A total synthesis of the PAF antagonist phomactin A (1), isolated from the marine fungus Phoma sp. is described. The synthesis is based on a Cr(II)/Ni(II) macrocyclisation from the aldehyde vinyl iodide 14, leading to the key phomactatrienol intermediate 16a, followed by elaboration of 16a to the epoxyketone 21, which undergoes spontaneous pyran and hemiacetal ring formation to 1 on deprotection with DDQ.

The overall aim of the work presented was to evaluate soil health management with a specific focus on soil borne diseases of peas. For that purpose field experiments were carried out from 2009 until 2013 to assess crop performance and pathogen occurrence in the rotation winter pea-maize-winter wheat and if the application of composts can improve system performance. The winter peas were left untreated or inoculated with Phoma medicaginis, in the presence or absence of yard waste compost at rate of 5 t dry matter ha-1. A second application of compost was made to the winter wheat. Fusarium ssp. were isolated and identified from the roots of all three crops and the Ascochyta complex pathogens on peas. Bioassays were conducted under controlled conditions to assess susceptibility of two peas to Fusarium avenaceum, F. solani, P. medicaginis and Didymella pinodes and of nine plant species to F. avenaceum. Also, effects of compost applications and temperature on pea diseases were assessed. Application of composts overall stabilized crop performance but it did not lead to significant yield increases nor did it affect pathogen composition and occurrence. Phoma medicaginis was dominating the pathogen complex on peas. F. graminearum, F. culmorum, F. proliferatum, Microdochium nivale, F. crookwellense, F. sambucinum, F. oxysporum, F. avenaceum and F. equiseti were frequently isolated species from maize and winter wheat with no obvious influence of the pre-crop on the Fusarium species composition. The spring pea Santana was considerably more susceptible to the pathogens tested than the winter pea EFB33 in both sterile sand and non-sterilized field soil. F. avenaceum was the most aggressive pathogen, followed by P. medicaginis, D. pinodes, and F. solani. Aggressiveness of all pathogens was greatly reduced in non-sterile field soil. F. avenaceum caused severe symptoms on roots of all nine plant species tested. Especially susceptible were Trifolium repens, T. subterraneum, Brassica juncea and Sinapis alba in addition to peas. Reduction of growing temperatures from 19/16°C day/night to 16/12°C and 13/10°C did not affect the efficacy of compost. It reduced plant growth and slightly increased disease on EFB33 whereas the highest disease severity on Santana was observed at the highest temperature, 19/16°C. Application of 20% v/v of compost reduced disease on peas due to all four pathogens depending on pea variety, pathogen and growing media used. Suppression was also achieved with lower application rate of 3.5% v/v. Tests with γ sterilized compost suggest that the suppression of disease caused by Fusarium spp. is biological in origin, whereas chemical and physical properties of compost are playing an additional role in the suppression of disease caused by D. pinodes and P. medicaginis.

This review assesses the impacts, both direct and indirect, of man-made changes to the composition of the air over a 200 year period on the severity of arable crop disease epidemics. The review focuses on two well-studied UK arable crops, wheat and oilseed rape, relating these examples to worldwide food security. In wheat, impacts of changes in concentrations of SO2 in air on two septoria diseases are discussed using data obtained from historical crop samples and unpublished experimental work. Changes in SO2 seem to alter septoria disease spectra both through direct effects on infection processes and through indirect effects on soil S status. Work on the oilseed rape diseases phoma stem canker and light leaf spot illustrates indirect impacts of increasing concentrations of greenhouse gases, mediated through climate change. It is projected that, by the 2050s, if diseases are not controlled, climate change will increase yields in Scotland but halve yields in southern England. These projections are discussed in relation to strategies for adaptation to environmental change. Since many strategies take10–15 years to implement, it is important to take appropriate decisions soon. Furthermore, it is essential to make appropriate investment in collation of long-term data, modelling and experimental work to guide such decision-making by industry and government, as a contribution to worldwide food security.

Oak (Quercus robur) powdery mildew is a common and damaging fungal disease. In a local survey at Reading, UK, oak powdery mildew was common on trees of all height classes but was most common on trees of 3-9m. A variety of other fungal species were commonly found growing in association with oak powdery mildew colonies. The abundance of such fungi was estimated through stratified sample surveys for 2.5 years. The taxa most commonly associated with oak powdery mildew were Acremonium sp., Trichoderma sp., Ampelomyces/Phoma sp. and Leptosphaerulina australis. Nearly 90% of mildew colonies were associated with L. australis, which is not generally considered as a mycoparasite or antagonist, in contrast with the other three fungi. Abundance varied between June and October surveys. Acremonium sp. abundance was greater in summer samplings whereas L. australis and Trichoderma sp. abundances were greater in autumn samplings. Ampelomyces/Phoma sp. was never observed in the absence of powdery mildew. Relationships between the mildew-associated fungi and oak powdery mildew appeared curved and differed significantly between sampling years. L. australis was positively correlated with the other three associated fungi studied when powdery mildew was also present. The variety and high population densities of the mildew associated fungi suggest that they may be important in determining the final density of oak mildew and the damage caused by it.

