Discovery and development of new pharmaceuticals from marine organisms are attracting increasing interest. Several agents derived from marine organisms are under preclinical and clinical evaluation as potential anticancer drugs. We extracted and purified a novel anti-tumor protein from the coelomic fluid of Meretrix meretrix Linnaeus by ammonium sulphate fractionation, ion exchange and hydrophobic interaction chromatography. The molecular weight of the highly purified protein, designated MML, was 40 kDa as determined by SDS-PAGE analysis. MML exhibited significant cytotoxicity to several cancer cell types, including human hepatoma BEL-7402, human breast cancer MCF-7 and human colon cancer HCT116 cells. However, no inhibitory effect was found when treating murine normal fibroblasts NIH3T3 and benign human breast MCF-10A cells with MML. The cell death induced by MML was characterized by cell morphological changes. The induction of apoptosis of BEL-7402 cells by MML was weak by DNA ladder assay. The possible mechanisms of its anti-tumor effect might be the changes in cell membrane permeability and inhibition of tubulin polymerization. MML may be developed as a novel, highly selective and effective anti-cancer drug.

在0.05 mol/L Tris,0.15 mol/L NaCl溶液中,用吸附伏安法研究长春新碱(VCR),其峰电位在-1.68 V(vs. Ag/AgCl),峰电流与1.0*10^{-8}~2.0*10^{-7}mol/L VCR浓度成正比,检测限为7.0*l0^{-9} mol/L,用常规脉冲极谱法、线性扫描和循环伏安法等研究该体系的电化学行为,实验表明,电极还原过程为具有吸附特征的不可逆过程。VCR的吸附符合Frumkin吸附等温式。也研究了VCR与微管蛋白的相互作用。实验表明,VCR与微管蛋白形成一电活性的结合物,这一结合物具有吸附性,且还原过程也为不可逆过程。

Microtubules (MT) are composed of 13 protofilaments, each of which is a series of two-state tubulin dimers. In the MT wall, these dimers can be pictured as "lattice" sites similar to crystal lattices. Based on the pseudo-spin model, two different location states of the mobile electron in each dimer are proposed. Accordingly, the MT wall is described as an anisotropic two-dimensional (2D) pseudo-spin system considering a periodic triangular "lattice". Because three different "spin-spin" interactions in each cell exist periodically in the whole MT wall, the system may be shown to be an array of three types of two-pseudo-spin-state dimers. For the above-mentioned condition, the processing of quantum information is presented by using the scheme developed by Lloyd.

