Bdellovibrio bacteriovorus cells have a single polar flagellum whose helical pitch and diameter characteristically change near the midpoint, resulting in a tapered wave. There are six flagellin genes in the genome: fliC1 to fliC6. Accordingly, the flagellar filament is composed of several similar flagellin species. We have used knockout mutants of each gene and analyzed the mutational effects on the filament length and on the composition and localization of each flagellin species in the filament by electron microscopy and one- and two-dimensional polyacrylamide gel electrophoresis. The location and amounts of flagellins in a filament were determined to be as follows: a small amount of FliC3 at the proximal end, followed by a large amount of FliC5, a large amount of FliC1, a small amount of FliC2 in this order, and a large amount of FliC6 at the distal end. FliC4 was present at a low level, but the location was not determined. Filament lengths of newly born progeny cells increased during prolonged incubation in nutrient-deficient buffer. The newly formed part of the elongated filament was composed of mainly FliC6. Reverse transcription PCR analysis of flagellar gene expression over 5 days in buffer showed that fliC gene expression tailed off over 5 days in the wild-type cells, but in the fliC5 mutant, expression of the fliC2, fliC4, and fliC6 genes was elevated on day 5, suggesting that they may be expressed to compensate for the absence of a major component, FliC5.

Metabolic alterations have been identified as a frequent event in cancer. This is often associated with increased flux through glycolysis, and also a secondary pathway to glycolysis, hexosamine biosynthetic pathway (HBP). HBP provides substrate for N-linked glycosylation, which occurs in the endoplasmic reticulum and the Golgi apparatus. N-linked glycosylation supports protein folding and correct sorting of proteins to plasma membrane and secretion. This process generates complex glycoforms, which can be recognized by other proteins and glycosylation of receptor tyrosine kinases (RTK) can also regulate their plasma-membrane retention time. Of special interest for experimental biologists, plants produce proteins, termed lectins, which bind with high specificity to glyco-conjugates. For the purposes of molecular biology, plant lectins can be conjugated to different moieties, such as agarose beads, which enable precipitation of specifically glycosylated proteins. In this chapter, we describe in detail how to perform pull-down experiments with commercially available lectins to identify changes in the glycosylation of RTKs.

Introduction: Transient receptor potential (TRP) channels are widely, but not uniformly, distributed in tissues. To date the dominant focus of attention has been on TRP expression and functionality in neurons. However, their expression and activation in selected non-neuronal cells suggest TRPs have a potential role in coordinating cross-talk during the inflammatory process. Fibroblasts comprise the major cell type in the dental pulp and play an important role in pulpal inflammation. Objectives: The aim of this study was to investigate the expression and functionality of the TRP channels TRPA1, TRPM8, TRPV4 and TRPV1 in human dental pulp fibroblasts. Methods: Dental pulp fibroblasts were derived by explant culture of pulps removed from extracted healthy teeth. Fibroblasts were cultured in DMEM supplemented with 10% FCS, 100U/ml penicillin and 100µg/ml streptomycin. Protein expression of TRP channels was investigated by SDS- polyacrylamide gel electrophoresis and Western blotting of cell lysates from fibroblast cells in culture. TRPA1, TRPM8, TRPV4 and TRPV1 expression was determined by specific antibodies, detected using appropriate anti-species antibodies and chemiluminescence. Functionality of TRP channels was determined by Ca2+ microfluorimetry. Cells were grown on cover slips and incubated with Fura 2AM prior to stimulation with icilin (TRPA1 agonist), menthol (TRPM8 agonist), 4 alpha-phorbol 12,13-didecanoate (4alphaPDD) (TRPV4 agonist) or capsaicin (TRPV1 agonist). Emitted fluorescence (F340/F380) was used to determine intracellular [Ca2+] levels. Results: Fibroblast expression of TRPA1, TRPM8, TRPV4 and TRPV1 was confirmed at the protein level by Western blotting. Increased intracellular [Ca2+] levels in response to icillin, methanol, 