The study of maturation and spawning of the oyster is part of a research program to investigate the summer mortalities of the oysters, Crassostrea gigas in Marennes-Oléron Bay. Four maturity stages were simultaneously obtained by diet and thermal conditioning (immature, low maturation, mature and post-spawning stages). Measurements of clearance, filtration, absorption and respiration rates allowed a calculation of the scope for growth and hence an estimation of the oyster's energetic budget at various maturity stages. Male and female oysters had similar physiological responses. The filtration rate ranged from 2.4 to 2.6 1.h(-1) at the early stages of maturation and decreased to 1.8 1.h.' during the maturity stage. Growth rate resulting from gonad development did not induce filtration rate changes. Mature 2.5 and 1.5-year-old oysters showed a negative energy budget reaching -15 and -90 J.h(-1) respectively. By contrast, non-ripe oysters had scope for growth in the range 110 to 170 J.h(-1). A negative energy budget during the high maturation stage resulted from a reduced absorption efficiency. A new allometric relationship for the respiration model of C. gigas was defined during vitellogenesis with a 0.574 coefficient value. Based on Our results, the oyster's physiological weakness during vitellogenesis should be considered as a part of explanation for spring and summer mortalities of cultured oysters in Marennes-Oléron Bay.

The study of sexual maturation and spawning in the Pacific oyster (Crassostrea gigas) is part of a vast research programme that endeavours to understand the causes of mortality that occur sporadically during the spring and summer seasons in the Marennes-Oléron Bay. Thermal and diet conditioning were used to obtain oysters at each stage of maturity simultaneously. Using the measured rates of clearance, consumption, absorption and respiration provided estimates of growth potential and gave the energetic budget of oysters at different stages of sexual maturity. Physiological responses were similar for males and females. Filtration decreased from 2.4 to 2.6 l.h (-1) to 1.8 l.h (-1) with increasing maturity. Weight gain was associated with gonad development and did not appear to have an effect on the clearance rate. Oysters 2.5 years old showed a negative energy budget (-15 J h (-1)) at later maturity stages. This deficit was confirmed (90 J.h (-1)) in oysters 1.5 years old at the same stage of maturity. On the contrary, immature oysters, in the early stages of maturity or post-spawning, had a growth potential of 110 to 170 J.h (-1). The energy deficit observed at later stages of maturity was primarily due to absorption, which decreased sharply during peak gametogenesis. Using measured respiration rates, an allometric relationship specific to gonad growth was determined with a coefficient of 0.574. Low physiological performance of oysters, observed at later stages of sexual maturity, must be taken into account in research on the factors responsible for spring and summer mortalities affecting oyster farms in Marennes-Oléron.

Effects of a remarkably high overall lipid Tisochrysis lutea strain (T+) upon gross biochemical composition, fatty acid (FA), sterol and lipid class composition of Crassostrea gigas larvae were evaluated and compared with a normal strain of Tisochrysis lutea (T) and the diatom Chaetoceros neogracile (Cg). In a first experiment, the influence of different single diets (T, T+ and Cg) and a bispecific diet (TCg) was studied, whereas, effects of monospecific diets (T and T+) and bispecific diets (TCg and T+Cg) were evaluated in a second experiment. The strain T+ was very rich in triglycerides (TAG: 93â95% of total neutral lipids), saturated FA (45%), monounsaturated FA (31â33%) and total fatty acids (4.0â4.7 pg cellâˆ1). Larval oyster survival and growth rate were positively correlated with 18:1n-7 and 20:1n-7, in storage lipids (SL), and negatively related to 14:0, 18:1n-9, 20:1n-9, 20:4n-6 and trans-22-dehydrocholesterol in membrane lipids (ML). Surprisingly, only the essential fatty acid 20:5n-3 in SL was correlated positively with larval survival. Correlations suggest that physiological disruption by overabundance of TAG, FFA and certain fatty acids in larvae fed T+ was largely responsible for the poor performance of these larvae. â˜High-lipidâ strains of microalgae, without regard to qualitative lipid composition, do not always improve bivalve larval performance.

