Polyunsaturated fatty acids (PUFA) are essential structural components of the central nervous system. Their role in controlling learning and memory has been well documented. A nutrigenomic approach with high-density microarrays was used to reveal brain gene-expression changes in response to different PUFA-enriched diets in rats. In aged rats fed throughout life with PUFA-enriched diets, genes with altered expressions included transthyretin, α-synuclein, and calmodulins, which play important roles in synaptic  plasticity and learning. The effect of perinatal omega-3 PUFA supply on gene expression later in life also was studied. Several genes showed similar changes in expression in rats fed omega-3-deficient diets in the perinatal period, regardless of whether they or their mothers were fed omega-3 PUFA-sufficient diets after giving birth. In this experiment, among the down-regulated genes were a kainate glutamate receptor and a DEAD-box polypeptide. Among the up-regulated genes were a chemokine-like factor, a tumor necrosis factor receptor, and cytochrome c. The possible involvement of the genes with altered expression attributable to different diets in different brain regions in young and aged rats and the possible mode of regulatory action of PUFA also are discussed. We conclude that PUFA-enriched diets lead to significant changes in expression of several genes in the central nervous tissue, and these effects appear to be mainly independent of their effects on membrane composition. The direct effects of PUFA on transcriptional modulators, the downstream developmentally and tissue-specifically activated elements might be one of the clues to understanding the beneficial effects of the omega-3 PUFA on the nervous system.

RNA polymerase II transcribes genes encoding proteins and a large number of small stable RNAs. While pre-mRNA 3'-end formation requires a machinery ensuring tight coupling between cleavage and polyadenylation, small RNAs utilize polyadenylation-independent pathways. In yeast, specific factors required for snRNA and snoRNA 3'-end formation were characterized as components of the APT complex that is associated with the core complex of the cleavage/polyadenylation machinery (core-CPF). Other essential factors were identified as independent components: Nrd1p, Nab3p and Sen1p. Here we report that mutations in the conserved box D of snoRNAs and in the snoRNP-specific factor Nop1p interfere with transcription and 3'-end formation of box C/D snoRNAs. We demonstrate that Nop1p is associated with box C/D snoRNA genes and that it interacts with APT components. These data suggest a mechanism of quality control in which efficient transcription and 3'-end formation occur only when nascent snoRNAs are successfully assembled into functional particles.

A young man stabs a defenceless elderly man to death, and remarks “I’ll lug the guts into the neighbour room.” Martha Grace Duncan has argued that such apparent remorselessness and other forensic features must be interpreted differently in children and young people as compared to adult defendants, because of developmental effects. Professor Duncan discusses a range of fictional as well as real examples in pressing her claim, and also appeals to psychiatric, psychological and psychoanalytic expertise. In order to examine the general validity of her argument, it is hypothesised here that a Duncanian adolescence defense has been presented for Prince Hamlet who, miraculously revived, now stands his trial for murder. It is argued that the “adolescence defense” is unsound in principle and that children and youth (whether or not as superannuated as the Prince of Denmark) should be treated in the same forensic manner as adults. If we respect children and youth, we must respect their autonomy however uncomfortable for us this may be: “So young, my lord, and true.”

Plagiarism is viewed by many academics as a kind of Pandora's box—the elements contained inside are too frightening to allow escape for fear of the havoc that may result. Reluctance by academic members of staff to discuss student plagiarism openly may contribute to the often untenable situations we, as teachers, face when dealing with student plagiarism issues. In this article, I examine the dilemmas English for Academic Purposes (EAP) staff face when dealing with student plagiarism in the tertiary classroom. The perceptions of all 11 teachers involved in teaching a first year EAP writing subject at South-Coast University are detailed in light of the university's policy on plagiarism. My research indicates that not only is an agreed definition of plagiarism difficult to reach by members of staff teaching the same subject, but plagiarism is a multi-layered phenomenon encompassing a spectrum of human intention. Evaluating the spectrum can lead to differences in the implementation of university plagiarism policy, the result of which embodies issues of equity. The aim of the article is to encourage policy-makers and academic staff to acknowledge the concerns about implementation of plagiarism policy. Collaborative, cross-disciplinary re-thinking of plagiarism is needed to reach workable solutions.

