DNA in canned tuna is degraded into short fragments of a rew hundred base pairs. The polymerase chain reaction (PCR) was used to amplify short sequences of mitochondrial DNA, which were denatured and analysed by polyacrylamide gel electrophoresis (native PAGE) for detection of single strand conformation polymorphisms. Species specific patterns of DNA bands were obtained for a number of tuna and bonito species. DE: In Thunfischkonserven liegt die DNA in Form kurzkettiger Fragmente von wenigen Hundert Basenpaaren Länge vor. Mit Hilfe der Polymerase-Kettenreaktion (PCR) wurden kurze Sequenzen der mitochondrialen DNA vervielfältigt. Anschließend wurde die gebildete DNA in Einzelsträngen überführt, die durch eine native Polyacrylamidgel-Elektrophorese (PAGE) aufgetrennt wurde. Für eine Reihe von Thunfischen und Boniten ergaben die Einzelstränge artspezifische Bandenmuster, die auf unterschiedliche Konformationen der DNA-Stränge der einzelnen Fischarten zurückzuführen sind.

Molecular-based approaches for shark species identification have been driven largely by issues specific to the fishery. In an effort to establish a more comprehensive identification data set, we investigated DNA sequence variation of a 1.4-kb region from the mitochondrial genome covering partial sequences from the 12S rDNA, 16S rDNA, and the complete valine tRNA from 35 shark species from the Atlantic fishery. Generally, within-species variability was low in relation to interspecific divergence because species haloptypes formed monophyletic groups. Phylogenetic analyses resolved ordinal relationships among Carcharhiniformes and Lamniformes, and revealed support for the families Sphyrnidae and Triakidae (within Carcharhiniformes) and Lamnidae and Alopidae (within Lamniformes). The combination of limited intraspecific variability and sufficient between-species divergence indicates that this locus is suitable for species identification.

The anatomical and morphometric (shape indices, contour descriptors and otolith weight) characterizations of sagittal otoliths were investigated in 13 species of Lutjanus spp. inhabiting the Persian Gulf. This is the first study that compares the efficiency of three different image analysis techniques for discriminating species based on the shape of the outer otolith contour, including elliptical Fourier descriptors (EFD), fast Fourier transform (FFT) and wavelet transform (WT). Sagittal otoliths of snappers are morphologically similar with some small specific variations. The use of otolith contour based on wavelets (WT) provided the best results in comparison with the two other methods based on Fourier descriptors, but only the combination of the all three methods (EFD, FFT and WT) was useful to obtain a robust classification of species. The species prediction improved when otolith weight was included. In relation to the shape indices, only the aspect ratio provided a clear grouping of species. Also, another study was carried on to test the possibility of application of shape analysis and comparing otolith contour of otoliths of Lutjanus johnii from Persian Gulf and Oman Sea to identify potential stocks. The results showed the otoliths have differences in contour shape and can be contribute to two different stocks.

This study describes the molecular identification of sixteen fish species present in processed products imported into Iran for human consumption. DNA barcoding using direct sequencing of about 650 bp of the mitochondrial Cytochrome Oxidase subunit I gene revealed incorrect labeling (31.25%). Substitution of fish species constitutes serious economic fraud, and our results increase concern regarding the trading of processed fish products in Iran from both health and conservation points of view.

