In this study, we investigated the efficacy of an LNA (locked nucleic acid)-modified DNA aptamer named RNV66 targeting VEGF against various breast cancer cell lines. Our results demonstrate that RNV66 efficiently inhibits breast cancer cell proliferation both in vitro and in vivo. Introduction of LNA nucleotides were crucial for higher efficacy. Furthermore, the binding interaction of RNV66 with VEGF was investigated using molecular dynamic simulations leading to the first computational model of the LNA aptamer-VEGF complex blocking its interaction with VEGF-receptor.

PURPOSE: Alzheimer's disease (AD) is one of the untreatable neurodegenerative diseases characterised by the pathologic amyloid plaque deposition and inflammation. The aim of this study is to evaluate the neuroprotective effects of nanoformulated SurR9-C84A, a survivin mutant belonging to the inhibitors of the apoptosis (IAP) protein family. The effect of SurR9-C84A was studied against the β-amyloid toxicity and various inflammatory insults in the differentiated SK-N-SH neurons. METHOD: SurR9-C84A loaded poly(lactic-co-glycolic acid) nanoparticles were prepared following the modified double emulsion technique. The neuroprotective effect of SurR9-C84A was evaluated against the amyloid-β (Aβ) peptide fragment, N-methyl-D-aspartate (NMDA) toxicity and the inflammatory assaults. To mimic the in vivo situation, a co-culture of neurons and microglia was also studied to validate these results. RESULTS: SurR9-C84A treatments showed improved neuronal health following Aβ, and NMDA toxicity in addition to inflammatory insults induced in mono and co-cultures. The neuroprotective effect was evident with the reduced neuronal death, accelerated expression of neuronal integrity markers (neurofilaments, beta-tubulin III etc.,) and the neuroprotective ERK/MAPK signalling. CONCLUSION: The current results demonstrated that the SurR9-C84A nanoformulation was very effective in rescuing the neurons and holds a potential future application against AD.

A chimeric aptamer-DNAzyme conjugate was generated for the first time using a nucleolin aptamer (NCL-APT) and survivin Dz (Sur_Dz) and exhibited the targeted killing of cancer cells. This proof of concept of using an aptamer for the delivery of DNAzyme can be applied to other cancer types to target survivin in cancer cells in a specific manner.

We investigated the anti-cancer activity of alginate coated chitosan nanoparticles (CHNP) encapsulating cell-permeable dominant negative survivin (SR9) with locked nucleic acid (LNA) aptamers targeting EpCAM and nucleolin (termed as "nanobullets") in vitro (2D and 3D cell culture models) and in vivo (colon cancer mouse xenograft model). We incorporated three LNA modifications in each sequence in order to enhance the stability of these aptamers. Confocal microscopy revealed binding of the LNA-aptamers to their specific markers with high affinity. The muco-adhesive nanobullets showed 6-fold higher internalization in cancer cells when compared to non-cancerous cells, suggesting a tumour specific uptake. A higher intensity of nanobullets was observed in both the periphery and the core of the multicellular tumour spheroids compared to non-targeted CHNP-SR9. The nanobullets were found to be the highly effective as they led to a 2.26 fold (p < 0.05) reduction at 24 h and a 4.95 fold reduction (p ≤ 0.001) in the spheroid size at 72 h. The tumour regression was 4 fold higher in mice fed on a nanobullet diet when compared to a control diet. The nanobullets were able to show a significantly high apoptotic (p ≤ 0.0005) and necrotic index in the tumour cell population (p ≤ 0.005) when compared to void NPs. Liquid plasma, produced by nanosecond pulses, provides an efficient and simple way to fabricate a nanocomposite architecture of Co3O4/CNTs from carbon nanotubes (CNTs) and clusters of Co3O4 nanoparticles in deionized water. The crucial feature of the composite's structure is that Co3O4 nanoparticle clusters are uniformly dispersed and anchored to CNT networks in which Co3O4 guarantees high electrochemical reactivity towards sodium, and CNTs provide conductivity and stabilize the anode structure. We demonstrated that the Co3O4/CNT nanocomposite is capable of delivering a stable and high capacity of 403 mA h g(-1) at 50 mA g(-1) after 100 cycles where the sodium uptake/extract is confirmed in the way of reversible conversion reaction by adopting ex situ techniques. The rate capability of the composite is significantly improved and its reversible capacity is measured to be 212 mA h g(-1) at 1.6 A g(-1) and 190 mA h g(-1) at 3.2 A g(-1), respectively. Due to the simple synthesis technique with high electrochemical performance, Co3O4/CNT nanocomposites have great potential as anode materials for sodium-ion batteries.

