Phospholipases A(2) are components of Bothrops venoms responsible for disruption of cell membrane integrity via hydrolysis of its phospholipids. A class of PLA(2)-like proteins has been described which despite PLA(2) activity on artificial substrate, due to a D49K mutation, is still highly myonecrotic. This work reports the X-ray structure determination of two Lys49-PLA(2)s from Bothrops neuwiedi pauloensis (BnSP-7 and BnSP-6) and, for the first time, the comparison of eight dimeric Lys49-PLA2s. This comparison reveals that there are not just two (open and closed) but at least six different conformations. The binding of fatty acid observed in three recent Lys49-PLA(2) structures seems to be independent of their quaternary conformation. Cys29 polarization by Lys122 is not significant for BnSP-7 and BnSP-6 or other structures not bound by fatty acids. These structures may be in an active state when nothing is bound to them and the Lys122/Cys29 interactions are weak or absent. (C) 2003 Elsevier B.V. All rights reserved.

One of the main components of snake venoms are the Asp49-phospholipases A(2), also known as svPLA(2)s. The study of these toxins is a matter of great scientific interest due to their wide variety of biological effects. In this work we present strong evidences found in literature and other aspects which strengthen the importance of quaternary assembly for understanding the activities and molecular evolution of svPLA(2)s.

The upper portion of the Rio Guaratuba, located in the Serra do Mar coastal range of Southeastern Brazil, shows biological and geological evidences of being captured and diverted away from its original course in the direction of the Parana River Basin to become a coastal river due to the Quaternary activity of NW-trending faults. Despite draining directly to the Atlantic Ocean, the upper portion of the Guaratuba still maintains an ichthyofauna that is typical of the adjoining Parana River Basin rather than the characteristic fish fauna of the Brazilian coastal drainages. The fish fauna of the upper Guaratuba is an evident testimony of the tectonic process that allowed the faunal interchange between the upland basins and the coastal drainages that probably has been taking place in Southeastern Brazil throughout the long geological history of the passive Brazilian continental margin.

Lotus tetragonolobus lectin (LTA) is a fucose-specific legume lectin. Although several studies report a diverse combination of biological activities for LTA, little is known about the mechanisms involved in L-fucosyl oligosaccharide recognition. The crystal structure of LTA at 2.0 angstrom resolution reveals a different legume lectin tetramer. Its structure consists of a homotetramer composed of two back-to-back GS4-like dimers arranged in a new mode, resulting in a novel tetramer. The LTA N-linked carbohydrate at Asn4 and the unusual LTA dimer-dimer interaction are related to its particular mode of tetramerization. In addition, we used small angle X-ray scattering to investigate the quaternary structure of LTA in solution and to compare it to the crystalline structure. Although the crystal structure of LTA has revealed a conserved metal-binding site, its L-fucose-binding site presents some punctual differences. Our investigation of the new tetramer of LTA and its fucose-binding site is essential for further studies related to cross-linking between LTA and complex divalent L-fucosyl carbohydrates. The Pantanal is a tectonic depression located at the left margin of the Upper Paraguay River. The Paraguay is the trunk river of an alluvial depositional tract composed by several large marginal alluvial fans, the Taquari fan being the largest one. The present landscape is a complex tropical wetland characterized by month-long floods every year, with geomorphic features derived from the present conditions and others inherited from successive Pleistocene and Holocene climates. Some areas containing ponds are landscape relicts generated by eolian deflation during the Last Glacial Maximum. Many ponds, closed depressions isolated from the superficial waters by vegetated crescent ridges of fine sands, were interpreted as salt pans bordered by lunette sand dunes. Initiation of the modern wetland has occurred during the Pleistocene/Holocene transition, with the change to a more humid climate and the individualization of lacustrine systems. Active tectonics has been playing an important role in the development of the Pantanal landscape. Nowadays, the Paraguay River meanders in a large flood plain with extensive swamp surfaces, being structurally constrained by faults in the west border of the basin. Sedimentation within the Pantanal wetland is also affected by tectonic activity, especially along faults associated with the Transbrasiliano Lineament. (C) 2003 Elsevier B.V. Ltd and INQUA. Two series of new chitosan derivatives were synthesized by reaction of deacetylated chitosan (CH) with propyl (CH-Propyl) and pentyl (CH-Pentyl) trimethylammonium bromides to obtain derivatives with increasing degrees of substitution (DS). The derivatives were characterized by 1H NMR and potentiometric titration techniques and their antifungal activities on the mycelial growth of Aspergillus flavus were investigated in vitro. The antifungal activities increase with DS and the more substituted derivatives of both series, CH-Propyl and CH-Pentyl, exhibited antifungal activities respectively three and six times higher than those obtained with commercial and deacetylated chitosan. The minimum inhibitory concentrations (MIC) were evaluated at 24, 48 and 72h by varying the polymer concentration from 0.5 to 16g/L and the results showed that the quaternary derivatives inhibited the fungus growth at polymer concentrations four times lower than that obtained with deacetylated chitosan (CH). The chitosans modified with pentyltrimethylammonium bromide exhibited higher activity and results are discussed taking into account the degree of substitution (DS). © 2012 Elsevier GmbH.
