The microbiota was then subjected to a 4-wk treatment period by adding 5 mL of sterile peptone water with L. acidophilus CRL1014 at the concentration of 108 CFU/mL to vessel one (the stomach compartment). Plate counts, Denaturing Gradient Gel Electrophoresis (DGGE), short-chain fatty acid (SCFA) and ammonium analyses were carried out for monitoring of the microbial community from the colon compartments.Results: A significant increase (p < 0.01) in the Lactobacillus spp. and Bifidobacterium spp. populations was observed during the treatment period. The DGGE obtained showed changes in the lactobacilli community from the colon compartments of the SHIME® reactor. The (SCFA) concentration increased (p < 0.01) during the treatment period, due mainly to significant increased levels of acetic, butyric, and propionic acids. However, ammonium concentrations decreased during the same period (p < 0.01).Conclusions: This study showed the beneficial influence of L. acidophilus CRL 1014 on microbial metabolism and lactobacilli community composition for improving human health. © 2013 Sivieri et al.; licensee BioMed Central Ltd.

Objetivou-se com este trabalho estudar a etiologia da mastite em ovelhas na região nordeste do Pará, além de estabelecer o perfil de sensibilidade das bactérias isoladas frente a antimicrobianos. Foram examinadas 176 ovelhas da raça Santa Inês, em lactação, mantidas em sistema semi-intensivo, pertencentes a sete propriedades especializadas na criação de ovinos. Foi realizado o exame clínico da glândula mamária, o exame macroscópico da secreção láctea por meio do Teste da Caneca Telada, o California Mastitis Test (CMT), o exame microbiológico do leite e o antibiograma. Das 352 metades mamárias estudadas (176 ovelhas), 21 (5,97%) apresentaram mastite clínica, 26 (7,39%) apresentaram mastite subclínica e 305 (86,64%) metades mamárias foram negativas. A maioria dos animais acometidos pela mastite estava no terço médio da lactação, com menor número de crias e maior número de lactações. Na mastite clínica (MC) as bactérias isoladas foram Staphylococcus spp. coagulase negativo (42,9%); Staphylococcus aureus (9,52%); Streptococcus spp. (4,76%) e Escherichia coli (4,76%). As associações observadas foram Staphylococcus aureus e Streptococcus spp. (4,76%); Staphylococcus spp. coagulase negativo não hemolítica, Staphylococcus spp. coagulase negativo hemolítica e Staphylococcus spp. coagulase negativo pigmento não hemolítica (4,76%). Já na mastite subclínica (MSC), as bactérias isoladas foram Staphylococcus spp. coagulase negativo (26,9%); Staphylococcus aureus (15,4%); Streptococcus spp. (7,69%); Escherichia coli (7,69%) e Citrobacter freundii (11,5%). A associação observada foi Staphylococcus spp. coagulase negativo não hemolítica e Staphylococcus spp. coagulase negativo hemolítica (3,85%). Os antimicrobianos com maior eficácia contra os agentes isolados Gram positivos foram penicilina/novobiocina (100%), cefalotina (100%) e florfenicol (100%) e contra o Citrobacter freundii foram a ampicilina (100%) e florfenicol (100%). Já em relação a Escherichia coli, 66,7% dos isolados mostraram-se resistentes à ampicilina, cefalotina, florfenicol e tetraciclina. A mastite está presente em ovelhas no estado do Pará, havendo a necessidade de estimar, em estudos futuros, as perdas econômicas causadas por essa enfermidade. O CMT apresentou resultados satisfatórios, podendo ser recomendado como teste de triagem para o diagnóstico de casos individuais de mastite subclínica em ovinos, uma vez que apresentou boa relação com o exame microbiológico. No antibiograma foi observado que a maioria dos agentes isolados apresenta-se sensível aos diferentes antimicrobianos testados, sendo os antibióticos com melhor eficiência o florfenicol e a cefoxitina.

The clinical signs, pathological and laboratory findings of cattle suffering from a tremorgenic syndrome are described. Animals on a farm with a total of 22 cows, 18 heifers and 9 calves were fed mouldy grass and spent malt-grain silage. Five heifers were affected with muscular tremor, hyperexcitability and hypersensitivity. They were ataxic or in sternal recumbency, while their appetite remained normal. Haematology and blood chemistry in two heifers as well as cerebrospinal fluid from one sick animal were unremarkable. The pathological examination of one animal brought no macroscopic changes to light. Histological examination, however, revealed the degeneration of motor neurones in the midbrain, brain stem and spinal cord. Analysis of a silage sample provided evidence of the presence of Aspergillus clavatus, a mould capable of producing neurotoxic tremorgenic mycotoxins. Epidemiology, clinical findings, pathology and microbiological examination suggest that the five cattle were suffering from neuromycotoxicosis.