花粉管是种子植物受精过程中的雄性生殖单位的载体,由于其生长依赖于胞内Ca2+梯度,并具有典型的顶端极性生长的特点,因而成为近年来研究植物细胞相互识别、胞内和胞外信号传导理想的模式系统。裸子植物花粉与被子植物相比具有萌发时间长、生长缓慢等特点。Ca2+在裸子植物花粉萌发和花粉管生长中的作用机制目前尚不明确。本研究以裸子植物白皮松(Pinus bungeana)的花粉为材料,运用不同浓度钙通道抑制剂Nifedipine(Nif)处理,对其花粉萌发和花粉管生长进行了细胞学研究和蛋白质组学分析,以探讨Ca2+对白皮松花粉生长的调控机制,为进一步揭示裸子植物花粉萌发和花粉管生长机理提供参考。 本论文首先研究了钙通道抑制剂Nif和Ca2+螯合剂EGTA对白皮松花粉萌发和花粉管生长的影响。结果表明,用Nif处理花粉后,花粉萌发和花粉管生长均受到明显抑制。经Ca2+荧光探针Fluo-3AM标记,对Nif处理后Ca2+在花粉管中的分布模式进行了观察,发现Ca2+在正常生长的花粉管中呈梯度分布,并在其顶端的荧光最强。与对照相比,处理后的花粉管荧光强度明显减弱,且顶端Ca2+梯度消失。通过EGTA漂洗后的花粉粒,在正常培养基上萌发率较高,但其生长速率受到抑制。由此说明了细胞壁钙库对花粉管生长的抑制效应显著高于对花粉萌发的影响,是花粉管生长的限速因子。同时外加钙调素还可逆转EGTA对其花粉萌发和花粉管生长的抑制。上述结果表明,白皮松花粉萌发及花粉管生长需要外源Ca2+的内流,以及胞内形成的Ca2+浓度梯度。 用FM4-64探针标记结果发现,正常花粉管中的胞吞作用主要发生在顶端和亚顶端区域,胞吞的小泡也集中分布在这两个区域。经Nif处理后,既不影响花粉管的胞吞过程,同时胞吞发生的位置也与对照相似,只是胞吞的小泡分散于整个花粉管中。电子显微镜观察表明,各种细胞器在白皮松正常生长的花粉管中分布与被子植物存在较大差异,例如前者无明显地分区现象,不具胼胝质塞。白皮松花粉管顶端和亚顶端的壁旁体与质膜融合现象频繁发生。花粉管经Nif处理后,线粒体出现不同程度的解体和液泡化,内质网液泡化和核糖体脱落,液泡大量聚集在花粉管顶端,壁旁体与膜融合现象减少,以及花粉管壁明显变薄等。此外,通过微丝特异性探针鬼笔环肽标记结果表明,正常生长花粉管的微丝呈长轴向排列, Nif处理后微丝断裂,其断裂程度与处理浓度有关。由此可见,外源Ca2+对花粉管的胞吞无明显抑制或促进作用,但对胞吞小泡重回收可能有影响。当胞内Ca2+梯度消失后,则明显抑制了微丝骨架的聚合,进而使胞吐作用减缓,高尔基体分泌小泡聚集,多种细胞器液泡化,引起花粉管顶端膨大,细胞壁变薄,继而抑制花粉管的正常生长。 运用免疫荧光标记技术显示,正常生长的花粉管壁含有纤维素、胼胝质、果胶质和阿拉伯半乳聚糖蛋白(AGPs),其中纤维素和胼胝质在细胞壁上呈均匀分布,而酸性果胶质只存在花粉管两侧壁上,酯化果胶分布于花粉管顶端。经过Nif处理后,胼胝质和酸性果胶质均在花粉管顶端累积,而AGPs和纤维素的分布却无明显变化。另外,傅里叶红外光谱分析结果也同样支持上述结论。通过花粉管壁蛋白的SDS-PAGE分析表明,Nif对细胞壁蛋白的合成也有较大的影响。 在花粉管的钙通道受到抑制后,应用蛋白质组学技术分析其蛋白质的表达图谱,通过双向电泳已分离出约1000个蛋白质斑点,经软件分析发现,除其中50个蛋白斑点外,大部分蛋白质的表达量均未发生变化。上述50个发生变化的蛋白斑点酶切后,再经过ESI-MS/MS鉴定和质谱数据库的搜索,共鉴定出28个蛋白,其中12个为上调蛋白,16个下调蛋白,根据其主要功能分为与代谢、细胞扩展、翻译后修饰以及信号相关的蛋白。经过Nif处理后,花粉管中碳水化合物代谢能力下降,ATP的产生受到抑制,参与细胞壁多糖合成及小泡运输的蛋白,如valosin containing protein(VCP)、reversibly glycolsylated polypeptide(RGP)、UDP-glucose dehydrogenase (UDPGDH)和α-tubulin表达下调。另外,通过上述方法还鉴定出与丝氨酸/苏氨酸激酶保守结构域同源的受体蛋白激酶等。 综上所述,白皮松花粉管钙通道受到抑制后,通过影响花粉管蛋白的表达,抑制微丝微管骨架的组装,致使胞吐速度变慢,花粉管壁酸性果胶质、胼胝质等多糖分布的变化及总多糖含量的减少,最终抑制了花粉管的正常生长。

