The efficient synthesis of 5-(5-bromovaleramido)-1,10-phenanthroline, 5-(6-bromohexanamido)-1,10-phenanthroline, and 5-(11-bromoundecanamido)-1,10-phenanthroline are described, which reacted with cis-Ru(bpy)(2)Cl-2. 2H(2)O and sodium hexafluorophosphate to form Ru(bpy)(2)[phen-NHCO(CH2)(n)Br](PF6)(2) (n = 4, 5 or 10; phen = 1,10-phenanthroline). The intricate H-1 NMR spectra at low field of these complexes were completely assigned in virtue of H-1-H-1 COSY technique. Cyclic voltammetry was used to study electrochemical behaviours of these complexes, and their luminescent properties were investigated with fluorescent spectra.

The electrochemiluminescence (ECL) of the Ru(bgy)(3)(2-)/S2O82- system in purely aqueous solution at a carbon paste electrode can be clearly seen with the naked eye for Ru(bpy)(3)(2+) concentrations higher than 1 mmol L-1. The log-log plot of the emmitted light intensity vs. Ru(bpy)(3)(2+) concentration is linear over the region 10(-3)-10(-7) mol L-1 with a correlation coefficient of 0.997. The ECL intensity increases linearly with the S2O82- concentration from 10(-6) mol L-1 up to 0.3 mmol L-1 and drops off sharply at concentrations higher than 1 mmol L-1. In addition, a weak ECL signal was obtained when the potential was biased more negative than -0.6 V even in the absence of S2O82-.

A novel method for the sensitive and selective determination of peroxydisulfate was proposed. It was based on an electrochemiluminescent method with Cr(bpy)(3)(3+). The applied potential was set at -0.5 V in Cr(bpy)(3)(3+) system which was much more positive than those in Ru(bpz)(3)(2+) and Ru(bpy)(3)(2+) systems. A linear calibration was obtained over the range of 7-100 mu mol l(-1) with a correlation coefficient of 0.993. The relative standard deviation was 2% for 10 replicate injections of 0.01 mol l(-1) S2O82- solution. The detection limit (S/N=3) was 1 mu mol l(-1). The method was successfully applied to determination of peroxydisulfate in phase transfer catalysis. (C) 2000 Elsevier Science B.V. All rights reserved.

An electrochemically stable monolayer of tris(2,2'-bipyridyl)ruthenium(II) was obtained for the first time. It was based on the electrostatic attachment of Ru(bpy)(3)(2+) to the benzene sulfonic acid monolayer film, which was covalently bound onto glassy carbon electrode by the electrochemical reduction of diazobenzene sulfonic acid. The surface-confined Ru(bpy)(3)(2+) underwent reversible surface process, and reacted with the coreactant, tripropylamine, to produce electrochemiluminescence. In view of the stability of the electrode, the results strongly suggested that light was emitted from the surface-confined Ru(bpy)(3)(2+), not from the detached Ru(bpy)(3)(2+). The Ru(bpy)(3)(2+) modified electrode was used to the determination of tripropylamine. It showed good linearity in the concentration range from 5 muM to 1 muM with a detection limit of 1 muM (S/N = 4). The good stability of the Ru(bpy)(3)(2+) modified electrode also showed that the benzene sulfonic acid monolayer film prepared can be served as an excellent support to construct multilayers. The electrochemiluminescence (ECL) of tris(2,2'-bipyridine)ruthenium(ii) [Ru(bpy)(3)(2+)] immobilized in poly(p-styrenesulfonate) (PSS)-silica-Triton X-100 composite films was investigated. The cooperative action of PSS, sol-gel and Triton X-100 attached Ru(bpy)(3)(2+) to the electrode strongly, and the presence of Triton X-100 prevented drying fractures of the sol-gel films during gelation and even on repeated wet-dry cycles. The modified electrode was used for the ECL detection of oxalate, tripropylamine (TPA) and NADH in a flow injection analysis (FIA) system with a newly designed flow cell. The detection scheme exhibited good stability, short response time and high sensitivity. Detection limits were 0.1, 0.1 and 0.5 mu mol L-1 for oxalate, TPA and NADH, respectively, and the linear concentration range extended from 0.001 to 1 mmol L-1 for the three analytes. Applications of the flow cell in ECL and electrochemical detection, as well as the immobilization of reagents based on the cooperative action, are suggested.

The recent progress in electrochemiluminescent (ECL) assay is reviewed. This review begins with the fundamental researches in ECL, including the discovery of new ECL-active species, such as biochemical, organic and metallorganic materials, digital modeling of ECL process, the flow cells used in ECL assay, and electrochemiluminescent sensor. The application of ECL in environmental analysis, immunoassay, nucleotide acid hybridization sensor. The applications of ECL in environmental analysis, immunooassay, nucleic acid hybridization assay, and other aspects are reviewed with the latest references in detail. Finally, the main problems in the further investigation are outlooked, so are its prospects.