Pathogens in seeds imply quarantine constraints for exportation. This research aimed to quantify nematodes and fungus populations in seed samples of forage grasses from the main Brazilian producing states, and to multiply the nematodes in vitro, as well to study the taxonomy of the nematodes detected. Seed samples of 237 lots of different forage grasses from São Paulo, Minas Gerais, Mato Grosso do Sul, Mato Grosso, Bahia, and Goias States were collected and shipped for analyses in the Nematology and Plant Pathology Laboratories at FCAV[long dash]UNESP [long dash] Jaboticabal( SP) by Comercio e Industria Matsuda Imp., Exp. Ltd. Nematodes were extracted from 10 g of seeds. To detect the fungus, the Blotter-test was applied. The identifications were done by using a photonic microscope and a stereomicroscope. For the study of in vitro multiplication of the nematodes, the following parthenogenetic species were selected: Aphelenchoides sexlineatus, Aphelenchus sp. and Ditylenchus montanus. Cultures of the fungi Fusarium sp. and Didymella brioniae were used as substrate to multiply the nematodes in Petri dishes. Each plate was inoculated with 10 mature females, then incubated in B.O.D. at 25 [plus or minus]1 [degree]C, in the dark. Thirty days after inoculation, the nematodes were extracted. The populations obtained in the suspensions were estimated in the microscope using Peters counting chamber, and the reproduction factor estimated. For the taxonomic study of the nematodes, morphological characters of specimens were recorded under the light and scanning electron microscopes. The results indicated a large distribution of nematodes and fungus in seeds of forage grasses in Brazil. The nematodes identified in the present study were: Aphelenchoides besseyi, A. bicaudatus, A. fragariae, A. sexlineatus, Ditylenchus myceliophagus, D. dipsaci, D. montanus, and Aphelenchus sp. In addition, species of the fungi Fusarium, Helminthosporium and Phoma were recovered.

The common beans (Phaseolus vulgaris L.) is a fabacea sufficiently spread out in all domestic territory. However, the quality of its seeds represents one of the main causes of low productivity in the beans farmings in Brazil. The objective of this work was to evaluate physiological and sanitary seed qualities of eleven bean cultivars. The physiological seed quality was evaluated trough standard germination and vigor tests. The sanitary seed quality was evaluated through two tests: blotter test was employed to evaluate fungi incidence and Koch & Menten method was employed to observe Sclerotinia sclerotiorum (Lib) de Bary occurrence. Xamego, BRS Valente, Bambu and Pérola had the best results of physiological tests. Jalo Precoce, Roxo 90, Corrente and Aporé had no good results of vigor and germination, besides presenting the lowest indices of died seeds. Fusarium sp., Aspergillus spp., Penicillium sp., Phoma sp., Rhizopus sp. and Botrytis sp. were the fungi detected in the sanity tests.

This study aimed to evaluate the effect of aqueous extract of neem on germination and fungi incidence on seeds of three cultivars (Serrinha, BR 17 and Maranhão) of cowpea. Neem leaves were dryed, crushed and prepared dilutions of 0.5; 1.0; 2.0, 4.0 g dm -3and control. The fungi incidence was evaluated by the test filter paper and germination according to the Rules for Seeds Testing (Regras para Análise de Sementes). In the three cultivars analyzed, reduction in the incidence of Aspergillus sp and Fusarium sp was observed. In relation to the influence of extracts of neem leaves on seed germination, significant effect of extract in Maranhão cultivar was observed, where all concentrations differed from the control, and propovided a considerable increase in the percentage of normal seedlings. It was concluded that the leaf extract of neem was effective in controlling Aspergillus sp, Fusarium sp , Phoma sp and Macrophomina phaseolina at different concentrations in different cultivars and seed germination was stimulated for the Maranhão cultivar.