高等植物基因表达过程中的信号传导是目前植物分子生物学研究的前沿和热点之一。不少研究者已将脱落酸、乙烯、细胞分裂素及其它植物激素的作用一起归之于植物基因表达的信号传导系统。细胞分裂素作为—类重要的植物激素在植物的生长和发育过程中起重要的调节控制作用。因此研究细胞分裂素的基因与植物发育过程的关系是十分重要的。 为研究细胞分裂素对植物基因表达的调节,本文从转录和翻译水平上测定了黄瓜子叶在外源细胞分裂素诱导下微管蛋白基因表达的活性。发现经BAP处理的黄瓜子叶中α,β-tubulin mRNA迅速积累,微管蛋白的含量迅速增加。这表明外源细胞分裂素在诱导黄瓜子叶膨大的过程中激活了微管蛋白基因的表达。 为探索不同启动子驱动下的细胞分裂素基因转入植物后的表达对转基因植物生长发育的调控,本文将来自根癌农杆菌的细胞分裂素基因(T-cyt)分别置于CaMV 35S启动子,rbc S启动子和T-cyt基因自身启动子的调控下,构建了嵌合表达质粒,分别转化烟草和马铃薯。转基因烟草和马铃薯的PCR检测和Southern杂交鉴定均证实T-cyt基因已分别整合进烟草和马铃薯的核基因组中。标志基因NPTⅡ的酶活性测定表明有外源基因的表达。转基因烟草的Northern分析表明:CaMV 35s启动子驱动的T-cyt基因的mRNA在叶、茎和根中均有表达;rbc S启动子指导的T-cyt基因在叶中表达最强,茎中较弱,在根中几乎没有表达。转细胞分裂素基因的烟草在生长发育上与未转化的对照相比有明显不同。转基因烟草中叶绿素a,b含量明显增加,叶面积减小,叶衰老迟缓。T-cyt基因转化的烟草顶端优势受到抑制,侧芽生长旺盛;与对照相比,其节间短,株高降低,根生长受抑制。 本文还构建了T-cyt基因自身启动子与报告基因GUS编码区的嵌合表达质粒,转化烟草和马铃薯以研究T-cyt启动子在植物中的表达。组织化学定位测定表明,T-cyt启动子在植物的茎,叶中的表达较强,特别是在腋芽的生长点有很高的表达活性,但在根中的表达较弱。诱导性表达试验表明,T-cyt启动子的表达强度受细胞分裂素的诱导,而生长素对T-cyt启动子的表达无明显影响。这提示T-cyt启动子是一个细胞分裂素诱导性表达的启动子。 总之,将T-cyt基因转入植物,作为调节内源细胞分裂素的一种手段,可以对植物的生长发育进行调控。尤其是利用发育阶段特异性和各种器官特异性表达的启动子可以调节T-cyt基因的表达活性,有可能创造出具有经济价值的、具有新遗传特性的植物。

The origin of cytoskeleton and the origin of relevant intracellular transportation system are big problems for understanding the emergence of eukaryotic cells. The present article summarized relevant information of evidences and molecular traces on the origin of actin, tubulin, the chaperonin system for folding them, myosins, kinesins, axonemal dyneins and cytoplasmic dyneins. On this basis the authors proposed a series of works, which should be done in the future, and indicated the ways for reaching the targets. These targets are mainly: 1) the reconstruction of evolutionary path from MreB protein of archaeal ancestor of eukaryotic cells to typical actin; 2) the finding of the MreB or MreB-related proteins in crenarchaea and using them to examine J. A. Lake's hypothesis on the origin of eukaryote from "eocytes" (crenarchaea); 3) the examinations of the existence and distribution of cytoskeleton made of MreB-related protein within coccoid archaea, especially in amoeboid archaeon Thermoplasm acidophilum; 4) using Thermoplasma as a model of archaeal ancestor of eukaryotic cells; 5) the searching for the homolog of ancestral dynein in present-day living archaea. During the writing of this article, Margulis' famous spirochaete hypothesis on the origin of flagella and cilia was unexpectedly involved and analyzed from aspects of tubulins, dyneins and spirochaetes. Actually, spirochaete cannot be reasonably assumed as the ectosymbiotic ancestor of eukaryotic flagella and cilia, since their swing depends upon large amount of bacterial flagella beneath the flexible outer wall, but not depends upon their intracellular tubules and the assumed dyneins. In this case, if they had "evolved" into cilia and lost their bacterial flagella, they would immediately become immobile! In fact, tubulin and dynein-like proteins have not been found in any spirochaete.