4alphaPDD and capsacin, indicated functional expression of TRPA1, TRPM8, TRPV4 and TRPV respectively. Conclusions: The presence and functionality of TRP channels on dental pulp fibroblasts suggests a potential role for these cells in the pulpal neurogenic inflammatory response. Introduction: Accumulating evidence supports a role for odontoblasts in initiating tooth pain, however direct ionic mechanisms underlying dentine nociceptive function remain unclear. The transient receptor potential (TRP) ion channels are directly related to cellular mechanisms of nociception and thermo-sensitive function but their expression by human odontoblasts remains to be determined. Objectives: To investigate the expression and functionality of the thermo-sensitive TRP channels TRPV1, TRPV4, TRPM8 and TRPA1 in human odontoblasts. Methods: Human odontoblasts were derived from dental pulp of immature permanent third molars by explant method. Cell lysates of odontoblasts were subject to SDS- polyacrylamide gel electrophoresis and proteins were blotted onto nitrocellulose membranes. Blots were probed with primary antibodies to TRPA1, TRPM8, TRPV4 and TRPV1. Detection of bound primary antibodies was achieved using appropriate anti-species antibody conjugates and chemiluminescent substrates. Functionality of the channels was determined with Ca2+ microfluorimetry, where cells grown in cover slips and incubated with Fura 2AM prior to stimulation with capsaicin (TRPV1 agonist), 4 alpha-phorbol 12,13-didecanoate (4áPDD) (TRPV4 agonist), icilin (TRPA1 agonist) and menthol (TRPM8 agonist). Emitted fluorescence was measured and the fluorescence ratio (R) was calculated as F340/F380 to determine the level of [Ca2+]i. Results: Western blotting confirmed the molecular localisation of thermo-sensitive TRP channels in human odontoblasts. Functionality assays revealed increase in [Ca2+]i in response to capsacin, icillin, methanol and 4áPDD indicating functional expression of TRPV1, TRPA1, TRPM8 and TRPV4 respectively. Conclusions: Functional expression of thermo-sensitive TRP channels in human odontoblasts may indicate a crucial role for odontoblasts in thermally induced dental pain. O trabalho de investigação apresentado nesta dissertação foi desenvolvido tendo como objectivo a síntese e funcionalização de meso-triarilcorróis para utilização como quimiossensores. Este trabalho encontra-se apresentado ao longo de cinco capítulos. No primeiro capítulo são apresentadas as características gerais, as metodologias de síntese e de funcionalização de macrociclos de tipo corrólico, e descrevemse algumas aplicações em que têm sido utilizados. São ainda abordadas algumas das propriedades e características dos quimiossensores e os mecanismos de deteção de diversos analítos. No segundo capítulo, após uma pequena introdução às reações de Wittig e de Diels-Alder, escolhidas para a funcionalização do macrociclo corrólico, descreve-se o estudo efectuado para a obtenção do complexo de gálio(III) do 3- vinil-5,10,15-tris(pentafluorofenil)corrol e o seu comportamento como dieno, em reações de Diels-Alder na presença dos dienófilos 1,4-benzoquinona e 1,4- naftoquinona. Desses estudos resultaram dois aductos cuja habilidade sensorial, bem como a dos seus precursores, foi estudada, em solução, na presença de aniões esféricos (F-, Br-, Cl-), lineares (CN-) e volumosos (CH3COO-, H2PO4 -). Dos macrociclos estudados verificou-se que o corrol base-livre 5,10,15-tris(pentafluorofenil)corrol apresenta uma elevada sensibilidade para o anião fluoreto (F-), e que a coordenação do núcleo corrólico com gálio(III) diminui a afinidade para este anião. Em geral, todos os compostos mostraram afinidade para o anião cianeto (CN-) mesmo quando em suportes poliméricos. O gel de poliacrilamida revelou-se muito promissor na determinação de CN- em amostras de água. No terceiro capítulo é avaliada a reatividade do complexo de gálio(III) do 3- vinil-5,10,15-tris(pentafluorofenil)corrol ainda como dieno mas agora na presença de um dienófilo linear, o acetilenodicarboxilato de dimetilo. Desse estudo resultaram dois novos derivados corrólicos. A habilidade sensorial dos mesmos perante os aniões fluoreto, cianeto, acetato, e fosfato foi avaliada por espectroscopia de absorção e emissão tendo um dos aductos mostrado ser colorimétrico para o anião cianeto. No quarto capítulo descreve-se a síntese e caracterização