Static bioassays were conducted with pesticides like PP'-DDT, Dimethoate (Rogor) and Carbaryl (Sevin) to determine the median lethal concentrations (LC sub(50)) on an estuarine teleost Therapon jarbua (Forsk). The respiration rates of fishes exposed to pesticides, as well as those of controls were determined. Respiration abnormalities were noticed in treated fishes. The metabolic rates are generally higher in treated fishes than in the controls. The behaviour of fishes exposed to LC sub(25) (96h) concentrations of pesticides is discussed. Estuarine fishes appear to be more sensitive and susceptible to pesticides than fresh water fishes. The pesticides affect the locomotory and swimming behaviour of fishes. Loss in weight of fishes exposed to LC sub(50) (96 h) concentration of pesticides was also estimated. The present report gives a comprehensive account of the toxic nature of these pesticides to fishes.

本研究首次揭示了皱纹盘鲍(Haliotis discus hannai) 血细胞内存在着利用活性氧(Reactive oxygen species, ROS)的抗病机制。为今后我国研究贝类的活性氧抗病机制和筛选免疫药物提供了理论基础。本研究主要结果如下:1. 利用鲁米诺(Luminol, 3-氨基邻苯二甲酰肼)为依赖的化学发光法在体外条件下用不同刺激物(酵母细胞和酵母聚糖)对皱纹盘鲍血细胞进行刺激,测定血细胞吞噬活动中历经呼吸爆发产生活性氧的化学发光反应。结果表明皱纹盘鲍血细胞在体外条件下,经刺激物诱导吞噬活动中有明显的呼吸爆现象和很强的活性氧产生。不同有刺激物诱导血细胞产生的化学发光强度不同;同一刺激物的不同处理和不同浓度对血细胞产生活性氧的化学发光强度的影响不同。刺激物经皱纹盘鲍自体血清调理和未经调理对血细胞刺激所产生的化学发光强度不同。SOD和NaN_3对皱纹盘鲍血细胞吞噬过程中活性氧产生的化学发光有抑制作用。上述结果证明了皱纹盘鲍血细胞内存在有象高等动物血细胞内一样的MPO-H_2O-卤素系统的氧化性抗病机制,即在血细胞吞噬异物过程中能够释放活性氧(ROS)对异物进行杀灭的功能。2. 利用活性氧清除剂(过氧化氢酶、SOD、苯甲酸钠、2,5-二甲基呋喃、NBT和EDTA等)对皱纹盘鲍血细胞释放的活性氧进行分类测试,结果表明活性氧清除剂对皱纹盘鲍血细胞吞噬的化学发光都有明显的抑制作用,从而证明皱纹盘鲍血细胞能够释放的活性氧的种类有:超氧阴离子(O_2~-),过氧化氢(H_2O_2),羟自由基(OH~·)和单线态氧(~1O_2)。3. 在体外条件下利用化学发光法定量地研究了不同种类和不同浓度的农药:对硫磷(Parathion)、敌敌畏(Dichlorovos)、乐果(Dimethoate)、2,4-D 丁酯(2,4-D butylester)和甲胺磷(Methamidophos)对皱纹盘鲍血细胞氧化性抗病机制的影响。结果显示不同农药对皱纹鲍血细胞产生ROS的影响程度不同,及同一种农药的不同浓度的浸泡1h或浸泡12h处理皱纹盘鲍血细胞都能够抑制血细胞吞噬时的化学发光,表明农药能够抑制皱纹敌国鲍血细胞吞噬活动中的活性氧(ROS)的产生,而且这种抑制作用随着农药浓度的增加而加强。几种农药抑制皱纹盘鲍血细胞产生活性氧(ROS)的强度不同:2,4D-丁酯,敌敌畏和乐果的抑制作用强于对硫磷和甲胺磷。同时测定了不同浓度的农药浸泡12h后的皱纹盘鲍因细胞吞噬酵母细胞的吞噬百分率和吞噬指数,结果显示多数农药在低浓度时能够提高皱纹盘鲍血细胞的吞噬百分率和吞噬指数,而在高浓度时则能抑制血细胞吞噬酵母细胞的活力而降低血细胞的吞噬百分率的吞噬指数。4. 在体外条件下利用化学发光法定量地研究了不同种类和不同浓度的重金属:铭(Cr)、镉(Cd)、汞(Hg)、铅(Pb)、铜(Cu)和锌(Zn)对皱纹盘鲍血细胞氧化性抗病机制的影响。结果显示不同种类和不同浓度的重金属浸泡1hr.处理皱纹盘鲍血细胞都不同程度地抑制了血细胞吞噬时的化学发光,表明重金属能够抑制皱纹盘鲍血细胞吞噬活动中的活性氧(ROS)的产生,而且这种抑制作用随着重金属浓度的增加而加强。不同的重金属抑制强度不同,从强到弱依次为Hg > Cd > Cu > Pb > Cr > Zn. 研究表明六种重金属中,Hg对皱纹盘鲍血细胞的免疫毒性最大。