A novel method for the highly sensitive determination of perchlorate was proposed. It was based on solvent extraction in the presence of Ru(bpy)(3)(2+) followed by Ru(bpy)(3)(2+) electrochemiluminescent determination. A linear calibration was obtained over the range of 0.1 to 10 mu mol l(-1) with a correlation coefficient of 0.998. The detection limit (S/N = 3) was 5.0 x 10(-8) mol l(-1). The relative standard deviation for 10 replicates of 1 mu mol l(-1) perchlorate was 1.6%. Interference studies suggest that this method is selective for the determination of perchlorate. Application of this method to the highly sensitive determination of other anions is suggested. (C) 1999 Elsevier Science S.A. All rights reserved.

The effects of heteropoly acids and Triton X-100 on electrochemiluminescence (ECL) of Ru(bpy)(3)(2+) are investigated. Triton X-100 prevents the oxidation of oxalate and results in an increase of the ECL signal. H5SiW11VO40 prevents the direct oxidation of oxalate and makes the electrochemical behavior of Ru(bpy)(3)(2+) less reversible, which leads to a decrease of the ECL signal. In contrast, H3PMo12O40 has negligible effect on ECL intensity. Some possible reasons for the effects on the ECL of Ru(bpy)(3)(2+) are discussed based on the adsorption of SiW11VO405- on electrode surface and the ion association between SiW11VO405- and Ru(bpy)(3)(2+). The signal of ECL decreases linearly with the concentration of heteropoly acid in the range from 2x10-6 to 1x10(-4) mol l(-1). The results indicate that ECL of RU(bpy)(3)(2+) is a potential sensitive and selective detection method for heteropoly acids and hence for the elements comprised in them.

Ethylenediaminetetraacetate (EDTA) was chosen for the investigation of the effect of metal ions on Ru(bpy)(3)(2+) electrochemiluminescence (ECL). Metal ions that are preferentially bound to the oxygen atoms (MIO) have no effect on the intensity of ECL except for Al3+ and Y3+, whereas metal ions that are preferentially bound to the nitrogen atoms (MIN) preclude the oxidation of EDTA and decrease the ECL intensity.

Oligodeoxynucleotides (ODNs) containing unmethylated CpG motifs in certain contexts are known to be immunostimulatory in vertebrate systems. CpG ODNs with immune effects have been identified for many fish species but, to our knowledge, not for turbot. In this study, a turbot-effective CpG ODN, ODN 205, was identified and a plasmid, pCN5, was constructed which contains the CpG motif of ODN 205. When administered into turbot via intraperitoneal (i.p.) injection, both ODN 205 and pCN5 could (i) inhibit bacterial dissemination in blood in dose and time dependent manners, and (ii) protect against lethal bacterial challenge. Immunological analyses showed that in vitro treatment with ODN 205 stimulated peripheral blood leukocyte proliferation, while i.p. injection with ODN 205 enhanced the respiratory burst activity, chemiluminescence response, and acid phosphatase activity of turbot head kidney macrophages. pCN5 treatment-induced immune responses similar to those induced by ODN 205 treatment except that pCN5 could also enhance serum bactericidal activity in a calcium-independent manner. To examine whether ODN 205 and pCN5 had any effect on specific immunity, ODN 205 and pCN5 were co-administered into turbot with a Vibrio harveyi subunit vaccine, DegQ. The results showed that pCN5, but not ODN 205, significantly increased the immunoprotective efficacy of DegQ and enhanced the production of specific serum antibodies in the vaccinated fish. Further analysis indicated that vaccination with DegQ in the presence of pCN5 upregulated the expression of the genes encoding MHC class II alpha, IgM, Mx, and IL-8 receptor. Taken together, these results demonstrate that ODN 205 and pCN5 can stimulate the immune system of turbot and induce protection against bacterial challenge. In addition, pCN5 also possesses adjuvant property and can potentiate vaccine-induced specific immunity. (C) 2010 Elsevier Ltd. All rights reserved.

Streptococcus iniae is a severe aquaculture pathogen that can also infect humans and animal. A putative secretory antigen, Slat 0, was identified from a pathogenic S. iniae strain by in vivo-induced antigen technology. Using turbot as an animal model, the immunoprotective effect of Sia10 was examined as a DNA vaccine in the form of plasmid pSia10, which expresses sia10 under the cytomegalovirus immediate-early promoter. In fish vaccinated with pSia10, transcription of sia10 was detected in muscle, liver, spleen, and kidney at 7, 14, 21, 28, 35, 42, and 49 days post-vaccination. In addition, production of Sia10 protein was also detected in the muscle tissues of pSia10-vaccinated fish. Fish vaccinated with pSia10 exhibited a relative percent survival (RPS) of 73.9% and 92.3%, respectively, when challenged with high and low doses (producing a cumulative mortality of 92% and 52%, respectively, in the control groups) of S. iniae. Immunological and transcriptional analyses showed that vaccination with pSia10(i) induced much stronger chemiluminescence response and significantly higher levels of nitric oxide production and acid phosphatase activity in head kidney macrophages; (ii) caused the production of specific serum antibodies, which afforded apparent immunoprotection when transferred passively into naive fish; and (iii) upregulated the expression of the genes encoding proteins that are possibly involved in both innate and adaptive immune responses. Taken together, these results indicated that pSia10 is an effective vaccine candidate and may be used in the control of S. iniae infection in aquaculture. (C) 2010 Elsevier Ltd. All rights reserved.