Incorporating phenylpyridine- and triazolylpyridine-based ligands decorated with methylsulfonate or tetraethylene glycol (TEG) groups, a series of iridium(III) complexes has been created for green and blue electrogenerated chemiluminescence under analytically useful aqueous conditions, with tri-n-propylamine as a coreactant. The relative electrochemiluminescence (ECL) intensities of the complexes were dependent on the sensitivity of the photodetector over the wavelength range and the pulse time of the applied electrochemical potential. In terms of the integrated area of corrected ECL spectra, with a pulse time of 0.5 s, the intensities of the Ir(III) complexes were between 18 and 102 % that of [Ru(bpy)3 ](2+) (bpy=2,2'-bipyridine). However, when the intensities were measured with a typical bialkali photomultiplier tube, the signal of the most effective blue emitter, [Ir(df-ppy)2 (pt-TEG)](+) (df-ppy=2-(2,4-difluorophenyl)pyridine anion, pt-TEG=1-(2-(2-(2-(2-hydroxyethoxy)ethoxy)ethoxy)ethyl)-4-(2-pyridyl)-1,2,3-triazole), was over 1200 % that of the orange-red emitter [Ru(bpy)3 ](2+) . A combined experimental and theoretical investigation of the electrochemical and spectroscopic properties of the Ir(III) complexes indicated that the greater intensity from [Ir(df-ppy)2 (pt-TEG)](+) relative to those of the other Ir(III) complexes resulted from a combination of many factors, rather than being significantly favored in one area.

Fluorescence microscopy studies using 4-morpholinoscriptaid (4MS) demonstrated rapid cellular uptake of this scriptaid analogue into the cytoplasm but no nuclear penetration. As 4MS and scriptaid have the same in vitro activity against HDACs and KASUMI-1 cells; 4MS exemplifies a rational approach to subtly modify ‘profluorogenic’ substrates for intracellular studies.

The ability to spatiotemporally identify the formation of specific anionic species, or track changes in their concentration inside living systems, is of critical importance in deciphering their exact biological roles and effects. The development of probes (also called bioimaging agents and intracellular sensors) to achieve this goal has become a rapidly growing branch of supramolecular chemistry. In this critical review the challenges specific to the task are identified and for a select range of small anions of environmental and biological relevance (fluoride, chloride, iodide, cyanide, pyrophosphate, bicarbonate, hydrosulphide, peroxynitrite, hypochlorite and hypobromite) a comprehensive overview of the currently available in vitro and in vivo probes is provided.

A modular approach has been developed for the synthesis of rigid linear di- and tritopic ligands based on a fused [6]polynorbornane scaffold. The design provides up to three sites for installing functionality, including both "ends" and a "central" position with the advantage that each region can be independently addressed during synthesis. To illustrate the utility of the approach, both pyridyl and picolyl units were incorporated to provide six new ligands, with centers and ends either matched or mismatched. Indeed, both [M2 L4 ] cages with endohedral functionality and [M3 L4 ] complexes were cleanly produced from these ligands with assembled structures confirmed by using (1) H NMR spectroscopy, HRMS, and molecular modelling.

The use of modified Job plot methodology provides a quick and easy means for evaluating host-guest stoichiometry and requires less material than the original method. In this short paper, the results of a recent anion recognition study using thiourea-functionalised norbornanes and [n]polynorbornanes are presented. A significant difference between the plots constructed using the original and modified method was observed and, as such, caution is advised when the modified Job's method is solely used for the determination of host-guest stoichiometry.

