OBJECT: Glycerol is considered to be a marker of cell membrane degradation and thus cellular lysis. Recently, it has become feasible to measure via microdialysis cerebral extracellular fluid (ECF) glycerol concentrations at the patient's bedside. Therefore the aim of this study was to investigate the ECF concentration and time course of glycerol after severe traumatic brain injury (TBI) and its relationship to patient outcome and other monitoring parameters. METHODS: As soon as possible after injury for up to 4 days, 76 severely head-injured patients were monitored using a microdialysis probe (cerebral glycerol) and a Neurotrend sensor (brain tissue PO2) in uninjured brain tissue confirmed by computerized tomography scanning. The mean brain tissue glycerol concentration in all monitored patients decreased significantly from 206 +/- 31 micromol/L on Day 1 to 9 +/- 3 micromol/L on Day 4 after injury (p < 0.0001). Note, however, that there was no significant difference in the time course between patients with a favorable outcome (Glasgow Outcome Scale [GOS] Scores 4 and 5) and those with an unfavorable outcome (GOS Scores 1-3). Significantly increased glycerol concentrations were observed when brain tissue PO2 was less than 10 mm Hg or when cerebral perfusion pressure was less than 70 mm Hg. CONCLUSIONS: Based on results in the present study one can infer that microdialysate glycerol is a marker of severe tissue damage, as seen immediately after brain injury or during profound tissue hypoxia. Given that brain tissue glycerol levels do not yet add new clinically significant information, however, routine monitoring of this parameter following traumatic brain injury needs further validation.

BACKGROUND: Acute epidural and subdural haematomas remain among the most common causes of mortality and disability resulting from traumatic brain injury. In the last three decades improvements in rescue, neuromonitoring and intensive care have led to better outcomes. The purpose of this study was to evaluate the impact of these strategies on outcome in patients treated in a single institution in Switzerland. METHODS: A total of 76 consecutive patients who underwent emergency craniotomy for acute traumatic epidural and subdural haematoma at University Hospital Bern between January 2000 and December 2003 were included in this study. RESULTS: Thirty-seven patients presented with an epidural haematoma and 46 with a subdural haematoma. In seven patients both haematomas could be documented. The median age was 54 years (IQR 28). The median initial GCS score was 7 (IQR 6). The median time from primary injury to surgery was 3 hours (IQR 2.5 hours). The median stay in the ICU was 3 days (IQR: 3 days). The outcome was favourable (GOS 4 and 5) in 43 patients (57%). Thirteen patients (17%) remained severely or moderately disabled (GOS 3). Finally, a total of 21 patients (28%) died or remained in a persistent vegetative state (GOS 1 and 2). Mortality was 41% for acute subdural haematoma (19/46) and 3% (1/37) for patients with epidural haematoma. Only age, GCS at admission and pupil abnormalities seemed to be associated with outcome. Time to surgery was not. CONCLUSION: In patients admitted with acute traumatic epidural and subdural haematomas that are treated within a median of 3 hours after primary injury, factors such as age, initial GCS and pupil abnormalities still appear to be the most important factors correlating with outcome.

Ensuring sustainable use of natural resources is crucial for maintaining the basis for our livelihoods. With threats from climate change, disputes over water, biodiversity loss, competing claims on land, and migration increasing worldwide, the demands for sustainable land management (SLM) practices will only increase in the future. For years already, various national and international organizations (GOs, NGOs, donors, research institutes, etc.) have been working on alternative forms of land management. And numerous land users worldwide – especially small farmers – have been testing, adapting, and refining new and better ways of managing land. All too often, however, the resulting SLM knowledge has not been sufficiently evaluated, documented and shared. Among other things, this has often prevented valuable SLM knowledge from being channelled into evidence-based decision-making processes. Indeed, proper knowledge management is crucial for SLM to reach its full potential. Since more than 20 years, the international WOCAT network documents and promotes SLM through its global platform. As a whole, the WOCAT methodology comprises tools for documenting, evaluating, and assessing the impact of SLM practices, as well as for knowledge sharing, analysis and use for decision support in the field, at the planning level, and in scaling up identified good practices. In early 2014, WOCAT’s growth and ongoing improvement culminated in its being officially recognized by the UNCCD as the primary recommended database for SLM best practices. Over the years, the WOCAT network confirmed that SLM helps to prevent desertification, to increase biodiversity, enhance food security and to make people less vulnerable to the effects of climate variability and change. In addi- tion, it plays an important role in mitigating climate change through improving soil organic matter and increasing vegetation cover. In-depth assessments of SLM practices from desertification sites enabled an evaluation of how SLM addresses prevalent dryland threats. The impacts mentioned most were diversified and enhanced production and better management of water and soil degradation, whether through water harvesting, improving soil moisture, or reducing runoff. Among others, favourable local-scale cost-benefit relationships of SLM practices play a crucial role in their adoption. An economic analysis from the WOCAT database showed that land users perceive a large majority of the technologies as having benefits that outweigh costs in the long term. The high investment costs associated with some practices may constitute a barrier to adoption, however, where appropriate, short-term support for land users can help to promote these practices. The increased global concerns on climate change, disaster risks and food security redirect attention to, and trigger more funds for SLM. To provide the necessary evidence-based rationale for investing in SLM and to reinforce expert and land users assessments of SLM impacts, more field research using inter- and transdisciplinary approaches is needed. This includes developing methods to quantify and value ecosystem services, both on-site and off-site, and assess the resilience of SLM practices, as currently aimed at within the EU FP7 projects CASCADE and RECARE.

