The 23S ribosomal RNA (rRNA) gene has been sequenced in strains of the fish pathogens Photobacterium damselae subsp. damselae (ATCC 33539) and subsp. piscicida (ATCC 29690), showing that 3 nucleotide positions are clearly different between subspecies. In addition, the 5S rRNA gene plus the intergenic spacer region between the 23S and 5S rRNA genes (ITS-2) were amplified, cloned and sequenced for the 2 reference strains as well as the field isolates RG91 (subsp. damselae) and DI21 (subsp. piscicida). A 100% similarity was found for the consensus 5S rRNA gene sequence in the 2 subspecies, although some microheterogeneity was detected as inter-cistronic variability within the same chromosome. Sequence analysis of the spacer region between the 23S and 5S rRNA genes revealed 2 conserved and 3 variable nucleotide sequence blocks, and 4 different modular organizations were found. The ITS-2 spacer region exhibited both inter-subspecies and inter-cistronic polymorphism, with a mosaic-like structure. Whereas the predominance of El Niño Southern Oscillation (ENSO) mode in the tropical Pacific sea surface temperature (SST) variability is well established, no such consensus seems to have been reached by climate scientists regarding the Indian Ocean. While a number of researchers think that the Indian Ocean SST variability is dominated by an active dipolar-type mode of variability, similar to ENSO, others suggest that the variability is mostly passive and behaves like an autocorrelated noise. For example, it is suggested recently that the Indian Ocean SST variability is consistent with the null hypothesis of a homogeneous diffusion process. However, the existence of the basin-wide warming trend represents a deviation from a homogeneous diffusion process, which needs to be considered. An efficient way of detrending, based on differencing, is introduced and applied to the Hadley Centre ice and SST. The filtered SST anomalies over the basin (23.5N-29.5S, 30.5E-119.5E) are then analysed and found to be inconsistent with the null hypothesis on intraseasonal and interannual timescales. The same differencing method is then applied to the smaller tropical Indian Ocean domain. This smaller domain is also inconsistent with the null hypothesis on intraseasonal and interannual timescales. In particular, it is found that the leading mode of variability yields the Indian Ocean dipole, and departs significantly from the null hypothesis only in the autumn season.

A high-resolution crystal structure is reported for d(TpA)*, the intramolecular thymine-adenine photoadduct that is produced by direct ultraviolet excitation of the dinucleoside monophosphate d(TpA). It confirms the presence of a central 1,3-diazacyclooctatriene ring linking the remnants of the T and A bases, as previously deduced from heteronuclear NMR measurements by Zhao et al. (The structure of d(TpA)*, the major photoproduct of thymidylyl-(3'-5')-deoxyadenosine. Nucleic Acids Res., 1996, 24, 1554-1560). Within the crystal, the d(TpA)* molecules exist as zwitterions with a protonated amidine fragment of the eight-membered ring neutralizing the charge of the internucleotide phosphate monoanion. The absolute configuration at the original thymine C5 and C6 atoms is determined as 5S,6R. This is consistent with d(TpA)* arising by valence isomerization of a precursor cyclobutane photoproduct with cis-syn stereochemistry that is generated by [2 + 2] photoaddition of the thymine 5,6-double bond across the C6 and C5 positions of adenine. This mode of photoaddition should be favoured by the stacked conformation of adjacent T and A bases in B-form DNA. It is probable that the primary photoreaction is mechanistically analogous to pyrimidine dimerization despite having a much lower quantum yield.

The chiral stabilised azomethine ylide formed from condensation of the dimethyl acetal of acetone with (5S)-5-phenylmorpholinone undergoes stereoselective exo-cycloaddition reactions with a range of doubly and singly activated dipolarophiles when generated in the presence of excess (MgBr2OEt2)-O-.. The cycloadducts can be degraded to yield enantiomerically pure proline derivatives.

Research interest in oats has focussed on their nutritional value, but there have been few studies of their food processing. Heat treatment is characteristic of oat processing, as it is needed to inactivate lipase and to facilitate flaking. A Texture Analyser was used to characterise the mechanical properties of unkilned and kilned oat groats after steaming and tempering in an oven for 30, 60 and 90 min at 80, 95 and 110 degrees C. Maximum force, number of peaks before maximum and final force after 5s hold were used to characterise the behaviour of the groats during compression. Kilned groats were larger and softer before steaming. After steaming and tempering, the moisture content of the kilned groats was higher than for unkilned groats. Hot, steamed oats were softer than cold, unsteamed groats, indicated by a decrease in maximum force from 59 to 55 N, and there was no significant difference between kilned and unkilned groats. However, higher temperatures during tempering increased maximum force. These results suggest that mild steam treatment yields softer oat groats, whereas cold or over-treated groats tend to be harder. Irradiation of 5S-5-O-tert-butyldimethylsiloxymethylfuran-2(5H)-one 1 in acetonitrile yields the C2-symmetric bis(lactone), 1S,2S,6S,7S-[3S,10S]-bis-tert-butyldimethylsiloxymethyl-4,9-dioxatricyclo[5.3.0.02,6]deca-5,8-dione 6, and a 3-substituted intramolecular product resulting from an apparent 8-endo-trig cyclisation.