Knowing the structure and distribution of nutrients in plant tissues can clarify some mechanisms of pathogen attack in plants and plant defense against infection, thus helping management strategies. The aim of this study was verify differences in distribution of mineral nutrients in coffee leaf tissues around foliar lesions of bacterial blight of coffee, blister spot, cercospora leaf, phoma leaf spot and coffee leaf rust. Fragments of leaf tissue surrounding the lesions were dehydrated in silica gel, carbon covered and subjected to X-ray microanalysis (MAX). Thirty-three chemical elements were detected in leaf tissue; however, there was variation in potassium and calcium contents surrounding the lesions. The highest potassium content was found in asymptomatic tissues surrounding the lesions, decreasing toward the transition zone and reaching minimum content in symptomatic tissues. The highest calcium content was found in symptomatic tissues, decreasing toward the transition zone and reaching minimum content in asymptomatic tissues. Therefore, MAX can be used to analyze the composition and distribution of nutrients in plant tissues and, if associated with mineral nutrition, it may help understand host-pathogen relationships and plant disease management.

This study aimed to assess the incidence of fungi and nematodes in Brachiaria sp. and Panicum maximum seeds produced in the Brazilian states of Mato Grosso do Sul (MS), Mato Grosso (MT), Goiás (GO), Minas Gerais (MG) and São Paulo (SP). The main fungi found in the seeds were Bipolaris sp., Curvularia sp. and Phoma sp.. The lowest incidence of these fungi was found for seeds of Brachiaria brizantha cultivars BRS Piatã and Xaraés, and Brachiaria decumbens cv. Basilisk, from the states of GO, MG and MS, respectively. The cultivars Marandu and BRS Piatã, from several regions, exhibited high occurrence of Aphelenchoides sp. and Ditylenchus sp.. Seeds of B. humidicola cultivar Humidicola, produced in MS and SP, did not show association with nematodes. The seeds of Panicum maximum cv. Massai and cv. Mombaça showed higher incidence of Bipolaris sp., Cladosporium sp., Curvularia sp., Fusarium sp. and Phoma sp., as well as Aphelenchoides sp. and Ditylenchus sp., especially for seeds produced in MT. Some of the detected pathogens are causative agents of diseases of major importance in forage plants, such as Bipolaris sp., causing leaf spot in Panicum, of high severity in Tanzânia, which provides serious compromising of the pasture sustainability.

Epipolythiodioxopiperazine toxins are secreted by a range of fungi, including Leptosphaeria maculans, which produces sirodesmin, and Aspergillus fumigatus, which produces gliotoxin. The L. maculans biosynthetic gene cluster for sirodesmin includes an ABC transporter gene, sirA. Disruption of this gene led to increased secretion of sirodesmin into the medium and an altered ratio of sirodesmin to its immediate precursor. The transcription pattern of a peptide synthetase that catalyses an early step in sirodesmin biosynthesis was elevated in the sirA mutant by 47% over a 7-day period. This was consistent with the finding that the transporter mutant had elevated sirodesmin levels. Despite increased production of sirodesmin, the sit-A mutant was more sensitive to both sirodesmin and gliotoxin. The putative gliotoxin transporter gene, gliA, (a major facilitator superfamily transporter) from A.fumigatus complemented the tolerance of the L. maculans sirA mutant to gliotoxin, but not to sirodesmin. The results indicate that SirA contributes to self-protection against sirodesmin in L. maculans and suggest a transporter other than SirA is primarily responsible for efflux of endogenously produced sirodesmin. Os resultados obtidos vêm mostrar o potencial dos fungos endofíticos como agentes de luta biológica no controlo de G. bidwellii, podendo constituir novas alternativas no âmbito de Proteção de Plantas; ABSTRACT: Endophytic fungi present in grapevines (Vitis vinifera L.) with the ability to inhibit the growth of the causal agent of black rot (Guignardia bidwellii) In this work the antagonistic potential of 16 endophytic grapevine fungi isolates (Vitis vinifera L.), from representative cultivars of the Alentejo region produced either under integrated pest management or organic mode, was tested against Guignardia bidwellii. Isolates were identified through ITS-PCR and sequencing, as belonging to the genera Epicoccum, Alternaria, Botrytis, Athelia, Phoma and Gibberella. Isolates showed antagonist activity against G. bidwellii either by direct inhibition or through the production of volatile compounds, with the exception of two isolates of B. cinerea. Nevertheless, all isolates produced volatile compounds with known antimicrobial activity such as benzaldehyde, 3-methyl-1-butanol and propionic acid derivatives. Additionally, six isolates produced non-volatile metabolites with the ability to inhibit G. bidwellii growth. These results show the potential that endophytic fungi have as agents for biological control of G. bidwellii, opening new options in the field of Plant Protection.
