The Tahiti lime appears very susceptible to attack by post-harvest diseases, primarily by the fungi Penicillium and Phomopsis, and also because of its high sensitivity to storage at low temperatures. In order to reduce such damage, the present study aimed to verify the efficiency of heat treatment and disinfection of pathogens in the prevention of post-harvest chilling injury of this cultivar and to compare this treatment with other products using the conventional fungicides. The heat treatments were studied with hot-water temperatures ranging between 48 and 56° C. Water at room temperature was used as a control treatment. After treatment, the fruits were kept under cold temperature at 10° C and RH 90% for about 45 days. For comparison, three other treatments were carried out simultaneously, one using imazalil, one with baking soda, and a third with sodium carbonate, these three products being applied by baths in cold water. Two groups of fruit were evaluated, one treated by immersion considering pathogens coming from the field and another by inoculation with spores of the previously isolated pathogens. For the evaluation of physical and chemical parameters of fruits, determinations were made of the skin color, texture, weight loss, size, juice yield, soluble solids, total acidity and vitamin C content. The determination of the sensitivity of the fruit to cold was made by their exposure at temperatures inducing cold damage. The design was a randomized block design with nine treatments, analyzed by the Statgraphics statistical package. Heat treatments, especially at 52° C, were shown to be more promising in the control of pathogenic fungi and cold damage, surpassing the conventional fungicides. No changes were found in the intrinsic and extrinsic parameters in relation to the application of the different treatments.

Pinus taeda is one of the main timber trees in Brazil, occupying 1.8 million ha with an annual productivity of 25-30 m(3) ha(-1). Another important species is Araucaria angustifolia, belonging to the fragile Rainforest biome, which for decades has been a major source of timber in Brazil. Some diseases that affect the roots and/or the stem of these trees and cause "damping-off" of the seedlings, with economic and environmental losses for the forest sector, are caused by the plant pathogenic fungi Fusarium sp. or Armillaria sp. This research project intended to isolate actinobacteria from the Araucaria rhizosphere, which present an antagonistic effect against these fungi. After the selection of the best pathogen inhibitors, morphologic characteristics, enzyme production, and their effect on the growth of Pinus taeda were studied. The actinobacteria were tested for their antagonistic capacity against Fusarium sp. in Petri plates with PDA as substrate. The inhibition zone was measured after 3, 5, 7, and 10 days. Of all the isolates tested, only two of them maintained inhibition zones up to 4 mm for 10 days. The inhibition of Armillaria sp. was tested in liquid medium and also in Petri dishes through the evaluation of the number of the fungal rhizomorphs in dual culture with the actinobacteria. It was found that all five isolates were able to inhibit the rhizomorph production, with the best performance of the isolate A43, which was capable of inhibiting both fungi, Fusarium and Armillaria. In a greenhouse experiment, the effect of five isolates on the growth of Pinus taeda seedlings was tested. Plant height, stem diameter, root and shoot dry matter were determined. The Streptomyces isolate A43 doubled plant growth. These results may lead to the development of new technologies in the identification of still unknown bacterial metabolites and new management techniques to control forest plant diseases.

Ergosterol peroxide, a presumed product of the H2O2-dependent enzymatic oxidation of ergosterol, has been isolated from yeast from yeast forms of the pathogenic fungus Sporothrix schenckii. The substance, which may have a role in fungal virulence, has been characterized mainly using spectroscopic methods (1H and 13C nuclear magnetic resonance and high resolution mass spectra). The purified compound showed a molecular formula of C28H44O3, displaying characteristic features of epidioxy sterols and was reverted to ergosterol when submitted to S. schenckii enzymatic extract.

O isolamento e a manutenção de fungos basidiomicetos simbiontes de formigas da tribo Attini tem sido dificultado pela baixa velocidade de crescimento desses fungos, bem como pela presença de muitos microrganismos que vivem na superfície do material que as formigas mantêm no interior nos ninhos como substrato para o crescimento dos seus fungos simbiontes. No presente trabalho nós descrevemos um método que aumenta em mais de sete vezes a eficiência de isolamento desses fungos, quando comparada àquela obtida por procedimentos tradicionais. Ninhos subterrâneos de formigas atíneas dos gêneros Atta, Acromyrmex, Trachymyrmex e Mycetarotes foram localizados e deles foram coletadas amostras contendo fungos simbiontes e formigas, que foram transportadas para o laboratório, onde as formigas foram capazes de limpar a cultura do fungo e estimular o seu crescimento. em seguida, porções dos micélios foram assepticamente coletadas e transferidas para meio Yeast Nitrogen Base contendo glicose e cloranfenicol. Para facilitar a manutenção dos isolados em culturas de laboratório, diferentes nutrientes foram analisados para a elaboração de um meio de cultivo complexo, que possibilitou aumentar a velocidade de crescimento dos fungos e estocá-los por longos períodos. O método foi aplicado com sucesso para os fungos simbiontes de todos os gêneros de formigas estudados, gerando, assim, um procedimento extremamente útil para a formação e manutenção de uma coleção representativa de diferentes fungos simbiontes de formigas da tribo Attini.