The purpose of the cruise was to map the manganese rich pavements of the Blake Plateau area which had been extensively investigated by ships of the Woods Hole Oceanographic Institution since 1956. From August until September 1965, 112 sites were sampled from R/V Gosnold on a joint USGS-WHOI expedition. At most of the stations on the Blake pavement, large slabs of manganese were recovered along with phosphate rich nodules.

Esta tesis doctoral propone una metodología de estimación de demanda de tráfico de Internet basada en la caracterización de perfiles de usuario de Internet, con el objetivo de analizar el rendimiento y dimensionamiento de una red de acceso. Se realiza un exhaustivo análisis del estado del arte clasificado en tres partes. La primera parte se encuentra relacionada con la caracterización de usuarios en Internet. Incluye un estudio de las metodologías de extracción de conocimiento basado en técnicas de minería de datos, y un análisis de modelos teóricos y estudios previos de usuarios de Internet. En la segunda parte, se incluye un análisis de modelos teóricos para caracterizar fuentes de tráfico de aplicaciones de Internet, así como un estudio comparativo de los modelos de tráfico ON/OFF para un conjunto de aplicaciones representativas de Internet. En la última parte, se incluye un estudio de las arquitecturas de redes de acceso más relevantes y se propone un modelo genérico de arquitectura de red de acceso. Esta tesis doctoral define un marco metodológico basado en Procesos de Descubrimiento de Conocimiento (KDPs), con el que extraer, identificar y caracterizar, a los usuarios de Internet a partir de fuentes de información estadística. Se ha aplicado esta metodología a los usuarios residenciales en España y se ha identificado una distinción clara entre No-Usuarios (47%) y Usuarios de Internet (53%). Dentro de los usuarios de Internet se han extraído 4 perfiles de usuarios: Esporádicos (16%), Instrumentales (10%), Sociales (14%) y Avanzados (13%). Esta metodología también ha sido aplicada a años anteriores con el fin de realizar un pronóstico de la evolución de la tipología de usuarios de Internet en España. A continuación, se propone un método de estimación de demanda de tráfico basado en los perfiles de usuario de Internet identificados, con el objetivo de analizar el rendimiento de la red de acceso subyacente. Esta metodología se encuentra basada en 3 modelos: red de acceso, tráfico de red y perfiles de usuario y aplicaciones. Por último, la tesis presenta un modelo y una herramienta de simulación con la que se implementa el método de estimación de demanda anteriormente descrito. El modelo y la herramienta de simulación han sido validados frente a un modelo analítico mediante el uso de un escenario simplificado basado en fuentes de tráfico ON/OFF homogéneas. Mediante el uso de la herramienta de simulación desarrollada, se aplica la metodología de estimación de demanda a dos casos de uso, que se corresponden a dos escenarios de redes de acceso idénticas, a excepción de la caracterización de los usuarios de la misma. En el primer caso de uso, la red de acceso se caracteriza por los perfiles de usuario residenciales de Internet identificados para el año 2012, y en el segundo caso de uso, se utiliza el pronóstico de evolución de perfiles de usuario de Internet para el año 2017. Se concluye con una comparación del rendimiento de la red de acceso para ambos casos de uso, a partir del análisis del Grado de Servicio (GoS) de ambos escenarios.