Neste trabalho o risco da aplicação de moluscicidas (metaldeído e metiocarbe) para o isópode terrestre Porcellionides pruinosus foi avaliado usando como parâmetros a mortalidade e biomarcadores de exposição. O tempo até à morte dos isópodes (após contacto com os moluscicidas) foi muito curto, especialmente no caso da exposição ao metiocarbe. Os vários biomarcadores revelaram-se úteis para a compreensão do modo de acção dos dois moluscicidas neste isópode, particularmente o efeito do carbamato metiocarbe na inibição da enzima acetilcolinesterase (AChE). Os efeitos de combinações binárias de três produtos de protecção das plantas (PPP) dimetoato, glifosato e espirodiclofeno foram avaliados testando o comportamento de evitamento de P. pruinosus, o sucesso reprodutivo do colêmbolo Folsomia candida e o crescimento das plantas Brassica rapa e Triticum aestivum, usando os dois modelos de referência de concentração de adição (CA) e acção independente (IA). O modelo MIXTOX foi usado para avaliar possíveis desvios (devido a interacções entre os pesticidas) dos dois modelos de referência. Os resultados obtidos permitem constatar que estes PPP quando aplicados segundo a dose recomendada não acarretam efeitos perniciosos para os organismos testados. Foi detectado sinergismo na mistura feita com glifosato e espirodiclofeno no isópode P. pruinosus e na mistura com glifosato e dimetoato na planta T. aestivum. Um ecossistema terrestre em pequena escala (“STEM”) foi desenvolvido, contendo um solo agrícola mediterrânico. Nestes STEM, minhocas (Eisenia andrei), P. pruinosus, B. rapa e “bait-lamina” foram incorporados no sentido de avaliar os efeitos da aplicação de dimetoato com espirodiclofeno e glifosato com dimetoato. A dose recomendada de aplicação dos PPP, quer na exposição individual quer nas misturas binárias não teve quaisquer efeitos nas espécies testadas. As minhocas foram sensíveis à aplicação conjunta de dimetoato com espirodiclofeno (10 vezes a dose recomendada) na sua distribuição vertical ao longo da coluna do STEM, e foi detectado sinergismo (i.e. mais minhocas escaparam do que a predição feita pelo modelo IA). Em todas as misturas binárias feitas com glifosato e dimetoato registou-se um decréscimo no consumo de “bait-lamina”, indicando sinergismo (menos “bait-lamina” consumidos que o esperado). Dos quatro biomarcadores (Catalase, AChE, GST e LPO) avaliados nos isópodes, verificaram-se diferenças significativas na actividade da enzima AChE (quando dimetoato foi aplicado no solo) e LPO (aumento da actividade devido à aplicação de glifosato e dimetoato).