Leaf-cutting ants of the genera Atta and Acromyrmex (tribe Attini) are symbiotic with basidiomycete fungi of the genus Leucoagaricus (tribe Leucocoprineae), which they cultivate on vegetable matter inside their nests. We determined the variation of the 28S, 18S, and 5.8S ribosomal DNA (rDNA) gene loci and the rapidly evolving internal transcribed spacers 1 and 2 (ITS1 and ITS2) of 15 sympatric and allopatric fungi associated with colonies of 11 species of leafcutter ants living up to 2,600 km apart in Brazil. We found that the fungal rDNA and ITS sequences from different species of ants were identical (or nearly identical) to each other, whereas 10 GenBank Leucoagaricus species showed higher ITS variation. Our findings suggest that Atta and Acromyrmex leafcutters living in geographic sites that are very distant from each other cultivate a single fungal species made up of closely related lineages of Leucoagaricus gongylophorus. We discuss the strikingly high similarity in the ITS1 and ITS2 regions of the Atta and Acromyrmex symbiotic L. gongylophorus studied by us, in contrast to the lower similarity displayed by their non-symbiotic counterparts. We suggest that the similarity of our L. gongylophorus isolates is an indication of the recent association of the fungus with these ants, and propose that both the intense lateral transmission of fungal material within leafcutter nests and the selection of more adapted fungal strains are involved in the homogenization of the symbiotic fungal stock.

Estudou-se o crescimento micelial de dez isolados de Diaporthe citri, utilizando-se seis meios de cultura (aveia-ágar, maltose-peptona-ágar, batata-dextrose-ágar, folha de laranja-dextrose-ágar, folha de limão-dextrose-ágar, milho-ágar) à temperatura de 22 ± 2 °C e fotoperíodo de 12 h claro/12 h escuro. O cultivo em meio de batata-dextrose-ágar (BDA) foi conduzido em cinco temperaturas diferentes (10, 15, 20, 25 e 30 °C). Três diferentes regimes de luminosidade (12 h claro/12 h escuro, claro contínuo, escuro contínuo) foram utilizados para verificar o crescimento do fungo. Foram observadas variações na produção de picnídios e de massa micelial nos diferentes meios de cultura, temperaturas e regimes de luminosidade testados, sendo que, para a maioria dos isolados, o meio de cultura de aveia-ágar, a faixa de 20 a 25 °C e o regime de claro contínuo induziram maior crescimento micelial. A produção de picnídios foi maior para o regime de luz contínua. O teste de patogenicidade foi feito por inoculação de discos de micélio de 5 mm de diâmetro em ferimentos em ramos e caule de limão 'Feminelo' (Citrus limon) enxertado em citrumelo 'Swingle'(Poncirus trifoliolata x Citrus paradisi) e plantas de limão 'Cravo' (C. limonia) enxertados com laranja 'Valência' (C. sinensis). Após sete dias, houve o aparecimento de exsudação de goma nas plantas inoculadas com os isolados, mas não na testemunha. Todos os isolados mostraram-se patogênicos, sendo os isolados PC2 e PC5, os que causaram comprimento de lesão maior nas plantas.

Tem sido atribuído a vários patógenos reduções na qualidade de sementes de soja. Com o objetivo de avaliar o transporte dos fungos pelas sementes provenientes de plantas de soja, artificialmente inoculadas em condições de campo com dois isolados de Colletotrichum dematium var. truncata e um isolado de Phomopsis sojae, e seu efeito sobre a qualidade sanitária e fisiológica das mesmas, quatro variedades de soja (BRS 133, MSoy 6101, Conquista e Liderança) foram manualmente semeadas no campo, e as plantas obtidas foram inoculadas com C. dematium var. truncata isolado 1 e isolado 2, e P. sojae. A testemunha permaneceu sem inoculação. As sementes foram colhidas manualmente e submetidas a testes para avaliação da sanidade, teor de água, peso de 100 sementes, germinação em papel e emergência em areia. A desinfestação das sementes com hipoclorito de sódio a 1% por 3 minutos diminuiu a incidência dos patógenos nas mesmas. As sementes apresentaram uma alta infecção por Fusarium spp.. Phomopsis sojae foi encontrada em todos os tratamentos, e sua incidência nas sementes esteve relacionada com a incidência de Fusarium sp.. Há diferença na patogenicidade entre isolados de C. dematium var. truncata, sendo o isolado 1 o mais patogênico, com destaque para a variedade Liderança (25% de infecção por C. dematium var. truncata). Fusarium sp. influenciou negativamente a germinação de sementes da variedade MSoy 6101, independente do tratamento, e C. dematium var. truncata afetou negativamente a germinação das sementes das variedades Conquista e Liderança. Os valores de emergência em areia apresentaram-se significativamente superiores à germinação em papel, para as variedades MSoy 6101, Conquista e Liderança.

Brazilian soils predominantly consist of iron and aluminum oxides and have a low phosphorus content. The present study was carried out in order to assess the status of phosphate fractions in pasture, forest and agricultural soils and the ability of soil fungi to solubilize iron and aluminum phosphates. The abundance of P fractions in the soils studied occurred in the following order: Fe-P > reductant-soluble Fe-P > occluded Fe-P > occluded Al-P > Al-P > Ca-P. of the 481 fungi isolated, 33 showed the ability to solubilize the inorganic phosphates in culture. of these, 14 were considered to be high or very high solubilizers based on a solubilization capacity > 1000 mu g PO43- ml(-1). Isolate F-111 was the only one that dissolved all the insoluble phosphates used. Nine isolates solubilized both Al-P and Ca-P, and four other isolates only solubilized Ca-P. The highest number of isolates with high solubilization capacity were detected in pasture soil, followed by tropical rain forest and forest patch soils. Pasture soil presented both the largest contents of insoluble phosphates and the largest number of fungal isolates with phosphate-solubilizing ability. The range and size of P fractions influenced the number of fungi and their ability to solubilize hardly soluble phosphates. (c) 2004 Elsevier B.V. All rights reserved.