Feeding strategies and digestive capacities can have important implications for variation in energetic pathways associated with ecological and economically important traits, such as growth or reproduction in bivalve species. Here, we investigated the role of amylase in the digestive processes of Crassostrea gigas, using in vivo RNA interference. This approach also allowed us to investigate the relationship between energy intake by feeding and gametogenesis in oysters. Double-stranded (ds)RNA designed to target the two α-amylase genes A and B was injected in vivo into the visceral mass of oysters at two doses. These treatments caused significant reductions in mean mRNA levels of the amylase genes: âˆ50.7% and âˆ59% mRNA A, and âˆ71.9% and âˆ70.6% mRNA B in 15 and 75âµg dsRNA-injected oysters, respectively, relative to controls. Interestingly, reproductive knock-down phenotypes were observed for both sexes at 48âdays post-injection, with a significant reduction of the gonad area (âˆ22.5% relative to controls) and germ cell under-proliferation revealed by histology. In response to the higher dose of dsRNA, we also observed reductions in amylase activity (âˆ53%) and absorption efficiency (âˆ5%). Based on these data, dynamic energy budget modeling showed that the limitation of energy intake by feeding that was induced by injection of amylase dsRNA was insufficient to affect gonadic development at the level observed in the present study. This finding suggests that other driving mechanisms, such as endogenous hormonal modulation, might significantly change energy allocation to reproduction, and increase the maintenance rate in oysters in response to dsRNA injection.

Massive mortality outbreaks in cultured bivalves have been reported worldwide and they have been associated with infection by a range of viral and bacterial pathogens. Due to their economic and social impact, these episodes constitute a particularly sensitive issue in Pacific oyster (Crassostrea gigas) production. Since 2008, mortality outbreaks affecting C. gigas have increased in terms of intensity and geographic distribution. Epidemiologic surveys have lead to the incrimination of pathogens, specifically OsHV-1 and bacteria of the Vibrio genus, in particular Vibrio aestuarianus. Pathogen diversity may partially account for the variability in the outcome of infections. Host factors (age, reproductive statusâ¦) including their genetic background that has an impact on host susceptibility towards infection, also play a role herein. Finally, environmental factors have significant effects on the pathogens themselves, on the host and on the host-pathogen interaction. Further knowledge on pathogen diversity, classification, and spread, may contribute towards a better understanding of this issue and potential ways to mitigate the impact of these outbreaks.

Harmful algal blooms of Alexandrium spp. dinoflagellates regularly occur in French coastal waters contaminating shellfish. Studies have demonstrated that toxic Alexandrium spp. disrupt behavioural and physiological processes in marine filter-feeders, but molecular modifications triggered by phycotoxins are less well understood. This study analyzed the mRNA levels of 7 genes encoding antioxidant/detoxifying enzymes in gills of Pacific oysters (Crassostrea gigas) exposed to a cultured, toxic strain of A. minutum, a producer of paralytic shellfish toxins (PST) or fed Tisochrysis lutea (T. lutea, formerly Isochrysis sp., clone Tahitian (T. iso)), a non-toxic control diet, in four repeated experiments. Transcript levels of sigma-class glutathione S-transferase (GST), glutathione reductase (GR) and ferritin (Fer) were significantly higher in oysters exposed to A. minutum compared to oysters fed T. lutea. The detoxification pathway based upon glutathione (GSH)-conjugation of toxic compounds (phase II) is likely activated, and catalyzed by GST. This system appeared to be activated in gills probably for the detoxification of PST and/or extra-cellular compounds, produced by A. minutum. GST, GR and Fer can also contribute to antioxidant functions to prevent cellular damage from increased reactive oxygen species (ROS) originating either from A. minutum cells directly, from oyster hemocytes during immune response, or from other gill cells as by-products of detoxification.