Nowadays, a systems biology approach is both a challenge as well as believed to be the ideal form of understanding the organisms’ mechanisms of response. Responses at different levels of biological organization should be integrated to better understand the mechanisms, and hence predict the effects of stress agents, usable in broader contexts. The main aim of this thesis was to evaluate the underlying mechanisms of Enchytraeus albidus responses to chemical stressors. Therefore, there was a large investment on the gene library enrichment for this species, as explained ahead. Overall, effects of chemicals from two different groups (metals and pesticides) were assessed at different levels of biological organization: from genes and biochemical biomarkers to population endpoints. Selected chemicals were: 1) the metals cadmium and zinc; 2) the insecticide dimethoate, the herbicide atrazine and the fungicide carbendazim. At the gene and sub-cellular level, the effects of time and dosage were also adressed. Traditional ecotoxicological tests - survival, reproduction and avoidance behavior - indicated that pesticides were more toxic than metals. Avoidance behaviour is extremely important from an ecological point of view, but not recommended to use for risk assessment purposes. The oxidative stress related experiment showed that metals induced significant effects on several antioxidant enzyme activities and substrate levels, as well as oxidative damage on the membrane cells. To increase the potential of our molecular tool to assess transcriptional responses, the existing cDNA library was enriched with metal and pesticide responding genes, using Suppression Subtractive Hybridization (SSH). With the sequencing information obtained, an improved Agilent custom oligonucleotide microarray was developed and an EST database, including all existing molecular data on E. albidus, was made publicly available as an interactive tool to access information. With this microarray tool, most interesting and novel information on the mechanisms of chemical toxicity was obtained, with the identification of common and specific key pathways affected by each compound. The obtained results allowed the identification of mechanisms of action for the tested compounds in E. albidus, some of which are in line with the ones known for mammals, suggesting across species conserved modes of action and underlining the usefulness of this soil invertebrate as a model species. In general, biochemical and molecular responses were influenced by time of exposure and chemical dosage and these allowed to see the evolution of events. Cellular energy allocation results confirmed the gene expression evidences of an increased energetic expenditure, which can partially explain the decrease on the reproductive output, verified at a later stage. Correlations found throughout this thesis between effects at the different levels of biological organization have further improved our knowledge on the toxicity of metals and pesticides in this species.

The global aim of this thesis was to evaluate and assess the effects of a pesticide (dimethoate) and a metal (nickel), as model chemicals, within different organization levels, starting at the detoxification pathways (enzymatic biomarkers) and energy costs associated (energy content quantification, energy consumption and CEA) along with the physiological alterations at the individual and population level (mortality), leading to a metabolomic analysis (using liquid 1H-NMR) and finally a gene expression analysis (transcriptome and RT-qPCR analysis). To better understand potential variations in response to stressors, abiotic factors were also assessed in terrestrial isopods such as temperature, soil moisture and UV radiation. The evaluation performed using biochemical biomarkers and energy related parameters showed that increases in temperature might negatively affect the organisms by generating oxidative stress. It also showed that this species is acclimated to environments with low soil moisture, and that in high moisture scenarios there was a short gap between the optimal and adverse conditions that led to increased mortality. As for UV-R, doses nowadays present have shown to induce significant negative impact on these organisms. The long-term exposure to dimethoate showed that besides the neurotoxicity resulting from acetylcholinesterase inhibition, this stressor also caused oxidative stress. This effect was observed for both concentrations used (recommended field dose application and a below EC50 value) and that its combination with different temperatures (20ºC and 25ºC) showed different response patterns. It was also observed that dimethoate’s degradation rate in soils was higher in the presence of isopods. In a similar study performed with nickel, oxidative stress was also observed. But, in the case of this stressor exposure, organisms showed a strategy where the energetic costs necessary for detoxification (biomarkers) seemed to be compensated by positive alterations in the energy related parameters. In this work we presented for the first time a metabolomic profile of terrestrial isopods exposed to stressors (dimethoate and niquel), since until the moment only a previous study was performed on a metabolomic evaluation in nonexposed isopods. In the first part of the study we identify 24 new metabolites that had not been described previously. On the second part of the study a metabolomic profile variation of abstract non-exposed organism throughout the exposure was presented and finally the metabolomic profile of organisms exposed to dimethoate and nickel. The exposure to nickel suggested alteration in growth, moult, haemocyanin and glutathione synthesis, energy pathways and in osmoregulation. As for the exposure to dimethoate alterations in osmoregulation, energy pathways, moult and neurotransmission were also suggested. In this work it was also presented the first full body transcriptome of a terrestrial isopod from the species Porcellionides pruinosus, which will complement the scarce information available for this group of organisms. This transcriptome also served as base for a RNA-Seq and a RT-qPCR analysis. The results of the RNA-Seq analysis performed in organisms exposed to nickel showed that this stressor negatively impacted at the genetic and epigenetic levels, in the trafficking, storage and elimination of metals, generates oxidative stress, inducing neurotoxicity and also affecting reproduction. These results were confirmed through RT-qPCR. As for the impact of dimethoate on these organisms it was only accessed through RT-qPCR and showed oxidative stress, an impact in neurotransmission, in epigenetic markers, DNA repair and cell cycle impairment. This study allowed the design of an Adverse Outcome Pathway draft that can be used further on for legislative purposes.