de dois conjugados do tipo corrol-cumarina, resultantes de reações de Hetero-Diels-Alder entre o 3-vinil-5,10,15-tris(pentafluorofenil)corrolatogálio(III)(piridina) e orto-quinonasmetídeos gerados in situ a partir de reacções de Knoevenagel entre cumarinas e paraformaldeído. Realizaram-se estudos de afinidade sensorial para aniões e catiões com estes macrociclos, bem como com conjugados porfirinacumarina análogos. A inserção de uma unidade cumarina conferiu uma excepcional solubilidade tendo os novos derivados apresentado solubilidade em etanol. No quinto e último capítulo desta dissertação é avaliada a capacidade sensorial do 5,10,15-tris(pentafluorofenil)corrol e da sua espécie monoaniónica, para os catiões metálicos Na+, Ca2+, Cu2+, Cd2+, Pb2+, Hg2+, Ag+, Al3+, Zn2+, Ni2+, Cr3+, Ga3+, Fe3+ em tolueno e acetonitrilo. Os macrociclos corrólicos mostraram ser selectivos e colorimétricos para o catião Hg2+. Neste trabalho descreve-se ainda a síntese do derivado -iminocorrol, que após funcionalização com o 3-isocianatopropiltrimetoxisilano originou um derivado do tipo alcoxisilano, que foi, posteriormente, ancorado a nanopartículas comerciais de sílica. As novas nanopartículas ancoradas com o alcoxisilano corrol foram estudadas na presença de Cu2+, Hg2+ e Ag+. Na presença do catião Ag+ assistiu-se a uma mudança de cor, de verde para amarelo.

Photodynamic inactivation (PDI) is defined as the process of cell destruction by oxidative stress resulting from the interaction between light and a photosensitizer (PS), in the presence of molecular oxygen. PDI of bacteria has been extensively studied in recent years, proving to be a promising alternative to conventional antimicrobial agents for the treatment of superficial and localized infections. Moreover, the applicability of PDI goes far beyond the clinical field, as its potential use in water disinfection, using PS immobilized on solid supports, is currently under study. The aim of the first part of this work was to study the oxidative modifications in phospholipids, nucleic acids and proteins of Escherichia coli and Staphylococcus warneri, subjected to photodynamic treatment with cationic porphyrins. The aims of the second part of the work were to study the efficiency of PDI in aquaculture water and the influence of different physicalchemical parameters in this process, using the Gram-negative bioluminescent bacterium Vibrio fischeri, and to evaluate the possibility of recycling cationic PS immobilized on magnetic nanoparticles. To study the oxidative changes in membrane phospholipids, a lipidomic approach has been used, combining chromatographic techniques and mass spectrometry. The FOX2 assay was used to determine the concentration of lipid hydroperoxides generated after treatment. The oxidative modifications in the proteins were analyzed by one-dimensional polyacrylamide gel electrophoresis (SDS-PAGE). Changes in the intracellular nucleic acids were analyzed by agarose gel electrophoresis and the concentration of doublestranded DNA was determined by fluorimetry. The oxidative changes of bacterial PDI at the molecular level were analyzed by infrared spectroscopy. In laboratory tests, bacteria (108 CFU mL-1) were irradiated with white light (4.0 mW cm-2) after incubation with the PS (Tri-Py+-Me-PF or Tetra-Py+-Me) at concentrations of 0.5 and 5.0 μM for S. warneri and E. coli, respectively. Bacteria were irradiated with different light doses (up to 9.6 J cm-2 for S. warneri and up to 64.8 J cm-2 for E. coli) and the changes were evaluated throughout the irradiation time. In the study of phospholipids, only the porphyrin Tri-Py+-Me-PF and a light dose of 64.8 J cm-2 were tested. The efficiency of PDI in aquaculture has been evaluated in two different conditions: in buffer solution, varying temperature, pH, salinity and oxygen concentration, and in aquaculture water samples, to reproduce the conditions of PDI in situ. The kinetics of the process was determined in realtime during the experiments by measuring the bioluminescence of V. fischeri (107 CFU mL-1, corresponding to a level of bioluminescence of 105 relative light units). A concentration of 5.0 μM of Tri-Py+-Me-PF was used in the experiments with buffer solution, and 10 to 50 μM in the experiments with aquaculture water. Artificial white light (4.0 mW cm-2) and solar irradiation (40 mW cm-2) were used as light sources.