The explosion of the Deepwater Horizon (DWH) oil platform resulted in large amounts of crude oil and dispersant Corexit 9500A® released into the Gulf of Mexico and coincided with the spawning season of the oyster, Crassostrea virginica. The effects of exposing gametes and embryos of C. virginica to dispersant alone (Corexit), mechanically (HEWAF) and chemically dispersed (CEWAF) DWH oil were evaluated. Fertilization success and the morphological development, growth, and survival of larvae were assessed. Gamete exposure reduced fertilization (HEWAF: EC201 h = 1650 μg tPAH50 L∠1; CEWAF: EC201 h = 19.4 μg tPAH50 L∠1; Corexit: EC201 h = 6.9 mg L∠1). CEWAF and Corexit showed a similar toxicity on early life stages at equivalent nominal concentrations. Oysters exposed from gametes to CEWAF and Corexit experienced more deleterious effects than oysters exposed from embryos. Results suggest the presence of oil and dispersant during oyster spawning season may interfere with larval development and subsequent recruitment.

Aim The spread of non-indigenous species in marine ecosystems world-wide is one of today's most serious environmental concerns. Using mechanistic modelling, we investigated how global change relates to the invasion of European coasts by a non-native marine invertebrate, the Pacific oyster Crassostrea gigas. Location Bourgneuf Bay on the French Atlantic coast was considered as the northern boundary of C. gigas expansion at the time of its introduction to Europe in the 1970s. From this latitudinal reference, variations in the spatial distribution of the C. gigas reproductive niche were analysed along the north-western European coast from Gibraltar to Norway. Methods The effects of environmental variations on C. gigas physiology and phenology were studied using a bioenergetics model based on Dynamic Energy Budget theory. The model was forced with environmental time series including in situ phytoplankton data, and satellite data of sea surface temperature and suspended particulate matter concentration. Results Simulation outputs were successfully validated against in situ oyster growth data. In Bourgneuf Bay, the rise in seawater temperature and phytoplankton concentration has increased C. gigas reproductive effort and led to precocious spawning periods since the 1960s. At the European scale, seawater temperature increase caused a drastic northward shift (1400 km within 30 years) in the C. gigas reproductive niche and optimal thermal conditions for early life stage development. Main conclusions We demonstrated that the poleward expansion of the invasive species C. gigas is related to global warming and increase in phytoplankton abundance. The combination of mechanistic bioenergetics modelling with in situ and satellite environmental data is a valuable framework for ecosystem studies. It offers a generic approach to analyse historical geographical shifts and to predict the biogeographical changes expected to occur in a climate-changing world.

Chemical pollution by pesticides has been identified as a possible contributing factor to the massive mortality outbreaks observed in Crassostrea gigas for several years. A previous study demonstrated the vertical transmission of DNA damage by subjecting oyster genitors to the herbicide diuron at environmental concentrations during gametogenesis. This trans-generational effect occurs through damage to genitor-exposed gametes, as measured by the comet-assay. The presence of DNA damage in gametes could be linked to the formation of DNA damage in other germ cells. In order to explore this question, the levels and cell distribution of the oxidized base lesion 8-oxodGuo were studied in the gonads of exposed genitors. High-performance liquid chromatography coupled with UV and electrochemical detection analysis showed an increase in 8-oxodGuo levels in both male and female gonads after exposure to diuron. Immunohistochemistry analysis showed the presence of 8-oxodGuo at all stages of male germ cells, from early to mature stages. Conversely, the oxidized base was only present in early germ cell stages in female gonads. These results indicate that male and female genitors underwent oxidative stress following exposure to diuron, resulting in DNA oxidation in both early germ cells and gametes, such as spermatozoa, which could explain the transmission of diuron-induced DNA damage to offspring. Furthermore, immunostaining of early germ cells seems indicates that damages caused by exposure to diuron on germ line not only affect the current sexual cycle but also could affect future gametogenesis.

