Redox processes are involved in the mechanism of action of NADPH oxidase inhibitors such as diphenyleneiodonium and apocynin. Here, we studied the structure-activity relationship for apocynin and analogous ortho-methoxy-substituted catechols as inhibitors of the NADPH oxidase in neutrophils and their reactivity with peroxidase. Aiming to alter the reduction potential, the ortho-methoxy-catechol moiety was kept constant and the substituents at para position related to the hydroxyl group were varied. Two series of compounds were employed: methoxy-catechols bearing electron-withdrawing groups (MC-W) such as apocynin, vanillin, 4-nitroguaiacol, 4-cyanoguaiacol, and methoxy-catechol bearing electron-donating groups (MC-D) such as 4-methylguaiacol and 4-ethylguaiacol. We found that MC-D were weaker inhibitors compared to MD-W. Furthermore, the radicals generated by oxidation of MC-W via MPO/H(2)O(2), but not for MC-D, were able to oxidize glutathione (GSH) as verified by the formation of thiyl radicals, depletion of GSH, and recycling of the ortho-methoxy-catechols during their oxidations. The capacity of oxidizing sulfhydryl (SH) groups was also verified when ovalbumin was incubated with MC-W, but not for MC-D. Since the effect of apocynin has been correlated with inactivation of the cytosolic fractions of the NADPH oxidase complex and its oxidation during the inhibitory process develops a special role in this process, we suggest that the close relationship between the reactivity of the radicals of MC-W compounds with thiol groups and their efficacy as NADPH oxidase inhibitor could be the chemical pathway behind the mechanism of action of apocynin and should be taken into account in the design of new and specific NADPH oxidase inhibitors. Reactive oxygen species (ROS) are constantly produced by cells that promote cellular oxidative damage and are neutralized by an antioxidant system including superoxide dismutase, glutathione, peroxidase and catalase. Male volunteers were exercised for 20 minutes, three days (60, 70 and 80% of maximum heart rate). Catalase activity and plasma malondialdehyde concentration were measured. The mean age of the volunteers was 25 +/- 7 years, with body mass index 2 of 24.03 +/- 4.32 kg/m(2). Acute exercise training produced an increase of malondialdehyde concentration that was exercise intensity-dependent in young volunteers. However, catalase activity shows a great variability at baseline and the percentual of reduction was exercise intensity-independent in this particular population. Therefore, our study shows that acute cycling exercise promotes an increase of oxidative stress that was exercise intensity-dependent in young volunteers. Furthermore, the antioxidant system measured by catalase activity was effective to counterbalance the ROS production showing a saturation behavior at an intensity of 70% of maximum heart rate.

The behaviour of nitrobenzenesulfonamide anion radicals generated from the electrochemical reduction of aliphatic and aromatic amines protected by nitrobenzenesulfonyl (nosyl) groups in N,N-' dimethylformamide has been reported. The species have been characterized by voltammetry and optical and electron spin resonance spectroscopies. The visible spectra of the anion radicals were recorded and the hyperfine splitting constants were assigned to specific proton positions and nitrogen nuclei of the molecule. The stabilities of the anion radicals are affected by electronic properties of the protecting group and specific features of the amines, which show direct influence on the route of cathodic cleavage of the nitrobenzenesulfonamides.

This paper reports the construction of an axisymmetric nonpremixed piloted jet burner, with well-defined initial and boundary conditions, known as the Delft burner, to assess turbulence-chemistry interaction in non-premixed turbulent flames. Detailed experimental information is described, involving hot-wire anemometry, thin-wire thermocouples and chemiluminescence visualization measurements. Radial profile of the axial mean velocity indicates excellent agreement between flow patterns developed within Delft installation and the one described herein. Chemiluminescence emissions from CH and C2 free-radicals were acquired with a CCD camera. Tomography reconstruction analysis was utilised to compare radical emissions and temperature spatial distributions. There was a strong dependence between temperature and CH/C 2 emissions. This is an indication that these radicals can be used in flame front studies.