A novel self-adapting retarder itaconic acid/acrylamide/sodium styrene sulfonate (IA/AM/SSS, hereinafter referred to as PIAS) was synthesized by free-radical, aqueous-solution polymerization and characterized by FTIR and TG. The optimum reaction conditions of polymerization were obtained from orthogonal experiments (L33) and subsequent data analysis. According to the evaluation as a retarder, the PIAS made it possible to obtain both a long thickening time and a swift compressive strength development for cement slurry, and therefore the applicable range of bottom hole circulation temperatures to the cement slurry has been widened to 60-180°C. Moreover, the working mechanism of the self-adapting retarder PIAS was found to rely on the change of spatial structure of the molecules to retard the hydration of the cement. This paper also expounds that the delayed coagulation of the cement slurry is attributed to adsorption, chelation and "poisoning" effects of the PIAS molecules on the surface of hydrated particles or ions through XRD and SEM analyses.

Nanofibres prepared by electrospinning have shown enormous potential for various applications. They are obtained predominantly in the form of nonwoven fibre webs. The 2-dimensional nonwoven feature and fragility have considerably confined their further processing into fabrics through knitting or weaving. Nanofibre yarns, which are nanofibre bundles with continuous length and a twist feature, show improved tensile strength, offering opportunities for making 3-dimensional fibrous materials with precisely controlled fibrous architecture, porous features and fabric dimensions. Despite a few techniques having been developed for electrospinning nanofibre yarns, they are chiefly based on the needle electrospinning technique, which often has low nanofibre productivity. In this study, we for the first time report a nanofibre yarn electrospinning technique which combines both needle and needleless electrospinning. A rotating intermediate ring collector was employed to directly collect freshly-electrospun nanofibres into a fibrous cone, which was further drawn and twisted into a nanofibre yarn. This novel system was able to produce high tenacity yarn (tensile strength 128.9 MPa and max strain 222.1%) at a production rate of 240 m h-1, with a twist level up to 4700 twists per metre. The effects of various parameters, e.g. position of the electrospinning units, operating conditions and polymer concentration, on nanofibre and yarn production were examined.

Hydrogel nanofibers with high water-absorption capacity and excellent biocompatibility offer wide use in biomedical areas. In this study, hydrogel nanofibers from polyvinylpyrrolidone (PVP) and PVP/poly(acrylic acid) (PAA) blend were prepared by electrospinning and by subsequent heat treatment. The effects of post-electrospinning heat treatment and PVP/PAA ratio on hydrogel properties of the nanofibers were examined. Heat treatment at a temperature above 180°C was found to play a key role in forming insoluble and water-absorbent nanofibers. Both PVP and PVP/PAA nanofibers showed high morphology stability in water and excellent water retention capacity. The swelling ratio of PVP/PAA nanofibers declined with increasing heating temperature and decreasing PVP/PAA unit ratio. In comparison with dense casting films, these nanofiber membranes showed nearly doubled swelling ratio.

Superhydrophobic cotton fabrics with a very low contact angle hysteresis were prepared using a single-pot coating solution comprising SU-8 (a negative photoresist), a fluorinated alkyl silane and silica nanoparticles. The fabric was treated using a dip-coating technique and subsequently cured under UV light. The coated fabric showed excellent superhydrophobicity with a water contact angle as high as 163° and a sliding angle as low as 2°. The coating was durable enough to withstand 100 laundry cycles. It also had excellent stability against long immersion times in organic solvents, and acid and base solutions.

Herein, a new graphene/Cu nanoparticle composite was prepared via the in situ reduction of GO in the presence of Cu nanoparticles which was then utilized as a sacrificing template for the formation of flexible and porous graphene capacitor electrodes by the dissolution of the intercalated Cu nanoparticle in a mixed solution of FeCl3 and HCl. The porous RGO electrode was characterized by atomic force microscopy (AFM), Fourier transform infrared (FTIR) spectroscopy, X-ray diffraction (XRD), Raman spectroscopy, X-ray photoelectron spectroscopy (XPS) and thermogravimetric analysis (TGA). The as-prepared graphene/Cu nanoparticle composite and the pure graphene film after removal of Cu nanoparticles possessed high conductivity of 3.1 × 103 S m-1 and 436 S m-1 respectively. The porous RGO can be used as the electrode for the fabrication of supercapacitors with high gravimetric specific capacitances up to 146 F g-1, good rate capability and satisfactory electrochemical stability. This environmentally friendly and efficient approach to fabricating porous graphene nanostructures could have enormous potential applications in the field of energy storage and nanotechnology.