Introducción: El uso de sustancias químicas como protección contra plagas es una práctica común en las floriculturas, la aplicación de los conceptos de manejo integrado de éstas resguarda los cultivos asegurando la cantidad y la calidad necesaria del producto neto requerido para satisfacer la demanda internacional. El biomonitoreo se ha utilizado en una variedad de estudios ocupacionales y ambientales para determinar exposiciones a plaguicidas. Materiales y Métodos: Se realizó un estudio de corte transversal en 300 trabajadores vinculados a 15 empresas, con el objetivo de caracterizar la exposición al plaguicida Metamidofos en trabajadores de empresas de flores en la Sabana de Bogotá (Cundinamarca) y Rionegro (Antioquia). Se aplicó una encuesta, la cual incluyó las variables socio demográficas, ocupacionales y toxicológicas y se recolectaron muestras de orina para determinar el plaguicida metamidofos. Resultados: Se observó que la fuerza laboral pertenecía en su gran mayoría a la zona urbana (88.5%) y se resalta la participación del género femenino en edades de entre 19 y 58 años con un promedio de 36,7 años. El oficio en el que se encontraba la mayor parte de los trabajadores fue cultivo y corte con el 52.5%. El tiempo de trabajo en el sector floricultor reportado fue mínimo de 12 (3.3%) y máximo de 22 años (6.1%). En cuanto a los elementos de protección personal, el 96.1% (344) afirma utilizarlos en su trabajo. El promedio de metamidofos al inicio de la jornada fue de 29,12 μg/l y al finalizar 15,70 μg/l. Conclusiones: Este trabajo permitió conocer el panorama de la exposición al plaguicida Metamidofos y constituye un aporte para continuar investigando sobre el tema y a la vez hacer seguimiento a los trabajadores por medio de su inclusión en programas de vigilancia epidemiológica.

1. We tested three pesticides used for field manipulations of herbivory for direct phytoactive effects on the germination and growth of 14 herbaceous plant species selected to provide a range of life-history strategies and functional groups. 2. We report three companion experiments: (A) Two insecticides, chlorpyrifos (granular soil insecticide) and dimethoate (foliar spray), were applied in fully-factorial combination to pot-germinated individuals of 12 species. (B) The same fully-factorial design was used to test for direct effects on the germination of four herbaceous legumes. (C) The molluscicide, metaldehyde, was tested for direct effects on the germination and growth of six plant species. 3. The insecticides had few significant effects on growth and germination. Dimethoate acted only on growth stimulating Anisantha sterilis, Sonchus asper and Stellaria graminea. In contrast, chlorpyrifos acted on germination increasing the germination of Trifolium dubium and Trifolium pratense. There was also a significant interactive effect of chlorpyrifos and dimethoate on the germination of T pratense. However, all. effects were relatively small in magnitude and explanatory power. The molluscicide had no significant effect on plant germination or growth. 4. The small number and size of direct effects of the pesticides on plant performance is encouraging for the use of these pesticides in manipulative experiments on herbivory, especially for the molluscicide. However, a smatt number of direct (positive) effects of the insecticides on some plant species need to be taken into account when interpreting field manipulations of herbivory with these compounds, and emphasises the importance of conducting tests for direct phyto-active effects. The influence of the sample matrix in the CC-electron-capture detection analysis of the pesticides dimethoate, diazinon, chlorothalonil.. parathion methyl and fenitrothion in fruits samples has been studied. Experiments have been carried out where the pesticide responses in standard solutions prepared in selected solvent were compared with their response when present in apple, mango, papaya, banana, pineapple and melon extracts. The presence of matrix effects (MEs) and their extent were shown to be simultaneously influenced by several factors (matrix concentration, matrix type, pesticide concentration, analytical range). Pronounced MEs were observed particularly for dimethoate and diazinon in all matrices tested; in lower concentrations, all pesticides presented significant ME. The other pesticides presented variable ME. Higher ME enhancement was detected at lower pesticide concentration levels of and/or at higher matrix concentration solutions. The ME detected for fenitrothion, in the analytical range evaluated, were dependent on matrix type. For each pesticide, solvent and matrix-matched calibrations were compared for all fruit samples, and it could be concluded that quantitation based on standard solutions prepared in blank matrix extract (matrix-matched calibration) should be used to compensate the MEs and to obtain more accurate results for the pesticides studied.