Australia's box-ironbark forests and woodlands once covered about 14 per cent of the State of Victoria on the riverine plains and foothills of the Great Dividing Range. But approximately 83 per cent of the total original habitat has been destroyed and what remains of this significant ecosystem is now highly fragmented and vulnerable to further degradation. Moreover, only 14 per cent of the area remaining is on public land. A 10 year campaign on the part of the environmental movement eventually succeeded in forcing the State government to conduct an independent inquiry into this ecosystem and make recommendations on future management. This paper outlines the innovative public participation process adopted by the Victorian State government and the outcomes of the inquiry. A subsequent compensation package for commercial operations disadvantaged by the proclamation of a series of new national parks is also discussed, as are the shortcomings of a process that can have little or no impact on what happens on private land.

The most difficult aspect of teaching editing at a university tutorial is imparting to students a sensitivity to the appropriate relationship between the editor and author and, more particularly, between the editor and the author's work. Students are tempted to see themselves as critics or assessors of the author's work rather than assistants in the writing process. This paper discusses where teaching editing is difficult in a classroom, arguing that such difficulty is a symptom of both problems experienced in the editing profession and limitations of university teaching.

TATA box is one of the most important transcription factor binding sites. But the exact sequences of TATA box are still not very clear yet. In this study, we conducted a dedicated analysis on the frequency distribution of TATA Box and its extension sequences on human promoters. Sixteen TATA elements derived from TATA Box motif, TATAWAWN, were classified into three distribution patterns: peak, bottom-peak and bottom. Fourteen TATA extension sequences (up to two base extensions) were predicted to be the new TATA Box elements because of their high motif factors, which indicate their statistical significance. Statistical analysis on the promoters of mouse, zebrafish and drosophila melanogaster verified seven of these elements. It was also observed that the distribution of TATA elements on the promoters of housekeeping genes are very similar with their distribution on the promoters of tissue specific genes in human.

RNA interference (RNAi) mediated by DNA-based expression of short hairpin RNA (shRNA) is a powerful method of sequence-specific gene knockdown. A number of vectors for expression of shRNA have been developed that feature promoters from RNA polymerase III (pol III)-transcribed genes of mouse or human origin. To advance the use of RNAi as a tool for functional genomic research and for future development of specific therapeutics in the bovine species, we have developed shRNA expression vectors that feature novel bovine RNA pol III promoters. We characterized two bovine U6 small nuclear RNA (snRNA) promoters (bU6-2 and bU6-3) and a bovine 7SK snRNA promoter (b7SK). We compared the efficiency of each of these promoters to express shRNA molecules. Promoter activity was measured in the context of RNAi by targeting and suppressing the reporter gene encoding enhanced green fluorescent protein. Results show that the b7SK promoter induced the greatest level of suppression in a range of cell lines. The comparison of these bovine promoters in shRNA expression is an important component for the future development of bovine-specific RNAi-based research.

Identical single jersey wool knit fabrics produced inconsistent results when tested on the same type of ICI pill box in 4 independence testing laboratories.  The hypothesis formulated to investigate this event was that continued pilling testing in the ICI pill box changes the frictional properties of the cork liners, resulting in the difference pilling ratings.  The aim of this study was to decrease the inconsistencies in the pilling results by replacing the conventional cork lined pill box with a PerspexTM pill box.  The smae or similar pilling results found for several of the knit facots tested in the two difference pill boxes, while for others there were some significant differences found.  In addition, while using the PerspexTM pill box observations of the movement of the pilling tubes during testing provided evidence of greater fabric-to-fabric abrasion, rather the expected high degree of fabric-to-cork abrasion (or fabric-to-PerspexTM) during pilling testing.

Webcasting, sometimes labeled the "last frontier" or "the next stage of evolution" of the internet era, has been slow in its diffusion in Australia, due to technical constraints related to internet transmission speeds and bandwidth (Morrissey, 2003, cited in Lin, 2004, p. 446). According to Craig Preston, Operations Coordinator for New Media and Digital Services at the Australian Broadcasting Corporation (or ABC, a national broadcaster), webcasting in Australia is "pretty dead, really" (c. Preston, personal communication, August 23, 2005). This statement will be examined in this chapter, especially since the Australian Media Guide does not list "web casting" or "streaming" as a category or include the terms among its long list of entries (Gee, 2005)

The diet of a Southern Boobook Ninox novaeseelandiae in a Box-Ironbark woodland remnant in central Victoria was studied. An analysis of 42.5 pellets found invertebrates to represent 82% of the total prey items, but vertebrates, in the form of the House Mouse Mus domesticus, to represent 88% of the biomass. The proportion of spiders as prey items in the diet in this instance (43%) was significantly higher than in other studies on mainland Australia.