In the Pacific oyster, spermatozoa are characterized by a remarkably long movement phase (i.e., over 24 h) sustained by a capacity to maintain intracellular ATP level. To gain information on oxidative phosphorylation (OXPHOS) functionality during the motility phase of Pacific oyster spermatozoa, we studied 1) changes in spermatozoal mitochondrial activity, that is, mitochondrial membrane potential (MMP), and intracellular ATP content in relation to motion parameters and 2) the involvement of OXPHOS for spermatozoal movement using carbonyl cyanide m-chlorophenyl hydrazone (CCCP). The percentage of motile spermatozoa decreased over a 24 h movement period. MMP increased steadily during the first 9 h of the movement phase and was subsequently maintained at a constant level. Conversely, spermatozoal ATP content decreased steadily during the first 9 h postactivation and was maintained at this level during the following hours of the movement phase. When OXPHOS was decoupled by CCCP, the movement of spermatozoa was maintained 2 h and totally stopped after 4 h of incubation, whereas spermatozoa were still motile in the control after 4 h. Our results suggest that the ATP sustaining flagellar movement of spermatozoa may partially originate from glycolysis or from mobilization of stored ATP or from potential phosphagens during the first 2 h of movement as deduced by the decoupling by CCCP of OXPHOS. However, OXPHOS is required to sustain the long motility phase of Pacific oyster spermatozoa. In addition, spermatozoa may hydrolyze intracellular ATP content during the early part of the movement phase, stimulating mitochondrial activity. This stimulation seems to be involved in sustaining a high ATP level until the end of the motility phase.

Depuis 2008, des mortalités massives dâhuîtres creuses âgées de moins dâun an sont relevées sur le littoral français dès que la température de lâeau de mer atteint le seuil de 16°C. Ces mortalités de naissains sont associées à la détection du virus OsHV-1 μVar. Des travaux de qualification zoo-sanitaire menés depuis 2010 ont montré quâun lot de naissains issus du captage naturel sur deux était infecté par le virus bien avant lâapparition des mortalités dans le milieu naturel, mais quâa contrario les naissains dâécloserie expertisés présentaient une fréquence plus faible de portage latent. Dans ce contexte de crise zoo-sanitaire chronique, lâobjectif de notre étude consistait à obtenir une information du statut zoo-sanitaire OsHV-1 μVar des populations de naissains du captage 2013 à lâéchelle national. Ce travail avait pour finalité de permettre lâidentification précoce des zones de captage associées à un risque de mortalité due à lâinfection par OsHV-1 μVar. La méthodologie de lâépreuve thermique de laboratoire (ETL) correspond en une période de 1 mois dâisolement des naissains à évaluer en conditions contrôlées de laboratoire. Lors de cette période la température de lâeau de mer est maintenue constante à 21°C et la mortalité est relevée tous les 10 jours. La survie finale associée à des analyses qPCR permettent de qualifier le statut sanitaire en terme de portage OsHV-1 μVar de lâéchantillon (statut infecté ou non infecté). Entre février et mars 2014, vingt et un échantillons de naissains âgés de 5 à 7 mois ont été échantillonnés dans six sites du littoral français (étang de Thau, bassin dâArcachon, pertuis Charentais, baie de Bourgneuf, baie de Vilaine et rade de Brest) pour être qualifiés par ETL au site expérimental Ifremer dâArgenton. Les résultats de la campagne 2014 de qualification zoo-sanitaire confirment quâen période hivernale, dans le milieu naturel, des naissains de captage peuvent être infectés par OsHV-1 μVar sans développement apparent de maladies ni mortalité. En revanche en ETL, ces lots de naissains infectés par OsHV-1 μVar présentent des taux de mortalité importants. Ainsi, sur les 20 échantillons de naissains de captage étudiés, onze ont révélé des maladies en ETL avec des mortalités cumulées variant de 16 % à 80 %. Ce ratio dâun échantillon sur deux de naissains infectés par OsHV-1 μVar demeure proche de celui observé de 2010 à 2013 lors des travaux précédents qui avaient permis de définir lâETL. les six sites étudiés ont montré des résultats contrastés. En effet, les résultats se sont révélés favorables pour lâétang de Thau et la baie de Bourgneuf et défavorables dans le cas de la rade de Brest et la Baie de Vilaine. Pour ces deux derniers sites, tous les échantillons de naissains testés ont montré de fortes mortalités pendant lâETL. Des différences sâobservent également intra-site, notamment à Marennes Oléron avec 1 échantillon sur 3 infectés par OsHV-1 μVar ou dans pour Arcachon avec 3 échantillons sur 5. Dans le contexte préoccupant de lâépizootie actuelle, nos résultats confirment que certains lots de naissains de captage sont infectés par OsHV-1 μVar dès les mois de février-mars et pourrait être des acteurs actifs dans le déclenchement annuel du processus infectieux en milieu naturel quand la température de lâeau dépasse 16°C. Par ailleurs, notre étude démontre quâil serait possible chaque année dâobtenir une qualification sanitaire précoce des zones de captage. Cela permettrait dâaméliorer la gestion des risques liés aux transferts de naissains infectés par OsHV-1 μVar dans les différents bassins de production.