BACKGROUND: Australian farmers and their workers are exposed to a wide variety of pesticides. Organophosphate (OP) insecticides are a widely used class of pesticide used for animal husbandry practices (Naphthalophos for sheep dipping, jetting and drench), crop production for pest control (Dimethoate) and in public health (Maldison for head lice). Acute poisonings with this class of insecticide are reported among agricultural workers and children around the globe, due to the inhibition of acetylcholinesterase (AChE). Less is known about chronic exposures. Regular monitoring of erythrocyte AChE will enable farmers to identify potential exposure to organophosphate insecticides and take action to reduce exposures and improve their health and safety practices. This study aims to assess and improve the integration of AChE monitoring into routine point of care health clinics, and provide farming and non-farming people with a link between their AChE activity and their household chemical and agrichemical use. METHODS/DESIGN: The research will target individuals who work on mixed farming enterprises and routinely using OPs (n = 50) and non-farmers (n = 30). Baseline data are collected regarding demographic, health conditions and behaviours, Kessler 10 (K10) scores, chemical use and personal protection. Baseline anthropometric measures include height, weight, hip and waist circumference, body fat analysis and, biochemical analysis of fasted total serum cholesterol, triglycerides, low-density cholesterol (LDL), high-density cholesterol (HDL) and blood glucose. Analysis of erythrocyte cholinesterase (EAChE) activity is also conducted using a finger prick test. Testing of EAChE is then repeated in all participants every 3 weeks for a maximum of three times over a period 10 weeks. Participants are provided with full feedback and counselling about their EAChE activity after each reading and a detailed summary provided to all participants at the completion of the study. Data will be analysed using repeated measures within a general linear model. DISCUSSION: This work will provide an evidence base and recommendations for the integration of EAChE monitoring into Australian rural health clinics, leading to research which will further quantify pesticide exposure both on the farm and in the home, highlighting the importance of sustaining and providing a safe work and home environment for farming communities. Os ácaros predadores das famílias Phytoseiidae e Stigmaeidae constituem-se nos principais inimigos naturais de Brevipalpus phoenicis (Geijskes) em citros. Este ácaro-praga causa sérios prejuízos na produção, devido à transmissão do vírus da leprose dos citros (CiLV). Apesar do grande volume de informações sobre a sensibilidade de ácaros Phytoseiidae a agrotóxicos, praticamente não existem informações sobre o efeito desses compostos em ácaros Stigmaeidae no Brasil. Sendo assim, o trabalho teve por objetivo avaliar o efeito dos principais agrotóxicos utilizados em citros sobre o ácaro predador Agistemus brasiliensis Matioli, Ueckermann & Oliveira (Acari: Stigmaeidae), em condições de laboratório. Arenas de folhas de citros da variedade Pera, contendo 25 fêmeas adultas de A. brasiliensis, foram pulverizadas em torre de Potter. Avaliaram-se as mortalidades dos ácaros 72 horas após a aplicação. O efeito dos produtos na reprodução do acarino e a viabilidade dos ovos também foram avaliados. Quanto à seletividade, conforme proposta da Organização Internacional para o Controle Biológico (IOBC), os produtos foram classificados como: classe 1 - inócuo (E<30%), acrinathrin, bifenthrin, carbosulfan, deltamethrin; 2 - levemente nocivo (30%99%), calda sulfocálcica, cyhexatin, flufenoxuron, hexythiazox, óxido de fenbutatin, propargite, pyridaben e spirodiclofen. Estudos conduzidos em condições de campo ainda são necessários para se compreender melhor o efeito desses agrotóxicos sobre o ácaro predador.

