The advent of molecular biology has had a dramatic impact on all aspects of biology, not least applied microbial ecology. Microbiological testing of water has traditionally depended largely on culture techniques. Growing understanding that only a small proportion of microbial species are culturable, and that many microorganisms may attain a viable but non-culturable state, has promoted the development of novel approaches to monitoring pathogens in the environment. This has been paralleled by an increased awareness of the surprising genetic diversity of natural microbial populations. By targeting gene sequences that are specific for particular microorganisms, for example genes that encode diagnostic enzymes, or species-specific domains of conserved genes such as 16S ribosomal RNA coding sequences (rrn genes), the problems of culture can be avoided. Technical developments, notably in the area of in vitro amplification of DNA using the polymerase chain reaction (PCR), now permit routine detection and identification of specific microorganisms, even when present in very low numbers. Although the techniques of molecular biology have provided some very powerful tools for environmental microbiology, it should not be forgotten that these have their own drawbacks and biases in sampling. For example, molecular techniques are dependent on efficient lysis and recovery of nucleic acids from both vegetative forms and spores of microbial species that may differ radically when growing in the laboratory compared with the natural environment. Furthermore, PCR amplification can introduce its own bias depending on the nature of the oligonucleotide primers utilised. However, despite these potential caveats, it seems likely that a molecular biological approach, particularly with its potential for automation, will provide the mainstay of diagnostic technology for the foreseeable future.

The rate of sea level change has varied considerably over geological time, with rapid increases (0.25 cm yr-1) at the end of the last ice age to more modest increases over the last 4,000 years (0.04 cm yr-1; Hendry 1993). Due to anthropogenic contributions to climate change, however, the rate of sea level rise is expected to increase between 0.10 and 0.25 cm year-1 for many coastal areas (Warrick et al. 1996). Notwithstanding, it has been predicted that over the next 100 years, sea levels along the northeastern coast of North Carolina may increase by an astonishing 0.8 m (0.8 cm yr-1); through a combination of sea-level rise and coastal subsidence (Titus and Richman 2001; Parham et al. 2006). As North Carolina ranks third in the United States with land at or just above sea level, any additional sea rise may promote further deterioration of vital coastal wetland systems. This short paper records some measurements made on the Little Sea, a shallow, coastal, acidic lake on Studland Heath, Dorset. The lake, formed about 100 years ago by dunes cutting off a sea inlet, has not received any input of agricultural fertilizers or other waste products for at least the last 30 years. It is a Site of Special Scientific Interest (SSSI). Samples of surface water were taken from the northern and southern ends of the lake at 3-monthly intervals, from July 1995 to April 1996. The first samples in July 1995 were taken during a period of drought; rain, sometimes very heavy, came in late September. With the exception of silicate, potassium and phosphate, there were no large changes in plant nutrient concentrations during the year. The concentration of nitrate-nitrogen was very low (close to the limits of analytical detection), but total phosphorus at ca. 30 mu g per litre was similar to concentrations found in some of the Cumbrian eutrophic lakes. The large number of algal species at low cell/colony concentrations suggested that the lake is mesotrophic. Sodium, chloride and magnesium in the lake water were close to the same proportions as those found in sea water. Dry and wet deposition of sea-salts on the lake surface and its catchment area probably is the major source of sodium, magnesium and chloride ions in the lake, and also accounts for about half of the mean potassium and sulphate concentrations.

The use of reproductive and genetic technologies can increase the efficiency of selective breeding programs for aquaculture species. Four technologies are considered, namely: marker-assisted selection, DNA fingerprinting, in-vitro fertilization, and cryopreservation. Marker-assisted selection can result in greater genetic gain, particularly for traits difficult or expensive to measure, than conventional selection methods, but its application is currently limited by lack of high density linkage maps and by the high cost of genotyping. DNA fingerprinting is most useful for genetic tagging and parentage verification. Both in-vitro fertilization and cryopreservation techniques can increase the accuracy of selection while controlling accumulation of inbreeding in long-term selection programs. Currently, the cost associated with the utilization of reproductive and genetic techniques is possibly the most important factor limiting their use in genetic improvement programs for aquatic species.

The preliminary results obtained from the study of stomach contents of specimens of Gymnarchus niloticus of a size ranging from 56 cm (66 grams) to 152 cm (12.0 kg.) from August to December, 1982 indicate for these sizes an exclusively piscivorous diet. The predominant prey species in Lake Chad are Tilapia/Sarotherodon , and Clarias . The prey is often cut in two parts before it is swallowed. Small prey may be swallowed whole. The implication of this prey capture method on prey-predator relationship and therefore the impact of the predator is discussed. The breeding season of Gymnarchus niloticus in Lake Chad has been deduced from observation of changes in gonad maturity stages and the results indicate that breeding takes place between August and November. Data are also presented on the fecundity, size of ripe eggs and probable size at maturity
