Drosophila sturtevanti (37 strains) showed eighteen inversions, five new and thirteen previously described. Among these strains, 24 were maintained for seven to 21 years under laboratory conditions, eight for less than 1 year, and six were natural samples analysed in the first generation after collection. Flies from natural samples were the most polymorphic in the number of different inversions as well as in the frequency of flies bearing heterozygous inversions. In all cases, chromosome III presented the greatest number of inversions, and most of them occurred in strains from the Amazonian region. The data obtained were consistent with the hypothesis that the inversion variability of a species is proportional to the variability of its habitats.

Fertility (percentage of fertile crosses) and the degree of synapsis in salivary gland chromosomes in isofemale lines of Drosophila buzzatii, D. serido, D. koepferae and D. seriema were analysed. D. buzzatii was completely sterile in intercrosses with strains from the other species except for D. koepferae. The other species intercrossed to a greater or lesser degree, but also differed between crossing directions. Homologous pairing in salivary gland preparations of strains and hybrids conformed with the data on fertility. The lowest degree of synapsis was present in hybrids between D. koepferae and D. buzzatii, D. seriema and D. koepferae and D. koepferae and D. serido. These species also exhibited the lowest degree of reproductive compatibility. Hybrids between D. seriema and D. serido showed an intermediate degree of synapsis (pairing absent in the proximal and distal chromosome ends), as well as fertility greater than that found in the other interspecific crosses. Results of the fertility of crosses involving strains of a single species, compared with data in the literature, indicated that intraspecific divergence occurred in D. serido and D. koepferae.

The occurrence, number of insertion sites and antisense RNA expression of micropia transposable element were studied in 26 species that belong to three subgroups (mercatorum, mulleri and hydei) of repleta group of Drosophila. Under high specific PCR, micropia sequences were detected in 11 species, but under less stringent condition, this retrotransposon was detected in all species. The widespread distribution of micropia suggests that this element was already present at the common ancestor of the repleta group of Drosophila. Southern blot analysis showed a variation from 0 to 17 different insertion sites and the occurrence of male-specific sequences. We found that the expression of the 1.0 kb micropia antisense RNA is variable among the species and tissues (soma and testis), which suggests that more than one mechanism regulates transposition in these species. Variation of amplification by PCR and of antisense RNA expression, as well as divergence of nucleotide sequences among the species allow us to suggest that at least two subfamilies of micropia transposable element are harbored by the genome of this species group.

Few are studies on P elements that have addressed the saltans group. These studies had shown that species from the cordata and elliptica subgroups were devoid of any discernible P homologous sequences, while species from the parasaltans, sturtevanti, and saltans subgroups all contain P element sequences. Our analyses showed the presence of one to 15 P element insertion sites in species of the saltans group, including Drosophila neocordata and Drosophila emarginata (cordata and elliptica subgroups, respectively). From these species, only those from the parasaltans, sturtevanti, and saltans subgroups harbor canonical P elements and, only those of the last two subgroups seem to harbor putative full-sized elements. Due to the low similarity of the sequences found in D. neocordata and D. emarginata to those earlier described, we suggest that these sequences might be rudimental P element derivatives that were present in the ancestral of the subgenus Sophophora. Horizontal transfer ( HT), defined as the transfer of genetic material between species, is considered to be an essential step in the 'life cycle' of transposable elements. We present a broad overview of suspected cases of HT of transposable elements in Drosophila. Hundred-one putative events of HT have been proposed in Drosophila for 21 different elements (5.0% refer to non-long terminal repeat (LTR) retrotransposons, 42.6% to LTR retrotransposons and 52.4% to DNA transposons). We discuss the methods used to infer HT, their limits and the putative vectors of transposable elements. We outline all the alternative hypotheses and ask how we can be almost certain that phylogenetic inconsistencies are due to HT.

Esterases are known for their involvement in several physiological processes and high degree of polymorphism, in many organisms. Such polymorphism has been used to characterize species and species groups and to study genetic changes occurred in their evolutionary history. In the present study, the esterase patterns of 19 strains from 10 species representative of the five subgroups of the saltans species group were analyzed using polyacrylamide gel electrophoresis and alpha- and beta- naphthyl acetates as substrates. Fifty-one esterase bands were detected and classified as 31 alpha-esterases, 18 beta-esterases and two alpha/beta-esterases. on the basis of the inhibition patterns using Malathion and eserine sulfate, 34 bands were classified as carboxylesterases, 14 as acethylesterases and three as cholinesterases. Ten gene loci were tentatively established on the basis of data on band position in the gel, substrate preference and inhibition pattern. Twenty bands were species-specific, the remaining being shared by species from the same or different subgroups. Bands detected exclusively in males and bands with a different frequency or degree of expression between sexes were also detected. In the gels prepared for analysis of gene expression in the body parts (head, thorax and abdomen), the degree of expression of the beta-esterases was higher in the thorax, while the alpha-esterases were expressed predominantly in the abdomen and thorax. A global view of the data available at present on the esterases of the species from the saltans group and their degree of polymorphism are presented, as well as the possibility of using some beta-esterases, because of their characteristics in the gels, as markers for species identification.

Salivary gland cells of Drosophila mulleri/D. arizonensis aneuploid male hybrids carrying 3 microchromosomes exhibited morphological features which indicate heterochromatinization of one of the small polytene chromosomes. The process apparently changes the chromosome surface producing a coating with a net-like structure and a strong affinity for lacto-acetic orcein. The possibility of a dosage compensatory mechanism operating to counteract the effect of the extra chromosome is discussed on the basis of previous data which indicated that the microchromosomes of these species have ribosomal cistrons and are controlled by regulatory mechanisms especially evident in hybrids. © 1981 Dr W.