A presente pesquisa busca avaliar exegeticamente o texto que se encontra na Bíblia, especificamente no livro de Números capítulos 22-24 que relata sobre um personagem conhecido como Balaão. A pesquisa tem também como objeto o estudo sobre o panteão de divindades relatado no mesmo texto, assim como também o estudo dos textos descobertos em Deir Alla, na Jordânia, que apresentam um personagem designado como Balaão, possivelmente o mesmo personagem de Nm 22-24. A motivação que levou ao desenvolvimento dessa pesquisa foi o fato de se ter deparado com os conceitos dos diversos nomes divinos exibidos no texto, além da questão do profetismo fora de Israel, assim como as possibilidades hermenêuticas que se abrem para a leitura desse texto bíblico. O conceito geral sempre foi o de que Israel era a única nação onde existiam “verdadeiros” profetas e uma adoração a um único Deus, o “monoteísmo”. O que despertou interesse foi perceber, especialmente por meio da leitura dos livros bíblicos, que o profetismo não se restringiu somente a Israel. Ele antecede à formação do antigo Israel e já existia no âmbito das terras do antigo Oriente Médio, e que Israel ainda demorou muito tempo para ser monoteísta. Quem é esse Balaão, filho de Beor? Estudaremos sobre sua pessoa e sua missão. Examinaremos os textos de Deir Alla sobre Balaão e sua natureza de personagem mediador entre o divino e o humano. Esse personagem é apresentado como um grande profeta e que era famoso como intérprete de presságios divinos. Analisaremos a importante questão sobre o panteão de deuses que são apresentados na narrativa de Balaão nomeados como: El, Elyon Elohim e Shaddai, além de Yahweh. Entendemos, a princípio, que o texto possui uma conexão com a sociedade na qual foi criado e usando da metodologia exegética, faremos uma análise da narrativa em questão, buscando compreender o sentido do texto, dentro de seu cenário histórico e social. Cenário este, que nos apresentou esse profeta, não israelita, que profere bênçãos dos deuses sobre Israel e que, além disso, pronuncia maldições sobre os inimigos desse mesmo Israel. Percebemos que, parte do texto pesquisado é apresentado sob a ótica de Israel sobre as outras nações. A pesquisa defende, portanto, que o texto de Nm 22-24, além de nos apresentar um profeta fora de Israel igual aos profetas da Bíblia, defende que, o panteão de divindades também era adorado por Israel e que tais nomes são epítetos de uma mesma divindade, no caso YHWH. Defende, também, um delineamento de um projeto de domínio político e militar de Israel sobre as nações circunvizinhas.

How do secretory proteins and other cargo targeted to post-Golgi locations traverse the Golgi stack? We report immunoelectron microscopy experiments establishing that a Golgi-restricted SNARE, GOS 28, is present in the same population of COPI vesicles as anterograde cargo marked by vesicular stomatitis virus glycoprotein, but is excluded from the COPI vesicles containing retrograde-targeted cargo (marked by KDEL receptor). We also report that GOS 28 and its partnering t-SNARE heavy chain, syntaxin 5, reside together in every cisterna of the stack. Taken together, these data raise the possibility that the anterograde cargo-laden COPI vesicles, retained locally by means of tethers, are inherently capable of fusing with neighboring cisternae on either side. If so, quanta of exported proteins would transit the stack in GOS 28–COPI vesicles via a bidirectional random walk, entering at the cis face and leaving at the trans face and percolating up and down the stack in between. Percolating vesicles carrying both post-Golgi cargo and Golgi residents up and down the stack would reconcile disparate observations on Golgi transport in cells and in cell-free systems.

We report the results of two studies examining the genetic overlap between schizophrenia and velocardiofacial syndrome. In study A, we characterize two interstitial deletions identified on chromosome 22q11 in a sample of schizophrenic patients. The size of the deletions was estimated to be between 1.5 and 2 megabases. In study B, we examine whether variations in deletion size are associated with the schizophrenic phenotype in velocardiofacial syndrome patients. Our results show that a region of the genome that has been previously implicated by genetic linkage analysis can harbor genetic lesions that increase the susceptibility to schizophrenia. Our findings should facilitate identification and cloning of the schizophrenia susceptibility gene(s) in this region and identification of more homogeneous subgroups of patients.