O objectivo primordial deste trabalho foi efectuar a extracao e o estudo das proteinas da Goma da Alfarroba ou Locust Bean Gum (LBG) com vista a sua clarificacao e purificacao. Para atingir este objectivo em primeiro lugar estudou-se o teor de etanol que se deveria utilizar numa suspensao aquosa de LBG, uma vez que a LBG e um polissacarido que hidrata facilmente com agua e consequentemente origina solucoes altamente viscosas que impossibilitam a realizacao de tecnicas de extracao de proteinas. Para isso estudou-se o comportamento da LBG em suspensoes aquosas com 10% (p/v) de LBG e diferentes concentracoes de etanol, designadamente 10% (v/v), 20% (v/v), 30% (v/v) e 40% (v/v). Com este estudo concluiu-se que seria necessario utilizar uma concentracao de 40% (v/v) de etanol a 96% em cada suspensao de LBG a 10% (p/v) para conseguir evitar a sua hidratacao excessiva. Assim, todas as extracoes neste trabalho foram realizadas de acordo com estas condicoes. Posteriormente com vista a extracao das proteinas da LBG testaram-se diferentes tratamentos, designadamente tratamentos alcalinos, acidos e com detergentes. Segundo a bibliografia consultada para os tratamentos alcalinos, realizaram-se estas extracoes com hidroxido de sodio (NaOH) com concentracoes de 0,025 M, 0,05M, 0,1M e 0,2M. Estes tratamentos foram iniciados com uma concentracao de NaOH a 0,2M, contudo apos analise dos resultados verificou-se que esta originou uma LBG com uma coloracao mais amarela do que a LBG bruta (inicial) apresentava. Sendo a ideia final a clarificacao da goma este nao foi um resultado desejavel e como tal testaram-se concentracoes inferiores de NaOH, acima referidas. No final concluiu-se que todas estas extracoes efetuadas com NaOH originavam uma LBG mais amarela do que a LBG bruta e que assim este nao seria o tratamento mais adequado para o objectivo pretendido. Como tal prosseguiram-se os estudos com um tratamento acido, que atraves de consulta bibliografica se iniciou com acido sulfurico (H2SO4). O tratamento acido iniciou-se com uma concentracao de H2SO4 de 0,1M, testando-se posteriormente tambem uma extracao com H2SO4 com concentracao de 0,05M. No final destas extracoes verificou-se que a LBG obtida em ambas apresentava uma coloracao mais clara do que a LBG bruta, o que representava, numa primeira analise, um bom resultado. Por fim efectuaram-se extracoes solido-liquido com diferentes detergentes, designadamente com Tween 20, Tween 40, Tween 60, Tween 65, Tween 80, Tween 85, Triton X-100, Triton X-114 e SDS todos a uma concentracao de 0,1% (v/v) a excepcao do Tween 65 em que foi utilizada uma concentracao de 0,1% (p/v), porque a temperatura ambiente este detergente e solido. No final destas extracoes concluiu-se que na maioria dos casos a LBG apresentava uma coloracao mais clara do que a LBG bruta. Para quantificar as proteinas extraidas atraves de cada um dos tratamentos acima mencionados utilizaram-se dois metodos, o metodo de Bradford para quantificar as proteinas presentes nos sobrenadantes das extracoes e o metodo de Kjeldahl para quantificar as proteinas ainda presentes na LBG apos as extracoes. Analisando os resultados obtidos por estes metodos concluiu-se que a extracao efetuada com Tween 80, em que se efectuou uma lavagem adicional a LBG durante a filtracao com uma solucao de agua destilada e etanol a 40% (v/v), foi a que apresentou melhores resultados. Assim, foi com esta amostra proteica que se prosseguiram os estudos as proteinas. Para realizar os estudos as proteinas extraidas efectuou-se uma eletroforese em SDS-PAGE (sodium dodecyl sulfate polyacrylamide gel electrophoresis) com o intuito de verificar o estado da amostra. Analisando os geles obtidos constatou-se que a amostra se encontrava em boas condicoes. Deste modo realizou-se uma eletroforese bidimensional (2 – D) das proteinas extraidas da LBG. Nesta tecnica, como se desconheciam que tipo de proteinas se encontravam presentes na amostra utilizou-se uma banda de pH para a Focagem Isoelectrica (1a Dimensao) entre 3 e 10 e a separacao por Peso Molecular (2a Dimensao) foi efetuada em SDS-PAGE. Analisando os geles obtidos concluiu-se que a maioria das proteinas presentes nesta amostra apresentam pontos isoelectricos na gama de pH entre 5 e 6 e tem pesos moleculares entre 40 e 60 kDa. Pode-se entao concluir que o objectivo deste trabalho foi apenas parcialmente atingido uma vez que se conseguiu extrair e estudar algumas das caracteristicas das proteinas presentes na LBG, mas nao se alcancou a sua clarificacao.