The origin of eukaryotic flagella has long been a mystery. Here we review the possibility that flagella sprouted evolutionarily from the eukaryotic cell proper seems very unlikely because it is hard to imagine what function and benefit in natural selection the flagella would have provided to the cells when they first emerged as simple buds. Lynn Margulis' 1970 spirochete hypothesis, though popular still, has never been confirmed. Moreover, the absence of tubulin and axonemal dynein in the spirochetes and the incapability of the bacterial and eukaryotic membranes' making a continuum now suggest that the hypothesis is outdated. Tubulin genes were recently identified in a new bacteria division, verrucomicrobia, and microtubules have also been found in one of these species, epixenosomes, the defensive ectosymbionts. On the basis of these data, we propose a new symbiotic hypothesis: that the mid-ancestor of eukaryotic cells obtained epixenosomelike verrucomicrobia as defensive ectosymbionts and the ectosymbionts later became endosymbiotic. They still, however, protruded from the surface of their host to play their role. Later, many genes were lost or incorporated into the host genome. Finally, the genome, the bacterial membrane, and the endosymbiotic vesicle membrane were totally lost, and fingerlike protrusions with microtubules formed. As the cells grew larger, the defensive function of the protrusions eventually weakened and then vanished. Some of the protrusions took on a new role in cell movement, which led them to evolve into flagella. The key step in this process was that the dynein obtained from the host evolved into axonemal dyneins, attaching onto the microtubules and forming motile axonemes. Our hypothesis is unproven, but it offers a possible explanation that is consistent with current scientific thought. We hope that our ideas will stimulate additional studies on the origin of eukaryotic flagella and on investigations of verrucomicrobia. Whether such studies confirm, refine, or replace our hypothesis, they should nevertheless further our understanding of the origin of eukaryotic cells.

以斜带石斑鱼囊胚期胚胎和尾芽期胚胎分别作为检验组和驱动组,构建了石斑鱼囊胚期胚胎和尾芽期胚胎的抑制性差减杂交cDNA文库。以α-tubulin作为检测指标,显示差减效率分别高达28和27。分别取囊胚期胚胎和尾芽期胚胎各192和960个PCR阳性克隆进行斑点杂交,得到15个囊胚期和131个尾芽期的斑点杂交阳性克隆。测序和数据库比对分析表明,囊胚期15个阳性克隆中有11个已知基因的cDNA片段和没有同源性的4个cDNA片段;而在尾芽期的131个阳性克隆中,有123个已知基因的cDNA片段和8个没有同源性的cD

紫外线灭活的草鱼出血病病毒 (GCHV)能诱导鲫囊胚培养细胞 (CAB)产生高滴度的干扰素 ,从而诱导宿主细胞基因表达的改变并处于抗病毒状态。提取灭活病毒诱导未经病毒诱导的CAB细胞mRNA ,利用抑制性差减杂交技术 ,成功构建了鱼类培养细胞抗病毒基因差减cDNA文库。以鲫管家基因α tubulin和 β actin作为差减指标 ,检测差减cDNA文库的差减效率分别高达 2 15和 2 7倍 ,表明经过病毒诱导后的细胞中 ,某些差异表达基因的富集效率也接近 2 15倍。鱼类抗病毒基因差减cDNA文库的建立

C-Phycocyanin (C-PC) from blue-green algae has been reported to have various pharmacological characteristics, including antiinflammatory and anti-tumor activities. In this study, we expressed the beta-subunit of C-PC (ref to as C-POP) in Escherichia coli. We found that the recombinant C-PC/beta has anti-cancer properties. Under the treatment of 5 mu M of the recombinant C-PC/beta, four different cancer cell lines accrued high proliferation inhibition and apoptotic induction. Substantially, a lower response occurred in non-cancer cells. We investigated the mechanism by which C-PC/beta inhibits cancer cell proliferation and induces apoptosis. We found that the C-PC/beta interacts with membrane-associated beta-tubulin and glyceraldehyde-3-phosphate dehydrogenase (GAPDH). Under the treatment of the C-PC/beta, depolymerization of microtubules and actin-filaments were observed. The cells underwent apoptosis with an increase in caspase-3, and caspase-8 activities. The cell cycle was arrested at the G0/G1 phase under the treatment of C-PC/beta. In addition, the nuclear level of GAPDH decreased significantly. Decrease in the nuclear level of GAPDH prevents the cell cycle from entering into the S phase. Inhibition of cancer cell proliferation and induction of apoptosis may potentate the C-POP as a promising cancer prevention or therapy agent. (c) 2006 Elsevier Ireland Ltd. All rights reserved.