To ensure the authentication of fishery products lacking biological characters, rapid species identification methods are required. Two DNA- and protein-based methods, PCR-SSCP (polymerase chain reaction - single strand conformation polymorphism) of a 464 bp segment of the cytochrome b – gene and isoelectric focusing (IEF) of water-soluble proteins from fish fillets, were applied to identify fillets of (sub-) tropical fish species available on the European market. Among the samples analysed were two taxonomically identified species from the family Sciaenidae and one from Sphyraenidae. By comparison of DNA- and protein patterns of different samples, information about intra-species variability of patterns, and homogeneity of batches (e.g. fillet blocks or bags) can be obtained. PCR-SSCP and IEF may be useful for pre-checking of a large number of samples by food control laboratories. Zusammenfassung Zur Sicherstellung der Authentizität von Fischerei-Erzeugnissen ohne biologische Merkmale sind schnelle Verfahren zur Speziesidentifizierung hilfreich. Zwei Methoden der DNA- bzw. Protein-Analyse wurden eingesetzt, um Filets (sub-) tropischer Fischarten, die auf dem europäischen Markt angeboten werden, zu identifizieren. Bei diesen Methoden handelt es sich um die PCR-SSCP (Polymerase-Kettenreaktion – Einzelstrang-Konformationspolymorphismus) – Analyse der PCR-Produkte und die IEF (isoelektrische Fokussierung) der wasserlöslichen Fischmuskelproteine. Unter den untersuchten Proben waren zwei taxonomisch bestimmte Arten aus der Familie Sciaenidae und eine Spezies aus der Familie Sphyraenidae. Durch Vergleich der DNA- bzw. Proteinmuster lassen sich Informationen über die intra-spezifische Variabilität solcher Muster und die Einheitlichkeit von Partien (beispielsweise Filetblöcke oder Filetbeutel) gewinnen. PCR-SSCP und IEF können in Laboratorien der Lebensmittelüberwachung als Vortest gerade bei hohen Probenzahlen sinnvoll eingesetzt werden.

Pelagic juvenile rockfish (Sebastes spp.) collected in surveys designed to assess juvenile salmonids and other species in the Gulf of Alaska in 1998 and 2000–2003 provide an opportunity to document the occurrence of the pelagic juveniles of several species of rockfish. Often, species identification of rockfish is difficult or impossible at this stage of development (~20 to 60 mm), and few species indigenous to Alaska waters have been described. Use of mitochondrial DNA markers for rockfish species allowed unequivocal identification of ten species (S. aleutianus, S. alutus, S. borealis, S. entomelas, S. flavidus, S. melanops, S. pinniger, S. proriger, S. reedi, and S. ruberrimus) in subsamples from the collections. Other specimens were genetically assignable to groups of two or three species. Sebastes borealis, S. crameri, and S. reedi were identified using morphological data. Combining genetic and morphological data allowed successful resolution of the other species as S. emphaeus, probably S. ciliatus (although S. polyspinis cannot be totally ruled out), and S. polyspinis. Many specimens were initially morphologically indistinguishable from S. alutus, and several morphological groups included fish genetically identified as S. alutus. This paper details the characteristics of these pelagic juveniles to facilitate morphological identification of these species in future collections. (PDF file contains 32 pages.)

The species list is drawn from an analysis of catches taken by Sumalian and Russian trawlers in the Gulf of Aden and the Arabian Sea between 1985 and 1990. The southern coastline of the Republic of Yemen has been divided into 7 areas, including waters around Socotra Island. The average depth of each trawl was recorded in 50 m increments. Non-appearance of the species in the area does not mean that the species do not occur in that area or depth, merely that it was not recorded in any of the samples analyzed. Specimens that could not be identified to species level have been excluded. A total of 195 species from 75 families was recorded and is summarized. Most of the identification of species was from FAO species identification literature. Confirmation of some species and usage of common names is from ICLARM's FishBase and Al Sedfy, et al. (1982).

Thirty individuals of each species of Indian major carps, i.e., Catla catla, Cirrhinus cirrhosus (C. mrigala) and Labeo rohita, obtained from a nursery near Mymensingh, Bangladesh were analysed by means of allozyme electrophoresis. Twenty-one loci were studied. Several loci revealed significant deviation from Hardy-Weinberg expectations caused by deficiency of heterozygotes, indicating Wahlund effects due to problems with species identification. Moreover, bimodal distributions of individual heterozygosity within the three putative species indicated hybridisation. This was confirmed using analysis of individual admixture proportions, as individuals misidentified to species and hybrids between species were observed. Furthermore, factorial correspondence analysis to visualize genetic relationships among individuals revealed three distinct groups containing misclassified individuals, along with some intermediate individuals interpreted as hybrids. Ten per cent of all C. catla and L. rohita had been erroneously identified to species, and 40 per cent of all presumptive C. catla were hybrids between C. catla x C. cirrhosus and C. catla x L. rohita. In the case of C. cirrhosus, 37 per cent of the samples were C. cirrhosus x L. rohita hybrids. Thirty per cent of all presumptive L. rohita turned out to be hybrids between L. rohita x C. catla and L. rohita x C. cirrhosus. The high incidence of hybrids in C. catla might be responsible for slower growth of the fish in aquaculture.