A eritropoietina (EPO) é uma substância que estimula a produção de eritrócitos, aumentando a oxigenação muscular, sendo segregada de forma natural pelo organismo e excretada na urina em baixas concentrações. Devido às suas propriedades e características, a EPO foi rapidamente introduzida no mundo do desporto, como substância ilícita, proporcionando vantagens no rendimento desportivo. No início de 2000 foi desenvolvido um método de deteção direta de EPO Recombinante (rHuEPO) em urina humana por Lasne, baseado na focalização isoelétrica (IEF) em gel de poliacrilamida, seguido de duplo blote, tendo este sido publicado e validado. Em 2002, a Agência Mundial Antidopagem (AMA) implementou este mesmo método, sendo atualmente um dos métodos oficiais utilizado pelos laboratórios acreditados pela AMA. Desta forma, o ponto de partida para a realização deste trabalho consistiu na necessidade de implementar e validar o método de referência de IEF para a deteção de rHuEPO em urina humana. O trabalho foi realizado no Laboratório de Análises e Dopagem (LAD) do Instituto do Desporto de Portugal (IDP), atual Instituto Português do Desporto e Juventude (IPDJ). O principal objetivo deste trabalho consistiu no estudo/investigação de diferentes parâmetros de validação (especificidade/seletividade; capacidade de identificação; limite de deteção; exatidão e repetibilidade), de acordo com o protocolado no Procedimento Geral interno do Laboratório de Análises de Dopagem de Lisboa (LAD). O referido método de triagem e confirmação revelou possuir características de desempenho conformes com os requisitos aplicáveis, pelo que é considerado validado e apto.

Evidence concerning the presence or absence of common neuronglia lineages in the postnatal mammalian central nervous system is still a matter of speculation. We address this problem using optic nerve explants, which show an extremely long survival in culture. Morphological, immunocytochemical and immunochemical methods were applied. The results obtained from in vitro tissue were compared with optic nerves (ONs) and whole-brain samples from animals of different ages. Newborn rat ONs represented the starting material of our tissue culture; they are composed of unmyelinated axons, astrocytes and progenitor cells but devoid of neuronal cell bodies. At this age, Western blots of ONs were positively stained by neurofilament and synapsin I specific antibodies. These bands increased in intensity during postnatal in situ development. In explant cultures, the glia cells reach a stage of functional differentiation and they maintain, together with undifferentiated cells, a complex histotypic organization. After 6 days in vitro, neurofilaments and synapsin I could not be detected on immunoblots, indicating that 1) axonal degeneration was completed, and 2) neuronal somata were absent at the time. Surprisingly, after about 4-5 weeks in culture, a new cell type appeared, which showed characteristics typical of neurons. After 406 days in vitro, neurofilaments and synapsin I were unequivocally detectable on Western blots. Furthermore, both immunocytochemical staining and light and electron microscopic examinations corroborated the presence of this earlier-observed cell type. These in vitro results clearly show the high developmental plasticity of ON progenitor cells, even late in development. The existence of a common neuron-glia precursor, which never gives rise to neurons in situ, is suggested.

This report describes the partial purification and the characteristics of (Na+ + K+)-ATPase (ATP phosphohydrolase, EC 3.6.1.3) from an amphibian source. Toad kidney microsomes were solubilized with sodium deoxycholate and further purified by sodium dodecyl sulphate treatment and sucrose gradient centrifugation, according to the methods described by Lane et al. [(1973) J. Biol. Chem. 248, 7197--7200], Jørgensen [(1974) Biochim. Biophys. Acta 356, 36--52] and Hayashi et al. [(1977) Biochim. Biophys. Acta 482, 185--196]. (Na+ + K+)-ATPase preparations with specific activities up to 1000 mumol Pi/mg protein per h were obtained. Mg2+-ATPase only accounted for about 2% of the total ATPase activity. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis revealed three major protein bands with molecular weights of 116 000, 62 000 and 26 000. The 116 000 dalton protein was phosphorylated by [gamma-32P]ATP in the presence of sodium but not in the presence of potassium. The 62 000 dalton component stained for glycoproteins. The Km for ATP was 0.40 mM, for Na+ 12.29 mM and for K+ 1.14 mM. The Ki for ouabain was 35 micron. Temperature activation curves showed two activity peaks at 37 degrees C and at 50 degrees C. The break in the Arrhenius plot of activity versus temperature appeared at 15 degrees C.