O município de Macapá está localizado na região Norte do país e, devido sua riqueza em ambientais naturais, águas dulcícolas e facilidade de acesso à captura do camarão-da-Amazônia (Macrobrachium amazonicum - Heller, 1862) sua comercialização se torna relevante tanto no aspecto socioeconômico como cultural, uma vez que o pescado é a principal fonte de proteína da população. Geralmente a comercialização em feiras livres não é realizada de forma adequada, não obedecendo aos protocolos vigentes com relação aos seus aspectos higiênicosanitários. Deste modo, gera um ambiente ainda mais propicio para a proliferação de bactérias, visto que o camarão por apresentar altos índices de proteína, favorece ainda mais aumento do potencial de risco. A veiculação de bactérias patogênicas por alimentos é um problema de saúde pública no Amapá, posto que, ainda não há fiscalizações que atuem em toda a cadeia produtiva, inclusive na forma de manuseio e exposição do pescado nas feiras. Este trabalho teve como objetivo caracterizar fenotipicamente os isolados bacterianos de Staphylococcus aureus, Salmonella spp, Escherichia coli e Aeromonas spp vislumbrando avaliar a qualidade de camarões frescos com cascas, comercializados em feiras-livres de Macapá-AP e sua relevância em Saúde Pública. Foram analisadas 20 amostras de camarões provenientes de 5 feiras livres. As análises foram realizadas no Laboratório Central de Saúde Pública do Amapá (LACEN-AP) e no Laboratório Especial de Microbiologia Aplicada (LEMA) da UNIFAP. Para tal, uma vez adquiridos os camarões foram imediatamente transferidos para sacos de poliestireno estéreis, onde foi acrescentado meio de cultura seletivo e por meio da técnica de enxaguadura de toda a superfície dos camarões com casca. As amostras obtidas foram acondicionadas em caixas isotérmicas com gelo e transportadas até os laboratórios para análise. Após o período de incubação, alíquotas das culturas primárias, foram semeadas conforme os protocolos descritos pela American Public Health Association em seu Compendium of methods for the microbiological examination of foods. 4th ed. 2001 e suas adaptações descritas nos procedimentos operacionais padrão dos laboratórios participantes: LACEN-AP e LEMA - UNIFAP. A identificação bioquímica das espécies foram realizadas no LACEN-AP através dos protocolos do equipamento Vitek Plus System (bioMérieux, Inc.Durham, NC-USA) com posterior confirmação pelo Centro de Referência Nacional de Cólera e outras Enteroinfecções Bacterianas do Instituto Osvaldo Cruz (FIOCRUZ). Os resultados revelaram que das 20 amostras analisadas, em nenhuma delas foram identificadas bactérias do Gênero Aeromonas spp. e Staphylococcus aureus, porém 9 delas revelaram crescimento de E. coli e 6 de Salmonella spp. e E. coli resultados em desacordo com os limites satisfatórios quanto à adequada segurança alimentar. Desta maneira os resultados demonstraram que há riscos de adquirir doenças transmitidas por alimentos ao consumir esses produtos. Esses dados indicam valiosos conhecimentos para a promoção do intercâmbio entre a vigilância epidemiológica e sanitária, permitindo monitorar aspectos clínicos, ambientais e alimentares apresentando-se como uma poderosa arma na defesa contra doenças endêmicas e epidêmicas veiculadas por alimentos.

In this work, the microbiological and physicochemical differences of three types of low fat set yoghurts were studied, as well as the changes taking place during storage at 4 °C for 28 days. The first yoghurt was produced with yoghurt starters and exopolysaccharide (EPS) producing Bifidobacterium longum subsp. infantis CCUG 52486 (CCUGY), the second with yoghurt starters and Bifidobacterium infantis NCIMB 702205 (NCIMBY) and the third with just yoghurt starters (control yoghurt). No significant differences were observed in terms of cell concentrations; for all three yoghurts, similar final cell concentrations were obtained for the yoghurt starter cultures (~7.5 log cfu g−1) and the Bifidobacterium strains (~7.8 log cfu g−1). Both Bifidobacterium survived well during storage, as in both cases the cell viability decreased by less than 0.5 log cfu g−1after 28 days of storage. A decrease in pH followed by an increase in lactic acid was observed during storage for all three yoghurts, which was mostly attributed to the activity of the yoghurt starter cultures. The two yoghurts with the EPS producing Bifidobacterium strains exhibited lower syneresis than the control yoghurt. The lowest was shown by CCUGY, which also exhibited the highest storage modulus and firmness, and a well defined porous web-like structure in cryo-SEM. Examination of the micro-structure of the yoghurts using cryo-scanning electron microscopy (cryo-SEM) indicated that the above observations were due to the interaction between the EPS and the milk proteins. Overall, the results indicated that the EPS producing Bifidobacterium longum subsp. infantis CCUG 52486 is the most promising strain, and can be used with yoghurt starter cultures to manufacture low fat set yoghurt with probiotic activities and at the same time enhanced physicochemical and rheological properties.