Spindlin has been suggested to play an important role during the transition from oocyte maturation to embryo development in mouse, but its homolog similar to the mouse Spindlin in molecular and expression characterization has not been identified up to now in other vertebrates. In this study, a full length of cDNA sequence is cloned and sequenced from the gibel carp (Carassius auratus gibelio). It contains 1240 nucleotides with an open reading frame of 771 nt encoding 257 amino acids. Based on its amino acid sequence alignment and comparison analysis with the known Spin family proteins, the newly cloned Spin is named Carassius auratus gibelio Spindlin (CagSpin). Its product could be detected from mature eggs to blastula embryos, but its content decreased from the two-cell stage, and could not be detected after the gastrula stage. It suggests that the CagSpin should be a maternal protein that is expressed during oocyte maturation, and plays a crucial role in early cleavage of embryogenesis. CagSpin is the first homolog similar to mouse spindlin identified in fish, and also in other vertebrates. GST pull-down assay reveals the first biochemical evidence for the association of CagSpin and p-tubulin, the microtubule component. Therefore, CagSpin may play important functions by interacting with beta-tubulin and other spindle proteins during oocyte maturation and egg fertilization. (c) 2005 Elsevier Inc. The aim of this study was to determine the lowest concentration of nocodazole and colchicine to arrest blastomere division during the cleavage stage of loach embryos and to assess the reversibility and toxicity of the treatments in the treated embryos. Eight-cell loach embryos were incubated for 4, 8, 12, or 16 h in 1/10x Holtfreter supplemented with either nocodazole, an inhibitor of tubulin polymerization, or colchicine, an inhibitor of tubulin assembly. Complete arrest of cell cycle was observed, at a colchicine concentration of 0.996 mM and at a nocodazole concentration of 0.275 muM, respectively (the lowest effective concentration). No major morphological alteration in chromatin was observed. Reversibility and toxicity of both agents were dose and exposure period dependent. For both agents, prolonging cleavage arrest for more than 4 h (at the effective concentrations) is detrimental to development of embryos. Nocodazole treatment was less cytotoxic, whereas the concentrations of colchicine which induce cleavage arrest were detrimental to development beyond the blastula stage. Toxic effects beyond the blastula stage could be minimized for both agents by reducing the period of treatment and concentration.

BRUNOL 蛋白又称CELF(CUG-BP and ETR3 like factor)是一种典型的RNA 结合蛋白,它的N 端含有两个连续的RNA 识别结构域(RNA recognition motif,RRM), C 端有一个RRM 结构域。主要参与对可变剪切、翻译、降解和编辑等基因表达转录后水平的调节。迄今在人类中已发现6 个Brunol 基因家族成员,即Brunol1-6;在非洲爪蟾中已发现了5 个:Brunol1-5。近期,我们克隆了爪蟾的Brunol1-5 并研究了它们在非洲爪蟾早期胚胎发育过程中的时空表达图式。结果显示,与以往研究结果一致, Brunol1 基因高量、特异地在神经管中表达,提示Brunol1 基因可能对于爪蟾的神经系统的发生和发育发挥着重要的作用。本实验利用Morpholino 和过表达等手段研究了爪蟾Brunol1 基因对于爪蟾早期胚胎发育的影响。结果显示,在下调和过表达Brunol 1 基因的情况下都会导致胚胎出现体轴弯曲,眼睛和头部发育不全等表型。而将 Brunol1 基因特异的Morpholino 与它的mRNA 共注射时可以明显挽救这一表型。我们通过原位杂交实验,检测了一些爪蟾神经系统的标记基因在Brunol1 过表达胚胎中的表达情况,结果发现过表达Brunol1 基因能显著地下调Krox-20, N-tubulin, Lhx2, Pax6 等的表达,而Sox2 和Otx2 的表达却未受影响。这说明Brunol1 的异常表达确实影响到了神经系统发育过程的信号调控网络,导致胚胎发育的畸形。该结果将有助于阐述Brunol1 基因对于脊椎动物神经系统发生的意义。肌动蛋白是一种分布广泛而且在进化上十分保守的蛋白,它是构成细胞骨架的关键组分。通常人们将肌动蛋白分成肌肉型和胞质型两种类型,它们各自行使着不同的功能。在此,我们通过对古老的脊索动物文昌鱼的肌动蛋白基因家族进行系统的分析发现,文昌鱼中该基因家族成员多达30 多个,而且它们中很多都有连锁现象;进化分析的结果显示,文昌鱼的肌动蛋白基因家族通过串联重复序列的复制发生扩增;从结构上看,它们的基因结构多样化, 包含2-7 个外显子;同时,我们还克隆了两个不同的文昌鱼肌肉型的肌动蛋白基因,并进一步比较了它们在文昌鱼早期胚胎中的表达图式。结果显示,这两个基因在表达上有着细微的差别,这提示文昌鱼肌动蛋白基因家族成员在功能上的分化。该结论将有助于阐述肌动蛋白基因家族的进化以及它们在脊索动物发育的中所扮演的功能。