Reactive oxygen species (ROS) and free radical species have been implicated in initiating, accompanying or causing many diseases in living organisms; there is thus, a continual need for antioxidants molecules to inactivate ROS/free radicals. Many studies of plants crude extracts have demonstrated free-radical scavenging and antioxidant action. Maytenus species have long been used, in several countries, as traditional medicines against gastric ulcers, dyspepsia and others gastric problems and for their anti-inflammatory properties. In this study, Maytenus aquifolium (Celastraceae) root bark ethanol extract was assessed for its ability to scavenge free radicals and reactive oxygen species. The results were expressed as percentage inhibition of the active species. The extract was efficient against studied reactive species: DPPH radical (obtained inhibition = 35.5 ± 1.3 %), ABTS.+ (IC50 = 0.0036 ± 0.0003 mg/mL), HOCl (IC50 = 0.002 ± 0.0001 mg/mL), O2 .- (obtained inhibition = 36.0 ± 2.1 %), and NO. Biological activities of flavonoids have been extensively reviewed in literature. The biochemical profile of afzelin, kaempferitrin, and pterogynoside acting on reactive oxygen species was investigated in this paper. The flavonoids were able to act as scavengers of the superoxide anion, hypochlorous acid and taurine chloramine. Although flavonoids are naturally occurring substances in plants which antioxidant activities have been widely advertised as beneficial, afzelin, kaempferitrin, and pterogynoside were able to promote cytotoxic effect. In red blood cells this toxicity was enhanced, depending on flavonoids concentration, in the presence of hypochlorous acid, but reduced in the presence of 2,20 -azo-bis(2-amidinopropane) free radical. These flavonoids had also promoted the death of neutrophils, which was exacerbated when the oxidative burst was initiated by phorbol miristate acetate. Therefore, despite their well-known scavenging action toward free radicals and oxidants, these compounds could be very harmful to living organisms through their action over erythrocytes and neutrophils.

This study evaluated the effect of extract of Aloe vera in the transport water of matrinxã (Brycon amazonicus) fish on stress response and leukocyte respiratory activity. Fish was transported for 4 h in water containing Aloe at levels 0; 0.02; 0.2 and 2 mg/L, and sampled before transport 2, 4, 24 and 96 h after for determination of plasma glucose and respiratory activity of leukocytes. An additional in vitro assay was conducted with another fish species, pacu (Piaractus mesopotamicus), to test the respiratory burst of leukocytes exposed to Aloe extract (0.0, phosphate-buffered saline (PBS) only) at 0.1, 0.2, 0.5 and 1 mg/L). Plasma glucose increased after 2 and 4 h of transport and returned to control levels within 24 h, but the addition of Aloe in the transport water did not affect the level of blood glucose. However, at 2 h of transport, Aloe enhanced the respiratory activity of leukocytes in a dose-dependent way. The highest value of respiratory burst activity of leukocytes was observed in the fish transported in water containing Aloe at 2 mg/L. The enhancing effect of the plant extract on the production of oxygen radicals was confirmed in vitro in leukocytes of pacu incubated in Aloe at concentrations 0.1 and 0.2 mg/L. The results suggest that Aloe vera is a modulator of the immune system in fish improving the innate immune response tested.