Assim como a maioria dos cultivos, em cafeeiro, a associação de inimigos naturais com produtos fitossanitários seletivos é uma importante estratégia no manejo integrado de pragas (MIP). Dessa forma, este estudo objetivou avaliar a seletividade de agrotóxicos utilizados na cultura cafeeira sobre larvas de Cryptolaemus montrouzieri Mulsant, 1853 (Coleoptera: Coccinellidae), em laboratório. Os bioensaios foram realizados em condições controladas (25±2°C, UR de 70±10% e fotofase de 12 horas). Cada bioensaio consistiu em seis tratamentos e dez repetições, sendo cada parcela composta por duas larvas. Para as comparações das médias dos tratamentos, empregou-se um esquema fatorial inteiramente ao acaso de produtos x número de instares para os diferentes instares subsequentes à aplicação. Os inseticidas utilizados e suas respectivas dosagens de aplicação, em g de i.a. L-1 foram: tiametoxam (0,5), imidacloprido (0,7), óleo mineral (13,3), endossulfam (2,63) e dimetoato (0,48). A testemunha foi composta apenas por água destilada. A aplicação dos produtos foi realizada por meio de torre de Potter. Avaliaram-se a sobrevivência dos espécimes após serem contaminados com os produtos e os efeitos dos compostos sobre os parâmetros reprodutivos do predador. Tiametoxam (0,5), imidacloprido (0,7) e endossulfam (2,63) foram os mais prejudiciais, tendo sido observados, para os dois primeiros, 100% de mortalidade de larvas de primeiro e de segundo instares um dia após a aplicação e significativa redução na sobrevivência das larvas de terceiro e quarto instares. Dimetoato (0,48) foi nocivo para larvas de primeiro instar e levemente nocivo para larvas de quarto instar. em função da seletividade apresentada pelo óleo mineral (13,3), este pode ser recomendado em compatibilização com o predador C. montrouzieri em programas de MIP na cultura cafeeira, sem prejuízo ao desempenho do inimigo natural.

The fate of three pesticides (vinclozolin, dimethoate, cyproconazole) in plums, from field treatment to the drying process, was studied. Only vinclozolin showed measurable residue concentrations at harvest, while dimethoate was completely degraded after two weeks and cyproconazole was present at negligible levels just after treatment. During the drying process into prunes, the residues were not reduced during the fruit washing stage, but the drying stage led to complete elimination of vinclozolin residues.

The persistence of five pesticides (acephate, azinphos methyl, carbendazim, diazinon and dimethoate) in peach fruit was studied in real operating conditions. Pesticides showed different degradation rates. Diazinon was degraded with a half-life (t(1/2)) of 5.2 days, acephate, azinphos methyl and dimethoate with t(1/2) of ca 12 days and carbendazim with t(1/2) of 15.5 days. After the pre-harvest interval (PHI) all pesticide residues were within the limits established by Italian law.

Three insecticides (chlorpyrifos methyl, dimethoate and fenthion) were applied on clementine fruits with and without mineral oil to assess the effect of mineral oil on pesticide residues. In both experiments the residues on the fruits after the last treatment and at harvest time were not significantly different.

The decay rate of six insecticides (azinphos methyl, diazinon, dimethoate, methidathion, parathion methyl, and quinalphos) used to control Dacus oleae was studied. Degradation of pesticides showed pseudo-first-order kinetics with correlation coefficients ranging between -0.936 and -0.998 and half-lives between 4.3 days for dimethoate and 10.5 days for methidathion. Residues in olive oil were greater than on olives, with a maximum concentration factor of 7. Dimethoate was the only pesticide with lower residues in the oil than on the fruits. Olive washing affects pesticide residues ranging from no reduction to a 45% decrease. During 8 months of storage of the olive oil, diazinon, dimethoate, parathion methyl, and quinalphos did not show any remarkable difference, while methidathion and azinphos methyl showed a moderate decrease.