Partial sequences of cytochrome b (Cyt b) and 16S ribosomal RNA (16S rRNA) mitochondrial genes were used for species identification and estimating phylogenetic relationship among three commercially important Ompok species viz. O. Pabda, O. pabo and O. bimaculatus. The sequence analysis of Cyt b (1118bp) and 16S rRNA (569 & 570bp) genes revealed that O. pabda, O. pabo & 0. bimaculatus were genetically distinct species and they exhibited identical phylogenetic relationship. The present study discussed usefulness of mtDNA genes (Cyt b & 16S rRNA) in resolving taxonomic ambiguity and estimating phylogenetics relationship.

In Iranian Coastline water of Persian Gulf we researched about flat fishes and revision their Scientitic names, Investigated diversity species (Pleuronectiformes) around three provinces such as khouzestan, Bushehr and Hormozgen. The aim focuse on introducting major family and Species Identification for this Purpose 1311 Specimens collected. Then 36 investigation Parameters includes 18 Morphometric, 11 meristic, 7 distribution has been measured in Excel. The descriptive statistic in spss software would be able us to graph dendrograms by Clustering methods, then Design a standerd table and comparised all the datas with it six major family has been found in Iranian Coasline which named: Soleidae, Bothidae Paralichtidae, Cynoglossidae, Psettodidae, Citharidae. Some of the scientific species names such as Pseudorombus arsius, P. annulatus, P. elevatus, P.malayanus, P. triocellatus from Bothidae family Join it to Paralichtidae family and Eurylossa orientalis change It's name from Soleidae family to Brachirus onentalis. Cynoglossus, bilineatus, C.Puncticepts, C.durbaensis, Clachneri, one of the most Important Point that viewed in our samples, founding a new species (C.Sp) Which we couldn't identify it with all the known keys, then sent it to international Scientific references for this porpose. Khozestan has the maximus diversity speices (26 species) and the minimum is Hormozgan (15 Speices). 13 species of five family has never reported inpersian Gulf but we identify them in our studies for the first time. We abserved 29 Species of six family in our studies, comparision between are resualts with other researches shows that this use is the most compelet study in Iranian coastline water of Persian Gulf.

Identification problems are common for many sharks due to a general lack of meristic characteristics that are typically useful for separating species. Other than number of vertebrae and number and shape of teeth, identifications are frequently based on external features that are often shared among species. Identification problems in the field are most prevalent when live specimens are captured and releasing them with a minimum of stress is a priority (e.g., shark tagging programs). Identifications must be accurate and conducted quickly but this can be challenging, especially if specimens are very active or too large to be landed without physical damage. This field guide was designed primarily for use during field studies and presents a simplified method for identifying the 21 species of western North Atlantic Ocean sharks belonging to the family Carcharhinidae (carcharhinids). To assist with identifications a dichotomous key to Carcharhinidae was developed, and for the more problematic Carcharhinus species (12 species), separation sheets based on important distinguishing features were constructed. Descriptive text and illustrations provided in the species accounts were developed from field observations, photographs, and published references. (PDF file contains 36 pages.)

Sighting, stranding, and capture records of whales and dolphins for Venezuela were assembled and analyzed to document the Venezuelan cetacean fauna and its distribution in the eastern Caribbean. An attempt was made to confirm species identification for each of the records, yielding 443 that encompass 21 species of cetaceans now confirmed to occur in Venezuelan marine, estuarine, and freshwater habitats. For each species, we report its global and local distribution, conservation status and threats, and the common names used, along with our proposal for a Spanish common name. Bryde’s whale (Balaenoptera edeni) is the most commonly reported mysticete. The long-beaked common dolphin (Delphinus capensis) is the most frequent of the odontocetes in marine waters. The boto or tonina (Inia geoffrensis) was found to be ubiquitous in the Orinoco watershed. The distribution of marine records is consistent with the pattern of productivity of Venezuelan marine waters, i.e., a concentration at 63°07′W through 65°26′W with records declining to the east and to the west. An examination of the records for all cetaceans in the Caribbean leads us to conclude that seven additional species may be present in Venezuelan waters. (PDF file contains 61 pages.)