Ces mortalités de naissains sont associées à la détection du virus OsHV-1 μVar. Depuis 2010, des travaux de qualification zoo-sanitaire ont montré quâenviron un lot de naissains de captage sur deux était infecté par le virus avant lâapparition des mortalités dans le milieu naturel. En 2014, dans le cadre de la première action QUALIF, les résultats de la qualification zoo-sanitaire des naissains du captage de lâannée 2013 à lâéchelle nationale ont mis en évidence le caractère infectieux des maladies développées par ces naissains porteurs latents dâOsHV-1 μVar. Lâobjectif 2015 du second volet QUALIF consistait à reproduire le travail de qualification zoo-sanitaire OsHV-1 μVar pour les naissains du captage de lâannée 2014. Les résultats (2014 et 2015) permettront dâaborder la variation interannuelle du statut zoo-sanitaire des naissains pour chacun des sites étudiés. La méthodologie de lâépreuve de qualification zoo-sanitaire consiste en une période de 1 mois dâisolement des naissains en conditions contrôlées de laboratoire. Lors de cette épreuve, la température de lâeau de mer est maintenue constante à 21°C et la mortalité est relevée tous les 10 jours. La survie finale et les analyses qPCR permettent de qualifier le statut sanitaire en terme de portage OsHV-1 μVar de lâéchantillon (infecté ou non infecté). De début janvier à fin mars 2015, 39 échantillons de naissains âgés de 5 à 7 mois ont été prélevés dans 6 sites du littoral français (étang de Thau, bassin dâArcachon, bassin de Marennes Oléron, baie de Bourgneuf, embouchure de La Vilaine et rade de Brest) pour être testés en épreuve thermique de laboratoire (ETL) dans lâoutil expérimental Ifremer à Argenton. Les résultats montrent que 25 des 39 (soit 64 %) lots de naissains de captage testés ont développé des maladies en ETL associés à des mortalités cumulées variant de 6 à 72 %. Cette valeur de 64 % de lots détectés est plus élevée que celle précédemment observée en 2014 (55 %). Les résultats de cette seconde campagne de qualification zoo-sanitaire sont favorables pour les naissains de lâétang de Thau (absence de mortalité), moins favorables pour ceux dâArcachon, Marennes Oléron, et ceux de la baie de Bourgneuf (réponses contrastées en terme de mortalité intra-site) et très défavorables pour les naissains de La Vilaine et de la rade de Brest (mortalités observées sur tous les échantillons testés). La principale conclusion de cette seconde étude de qualification zoo-sanitaire est identique à celle de 2014, à savoir quâil est existe dans le milieu naturel en période hivernale des lots de naissains de captage infectés par OsHV-1 μVar sans développement apparent des maladies. Ces lots de naissains porteurs latents dâOsHV-1 μVar sont détectables en ETL et la présence dâOsHV-1 μVar peut alors être confirmée par qPCR. Dans le contexte actuel dâépizooties chroniques, nos résultats confirment à nouveau quâil est possible dâidentifier précocement les lots de naissains infectés ou non par OsHV-1 μVar. Ces animaux infectés sont un réservoir du virus en période hivernale. Dès que la température de lâeau de mer franchit le seuil de 16°C, ils participeront à la réémergence des maladies en milieu naturel. Par ailleurs, cette seconde étude de qualification zoo-sanitaire confirme la possibilité dâapprécier le risque sanitaire pour chaque zone de captage en fonction de la détection précoce des lots de naissains infectés par OsHV-1 μVar.