O presente trabalho foi desenvolvido com o objetivo de verificar a influência do uso contínuo de vários herbicidas residuais, sobre a distribuição das radicelas de laranjeiras Natal (Citrus sinensis (L.) Osbeck) enxertadas sobre limão Cravo (Cithus limonia Osbeck) e plantadas em janeiro de 1970. O experimento foi instalado no pomar de produção da Faculdade de Ciências Agrárias e Veterinárias do Campus de Jaboticabal (UNESP), em Latossol Roxo distrófico, com 1,6% de matéria orgânica e 51% de argila. O delineamento experimental utilizado foi o de blocos ao acaso, dentro de um esquema fatorial 11x 3x2x2, com três repetições. Os tratamentos utilizados, com as respectivas doses em kg do i.a/ha foram: fluometuron a 4,2; simazíne a 4,8; atrazine a 4,8; bromacil a 3,2;bromacil (40%) + diuron (40%) a 4,8; bromacil (53,3%) + diuron (27,6%) a 4,8; terbacil a 3,2; oxadiazon a 1,5; dichlobenil a 3,0 e 6,0; além de uma testemunha capinada. O estudo das radicel as foi feito pelo método do trado, tendo sido tomadas amostras de solo contendo radicelas, nas distâncias de 80, 160 e 240 cm do tronco, nas camadas de 0 a 15 cm e 15 a 30 cm de profundidade e em duas direções distintas (entrelinhas e entreplantas). Os resultados mostraram que as radicelas localizam-se superficialmente, com 70% delas nos primeiros 15 cm, do total encontrado na camada superficial de 0 a 30 cm de profundidade do solo. Cerca de 75% das radicelas situam-se até a distância de 160 cm do tronco. Na camada mais superficial do solo (0 a 15 cm), a quantidade de radicelas diminuiu de maneira acentuada, à medida que se afastou do tronco. Na camada de 15 a 30 cm de profundidade, ocorreu uma distribuição horizontal mais uniforme, comparada com a da camada de 0 a 15 cm. As radicelas das plantas do cultivar Natal distribuiram-se uniformemente, tanto na direção das plantas da linha vizinha, quanto nas das plantas de uma mesma linha, para o espaçamento de 7,0 m x 7,2 m. Não se detectaram diferenças estatisticamente significativas nas quantidades de radicelas entre as parcelas dos vários tratamentos com herbicidas, excluindo-se desta forma, qualquer efeito negativo de tais produtos químicos sobre o sistema radicular das laranjeiras.

Dietary modifications may significantly reduce cardiovascular disease (CVD) risk factors, including cholesterol and atherosclerosis. The present study addressed the effects of the crude extract from the pulp fruit of Tamarindus indica L. on lipid serum levels and early atherosclerotic lesions in hypercholesterolemic hamsters in vivo, and the extract's antioxidant action, in vitro. Animals were fed on either chow or atherogenic diet during 10 weeks and concomitantly received either water or T indica L. extract for drinking. Treatment of hypercholesterolemic hamsters with the T. indica pulp fruit extract (5%) led to a decrease in the levels of serum total cholesterol (50%), non-HDL cholesterol (73%) and triglyceride (60%), and to an increase of high-density lipoprotein (HDL) cholesterol levels (61%). In vitro, the extract presented radical scavenging ability, as assessed by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) and superoxide radicals assays, and led to decreased lipid peroxidation in serum, as assessed by the thiobarbituric acid reactive substances (TBARS) assay. In vivo, the extract improved the efficiency of the antioxidant defense system, as assessed by the superoxide dismutase, catalase and glutathione peroxidase activities. Together these results indicate the potential of tamarind extracts in diminishing the risk of atherosclerosis development in humans. (c) 2005 Elsevier Ltd. All rights reserved.

O objetivo deste trabalho foi avaliar o desenvolvimento e a produtividade de colmos de cana-de-açúcar, resultantes da aplicação de reguladores vegetais no iníco da safra. Ostratamentos consistiram na aplicação de três reguladores vegetais inibidores de crescimento - sulfometuron metil, glifosato e compostos de radicais carboxílicos orgânicos + glifosato - e na maturação natural como testemunha, em delineamento experimental em blocos ao acaso, com cinco repetições. Os maturadores retardam o processo de crescimento em altura das plantas, sem afetar o número e o diâmetro de colmos na colheita, e influenciam de forma e intensidade distintas a ocorrência do florescimento e chochamento. O glifosato proporciona elevados índices de brotação lateral e prejudica a rebrota da soqueira. Os maturadores induzem o aumento do teor de açúcares redutores totais, o que contribui para a melhoria da qualidade tecnológica da cana-de-açúcar.