Botrytis cinerea isolates collected from Niagara region were treated with different concentrations of the fiingicide, iprodione to test their sensitivity to this fungicide. These Botrytis cinerea isolates were divided into two groups according to their sensitivity to iprodione. Those isolates whose growth was inhibited by iprodione at concentrations < 2|i,g/nil were classified as sensitive isolates. Isolates that were able to show considerable growth at 2|j,g/ml iprodione were classified as resistant isolates. Resistant and sensitive isolates were compared for their morphological and growth characteristics, conidial germination, virulence on grape berries and protein banding profiles. The fungicide iprodione at a concentration of 2|xg/nil inhibited mycelial growth, sporulation and conidial germination of sensitive isolates but not those of resistant isolates. The inhibitory effect of the fungicide was greater on mycelial growth than on conidia germination of the sensitive isolates. Sensitive isolates produced no sclerotia whereas resistant isolates produced large number of sclerotia. The fungicide iprodione affected sclerotial production in the resistant isolates. The number of sclerotia was decreased by the increase of iprodione in the medium. Sporulation of resistant isolates was improved significantly in the presence of iprodione. The resistant isolates were as virulent as the sensitive isolates on grape berries. The sensitive and resistant isolates showed similar protein banding profiles in the absence of iprodione in polyacrylamide gel electrophoresis studies. Similar protein profiles were also observed when these isolates were grown in the presence of low iprodione concentration (0.5|ig/nil). However, in the presence of concentration (0.5|ig/nil). However, in the presence of iprodione at concentration of 5|Xg/nil, one protein band with approximate molecular weight of 83 KDa was present in the growing resistant isolates (and the controls) but was missing in the inhibited sensitive isolates.

Cell surface proteins obtained by alkaline extraction from isolated cell walls of Mortierella pusilla and M. candelabrum, host and nonhost, respectively, to the mycoparasite, Piptocephalis virginiana, were tested for their ability to agglutinate mycoparasite spores. The host cell wall protein extract had a high agglutinating activity (788 a.u. mg- t ) as compared with the nonhost extract (21 a.li. mg- t ). SDS-polyacrylamide gel electrophoresis of the cell wall proteins revealed four protein bands, a, b, c, and d (Mr 117, 100, 85 and 64 kd, respectively) at the host surface, but not at the nonhost surface, except for the faint band c. Deletion of proteins b or c from the host cell wall protein extract significantly reduced its agglutinating activity. Proteins band c, obtained as purified preparations by a series of procedures, were shown to be two glycoproteins. Carbohydrate analysis by gas chromatography demonstrated that glucose and Nacetylglucosamine were the major carbohydrate components of the glycoproteins. It was further shown that the agglutinating activity of the pure preparation containing both band c was 500-850 times that of the single glycoproteins, suggesting the involvement of both glycoproteins in agglutination. The results suggest that the glycoproteins band c are the two subunits of agglutinin present at the host cell surface. The two glycoproteins band c purified from the host cell wall protein extract were further examined after various treatments for their possible role in agglutination, attachment and appressorium formation by the mycoparasite. Results obtained by agglutination and attachment tests showed: (1) the two glycoprotein-s are not only an agglutinin responsible for the mycoparasite spore agglutination, but may also serve as a receptor for the specific recognition, attachment and appressorium formation by the mycoparasite; (2) treatment of the rnycoparasite spores with various sugars revealed that arabinose, glucose and N-acetylglucosamine inhibited the agglutination and attachment activity of the glycoproteins, however, the relative percentage of appressorium formation was not affected by the above sugars; (3) the two glycoproteins are relatively stable with respect to their agglutinin and receptor functions. The present results suggest that the agglutination and attachment may be mediated directly by certain sugars present at the host and mycoparasite cell surfaces while the appressorlum formation may be the response of complementary combinations of both sugar and protein, the two parts of the glycoproteins at the interacting surfaces of two fungi.