The temporal variability of delta(13)C in suspended particulate organic matter (POM) and oyster Crassostrea gigas along a salinity gradient was investigated from May 1992 to September 1993 within the estuarine bay of Marennes-Oleron (France). During this period the mean daily discharge of the Charente River exhibited large seasonal variation, with a high discharge from November 1992 to January 1993. Contrary to that at the river mouth and the marine littoral, delta(13)C in POM and in oysters at mid-estuary was affected by the high flood period. The delta(13)C values of POM decreased in mid-estuary and remained at low levels during the high discharge period, indicating an increasing contribution of terrestrial inputs to the estuarine POM pool. At the same site, a remarkable decrease of delta(13)C in oysters occurred between December 1992 and March 1993 (after a time lag compared to the ambient POM), indicating incorporation of terrestrial organic matter in oyster tissues during the high flood discharge. The lag between the delta(13)C decrease in POM and oysters is attributed to the time needed for oyster tissues to incorporate enough newly terrestrial light carbon to be recognized by the delta(13)C measure (about 1 to 2 mo). This time interval depends on tissue turnover time. The delta(13)C POM decrease (i.e. 1.3 parts per thousand) cannot explain entirely the decrease observed in oysters (i.e. 2.3 parts per thousand). In fact, the pattern exhibited by mid-estuarine oysters can be explained by the increasing contribution of terrestrial organic matter to their feeding, and the inability to preferentially utilize specific components of the estuarine POM that are C-13-enriched.

The linked concepts of 'microbial loop' and 'protozoan trophic link' have been very well documented in filter-feeding microzooplankton such as copepods, but have not been applied to energy transfer to benthic suspension-feeding macrofauna, with the exception of the recent demonstration of heterotrophic flagellate assimilation by mussels. The oyster Crassostrea gigas obtains energy resources by filtering microalgae (similar to 5 to 100 mu m). However, in turbid estuaries, light-limited phytoplanktonic production cannot entirely account for oyster energy requirements. Conversely, picoplankters (<2 mu m), which are main effecters of coastal energy flow and matter cycling, are not efficiently retained by oyster filtration. Ciliate protozoal as both micro-sized cells (similar to 5 to 100 run) and bacteria grazers, may represent a major intermediary in trophic transfer between picoplankton and metazoa. The ciliate Uronema was intensely cultured and labelled, using the cyanobacteria Synechococcus as an auto-fluorescent biomarker. The labelled ciliates were offered as potential prey to oysters. We report here the first experimental evidence of a significant retention and ingestion of ciliates by oysters, supporting the role of protozoa as a realistic trophic link between picoplankters and filter-feeding bivalves and thus enhancing their potential importance in estuarine microbial food webs.

Oysters play an important role in estuarine and coastal marine habitats, where the majority of humans live. In these ecosystems, environmental degradation is substantial, and oysters must cope with highly dynamic and stressful environmental constraints during their lives in the intertidal zone. The availability of the genome sequence of the Pacific oyster Crassostrea gigas represents a unique opportunity for a comprehensive assessment of the signal transduction pathways that the species has developed to deal with this unique habitat. We performed an in silico analysis to identify, annotate and classify protein kinases in C. gigas, according to their kinase domain taxonomy classification, and compared with kinome already described in other animal species. The C. gigas kinome consists of 371 protein kinases, making it closely related to the sea urchin kinome, which has 353 protein kinases. The absence of gene redundancy in some groups of the C. gigas kinome may simplify functional studies of protein kinases. Through data mining of transcriptomes in C. gigas, we identified part of the kinome which may be central during development and may play a role in response to various environmental factors. Overall, this work contributes to a better understanding of key sensing pathways that may be central for adaptation to a highly dynamic marine environment.