Presence of surface glycoprotein in Piptocephalis virginiana that recognizes the host glycoproteins band c, reported earlier from our laboratory, was detected by immunofluorescence microscopy. Germinated spores of P. virginiana treated with Mortierella pusilla cell wall protein extract, primary antibodies prepared against glycoproteins band c and FITC-goat anti-rabbit IgG conjugate showed fluorescence. This indicated that on the surfaces of the biotrophic mycoparasite P. virginiana , there might be a complementary molecule which recognizes the glycoproteins band c from M. pusilla. Immunobinding analysis identified a glycoprotein of Mr 100 kDa from the mycoparasite which binds with the host glycoproteins band c, separately as well as collectively. Purification of this glycoprotein was achieved by (i) 60% ammonium sulfate precipitation, (ii) followed by heat treatment, and (iii) Sephadex G-IOO gel filtration. The glycoprotein was isolated by preparative polyacrylamide gel electrophoresis by cutting and elution. The purity of the protein ·was ascertained by SDS-PAGE and Western blot analysis. Positive reaction to periodic acid-Schiff reagent revealed the glycoprotein nature of this 100 kDa protein. Mannose was identified as a major sugar component of this glycoprotein by using a BoehringerMannheim Glycan Differentiation Kit. Electrophoretically purified glycoprotein was used to raIse polyclonal antibody in rabbit. The specificity of the antibody was determined by dot-immunobinding test and western-blot analysis. Immunofluorescence mIcroscopy revealed surface localization of the protein on the germ tube of Piptocephalis virginiana. Fluorescence was also observed at the surfaceJ of the germinated spores and hyphae of the host, M. pusilla after treatment with complementary protein from P. virginiana, primary antibody prepared against the complementary protein and FITC-goat anti-rabbit IgG conjugate.

Several stresses to tissues including hyperthermia, ischemia, mechanical trauma and heavy metals have been demonstrated to affect the regulation of a subset of the family of heat shock proteins of70kOa (hsp70). In several organisms following some of these traumas, the levels of hsp70 mRNA and proteins are dramatically upregulated. However, the effects of the stress on limb and tail amputation in the newt Notophthalmus viridescens, involving mechanical tissue damage, have not adequately been examined. In the present study, three techniques were utilized to quantitate the levels of hsp70 mRNA and protein in the tissues of the forelimbs and tails of newts during the early post-traumatic events following surgical resection of these:: appendages. These included quantitative Western blotting of proteins separated by both one and twodimensional SDS-polyacrylamide gel electrophoresis and quantitative Northern blot analysis of total RNA. In tissues of both the limb and tail one hour after amputation, there were no significant differences in the levels of hsp70 protein measured by one-dimensional SOSPAGE followed by Western blotting, when compared to the levels measured in the unamputated limb. A 30 minute heat shock at 35°C failed to elicit an increase in the levels of hsp70 protein in these tissues. Further analysis using the more sensitive 20 PAGE separation of stump tissue proteins revealed that at least some of the five hsp70 isoforms of the newt may be differentially regulated in limbs and tails in response to trauma. It appears also that amputation of the tail and limb tissues leads to slight 3 elevation in the levels of HSP70 mRNA when compared to those of their respective unstressed tissues.

Black fly (Simuliidae) silk is produced by the larvae and pharate pupae and is used for anchorage and cocoon production. There exists limited information on simuliid silks, including protein composition and genetic sequences encoding such proteins. The present study aimed to expand what is known about simuliid silks by examining the silks of several simuliid species and by making comparisons to the silk of non-biting midges (Chironomidae). Silk glands were dissected out of larval and pupal simuliids, and protein contents were separated by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) and visualized with silver stain. Protein contents were compared by mass in kilodaltons (kDa) between life stages and among species. Polymerase chain reaction (PCR) was used to expand upon known gene sequence information, and to determine the presence of genes homologous to chironomid silk. SDS-PAGE of cocoons revealed the presence of a 56 kDa and a 67 kDa protein. Silk gland contained as many as 28 different proteins ranging from 319 kDa to 8 kDa. Protein profiles vary among species, and group into large (>200), intermediate(>100), and small (<100) protein classes as is found in chironomids.
